In addition to well-defined DNA repair pathways, all living organisms have evolved mechanisms to avoid cell death caused by replication fork collapse at a site where replication is blocked due to disruptive covalent m...In addition to well-defined DNA repair pathways, all living organisms have evolved mechanisms to avoid cell death caused by replication fork collapse at a site where replication is blocked due to disruptive covalent modifications of DNA. The term DNA damage tolerance (DDT) has been employed loosely to include a collection of mechanisms by which cells survive replication-blocking lesions with or without associated genomic instability. Recent genetic analyses indicate that DDT in eukaryotes, from yeast to human, consists of two parallel pathways with one being error-free and another highly mutagenic. Interestingly, in budding yeast, these two pathways are mediated by sequential modifications of the proliferating cell nuclear antigen (PCNA) by two ubiquitination complexes Rad6-Rad18 and Mms2-Ubc13-Rad5. Damage-induced monoubiquitination of PCNA by Rad6-Rad18 promotes translesion synthesis (TLS) with increased mutagenesis, while subsequent polyubiquitination of PCNA at the same K164 residue by Mms2-Ubc13-Rad5 promotes error-free lesion bypass. Data obtained from recent studies suggest that the above mechanisms are conserved in higher eukaryotes. In particular, mammals contain multiple specialized TLS polymerases. Among these, necroptosis, a regulatory caspase-independent, programmed cell death pathway, is supposed to be of importance in the pathogenesis of many diseases. The mechanism of regulating, in-ducing and blocking necroptosis is a complex process that involves expression and regulation of a series of molecules including receptor interacting protein kinase 1 (RIPK1), RIPK3, and mixed lineage kinase like protein. By blocking or downregulating expression of key molecules in the necroptotic pathway, intestinal inflammation can be affected to some extent. In this paper, we introduce the concept of necroptosis, its main pathway, and its impact on the pathogenesis ofinfammatory bowel disease (IBD) and other intestinal diseases, to explore new drug targets for intestinal diseases, including IBD.展开更多
In order to screen molecular markers linked to fertility restoring genes and further improve the breeding efficiency of restorer lines, in this study, wheat varieties 18A, 18B and 99AR144-1 were used as experimental m...In order to screen molecular markers linked to fertility restoring genes and further improve the breeding efficiency of restorer lines, in this study, wheat varieties 18A, 18B and 99AR144-1 were used as experimental materials to establish F2 fertility-segregating population. Plant quantitative trait "major gene + polygene mixed mo- del" separation analysis method and simple sequence repeat (SSR) molecular markers were adopted for genetic analysis of four generations, including the parents (P~ and P2), and hybrid (G and G) populations. The results show that AL-type fertility restoring gene is controlled by two pairs of additive-dominant-epistatic genes and addi- tive-dominant polygene; two primers linked to fertility restoring genes were selected by SSR molecular markers, including Xgwm95 on chromosome 2A and Barc61 on chromosome 1B, with the linkage distance of 15.0 cM and 18.0 cM, respectively. Based on verification, these two markers are reliable for distinguishing AL-type wheat ste- rile lines and restorer lines.展开更多

It also provides theoretical support for building an effective evaluation system. The basic idea is to use AHP and Grey Assessment to obtain the weights of the indicators, and count grey evaluation coefficients with whitening function. The compositive clustering coefficients are obtained by combining the weights and the grey evaluation coefficients. Evaluation result can be gotten from the compositive clustering coefficients.展开更多

The overbroken rock mass of gob areas is made up of broken and accumulated rock blocks compressed to some extent by the overlying strata. The beating pressure of the gob can directly affect the safety of mining fields...The overbroken rock mass of gob areas is made up of broken and accumulated rock blocks compressed to some extent by the overlying strata. The beating pressure of the gob can directly affect the safety of mining fields, formarion of road retained along the next goaf and seepage of water and methane through the gob. In this paper, the software RFPA'2000 is used to construct numerical models. Especially the Euler method of control volume is proposed to solve the simulation difficulty arising from plastically finite deformations. The results show that three characteristic regions occurred in the gob area: (1) a naturally accumulated region, 0-10 m away from unbroken surrounding rock walls, where the beating pressure is nearly zero; (2) an overcompacted region, 10-20 m away from unbroken walls, where the beating pressure results in the maximum value of the gob area; (3) a stable compaction region, more than 20 m away from unbroken walls and occupying absolutely most of the gob area, where the beating pressures show basically no differences. Such a characteristic can exolain the easy-seeoaged “O”-ring phenomena around mining fields very well.展开更多
Based on the historical observed data and the modeling results,this paper investigated the seasonal variations in the Taiwan Warm Current Water(TWCW)using a cluster analysis method and examined the contributions of th...Based on the historical observed data and the modeling results,this paper investigated the seasonal variations in the Taiwan Warm Current Water(TWCW)using a cluster analysis method and examined the contributions of the Kuroshio onshore intrusion and the Taiwan Strait Warm Current(TSWC)to the TWCW on seasonal time scales.The TWCW has obviously seasonal variation in its horizontal distribution,T-S characteristics and volume.The volume of TWCW is maximum(13746 km^3)in winter and minimum(11397 km^3)in autumn.As to the contributions to the TWCW,the TSWC is greatest in summer and smallest in winter,while the Kuroshio onshore intrusion northeast of Taiwan Island is strongest in winter and weakest in summer.By comparison,the Kuroshio onshore intrusion make greater contributions to the Taiwan Warm Current Surface Water(TWCSW)than the TSWC for most of the year,except for in the summertime(from June to August),while the Kuroshio Subsurface Water(KSSW)dominate the Taiwan Warm Current Deep Water(TWCDW).The analysis results demonstrate that the local monsoon winds is the dominant factor controlling the seasonal variation in the TWCW volume via Ekman dynamics,while the surface heat fl ux can play a secondary role via the joint ef fect of baroclinicity and relief.展开更多

The feasibility about the direct electro-reduction of NaV03 to metallic vanadi- um i...A novel and environmentally friendly route to directly prepare metallic vanadium from NaV03 by molten salt electrolysis is proposed. The feasibility about the direct electro-reduction of NaV03 to metallic vanadi- um is analyzed based on the thermodynamic calculations and experimental verifications. The theoretical decomposition voltage of NaV03 to metallic vanadium is only 0.47 V at 800 ℃ and much lower than that of the alkali and alkali earth metal chloride salts. The value is slightly higher than that of low-valence vanadium oxides such as V203, V305 and VO. However, the low-valence vanadium oxides can he further electro-reduced to metallic vanadium thermodynamically. The thermodynamic analysis is verified by the experimental results. The direct preparation of metallic vanadium from NaV03 by molten salt electrolysis is feasible.展开更多

The model of improved Support Vector Machine based on 1-norm(1-SVM) is provided from the optimization problem, yet it is a discrete programming. With the smoothing technique and optimality knowledge, the discrete programming is changed into a continuous programming. Experimental results show that the algorithm is easy to implement and this method can select and suppress the problem features more efficiently. Illustrative examples show that the 1-SVM deal with the linear or nonlinear classification well.展开更多
