[ Objective] The aim of this study was to provide a method for solving the problems in preparing BAC vector with High-copy plasmid pUC119- Bluelox BAC. [ Method ] With selecting a proper single restriction site, sequences of a single copy BAC vector plasmid were inserted into proper site of High-copy plasmid pUC119 vector. [ Result] The gene sequence of BAC vector lost control function of single copy number in new plasmid pUC119-BAC and was copied through High-copy form. The gene sequence of BAC vector basic function was completely cutted off through single enzyme digestion and the control function of single copy could be recovered by auto-connection. [ Conclusion] The High-copy pUC119-BAC plasmid was used to copy and amplify high copy of basic function gene sequence in BAC vector, besides that it could be used to construct transfer vector of molecular cloned recombinant virus or BAC library. 展开更多

Expanded bed adsorption(EBA),a promising and practical separation technique,has been widely studied in the past two decades.The development of adsorbents for EBA process is a challenging course,with the special design and preparation according to the target molecules and specific expanded bed systems.Many types of supporting matrices for expanded bed adsorbents have been developed,and their preparation methods are being consummated gradually.These matrices are activated and then coupled with ligands to form functionalized adsorbents,including ion-exchange adsorbents,affinity adsorbents,mixed mode adsorbents,hydrophobic charge induction chromatography adsorbents and others.In this review,the preparation of the matrices for EBA process is summa-rized,and the coupling of ligands to the matrices to prepare functionalized adsorbents is discussed as well. 展开更多

In this study,the sol-gel method was introduced to prepare the composite phase change material (CPCM). The CPCM was added to fabric with coating techniques and the thermal activity of modified fabric was studied. In addition,the thermal property and the microstructure of CPCM were also discussed in detail by means of polarization microscope and differential scanning calorimeter,respectively. According to the analysis of main influencial factors of the property of CPCM,the optimal preparing technique was determined. It was proved that CPCM could exhibit a good thermal property while phase transformation process took place,and a better appearance of the fabric modified with CPCM could be obtained due to the fact that in a warm circumstance,the liquid-state phase change material could be firmly enwrapped and embedded in the three-dimensional network all the time during the phase transformation. Besides,the fabric treated with CPCM had a high phase-transition enthalpy and an appropriate phase-transition temperature. As a result,a desirable temperature-adjustable function appeared. 展开更多

Novel Walking Stability-Based Gait Recognition Method for Functional Electrical Stimulation System Control

Gait recognition is the key question of functional electrical stimulation (FES) system control for paraplegic walking. A new risk-tendency-graph (RTG) method was proposed to recognize the stability information in FES-assisted walking gait. The main instrument was a specialized walker dynamometer system based on a multi-channel strain-gauge bridge network fixed on the walker frame. During walking process, this system collected the reaction forces between patient's upper extremities and walker and converted them into RTG morphologic curves of dynamic gait stability in temporal and spatial domains. To demonstrate the potential usefulness of RTG, preliminary clinical trials were done with paraplegic patients. The gait stability levels of two walking cases with 4- and 12-week FES training from one subject were quantified (0.43 and 0.19) from the results of temporal and spatial RTG. Relevant instable phases in gait cycle and dangerous inclinations of patient's body during walking process were also brought forward. In conclusion, the new RTG method is practical for distinguishing more useful gait stability information for FES system control. 展开更多

The contents about the central action mechanisms of needling Zusanli(ST 36) were sorted by summarizing the relevant literatures published in the past 10 years, and it was found that: by comparing acupuncture at Zusanli(ST 36), other acupoints or sham points, most studies showed that Zusanli(ST 36) had relatively specific characteristics in the brain function response; studies on the central action mechanisms of Zusanli(ST 36) were mainly focused on sequelae of cerebral apoplexy, pain, gastrointestinal diseases, weight loss and drug addiction withdrawal and so on; acupuncture manipulations, stimulation methods, individual differences, stimulation quantity, and stimulation duration, etc. could also induce different brain function responses; acupuncture at Zusanli(ST 36) had an after-working effect, also called long-term effect, but with poor repeatability. So, it was suggested that the future studies should focus on the factors that affect the clinical efficacies and experimental results, including the compatibility effects of the acupoints, reproducibility of research results, sample size, acupuncture methods, long-term effects and disease entities. 展开更多

When the five-axis CNC system executes the 3D cutter radius compensation function,the angle between two adjacent radius compensation vectors might become very large and the linear axes would move too fast if the tool orientation vector is close to the surface normal.The reason that results in this phenomenon is analyzed based on building the transmission relationship between the cutter contact point and the cutter location point.By taking the square-end tool as an example,an optimization algorithm to control the undesired movements is advanced.For the singular area where sudden change exists,the number of interpolation cycles is determined by the cutter feedrate,the limit speeds of machine axes and the maximum allowable angle between radius compensation vectors of adjacent NC blocks.The radius compensation vector of each interpolation cycle is obtained by a kind of vector rotation method.By maintaining the perpendicularity between the radius compensation vector and the tool orientation vector,the rapid movements of the linear axes are eliminated.A trial-cut experiment is performed to verify the correctness and the effectiveness of the proposed algorithm. 展开更多

A novel approach is designed to optimize the synthesis of sulfonic-functionalized silica material. Results from 29Si and 27AI NMR suggest that the AI acts as the bridging atom connecting the methanesulfonate and silica matrix. Further pyridine-FTIR spectra followed by catalytic activity tests demonstrate that compared with previous methods, our new approach results in higher Lewis acid site concentration, higher thermal stability and superior catalytic activity. Moreover, the whole catalysis preparation procedure is environmentally friendly. Specifically, the silica matrix is synthesized through hydrolysis of tetrae- thylorthosilicate employing formic acid as hydro-catalyst, in which no surfactant species or precursors were involved. 展开更多
