A reversed-phase high performance liquid chromatographic (RP-HPLC) method wasdeveloped and validated for the simultaneous deteimination of ceftazidime and tazobactam ininject-able powder. Methods Chromatography was carried out on Zorbax 300SB-C_(18) column using amixture of methanol and aqueous solution of phosphate buffer (pH = 5.6) as mobile phase. The UVdetection wavelength was 220 run. Results The linear ranges of ceftazidime and tazobactam were 0.62- 631.8 μg·mL^(-1) and 0.66 - 677.50 μg·mL^(-1), respectively. The average recoveries were 98.8%- 101.4% for ceftazidime, and 99,1% - 100.2% for tazobactam. The RSD values of inter-day andintra-day assays were lower than 1.5% for ceftazidime and 2.6% for tazobactam. Conclusion Thismethod is reproducible, simple, precise, and rapid for the quality control of ceftazidime andtazobactam in injectable powder. 展开更多

As the strict limitation of primary structure in traditional force method and displacement method in indeterminate analysis may lead to complicated high-order linear equations, a breakthrough of the limitation, i.e., the application of irregular force method and irregular displacement method, would be introduced in this paper to ease the difficulty of hand computations. By using hyperstatic primary structures and partly chained primary structures, the primary structures of force method and displacement method are reformed, and the order of the system is decreased. This paper considers a new form of the Steiner tree problem that is more practical and reliable,which we call Reliable Steiner Tree(RST)problem.The authors give a detailed definition for this new problem and design both an exact algorithm and an approximation algorithm for it.The definition is based on the reliability of full components instead of Steiner vertices.The task is thus to find the most reliable full components to make up an optimum reliable Steiner tree.The exact algorithm designed for this problem utilizes a dynamic programming frame.The approximation algorithm designed in this paper exploits a local search strategy that looks for the best full component according to a selection function at a time. 展开更多

A multiplexed targeted proteomic assay using a mTRAQ-MRM/MS-based approach was developed and assessed to systematically quantify the relative expressions of five candidate plasma apolipoproteins that have been previously shown to be dysregulated in neuropsychiatric disorders and cognitive dysfunction:apolipoprotein H(APOH),apolipoprotein J(APOJ),apolipoprotein A4(APOA4),apolipoprotein E(APOE),and apolipoprotein D(APOD).The peptides and transitions of each APO were carefully selected according to the tandem MS signals acquired on a TripleTOFTM 5600,followed by optimization of the declustering potential and collision energy voltages for transitions on a QTRAP 5500.Our results showed that the collision energies of mTRAQ-labeled peptides were approximately 15%–20%higher than corresponding non-labeled peptides.Through optimized transitions and parameters,we analyzed the relative abundances of the five APOs in human plasma with and without depletion of high abundant proteins.The results indicated that the MRM signals of four target APOs were significantly increased after depletion,while the MRM signal of one APO,APOD,was decreased.Furthermore,the relative abundances of the five target APOs in healthy human plasma were stable,and the ranking of these proteins according to their MS responses changed slightly.Therefore,we deduced that the rank order of the MS signals for these target proteins can be developed as a diagnostic signature for diseased plasma. 展开更多

Considering the coupled nonlinear Schr¨odinger system with multiply components, we provide a novel framework for constructing energy-preserving algorithms. In detail, based on the high order compact finite difference method, Fourier pseudospectral method and wavelet collocation method for spatial discretizations, a series of high accurate conservative algorithms are presented. The proposed algorithms can preserve the corresponding discrete charge and energy conservation laws exactly, which would guarantee their numerical stabilities during long time computations.Furthermore, several analogous multi-symplectic algorithms are constructed as comparison. Numerical experiments for the unstable plane waves will show the advantages of the proposed algorithms over long time and verify the theoretical analysis. 展开更多
