Phylogenetic analyses using the most parsimony method (MP), maximum likelihood method (ML), and neighbor joining method (NJ) showed strong support that the Pentatomomorpha lineage is a monophyly and the superfamily Aradoidea is a sister group to the remainder of the Pentatomomorpha (Trichophora). The Trichophora could be divided into two clades : one clade consisted of the monophyletic superfamilies Pentatomoidea and Pyrrhocoroidea; the other was mainly the polyphyletic superfamilies Lygaeoidea, Coreoidea and Idiostoloidea. The superfamilies Lygaeoidea and Coreoidea were both polyphyletic. Within Lygaeoidea, Piesmatidae was sister to Berytidae. They formed a clade locating at the basal of the Trichophora and distantly related to the other two families Lygaeidae and Rhyparochromidae. This research suggested that 18S rDNA was a proper marker to reconstruct the phylogeny of Pentatomomorpha that was accordant to morphological studies and the research of Li et al (2005). The Pyrrhocoroidea was further divided from the Coreoidea (s./at ). It was suggested that the Piesmatidae might be assigned as a superfamily of Pentatomomorpha rather than a family in Lygaeoidea.展开更多

Phylogenetic relationships among the superfamilies within this infraorder have been uncertain, especially for the Eutrichophora. The previous studies were based on morphological characters, or just mitochondrial or nuclear genes, or only partial 18 S rDNA and COI. In this study, we used maximum likelihood(ML) and Bayesian inference(BI) based on massive molecular datasets(18S rDNA, 28 S rDNA, Hox and mitochondrial genes totaling 21 loci and 12,538 characters) to infer a robust phylogeny for this terrestrial group. Results strongly support the monophyly of all superfamilies; the superfamily status of Aradoidea and the following relationships:(Aradoidea +(Pentatomoidea +(Coreoidea +(Lygaeoidea + Pyrrhocoroidea)))) in Pentatomomorpha, and(Coreoidea +(Lygaeoidea + Pyrrhocoroidea)) in Eutrichophora. Our results suggest that sampling greater numbers of genes is an effective tool for resolving phylogenetic problems.展开更多

PCR was performed with the primers and cDNA from the Triticum aestivum-Haynaldia villosa translo...Two pairs of degenerate primers were designed based on nucleotide-binding site (NBS) and serine/threonine kinase domain. PCR was performed with the primers and cDNA from the Triticum aestivum-Haynaldia villosa translocation line 6VS/6AL. Amplified products were cloned and sequenced. Nine clones with NBS and one with serine/threonine kinase domain were obtained. The NBS clones were classified to six groups according to their nucleotide sequence identities (90% or higher). These resistance gene analogs (RGAs) all have open reading frames (ORF), and their amino acid sequences show high similarity to Yr10 in wheat, Mla1 and Mla6 in barley, RPS2 in Arabidopsis and other resistance (R) genes with conserved motifs. They were preliminarily mapped on the chromosomes of homoeologous groups 1, 2 and 5 of common wheat by nulli-tetrasomic analysis. The 5'-end sequence of an RGA N5 was obtained by 5'-RACE PCR. It encodes six leucine zipper (LZ) and has high sequence similarity to RPS2.展开更多

During morphogenesis, three dorsal kinetics anlagen are primarily formed. Fragmentation of kinety 3 usually present, while sometimes secondarily lost. Dorsomarginal kineties are formed, while sometimes lost. Oxytrichids tend to have overlapping characters, e.g. cell shape and size, infraciliature, pellicle fea- tures. This makes a great problem for genera separation. In the present work, all typical 18 frontal-ventral-transverse-cirri Oxytrichi- dae s. 1. genera were revised systematically based on their living morphology, ciliature patterns and dorsal morphogenetic features. The outline of the genera, the schematic illustrations, and the key to typical 18 frontal-ventral-transverse-cirri genera of Oxytrichidae s. l. were clarified. Fifteen microsatellite loci were distinguishable and polymorphic with 6 types of allele-combinations. Null alleles were detected in 46.7% of loci, accounting for 11.7% of the total alleles. Four loci did not segregate in Mendelian Ratios. Three linkage groups were identified among 7 of the 15 segregating loci. Fluorescence-based automated capillary electrophoresis (ABI 310 Genetic Analyzer) that used to detect the microsatellite loci, has been proved a fast, precise, and reliable method in microsatellite genotyping.展开更多

Mycological analysis was comprised of 1,500 specimens of buds and petals a...The study of micromycetes infestation on flower buds, flower petals and seeds from 10 taxa of Rhododendron sp. was conducted during 2010-2012. Mycological analysis was comprised of 1,500 specimens of buds and petals and 500 seeds of Rhododendron sp.. It was shown that the necrosis and dieback of buds were caused by complex micromycetes (43 species), with dominants Pestalotiopsis sydowiana, Alternaria alternata, Truncatella truncata and Epicoccum nigrum. Watery brown spots on the flower petals, resulting in the dieback of flowers, were caused by 38 species, including the most common P. sydowiana, A. alternata and Trichoderma viride. The seeds were contaminated by 18 species, and in addition to the above, the following species were associated: Oidiodendron tennuissimum, Davidiella macrocarpa and Phoma leveillei. The results of the mycological analysis confirmed the diversity spectrum of micromycetes species that inhabit the infested Rhododendron buds, flowers and seeds. The research revealed which taxa attracted the largest number of colonies and species of fungi.展开更多
