采用本氏试剂与羟甲基纤维素水解还原糖反应的细胞化学方法,对玉米不定根淹水诱导形成通气组织过程中,逐步解体的皮层细胞内纤维素酶活性进行了定位观察。显示酶活性的沉淀颗粒首先出现在离根尖4mm的少数皮层细胞中,沿细胞壁分布;... 采用本氏试剂与羟甲基纤维素水解还原糖反应的细胞化学方法,对玉米不定根淹水诱导形成通气组织过程中,逐步解体的皮层细胞内纤维素酶活性进行了定位观察。显示酶活性的沉淀颗粒首先出现在离根尖4mm的少数皮层细胞中,沿细胞壁分布;在离根尖5~6mm的皮层组织内,有沉淀颗粒的皮层细胞数量增多,而且沉淀颗粒的密度也显著增大,沉淀颗粒仍沿细胞壁均匀分布;在离根尖7—8mm皮层细胞内,沉淀颗粒尽管仍主要沿细胞壁分布,但由原来的均匀分布变为凝集态的不连续分布。在离根尖9mm的皮层细胞内,沉淀颗粒则以凝集态分布在细胞壁上。在离根尖10mm的皮层组织内,部分细胞的细胞质被完全消化,细胞壁也被明显降解,呈残余态。随着局部位置的皮层细胞完全消化,在离根尖11mm的皮层组织内开始形成通气组织。研究结果显示纤维素酶活性的表达与皮层细胞的细胞壁降解紧密相联,支持前人提出的纤维素酶参与了通气组织形成过程的假说。 展开更多

以烟草(Nicotiana tabacumL.)花药为材料,通过4,’6-二脒基-2-苯基吲哚(DAPI)染色详细观察花粉发育过程,获得了花药发育时期与花蕾大小的对应关系;通过吲哚乙酸(IAA)单克隆抗体、免疫组织化学技术以及DR5∶∶GUS转基因植株的GUS活性对... 以烟草(Nicotiana tabacumL.)花药为材料,通过4,’6-二脒基-2-苯基吲哚(DAPI)染色详细观察花粉发育过程,获得了花药发育时期与花蕾大小的对应关系;通过吲哚乙酸(IAA)单克隆抗体、免疫组织化学技术以及DR5∶∶GUS转基因植株的GUS活性对花药和花粉发育过程中生长素的分布规律进行了研究。免疫酶标记结果表明,在不同的花药发育时期IAA水平呈现出明显的差别。小孢子母细胞时期,IAA在整个花药中均有分布,并且在小孢子母细胞发育晚期,IAA信号集中在小孢子母细胞的细胞核中;随着小孢子母细胞减数分裂后形成四分体,IAA信号逐渐减弱,四分体中几乎没有信号;单核花粉期的花药中IAA信号进一步减弱,仅存在于花药壁中;待小孢子继续发育为成熟二核期时,花粉和整个花药组织中均出现较强的IAA信号。GUS活性检测结果表明,烟草DR5∶∶GUS转基因植株中花药和花粉粒的GUS信号与IAA免疫酶定位结果基本一致。总的来说,IAA在烟草花药和花粉中的积累呈现出由强到弱、再由弱到强的分布规律,暗示IAA在被子植物花药和花粉发育过程中可能起着较为重要的作用。 展开更多

甲状腺手术特别是甲状腺全切加中央区淋巴结清扫的主要风险之一是甲状旁腺误切和(或)血运受损,可致暂时性或永久性甲状旁腺功能减退,引起低钙血症,目前已成为困扰甲状腺手术医生和患者的难题之一。原发性或肾性甲状旁腺功能亢进需切除... 甲状腺手术特别是甲状腺全切加中央区淋巴结清扫的主要风险之一是甲状旁腺误切和(或)血运受损,可致暂时性或永久性甲状旁腺功能减退,引起低钙血症,目前已成为困扰甲状腺手术医生和患者的难题之一。原发性或肾性甲状旁腺功能亢进需切除病变旁腺,而术中快速寻找或辨认旁腺有时较困难,特别是存在异位或额外旁腺时。目前对甲状旁腺的寻找辨认和保护主要依靠外科医师的经验,传统术中快速冰冻、纳米炭负显影和甲状旁腺激素(parathyroid hormone,PTH)试纸等辅助辨认方法仍有一定局限性。近红外自体荧光成像和光学相干断层成像可直观准确的进行术中甲状旁腺实时定位显示,吲哚菁绿荧光成像和激光散斑对比成像对术中甲状旁腺活性判断也有一定价值。本文就这些光学技术的应用原理、初步临床研究和应用前景进行综述。 展开更多

Comparing with conventional radioisotope based method, it was better in reproducibility and accuracy. Using this method, we found telomerase activities were absent in normal human liver cells, while detected in ail of four human hepatoma cell lines (BEL-7404, SMMC-7721, QGY-7903 and HCCM) without significant differences. 展开更多

Plasma membrane (PM) Ca2+-ATPase activity in poplar apical bud meristematic cells during short-day (SD)-induced dormancy development was examined by a cerium precipitation EM-cytochemical method. Ca2+-ATPase activity, indicated by the status of cerium phosphate precipitated grains, was localized mainly on the interior face (cytoplasmic side) of the PM when plants were grown under long days and reached a deep dormancy. A few reaction products were also observed on the nuclear envelope.When plant buds were developing dormancy after 28 to 42 d of SD exposure, almost no reaction products were present on the interior face of the PM. In contrast, a large number of cerium phosphate precipitated grains were distributed on the exterior face of the PM. After 70 d of SD exposure, when buds had developed a deep dormancy, the reaction products of Ca2+-ATPase activity again appeared on the interior face of the PM. The results seemed suggesting that two kinds of Ca2+-ATPases may be present on the PM during the SD-induced dormancy in poplar.One is the Ca2+-punlping ATPase, which is located on the interior face of the PM, for maintaining and restoring the Ca2+homeostasis. The other might be an ecto-Ca2+-ATPase, which is located on the exterior face of the PM, for the exocytosis of cell wall materials as suggested by the fact of the cell wall thickening during the dormancy developlnent in poplar. 展开更多

Objective To investigate the anti-tumor effects in vitro and in vivo distribution of the human/murine chimeric antibody (D2C). Methods The CD71 positive target cells (K562, GEM and SMMC7721) and the effector cells, freshly isolated human PBMC, with the ratio of target cells to effector cells 1:50, were incubated in various dilutions of D2C antibody ( Ab) . Antibody dependent cytotoxicity (AD-CC) was tested by using an LDH-release assay. Instead of effector cells, complement was added to the target cells (GEM, SMMC-7721) with various dilutions of D2C Ab. A method of counting death cells was used in complement dependent cytotoxicity (CDC) assay. Tumor localization and distribution of the chimeric antibody (D2C) were observed by labeling the chimeric Ab with radioiodine(131I) and injecting it into nude mice (Balb/c nu/nu) transplanted with human hepatocellular carcinoma cells (SMMC-7721).Results A significant ADCC was observed with the increased concentration of the D2C Ab. Cytolysis of CD71-positive target cells by the D2C Ab was found in the presence of fresh rabbit complement. Labeled D2C administered by intraperitoneal as well as tumor regional injection, was visualized by SPECT. The distribution of D2C Ab in murine organs and tissues showed that non-specific binding was lower following tumor regional administration than when the antibody was administered by an intraperitoneal injection. The human/murine chimeric antibody (D2C) has in vitro anti-tumor effects and can exert its effects in specific tumor localization. Its distribution and local effects in vivo can be detected by radioimmunoimaging.Conclusion CD71 human/murine chimeric antibody showed marked killing of tumor cells in vitro, and specific recognition and high affinity binding to tumor tissue in vivo 展开更多

We investigated the distribution of four enzymes involved in the immune response of Apostichopus japonicus. We collected samples of the tentacles, papillate podium, integument, respiratory tree, and digestive tract and stained them for acid phosphatase (ACP), alkaline phosphatase (AKP), non-specific esterase (NSE) and peroxidase (POD) activity. The distribution and content of ACE AKP, NSE, and POD differed among the tissues. The coelomic epithelium of the tentacle, papillate podium, and integument and the mucous layer of respiratory tree were positive for ACP activity. The coelomic epithelium and cuticular layer of the tentacle, papillate podium, and integument and the mucous layer and tunica externa of the respiratory tree and digestive tract stained positive or weakly positive for AKP activity. Almost all the epithelial tissues stained positive, strongly positive, or very strongly positive for NSE activity. The cuticular layer of the tentacle and integument and the mucous layer, tunica submucosa, and tunica externa of the respiratory tree and digestive tract stained positive for POD activity. We hypothesize that these enzymes play a role in the immune response in A. japonicus. 展开更多

Crustal Movement in the Northern Part of North China Associated with Zhangbei Earthquake

Crustal Deformation Monitoring Center, China Seismological Bureau, Tianjin 300180, China Crustal deformation in the northern part of North China associated with Zhangbei earthquake is analyzed using GPS data collected during 1992, 1996 and 1999, precise re-leveling data collected during 1992, 1998 and 2000, and INSAR result (September 22, 1997~May 6, 1998). The results indicate: ① The vertical deformation is not remarkable since 1992. The vertical crustal deformation in the central and northern part of North China in recent 10 years is of inheritance. The scope of the significant deformation is 15km×15km with a magnitude of 250mm when the event occurred. ② The horizontal deformation is not remarkable in any unit of North China. ③ Before and after the event (1992~1996 and 1996~1999), there is kinematic change of horizontal motion between different units. The activity mode along Yinshan tectonic zone changed from relative static state to left-lateral strike slip; The dominant extensional movement along Shanxi rift zone changed to right-lateral strike slip; Yanshan tectonic zone changed from left-lateral strike-slip with extension to left-lateral strike slip; Yanshan-Hebei transitional zone formed before the event disappeared after the event. ④ The scale of the deformation is closely related to the physical property of media and geological structure environment. Further analysis indicates that ① Zhangbei earthquake does not mean that the earthquake activity begins to be strengthened in North China; ② The crustal movement is normal at present; ③ Next stronger earthquake in North China might be located in Yanshan tectonic zone, especially at its both ends, and Shanxi tectonic zone. 展开更多

In the prostate cancer treatment, brachytherapy with iodine-125 seeds has been used. lodine-125 seeds are sealed radioactive sources, made by a titanium capsule containing the radioisotope inside. In the final phase of the seeds production, it is necessary to ensure that there is no leakage of the radioactive material. A leakage test is performed, immersing the seeds in water during 24 h and measuring the resulting activity in the water. This measurement is made in a sodium iodide detector. The immersion water is transferred to a plastic tube with a cap. The tube is placed by an automated positioning system, in the detector chamber. This study aims to determine the best positioning of the tube for the detection. It is also important to determine the influence of the positioning variation intrinsic of the automated positioning system during the iodine-125 seeds production. The results obtained will be used as a reference to adjust the equipment and process control system, in the production of the iodine-125 seeds. 展开更多

This article looks at how population movements are addressed by the Sendai Framework for Disaster Risk Reduction 2015–2030(SFDRR), and highlights some of the potential implications of the SFDRR on disaster risk reduction(DRR) and mobility management work. The article looks at the operational implications of the SFDRR text and covers issues of including migrants in DRR work;informing urban development about current and future mobility trends; managing relocations, evacuations, and displacement to prevent future risks and reduce existing ones; and preparing for and managing disaster-induced population movements to reduce the direct and indirect impacts of natural hazards. Overall, the references to human mobility within the SFDRR show an evolution in the way the issue is considered within global policy dialogues. Both the potential of population movements to produce risk and their role in strengthening the resilience of people and communities are now clearly recognized. This is an evolution of previously prevailing views of mobility as the consequence of disasters or as a driver of risk. While some implications of the DRR-mobility nexus might still be missing from DRR policy, population movements are now recognized as a key global risk dynamic. 展开更多

Against this background,we explored the unique ideas, properties, and standards of fracture repositioning in traditional Mongolian medicine.RESULTS: Fracture treatment with osteopathy in traditional Mongolian medicine in China is based on(a) the ideas of natural, sealed, self and dynamic repositioning of fractures;(b) the properties of structural continuity and functional completeness;(c) the standards of "integration of movement and stillness" and "force to force." CONCLUSION: The unique ideas, properties, and standards of fracture repositioning with osteopathy in traditional Mongolian medicine in China have resulted in the widespread use of such techniques and represents the future direction of the development of fracture repositioning. 展开更多

Saturated row-column designs are studied to eliminate positional effects in primary high- throughput screening experiments. Compared to designs currently used in practice, all compounds in the designs considered are comparable within each microplate in spite of the existence of row and column effects. The designs considered in this paper also have the maximum number of compounds arranged in each microplate. Two statistical methods are used to choose leading compounds in the designs considered. These two methods take full advantages of effect sparsity in primary screening. Simulation studies are carried out to compare the two statistical methods with two ad hoc methods in selecting active compounds. A method that maintains balanced false positives and false negatives is recommended. 展开更多
