Furthermore, filamentous bacteria in mud can be researched through fluorescence in situ hybridization, PCR-temperature denaturing gradient gel electrophoresis and PCR-single-stranded conformation polymorphism in order to determine the characters and states of active mud to achieve early warning of mud expansion.展开更多
Green fluorescent protein (GFP) fused to the F-actin binding domain of mouse talin labels the actin cytoskeleton in the living generative and sperm cells of a third generation transgenic rice (Oryza sativa L.) plant, ...Green fluorescent protein (GFP) fused to the F-actin binding domain of mouse talin labels the actin cytoskeleton in the living generative and sperm cells of a third generation transgenic rice (Oryza sativa L.) plant, A005-G-T-1-2. Observations were made on pollen at four major developmental stages, viz. I. uni-nucleate microspore stage; II. early bi-cellular pollen stage; III. late bi-cellular pollen stage; and IV. tri-cellular pollen stage. At each of these developmental stages vegetative nucleus, generative nucleus/ cell, and sperm cells were seen undergoing continuous and coordinated motion and migration. These movements seemed to be influenced by associated microfilament networks existing in the pollen. Based on these observations we propose that it is the interaction between the microfilament networks (usually one existing in the central cytoplasm and another in the cortex) that controls the dynamic movement of the vegetative nucleus, generative nucleus/cell and sperm cells. Furthermore, we have also observed that there is an array of microfilaments (oriented mostly parallel to the long axis of the cell) existing in the generative and sperm cells. As far as we are aware, this is the first report showing the existence of microfilaments in living generative and sperm cells of rice pollen. The implication and significance of the existence of microfilaments in generative and sperm cells in rendering self-propelled motion of these cells in relation to their passage and movement in the pollen tube and embryo sac for fertilization were discussed.展开更多

However, information on the fate of pencycuron in rice plant is lacking. The degradation of pencycuron in waterlogged tropic rice field was investigated.Pencycuron was applied at recommended field dose (187.5 g a.i./ha) and double recommended dose to cropped plots for three consecutive years. Pencycuron was rapidly degraded in rice plant at all doses of pencycuron application with first order half-lives of 1.57~2.77 d. In recent years,ternary heterostructures(HSs)composed of three semiconductors have attracted significant attention because of the effective separation and transfer of photogenerated electrons and holes in these materi...In recent years,ternary heterostructures(HSs)composed of three semiconductors have attracted significant attention because of the effective separation and transfer of photogenerated electrons and holes in these materials.In this work,new ternary Cd S/Zn S/Zn O(CZZ)HSs with one-dimensional(1D)nanofiber morphology have been successfully fabricated for the first time by a series of processes:electrospinning Zn O nanofibers,sulfurizing Zn O in situ to form Zn S/Zn O binary HSs,and depositing Cd S quantum dots in situ on the Zn S/Zn O HSs.Benefiting from the efficient separation and transfer of photoinduced charge carriers,the optimized CZZ ternary HSs exhibit a hydrogen evolution rate of 51.45 mmol h^-1 g^-1(quantum efficiency:26.88%at 420 nm)without any co-catalyst,which is 93.54 and 2.28 times higher than those exhibited by pristine Zn O and Zn S/Zn O binary HSs,respectively,under the same conditions.Furthermore,the rate of hydrogen evolution over the 1D CZZ nanofibers is significantly higher than that over 2D CZZ nanosheets(27.25 mmol h^-1 g^-1,in the presence of a Pt co-catalyst)prepared by the same sulfurization and deposition procedures.This can be ascribed to the significantly smaller geometric sizes of 1D nanofibers compared to those of 2D nanosheets,resulting in effectively suppressed recombination of photogenerated charge carriers and promotion of photocatalytic H2 evolution performance.展开更多

[Method] With the materials of rice root tips and anther, the specimen was prepared by the modified squash method, and microscopic observation of mitosis and meiosis in rice cells was also carried out. [ Result] Mitosis in rice cells included interphase, prophase, metaphase, anaphase and telophase. Chromosome in metaphase shortened to the minmum, which was a good time for observing and investigating chromosome. However, meiosis in rice cells included meiosis Ⅰ and meiosis Ⅱ. Chromosome replication appeared in meiosis Ⅰ, while cell division only appeared in meiosis Ⅱ. [ Conclusion] The modified squashing technique for rice chromosome can obtain accurate data of rice chromosome, which provides evidence for genetic breeding.展开更多

We tested the nutrient flux hypothesis by evaluating the ce...Turbulent mixing, in particular on a small scale, aff ects the growth of microalgae by changing diff usive sublayers and regulating nutrient fluxes of cells. We tested the nutrient flux hypothesis by evaluating the cellular stoichiometry and phosphorus storage of microalgae under dif ferent turbulent mixing conditions. A phanizomenon flos-aquae were cultivated in dif ferent stirring batch reactors with turbulent dissipation rates ranging from 0.001 51 m2/s 3 to 0.050 58 m 2/s 3, the latter being the highest range observed in natural aquatic systems. Samples were taken in the exponential growth phase and compared with samples taken when the reactor was completely stagnant. Results indicate that, within a certain range, turbulent mixing stimulates the growth of A. flos-aquae. An inhibitory ef fect on growth rate was observed at the higher range. Photosynthesis activity, in terms of maximum ef fective quantum yield of PSII(the ratio of F v/F m) and cellular chlorophyll a, did not change significantly in response to turbulence. However, Chl a/C mass ratio and C/N molar ratio, showed a unimodal response under a gradient of turbulent mixing, similar to growth rate. Moreover, we found that increases in turbulent mixing might stimulate respiration rates, which might lead to the use of polyphosphate for the synthesis of cellular constituents. More research is required to test and verify the hypothesis that turbulent mixing changes the dif fusive sublayer, regulating the nutrient flux of cells.展开更多

The L...Aspergillus oryzae 3042 mycelium pellets were immobilized by crosslinking method with reagents of gelatin and formaldehyde. An orthogonal design table was used to determine the optimal immobilization conditions. The L-aminoacylase activity of immobilized mycelium pellets under optimal conditions was assayed. The results show that the L-aminoacylase activity was 1350 U·g^-1 and the activity retention rate was 83%. The properties of the immobilized preparation were studied. Compared with free pellets, the appropriate pH of reaction system changed from 7.0 to 8.0, temperature changed from 52℃ to 63℃, and the ranges of the optimum reaction conditions were all improved. The effect of Co^2+ on immobilized mycelium pellets was investigated and the favorable concentration was determined. When the immobilized preparation was used for the resolution of D, L-alanine in a packed bed reactor with 10 mm in diameter, 200 mm in height, the operational stability was increased, and the half-life period was 53 d. Thermal stability analysis shows that the immobilized pellets were more stable than the free pellets.展开更多

When hyphae of Saprolegnia ferax were pre-fixed with paraformaldehyde, the TFP fluorescence showed a clear tip-high Ca(2+)-activated calmodulin (CaM) gradient. By indirect immunohistochemical staining, we proved that total CaM also mainly concentrated in the apex of hyphae.It is noticeble that when the hyphal tip growth ceased, the high intensity of TFP fluorescence usually disappeared in non-growing tips and became evident in the newly emerged growing tips, which implied that the concentration of CaM shifted in concord with the transfer of growing point in hyphae. 2×10(-5) mol/L CaM inhibitor TFP could reduce the tip growth rate and the percentage of growing hyphal tips to about 50%, and induce multibranching of hyphal tips. These findings strongly suggest that the high concentration of calmodulin in the hyphal tip may be involved in regulation of hyphal tip growth.展开更多

The influence of pH was significant:When pH is higher than 4.0, the high adsorption capacity is obtained,otherwise H+ ion inhibits the adsorption of heavy metal ions. The comparison of the chitin adsorbent with some other commercial adsorbents was made, in which that the adsorption behaviorchitin adsorbent is close to that of commercial cation exchange adsorbents, and its cost is much lower than those commercial adsorbents.展开更多

OsPMK1c, was isolated through screening of a cDNA library constructed from tillering materials. OsPI4Klc encoded a peptide of 608 amino acids with a calculated molecular mass of 68.4 kDa. The OsPI4Klc peptide shared high homology and possessed the highly conserved domains present in most isolated cloned PI4-kinases, i.e. a lipid kinase unique (LKU) domain and a catalytic (CAT) domain. A region with similarity to pleckstrin homology (PH) domain was present in OsPI4K1c as well. Further comparison with genomic sequences in databases revealed that OsPI4K1c is located at the 3'-end of a putative rice PI 4-kinase coding gene OsPI4K1, and its coding region corresponded to the C-terminal half of OsPI4Kl protein. Twelve exons (49-562 bp in size) and 11 introns (77-974 bp in size) were identified in OsPI4K1c. The recombinant protein expressed in Escherichia coli phosphorylates phosphatidylinositol at the D4 position of the inositol ring. OsPI4K1 transcript levels were detected in a low but constitutive manner in shoot, stem, leaf, spike and root tissues and did not change upon treatment with different hormones, calcium and jasmonic acid (JA). However, treatment with salicylic acid (SA) elevated the mRNA level of the OsPI4K1 gene, which suggested the involvement of OsPI4K1 in wounding responses.展开更多
The conventional silk fibroin(SF)films obtained by casting from SF aqueous solution are amorphous,and easily dissolved in water.This defect limits the practical use of SF films in biomedical field.A new water-insolubl...The conventional silk fibroin(SF)films obtained by casting from SF aqueous solution are amorphous,and easily dissolved in water.This defect limits the practical use of SF films in biomedical field.A new water-insoluble silk fibroin film(an SF-FA film)could be simply prepared by casting from SF formic acid solution.The physical properties and moisture permeability of SF-FA films were examined.The results show that the SF-FA film swollen in water exhibits outstanding pliability.The weight loss of SF-FA films in water is also very little.The unique physical properties for the SF-FA film in the wet relate to the intrinsic structure of the fibroin film.The cell viability of rat embryo dermal fibroblasts seeded on the control films and the SF-FA films was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay.Results show that the optical density(OD)value of the SF-FA film is almost the same as that of the conventional SF film treated with alcohol,therefore,the SF-FA film has good cell compatibility.In addition,the SF-FA film is also a biodegradable material.Because of their good transparency,it is helpful in study of clinical experiment with the SF-FA films.展开更多

The chain scission of DNA by Cu(Ⅱ)mitomycin C/ascorbic acid system,and the chemical kinetics of the Cu(Ⅱ) catalyzed aerobic oxidation of ascorbic acid in the presence of mitomycin C were also discussed. Experimental results showed that ascorbic acid and mitomycin C were synergetic in destroying Ehrlich ascites tumor cells.The effect is related to the breaking action of the Cu(Ⅱ)mitomycin C/ascorbic acid system on the DNA chain, and to the quantity of the system produced ·OH, and the speed of production which are both directly proportional to the concentration of ascorbic aci.These results imply that the ·OH produced in the aerobic oxidation of ascorbic acid,and·OH induced scission of DNA chain, are important factors to he synergetic cytocidal effect of ascorbic acid and mitomycin C on Ehrlich ascites tumor cells.展开更多

All the major stages of cell div...The pattern of change of the microtubule cytoskele-ton of the root-tip cells of rice during mitosis was studied using immunofluorescence technic and confocal laser scanning microscopy. All the major stages of cell division including preprophase, prophase, metaphase, anaphase and telophase were observed. The most significant finding was that in the preprophase cells microtubules radiating from the nuclear surface to the cortex were frequently seen. During development these microtubules became closely associated with the preprophase band and prophase spindle indicating that the microtubules radiating from the nuclear surface, the preprophase band and the prophase spindle were structurally and functionally closely related to each other. Granule-like anchorage sites for the radiating microtubules at the nuclear surface were often seen and the possibility that these granule-like anchorage sites might represent the microtubule organizing centres was discussed.展开更多

Compared with free pellets, the appropriate pH of reaction system changed from 7.0 to 8.0, temperature changed from 52℃ to 63℃, and the ranges of the optimum reaction conditions were all improved. The effect of Co^2+ on immobilized mycelium pellets was investigated and the favorable concentration was determined. When the immobilized preparation was used for the resolution of D, L-alanine in a packed bed reactor with 10 mm in diameter, 200 mm in height, the operational stability was increased, and the half-life period was 53 d. Thermal stability analysis shows that the immobilized pellets were more stable than the free pellets.
文摘In order to reduce the water shrinkage of rabbit hair knitted fabrics, a new method is developed, which is blending rabbit hairs with a little bit of blaze. The sericin on the blaze which can swell and melt in hot and wet condition can cohere the rabbit hairs through special processing. So the relative movement among fibers could be restricted. The testing results show that the water shrinkage of rabbit hair knitted fabrics can be greatly reduced after processed.
文摘Since direct staining of growing hyphae with trifluoperazine (TFP) could result in the migration of protoplasm in hyphae of water mold Saprolegnia feraX, an improved staining method for fixation of growing hyphae was developed. When hyphae of Saprolegnia ferax were pre-fixed with paraformaldehyde, the TFP fluorescence showed a clear tip-high Ca(2+)-activated calmodulin (CaM) gradient. By indirect immunohistochemical staining, we proved that total CaM also mainly concentrated in the apex of hyphae.It is noticeble that when the hyphal tip growth ceased, the high intensity of TFP fluorescence usually disappeared in non-growing tips and became evident in the newly emerged growing tips, which implied that the concentration of CaM shifted in concord with the transfer of growing point in hyphae. 2×10(-5) mol/L CaM inhibitor TFP could reduce the tip growth rate and the percentage of growing hyphal tips to about 50%, and induce multibranching of hyphal tips. These findings strongly suggest that the high concentration of calmodulin in the hyphal tip may be involved in regulation of hyphal tip growth.

文摘The adsorption properties of chitin adsorbent from mycelium of fermentation industries for the removal of heavy metal ions were studied. The result shows that the chitin adsorbent has high adsorption capacity for many heavy metal ions and Ni2+ in citric acid. The influence of pH was significant:When pH is higher than 4.0, the high adsorption capacity is obtained,otherwise H+ ion inhibits the adsorption of heavy metal ions. The comparison of the chitin adsorbent with some other commercial adsorbents was made, in which that the adsorption behaviorchitin adsorbent is close to that of commercial cation exchange adsorbents, and its cost is much lower than those commercial adsorbents.
文摘A partial rice (Oryza sativa L.) cDNA clone. OsPMK1c, was isolated through screening of a cDNA library constructed from tillering materials. OsPI4Klc encoded a peptide of 608 amino acids with a calculated molecular mass of 68.4 kDa. The OsPI4Klc peptide shared high homology and possessed the highly conserved domains present in most isolated cloned PI4-kinases, i.e. a lipid kinase unique (LKU) domain and a catalytic (CAT) domain. A region with similarity to pleckstrin homology (PH) domain was present in OsPI4K1c as well. Further comparison with genomic sequences in databases revealed that OsPI4K1c is located at the 3'-end of a putative rice PI 4-kinase coding gene OsPI4K1, and its coding region corresponded to the C-terminal half of OsPI4Kl protein. Twelve exons (49-562 bp in size) and 11 introns (77-974 bp in size) were identified in OsPI4K1c. The recombinant protein expressed in Escherichia coli phosphorylates phosphatidylinositol at the D4 position of the inositol ring. OsPI4K1 transcript levels were detected in a low but constitutive manner in shoot, stem, leaf, spike and root tissues and did not change upon treatment with different hormones, calcium and jasmonic acid (JA). However, treatment with salicylic acid (SA) elevated the mRNA level of the OsPI4K1 gene, which suggested the involvement of OsPI4K1 in wounding responses.

文摘The conventional silk fibroin(SF)films obtained by casting from SF aqueous solution are amorphous,and easily dissolved in water.This defect limits the practical use of SF films in biomedical field.A new water-insoluble silk fibroin film(an SF-FA film)could be simply prepared by casting from SF formic acid solution.The physical properties and moisture permeability of SF-FA films were examined.The results show that the SF-FA film swollen in water exhibits outstanding pliability.The weight loss of SF-FA films in water is also very little.The unique physical properties for the SF-FA film in the wet relate to the intrinsic structure of the fibroin film.The cell viability of rat embryo dermal fibroblasts seeded on the control films and the SF-FA films was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay.Results show that the optical density(OD)value of the SF-FA film is almost the same as that of the conventional SF film treated with alcohol,therefore,the SF-FA film has good cell compatibility.In addition,the SF-FA film is also a biodegradable material.Because of their good transparency,it is helpful in study of clinical experiment with the SF-FA films.
文摘The synergetic cytocidal effect of ascorbic acid and mitomycin C on Ehrlich as-cites tumor cells in culture was studied. The chain scission of DNA by Cu(Ⅱ)mitomycin C/ascorbic acid system,and the chemical kinetics of the Cu(Ⅱ) catalyzed aerobic oxidation of ascorbic acid in the presence of mitomycin C were also discussed. Experimental results showed that ascorbic acid and mitomycin C were synergetic in destroying Ehrlich ascites tumor cells.The effect is related to the breaking action of the Cu(Ⅱ)mitomycin C/ascorbic acid system on the DNA chain, and to the quantity of the system produced ·OH, and the speed of production which are both directly proportional to the concentration of ascorbic aci.These results imply that the ·OH produced in the aerobic oxidation of ascorbic acid,and·OH induced scission of DNA chain, are important factors to he synergetic cytocidal effect of ascorbic acid and mitomycin C on Ehrlich ascites tumor cells.
文摘The pattern of change of the microtubule cytoskele-ton of the root-tip cells of rice during mitosis was studied using immunofluorescence technic and confocal laser scanning microscopy. All the major stages of cell division including preprophase, prophase, metaphase, anaphase and telophase were observed. The most significant finding was that in the preprophase cells microtubules radiating from the nuclear surface to the cortex were frequently seen. During development these microtubules became closely associated with the preprophase band and prophase spindle indicating that the microtubules radiating from the nuclear surface, the preprophase band and the prophase spindle were structurally and functionally closely related to each other. Granule-like anchorage sites for the radiating microtubules at the nuclear surface were often seen and the possibility that these granule-like anchorage sites might represent the microtubule organizing centres was discussed.
