人类常常因误食被猪霍乱沙门氏菌感染的食物而引发中毒。因此,急需构建适用于快速、灵敏检测食物中猪霍乱沙门氏菌污染的新方法。鉴于此,本文构建了一种pH响应比色酶联免疫吸附法灵敏检测牛奶中猪霍乱沙门氏菌。该方法利用葡萄糖氧化酶... 人类常常因误食被猪霍乱沙门氏菌感染的食物而引发中毒。因此,急需构建适用于快速、灵敏检测食物中猪霍乱沙门氏菌污染的新方法。鉴于此,本文构建了一种pH响应比色酶联免疫吸附法灵敏检测牛奶中猪霍乱沙门氏菌。该方法利用葡萄糖氧化酶催化底物葡萄糖产生葡萄糖酸,导致溶液pH下降,进而引发溴甲酚紫(Bromocresol purple,BCP)溶液颜色由紫色变成亮黄色,随后通过记录BCP颜色变化(OD 430/OD 590)实现目标菌的定量检测。当菌浓度为2.54×103~6.17×10^5 CFU/mL,该方法定量检测猪霍乱沙门氏菌可用方程一表述:y 1=0.1051ln(x)+0.7024(R 2=0.7513);当菌浓度为6.17×10^5~1.67×10^7 CFU/mL,该方法定量检测猪霍乱沙门氏菌可用方程二表述:y2=1.3216ln(x)-15.797(R^(2)=0.9711);当菌浓度大于1.67×10^7 CFU/mL时,BCP溶液呈现明显亮黄色,OD 430/OD 590值趋于稳定,无法实现猪霍乱沙门氏菌定量检测。将六个不同浓度的猪霍乱沙门氏菌(2.5×103~5.6×10^6 CFU/mL)加标至牛奶中,检测结果显示回收率介于72.16%~103.58%,相对标准偏差介于7.54%~15.30%。总之,本研究所构建的比色ELISA方法适用于牛奶中不同浓度的猪霍乱沙门氏菌快速、灵敏定量检测。 展开更多

Effect of Leukemia Vaccine on the Macrophage of Mice

Objective: To evaluate the e?ect of the prepared leukemia vaccine on the cytotoxicity of macrophage of C57BL/6 mice. Methods: The model of mice bearing leukemia was established and three types of leukemia vaccine were prepared before they were administered on the mice respectively. The cy- totxicity of M? derived from the mice was measured after the active immunotherapy or prevention for 2–4 weeks later by using MTT colorimetric method and compared with the control group. Results: (1) With the growth of leukemia cells in the mice, the cytotoxicity of M? was seriously depressed; (2) The administration of leukemia vaccine prepared from inactivated leukemic cells, incomplete Freund’s adjuvant (IFA) and cytokines, such as rGM-CSF, rIL-2 and rIL-6, promoted the cellular immunity of mice bearing leukemia, more e?cient than leukemia vaccine from either inactivated leukemic cells and IFA or inactive leukemic cells per se. Conclusion: The leukemia vaccine prepared with inactive leukemic cells, IFA and cytokines could activate the non-speci?c cellular immunity such as M?, as well as, antigen present, immune surveillance and so on, and this activation constructed a base for further activate T lymphocyte. Naturally, this will certainly have a promising future in the therapy against hematopietic tumor. 展开更多

Herein, a multiwalled carbon nanotubes(MWNT)-based colorimetric probe was designed to discover and monitor the level of apolipoprotein-L1(Apo L1) in lumbar disc herniation(LDH) patients. Apo L1 could be easily found in human serum of the LDH group, but not obviously expressed in the normal control group(Ctrl), spine spondylolisthesis(SSP) group, spinal fracture(SFR) group, and spine scoliosis(SSC) group. Furthermore, the as-prepared MWNT-based probe was also used to track the recovery of LDH patients who have successful surgery operation. The P value of early diagnosis and recovery monitoring was <0.001 and >0.05 for the proposed method and a conventional enzyme-linked immunosorbent assay(ELISA), respectively, suggesting this detection strategy had significant differences compared with the traditional ELISA. All experimental results showed that Apo L1 might be a potential biomarker for early diagnosis of LDH. This proposed detection strategy has a potential application in discovering new biomarkers of diseases. 展开更多
