Preventive properties against lipid metabo...In this report, we reviewed recent literature on physiologically active substances from sea cucumbers (SCs) andtheir activities together with results obtained from our study. Preventive properties against lipid metabolism were reported inrats using a whole SC preparation with no particular constituent specified. Administration of the preparation lowered serumand hepatic cholesterol levels and improved the HDL/LDL ratio. These functions may be attributed to the stimulatory effectof the extract on the secretion of cholesterol in feces. Novel fucosylated chondroitin sulfates (FCSs) from Ludwigothureagrisea significantly induced fibroblast growth factor 2-dependent angiogenesis in human umbilical vein endothelial cells (HU-VECs). The proangiogenetic activity seemed attributable to the action of the sulfated fucose branches on the polysaccharide.SCs contain mycosporine-like amino acids (MAAs) that are capable of absorbing UV. A biogenetic precursor of MAAs wasfirst reported in SCs. The anti-proliferative effects of a branched chain fatty acid from a sea cucumber on prostate cancercells was reported with the activity of 5-lipoxygenase. Glycosphingolipid constituents in SCs have been systematically ana-lyzed over the past ten years. The results showed that the gangliosides in several SCs differed from those of mammals in thata sialic acid of SC gangliosides directly binded to glucose of cerebroside. Neuritogenic activity of the glycosphingolipids wasdemonstrated in vitro experiments and may lead to the development of therapeutic products for neurological disorders. Ourstudy also showed that sphingoid bases, the hydrolyzed products of glycosphingolipids from SCs, induced significant apoptosisin several tumor cell lines.展开更多

Ten SSR primers were used to asses...Sea cucumber Apostichopus japonicus samples were collected in Changdao, Penglai (PL), 27 individuals, and Lingshandao, Qingdao (QD), 30 individuals, in the Shandong Peninsula, China. Ten SSR primers were used to assess the genetic variation and relationship between and within the two stocks. Respectively, for each stock, the percentage of polymorphic bands was 85.2% and 86.9%; the gene diver- sity was 0.360 5 and 0.342 8; and the Shannon’s information index was 0.515 0 and 0.499 0. At species level, the percentage of polymorphic bands was 92.2%, the total gene diversity was 0.378 9 and the Shannon’s information index was 0.550 4. The coefficient of overall genetic differentiation and the ge- netic distances between the stocks were also calculated to be 0.073 0 and 0.079 6 using the POPGENE program. Results show that the genetic diversity of the two stocks is still large but the genetic distance between the two stocks is close. A dendrogram was constructed for the 57 individuals from the two stocks, showing that the genetic structure was unitary for PL stock but complex for QD stock.展开更多
The tribological behaviors of Cu-15Ni-8Sn/graphite composites with the graphite content of 38 vol.%against AISI321 stainless steel under dry-sliding,deionized water and sea water were investigated on a block-on-ring c...The tribological behaviors of Cu-15Ni-8Sn/graphite composites with the graphite content of 38 vol.%against AISI321 stainless steel under dry-sliding,deionized water and sea water were investigated on a block-on-ring configuration.The results indicated that the friction coefficient was the lowest under dry-sliding,and the highest in deionized water.The wear rate decreased to reach the minimum value of 1.39×10-15 m3/(N·m)in sea water and in deionized water,it increased to the maximum value of 5.56×10-15 m3/(N·m).The deionized water hindered the formation of tribo-oxide layer and lubricating film,which resulted in the largest friction coefficient and wear rate.In sea water,however,the corrosion products comprised of oxides,hydroxides and chlorides were found on the worn surface,and the compacted layer composed of corrosion products and graphite played an important role in keeping the excellent wear resistance.It was elucidated that the tribological behaviors of Cu-15Ni-8Sn/graphite composite were powerful influenced by the friction environments.展开更多

One new diketopiperazine brevianamide W(1a), as well as five known diketopiperazine alkaloids, diketopiperazine V(1b), brevianamide Q(2), brevianamide R(3), brevianamide K(4), and brevianamide E(5), were isolated from the Et OAc extract of the fermentation broth. Their structures were elucidated by spectroscopy techniques(NMR, MS). The six compounds exhibited moderate radical scavenging activity against DPPH with clearance ratio of 55.0%(1a and 1b), 53.7%(2), 46.2%(3), 61.4%(4) and 19.3%(5) at a concentration of 13.9 μmol L-1, respectively; while the positive control ascorbic acid showed a ratio of 70.3% at the concentration of 28.4 μmol L-1.展开更多

Marine natural product neolamellarins are a collection of lamellarin-like phenolic pyrrole compounds, which can inhibit hypoxia-induced HIF-1 activation. Many pyrrole-derived lamellarin-like alkaloids show potent MDR reversing activity. In this study, five permethylated derivatives of neolamellarin A were synthesized with their MDR reversing activity studied in order to identify new MDR reversal agents. A convergent strategy was adopted to synthesize the permethylated derivatives of neolamellarin A. Pyrrole was first converted into a corresponding N-trisisopropylsilyl (TIPS)-substituted derivative, then through iodination afforded 3,4-diiodinated pyrrole compound. The key intermediate, 3,4-disubstituent-lH-pyrrole, was obtained through desilylation of 3,4-disubstituent-l-TIPS pyrrole, which was prepared from 3,4-diiodinated pyrrole derivative and aryl boronic acid ester through Suzuki cross-coupling reaction between them. Then, the intermediate, 3,4-disubstituent-lH-pyrrole, reacted with fresh phenylacetyl chloride under n-BuLi/THF condition afforded the target compounds. Finally, we obtained five novel pyrrolic com- pounds, permethylated derivatives ofneolamellarin A 16a-e, in 30%-37% yield through five step reactions. The bioactivity testing of these compounds are in process.展开更多

Orobanche species cause severe yield reduction of many important crops. There are only very few herbicides which are able to selectively control broomrapes and different approaches have been put forward to develop natural product based pesticides to control Orobanche. Several phytopathogenic fungi were evaluated for their use as potential mycoherbicide and for ability to produce toxic metabolites which could be applied as herbicides. Using the alternative approach "suicidal germination", interesting results were obtained by testing two microbial metabolites (fusicoccins and ophiobolin A) especially with Orobanche species whose germination is not induced by the synthetic strigolactone GR24. From pea root exudates, peagol and peagoldione, close related to strigolactones, and three polyphenols, named peapolyphenols A-C, together with already well known polyphenol and a chalcone, were isolated. They showed a selective stimulation of Orobanche seed germination with the last two and peapolyphenol A showing a specific stimulatory activity on O. foetida. This review describes the most recent results achieved on Orobanche bio-control, mainly focusing on those regarding O. ramosa, O. crenata and O. foetida.展开更多
Caspase-1-mediated IL-1β production is generally controlled by two pathways. Toll-like receptors (TLRs) recognize pathogen-derived products and induce NF-KB-dependent pro-IL-1β transcription; NOD-like receptors (...Caspase-1-mediated IL-1β production is generally controlled by two pathways. Toll-like receptors (TLRs) recognize pathogen-derived products and induce NF-KB-dependent pro-IL-1β transcription; NOD-like receptors (NLRs) assemble caspase-l-activating inflammasome complexes that sense bacterial products/danger signals. Through a targeted chemical screen, we identify bromoxone, a marine natural product, as a specifc and potent inhibitor of the caspase-1 pathway. Bromoxone is effective over diverse inflammatory stimuli including TLR ligands plus ATP/nigeri- cin, cytosolic DNA, flagellin and Bacillus anthracis lethal toxin. Bromoxone also efficiently suppresses easpase-1 acti- vation triggered by several types of bacterial infection. Bromoxone acts upstream or at the level of the inflammasome in a transcription-independent manner. Bromoxone also inhibits pro-IL-1β expression by targeting components up- stream of IKK in the TLR-NF-kB pathway. The unique dual activities of bromoxone are shared by the known TAK1 inhibitor that specifically blocks Nalp3 inflammasome activation. Hinted from the mechanistic and pharmacological properties of bromoxone, we further discover that several known NF-KB inhibitors that act upstream of IKK, but not those targeting IKK or IKK downstream, are potent blockers of different NLRs-mediated caspase-1 activation. Our study uncovers a possible non-transcriptional molecular link between the NLR (Nalp3)-mediated inflammasome pathway and TLR-NF-kB signaling, and suggests a potential strategy to develop new anti-inflammatory drugs.展开更多

These activities can cause environmental problems and health prob...Throughout the world, the coastal zones of many countries are used increasingly for aquaculture in addition to other activities such as waste disposal. These activities can cause environmental problems and health problems where they overlap. The interaction between aquaculture and waste disposal, and their relationship with eutrophication are the subjects of this paper. Sewage discharge without adequate dispersion can lead to nutrient elevation and hence eutrophication which has clearly negative effects on aquaculture with the potential for toxic blooms. Blooms may be either toxic or anoxia-causing through the decay process or simply clog the gills of filter-feeding animals in some cases. With the development of aquaculture, especially intensive aquaculture, many environmental problems appeared, and have resulted in eutrophication in some areas. Eutrophication may destroy the health of whole ecosystem which is important for sustainable aquaculture. Sewage discharge may also cause serious public health problems. Filter-feeding shellfish growing in sewage-polluted waters accumulate micro-organisms, including human pathogenic bacteria and viruses, and heavy metal ion, presenting a significant health risk. Some farmed animals may also accumulate heavy metals from sewage. Bivalves growing in areas affected by toxic algae blooms may accumulate toxins (such as PSP, DSP) which can be harmful to human beings.展开更多
Diatoms and dinoflagellates are two major groups ofphytoplankton that flourish in the oceans, particularly in coastal zone and upwelling systems, and their contrasting production have been reported in several world se...Diatoms and dinoflagellates are two major groups ofphytoplankton that flourish in the oceans, particularly in coastal zone and upwelling systems, and their contrasting production have been reported in several world seas. However, this information is not available in the coastal East China Sea (ECS). Thus, to investigate and compare the decadal trends in diatoms and dinoflagellates, a sediment core, 47-cm long, was collected from the coastal zone of the ECS. Sediment chlorophyll-a (Chl-a), phytoplankton-group specifc pigment signatures of diatoms and dinoflagellates, and diatom valve concentrations were determined. The sediment core covered the period from 1961 to 2011 AD. The chlorophyll-a contents ranged from 2.32 to 73 gg/g dry sediment (dw) and averaged 9.81 gg/g dw. Diatom absolute abundance ranged from 29 152 to 177 501 valve/gram (v/g) dw and averaged 72 137 v/g dw. Diatom valve and diatom specific pigment marker concentrations were not significantly correlated. Peridinin increased after the 1980s in line with intensified use of fertilizer and related increases in nutrient inputs into the marine environment. The increased occurrence of dinoflagellate dominance after the 1980s can be mostly explained by the increase in nutrients. However, the contribution of dinoflagellates to total phytoplankton production (Chl-a) decreased during the final decade of this study, probably because of the overwhelming increase in diatom production that corresponded with the construction of the Three Gorges Dam (TGD) and related light availability. Similarly, the mean ratio of fucoxanthin/peridinin for the period from 1982 to 2001 was 6% less than for 1961 to 1982, while the ratio for 2001 to 2011 was 45.3% greater than for 1982 to 2001. The decadal variation in the fucoxanthirdperidinin ratio implies that dinoflagellate production had been gradually increasing until 2001. We suggest that the observed changes can be explained by anthropogenic impacts, such as nutrient loading and dam construction.展开更多

Xanthocillin X (1) was isolated for the first time from a marine fungus. In the bioassay, xanthocillin X (1) displayed remarkable antimicrobial activity against Staphylococcus aureus and Escherichia coli, and significant cytotoxicity against MCF-7, HepG2, H460, Hela, Du145, and MDA-MB-231 cell lines. Meleagrin (15) exhibited cytotoxicity against HepG2, Hela, Du145, and MDA-MB-231 cell lines. This is the first report of the cytotoxicity of xanthocillin X (1).展开更多

However, the accurate species identification and delim...Sparids are of considerable economic importance in marine fishery and aquaculture in China, and the species diversity of this group is considered relatively high. However, the accurate species identification and delimitation of sparids in China remain unaddressed. In this study, we used mitochondrial cytochrome oxidase subunit ?(COI) and 16S ribosomal RNA(16S) genes to conduct DNA barcoding and species delimitation in eleven sparid species from the coastal waters of China. Based on Kimura-2 parameter genetic distances, the mean intraspecific/interspecific variation for COI and 16S were calculated as 0.004/0.152 and 0.002/0.072, respectively. All the conspecific individuals formed monophyletic clusters in neighbour-joining trees of both markers. An obvious barcoding gap was detected for each species, and a common genetic threshold of 1.3% sequence divergence was defined for species delimitation in both markers. Although the sequence variation of 16S was generally lower than that of COI, the results indicated that sparid species could be ef fectively and accurately identified and delimited by COI as well as 16S. Thus, we propose that the COI gene serve as the standard DNA barcode for sparids, and that the 16S gene could also be an ideal candidate barcode. Moreover, each of the six sparid species( Argyrops spinifer, Rhabdosargus sarba, Dentex hypselosomus, Acanthopagrus latus, Acanthopagrus australis and Acanthopagrus berda) showed high intraspecific divergence(>1.3% genetic threshold) with the remarkable geographic lineages in the Indo-West Pacific oceans, which supported that potential unrecognized cryptic species were in them. The potential cryptic diversity revealed here might be primarily attributed to the allopatric divergences caused by the long-term geographic isolation between the Indian and West Pacific oceans or between the opposite sides of the Indian Ocean. The results further suggest that a revision of taxonomic status of these species is required, followed by development of a biodiversity conservation strategy.展开更多

Low-molecular-weight gelatin hydrolysate (LMW-GH) of 700-- 1700 Da was produced using an ultrafiltration membrane bioreaetor system. Chemiluminescence analysis revealed that LMW-GH scavenges high free radicals in a concentration-dependent manner; IC50 value for superoxide and hydroxyl radicals was 442 and 285 μgmL-1, respectively. LMW-GH exhibited excellent inhibitory characteristics against melanin synthesis and tyrosinase activity in B16 cells. Furthermore, LMW-GH notably increased in- traeellular glutathione (GSH), which in turn suppressed melanogenesis. LMW-GH performs antioxidation activity, holding the potential of being used as a valuable ingredient in function foods, cosmetics and pharmaceuticals or nutriceuticals.展开更多
A chemical investigation of the ethyl acetate extract of the fermentation broth of Alternaria tenuissima EN- 192, an endophytic fungus obtained from the stems of the marine mangrove plant Rhizophora stylosa, resulted ...A chemical investigation of the ethyl acetate extract of the fermentation broth of Alternaria tenuissima EN- 192, an endophytic fungus obtained from the stems of the marine mangrove plant Rhizophora stylosa, resulted in the isolation of nine known secondary metabolites, including four indole-diterpenoids: penijanthine A (1), paspaline (2), paspalinine (3), and penitrem A (4); three tricycloalternarene derivatives: tricycloalternarene 3a (5), tricycloalternarene lb (6), and tricycloalternarene 2b (7); and two alternariol congeners: djalonensone (8) and alternariol (9). The chemical structures of these metabolites were characterized through a combination of detailed spectroscopic analyses and their comparison with reports from the literature. The inhibitory activities of each isolated compound against four bacteria were evaluated and compounds 5 and 8 displayed moderate activity against the aquaculture pathogenic bacterium Vibrio anguillarum, with inhibition zone diameters of 8 and 9 mm, respectively, at 100 gg/disk. To the best of our knowledge, this is the first report on the secondary metabolites of mangrove-derived Alternaria tenuissima and also the first report of the isolation ofindole-diterpenoids from fungal genus Alternaria.展开更多
Dimethylsulfide(DMS) and dimethylsulfoniopropionate(DMSP) production by Scrippsiella trochoidea and Prorocentrum minimum was investigated to characterize the effects of physiological stage and salinity on DMS and DMSP...Dimethylsulfide(DMS) and dimethylsulfoniopropionate(DMSP) production by Scrippsiella trochoidea and Prorocentrum minimum was investigated to characterize the effects of physiological stage and salinity on DMS and DMSP pools of these two marine phytoplankton species.Axenic laboratory cultures of the two marine algae were tested for DMSP production and its conversion into DMS.The results demonstrated that both algal species could produce DMS,but the average concentration of DMS per cell in S.trochoidea(12.63 fmol/L) was about six times that in P.minimum(2.01 fmol/L).DMS and DMSP concentrations in algal cultures varied significantly at different growth stages,with high release during the late stationary growth phase and the senescent phase.DMS production induced by three salinities(22,28,34) showed that the DMS concentrations per cell in the two algal cultures increased with increasing salinity,which might result from intra-cellular DMSP up-regulation with the change of osmotic stress.Our study specifies the distinctive contributions of different physiological stages of marine phytoplankton on DMSP and DMS production,and clarifies the influence of salinity conditions on the release of DMS and DMSP.As S.trochoidea and P.minimum are harmful algal bloom species with high DMS production,they might play an additional significant role in the sulfur cycle when a red tide occurs.展开更多

Methods Column chromatography in association with semi-preparative HPLC were used for theisolation of compounds. 1D and 2D NMR, IR, UV, and MS were employed for structure elucidation.Results From the butanol fraction of the 95% EtOH extract of the starfish Asterias rollestoni, a newcompound N^7 -2'-deoxypseudoxanthosine (1), along with sixteen known compounds, 2'-0-methyl-inosine(2), 2'-deoxyinosine (3), 2'-0-methylguanosine (4), inosine (5); thymine (6), uracil (7), thymidine(8), deoxyuridine (9), 2'-0-methyluridine (10), ( ― )-(1S, 3S)-1-methyl-1, 2, 3,4-terrahydro-β-carboline-3-carboxyl-ic acid (11), ( ― )-(1R, 3S)-1-methyl-1, 2, 3,4-tetrahydro-β-carboline-3-carboxylic acid (12) , ( ― )-(3S)- 1, 2, 3,4-tetrahydro-β-carboline-3-carboxylic acid (13), L-tryptophan (14), L-phenylalanine (15), 3-carboxyindole (16), and p-hydroxybenzoic acid (17) , have been isolated. Conclusion Compound 1 is a newnatural product, and compounds 8, 9 and 10 are isolated from natural sources for the first time, andthe known compounds except 14 and 15 are first reported from starfish Asterias rollestoni.
文摘Two metabolites (A and B) were isolated from the mycelium of mangrove endophytic fungus Stysanus like sp. (#2492) from the South China Sea. Their structures were identified by spectral data as N-(2-hydroxytetracosyl)-2-amino-1,3,4-trihydroxyoctadecane (A) and γ -stearolactone (B). It is the first report that γ -stearolactone (B) is isolated from marine fungus as natural product.
文摘In this report, we reviewed recent literature on physiologically active substances from sea cucumbers (SCs) andtheir activities together with results obtained from our study. Preventive properties against lipid metabolism were reported inrats using a whole SC preparation with no particular constituent specified. Administration of the preparation lowered serumand hepatic cholesterol levels and improved the HDL/LDL ratio. These functions may be attributed to the stimulatory effectof the extract on the secretion of cholesterol in feces. Novel fucosylated chondroitin sulfates (FCSs) from Ludwigothureagrisea significantly induced fibroblast growth factor 2-dependent angiogenesis in human umbilical vein endothelial cells (HU-VECs). The proangiogenetic activity seemed attributable to the action of the sulfated fucose branches on the polysaccharide.SCs contain mycosporine-like amino acids (MAAs) that are capable of absorbing UV. A biogenetic precursor of MAAs wasfirst reported in SCs. The anti-proliferative effects of a branched chain fatty acid from a sea cucumber on prostate cancercells was reported with the activity of 5-lipoxygenase. Glycosphingolipid constituents in SCs have been systematically ana-lyzed over the past ten years. The results showed that the gangliosides in several SCs differed from those of mammals in thata sialic acid of SC gangliosides directly binded to glucose of cerebroside. Neuritogenic activity of the glycosphingolipids wasdemonstrated in vitro experiments and may lead to the development of therapeutic products for neurological disorders. Ourstudy also showed that sphingoid bases, the hydrolyzed products of glycosphingolipids from SCs, induced significant apoptosisin several tumor cell lines.

文摘Sea cucumber Apostichopus japonicus samples were collected in Changdao, Penglai (PL), 27 individuals, and Lingshandao, Qingdao (QD), 30 individuals, in the Shandong Peninsula, China. Ten SSR primers were used to assess the genetic variation and relationship between and within the two stocks. Respectively, for each stock, the percentage of polymorphic bands was 85.2% and 86.9%; the gene diver- sity was 0.360 5 and 0.342 8; and the Shannon’s information index was 0.515 0 and 0.499 0. At species level, the percentage of polymorphic bands was 92.2%, the total gene diversity was 0.378 9 and the Shannon’s information index was 0.550 4. The coefficient of overall genetic differentiation and the ge- netic distances between the stocks were also calculated to be 0.073 0 and 0.079 6 using the POPGENE program. Results show that the genetic diversity of the two stocks is still large but the genetic distance between the two stocks is close. A dendrogram was constructed for the 57 individuals from the two stocks, showing that the genetic structure was unitary for PL stock but complex for QD stock.

文摘The tribological behaviors of Cu-15Ni-8Sn/graphite composites with the graphite content of 38 vol.%against AISI321 stainless steel under dry-sliding,deionized water and sea water were investigated on a block-on-ring configuration.The results indicated that the friction coefficient was the lowest under dry-sliding,and the highest in deionized water.The wear rate decreased to reach the minimum value of 1.39×10-15 m3/(N·m)in sea water and in deionized water,it increased to the maximum value of 5.56×10-15 m3/(N·m).The deionized water hindered the formation of tribo-oxide layer and lubricating film,which resulted in the largest friction coefficient and wear rate.In sea water,however,the corrosion products comprised of oxides,hydroxides and chlorides were found on the worn surface,and the compacted layer composed of corrosion products and graphite played an important role in keeping the excellent wear resistance.It was elucidated that the tribological behaviors of Cu-15Ni-8Sn/graphite composite were powerful influenced by the friction environments.

文摘In order to obtain novel secondary metabolites, a deep sea inhabiting fungus Aspergillus versicolor CXCTD-06-6a was investigated. One new diketopiperazine brevianamide W(1a), as well as five known diketopiperazine alkaloids, diketopiperazine V(1b), brevianamide Q(2), brevianamide R(3), brevianamide K(4), and brevianamide E(5), were isolated from the Et OAc extract of the fermentation broth. Their structures were elucidated by spectroscopy techniques(NMR, MS). The six compounds exhibited moderate radical scavenging activity against DPPH with clearance ratio of 55.0%(1a and 1b), 53.7%(2), 46.2%(3), 61.4%(4) and 19.3%(5) at a concentration of 13.9 μmol L-1, respectively; while the positive control ascorbic acid showed a ratio of 70.3% at the concentration of 28.4 μmol L-1.

文摘The pyrrole-derived alkaloids with marine origin, especially their permethyl derivatives, have unique structures and promising biological activities. Marine natural product neolamellarins are a collection of lamellarin-like phenolic pyrrole compounds, which can inhibit hypoxia-induced HIF-1 activation. Many pyrrole-derived lamellarin-like alkaloids show potent MDR reversing activity. In this study, five permethylated derivatives of neolamellarin A were synthesized with their MDR reversing activity studied in order to identify new MDR reversal agents. A convergent strategy was adopted to synthesize the permethylated derivatives of neolamellarin A. Pyrrole was first converted into a corresponding N-trisisopropylsilyl (TIPS)-substituted derivative, then through iodination afforded 3,4-diiodinated pyrrole compound. The key intermediate, 3,4-disubstituent-lH-pyrrole, was obtained through desilylation of 3,4-disubstituent-l-TIPS pyrrole, which was prepared from 3,4-diiodinated pyrrole derivative and aryl boronic acid ester through Suzuki cross-coupling reaction between them. Then, the intermediate, 3,4-disubstituent-lH-pyrrole, reacted with fresh phenylacetyl chloride under n-BuLi/THF condition afforded the target compounds. Finally, we obtained five novel pyrrolic com- pounds, permethylated derivatives ofneolamellarin A 16a-e, in 30%-37% yield through five step reactions. The bioactivity testing of these compounds are in process.
文摘Orobanche spp. (broomrapes) are holoparasitic plants distributed predominantly in the Northern Hemisphere parasitizing the roots of a range of plant species mainly in wild ecosystems. Orobanche species cause severe yield reduction of many important crops. There are only very few herbicides which are able to selectively control broomrapes and different approaches have been put forward to develop natural product based pesticides to control Orobanche. Several phytopathogenic fungi were evaluated for their use as potential mycoherbicide and for ability to produce toxic metabolites which could be applied as herbicides. Using the alternative approach "suicidal germination", interesting results were obtained by testing two microbial metabolites (fusicoccins and ophiobolin A) especially with Orobanche species whose germination is not induced by the synthetic strigolactone GR24. From pea root exudates, peagol and peagoldione, close related to strigolactones, and three polyphenols, named peapolyphenols A-C, together with already well known polyphenol and a chalcone, were isolated. They showed a selective stimulation of Orobanche seed germination with the last two and peapolyphenol A showing a specific stimulatory activity on O. foetida. This review describes the most recent results achieved on Orobanche bio-control, mainly focusing on those regarding O. ramosa, O. crenata and O. foetida.
文摘Caspase-1-mediated IL-1β production is generally controlled by two pathways. Toll-like receptors (TLRs) recognize pathogen-derived products and induce NF-KB-dependent pro-IL-1β transcription; NOD-like receptors (NLRs) assemble caspase-l-activating inflammasome complexes that sense bacterial products/danger signals. Through a targeted chemical screen, we identify bromoxone, a marine natural product, as a specifc and potent inhibitor of the caspase-1 pathway. Bromoxone is effective over diverse inflammatory stimuli including TLR ligands plus ATP/nigeri- cin, cytosolic DNA, flagellin and Bacillus anthracis lethal toxin. Bromoxone also efficiently suppresses easpase-1 acti- vation triggered by several types of bacterial infection. Bromoxone acts upstream or at the level of the inflammasome in a transcription-independent manner. Bromoxone also inhibits pro-IL-1β expression by targeting components up- stream of IKK in the TLR-NF-kB pathway. The unique dual activities of bromoxone are shared by the known TAK1 inhibitor that specifically blocks Nalp3 inflammasome activation. Hinted from the mechanistic and pharmacological properties of bromoxone, we further discover that several known NF-KB inhibitors that act upstream of IKK, but not those targeting IKK or IKK downstream, are potent blockers of different NLRs-mediated caspase-1 activation. Our study uncovers a possible non-transcriptional molecular link between the NLR (Nalp3)-mediated inflammasome pathway and TLR-NF-kB signaling, and suggests a potential strategy to develop new anti-inflammatory drugs.

文摘Throughout the world, the coastal zones of many countries are used increasingly for aquaculture in addition to other activities such as waste disposal. These activities can cause environmental problems and health problems where they overlap. The interaction between aquaculture and waste disposal, and their relationship with eutrophication are the subjects of this paper. Sewage discharge without adequate dispersion can lead to nutrient elevation and hence eutrophication which has clearly negative effects on aquaculture with the potential for toxic blooms. Blooms may be either toxic or anoxia-causing through the decay process or simply clog the gills of filter-feeding animals in some cases. With the development of aquaculture, especially intensive aquaculture, many environmental problems appeared, and have resulted in eutrophication in some areas. Eutrophication may destroy the health of whole ecosystem which is important for sustainable aquaculture. Sewage discharge may also cause serious public health problems. Filter-feeding shellfish growing in sewage-polluted waters accumulate micro-organisms, including human pathogenic bacteria and viruses, and heavy metal ion, presenting a significant health risk. Some farmed animals may also accumulate heavy metals from sewage. Bivalves growing in areas affected by toxic algae blooms may accumulate toxins (such as PSP, DSP) which can be harmful to human beings.

文摘Diatoms and dinoflagellates are two major groups ofphytoplankton that flourish in the oceans, particularly in coastal zone and upwelling systems, and their contrasting production have been reported in several world seas. However, this information is not available in the coastal East China Sea (ECS). Thus, to investigate and compare the decadal trends in diatoms and dinoflagellates, a sediment core, 47-cm long, was collected from the coastal zone of the ECS. Sediment chlorophyll-a (Chl-a), phytoplankton-group specifc pigment signatures of diatoms and dinoflagellates, and diatom valve concentrations were determined. The sediment core covered the period from 1961 to 2011 AD. The chlorophyll-a contents ranged from 2.32 to 73 gg/g dry sediment (dw) and averaged 9.81 gg/g dw. Diatom absolute abundance ranged from 29 152 to 177 501 valve/gram (v/g) dw and averaged 72 137 v/g dw. Diatom valve and diatom specific pigment marker concentrations were not significantly correlated. Peridinin increased after the 1980s in line with intensified use of fertilizer and related increases in nutrient inputs into the marine environment. The increased occurrence of dinoflagellate dominance after the 1980s can be mostly explained by the increase in nutrients. However, the contribution of dinoflagellates to total phytoplankton production (Chl-a) decreased during the final decade of this study, probably because of the overwhelming increase in diatom production that corresponded with the construction of the Three Gorges Dam (TGD) and related light availability. Similarly, the mean ratio of fucoxanthin/peridinin for the period from 1982 to 2001 was 6% less than for 1961 to 1982, while the ratio for 2001 to 2011 was 45.3% greater than for 1982 to 2001. The decadal variation in the fucoxanthirdperidinin ratio implies that dinoflagellate production had been gradually increasing until 2001. We suggest that the observed changes can be explained by anthropogenic impacts, such as nutrient loading and dam construction.

文摘Bioassay-guided fractionation of the crude extract from Penicillium commune SD-118, a fungus obtained from a deep-sea sediment sample, resulted in the isolation of a known antibacterial compound, xanthocillin X (1), and 14 other known compounds comprising three steroids (2-4), two ceramides (5 and 6), six aromatic compounds (7-12), and three alkaloids (13-15). Xanthocillin X (1) was isolated for the first time from a marine fungus. In the bioassay, xanthocillin X (1) displayed remarkable antimicrobial activity against Staphylococcus aureus and Escherichia coli, and significant cytotoxicity against MCF-7, HepG2, H460, Hela, Du145, and MDA-MB-231 cell lines. Meleagrin (15) exhibited cytotoxicity against HepG2, Hela, Du145, and MDA-MB-231 cell lines. This is the first report of the cytotoxicity of xanthocillin X (1).

文摘Sparids are of considerable economic importance in marine fishery and aquaculture in China, and the species diversity of this group is considered relatively high. However, the accurate species identification and delimitation of sparids in China remain unaddressed. In this study, we used mitochondrial cytochrome oxidase subunit ?(COI) and 16S ribosomal RNA(16S) genes to conduct DNA barcoding and species delimitation in eleven sparid species from the coastal waters of China. Based on Kimura-2 parameter genetic distances, the mean intraspecific/interspecific variation for COI and 16S were calculated as 0.004/0.152 and 0.002/0.072, respectively. All the conspecific individuals formed monophyletic clusters in neighbour-joining trees of both markers. An obvious barcoding gap was detected for each species, and a common genetic threshold of 1.3% sequence divergence was defined for species delimitation in both markers. Although the sequence variation of 16S was generally lower than that of COI, the results indicated that sparid species could be ef fectively and accurately identified and delimited by COI as well as 16S. Thus, we propose that the COI gene serve as the standard DNA barcode for sparids, and that the 16S gene could also be an ideal candidate barcode. Moreover, each of the six sparid species( Argyrops spinifer, Rhabdosargus sarba, Dentex hypselosomus, Acanthopagrus latus, Acanthopagrus australis and Acanthopagrus berda) showed high intraspecific divergence(>1.3% genetic threshold) with the remarkable geographic lineages in the Indo-West Pacific oceans, which supported that potential unrecognized cryptic species were in them. The potential cryptic diversity revealed here might be primarily attributed to the allopatric divergences caused by the long-term geographic isolation between the Indian and West Pacific oceans or between the opposite sides of the Indian Ocean. The results further suggest that a revision of taxonomic status of these species is required, followed by development of a biodiversity conservation strategy.

文摘Gelatin extracted from the body wall of the sea cucumber (Stichopus japonicus) was hydrolyzed with flavourzyme. Low-molecular-weight gelatin hydrolysate (LMW-GH) of 700-- 1700 Da was produced using an ultrafiltration membrane bioreaetor system. Chemiluminescence analysis revealed that LMW-GH scavenges high free radicals in a concentration-dependent manner; IC50 value for superoxide and hydroxyl radicals was 442 and 285 μgmL-1, respectively. LMW-GH exhibited excellent inhibitory characteristics against melanin synthesis and tyrosinase activity in B16 cells. Furthermore, LMW-GH notably increased in- traeellular glutathione (GSH), which in turn suppressed melanogenesis. LMW-GH performs antioxidation activity, holding the potential of being used as a valuable ingredient in function foods, cosmetics and pharmaceuticals or nutriceuticals.

文摘A chemical investigation of the ethyl acetate extract of the fermentation broth of Alternaria tenuissima EN- 192, an endophytic fungus obtained from the stems of the marine mangrove plant Rhizophora stylosa, resulted in the isolation of nine known secondary metabolites, including four indole-diterpenoids: penijanthine A (1), paspaline (2), paspalinine (3), and penitrem A (4); three tricycloalternarene derivatives: tricycloalternarene 3a (5), tricycloalternarene lb (6), and tricycloalternarene 2b (7); and two alternariol congeners: djalonensone (8) and alternariol (9). The chemical structures of these metabolites were characterized through a combination of detailed spectroscopic analyses and their comparison with reports from the literature. The inhibitory activities of each isolated compound against four bacteria were evaluated and compounds 5 and 8 displayed moderate activity against the aquaculture pathogenic bacterium Vibrio anguillarum, with inhibition zone diameters of 8 and 9 mm, respectively, at 100 gg/disk. To the best of our knowledge, this is the first report on the secondary metabolites of mangrove-derived Alternaria tenuissima and also the first report of the isolation ofindole-diterpenoids from fungal genus Alternaria.

文摘Dimethylsulfide(DMS) and dimethylsulfoniopropionate(DMSP) production by Scrippsiella trochoidea and Prorocentrum minimum was investigated to characterize the effects of physiological stage and salinity on DMS and DMSP pools of these two marine phytoplankton species.Axenic laboratory cultures of the two marine algae were tested for DMSP production and its conversion into DMS.The results demonstrated that both algal species could produce DMS,but the average concentration of DMS per cell in S.trochoidea(12.63 fmol/L) was about six times that in P.minimum(2.01 fmol/L).DMS and DMSP concentrations in algal cultures varied significantly at different growth stages,with high release during the late stationary growth phase and the senescent phase.DMS production induced by three salinities(22,28,34) showed that the DMS concentrations per cell in the two algal cultures increased with increasing salinity,which might result from intra-cellular DMSP up-regulation with the change of osmotic stress.Our study specifies the distinctive contributions of different physiological stages of marine phytoplankton on DMSP and DMS production,and clarifies the influence of salinity conditions on the release of DMS and DMSP.As S.trochoidea and P.minimum are harmful algal bloom species with high DMS production,they might play an additional significant role in the sulfur cycle when a red tide occurs.
