The diversi...Relationships between phytoplankton community composition and environmental variables in the East China Sea (ECS) and Yellow Sea (YS) were investigated using geochemical and molecular microbiology methods. The diversity of phytoplankton was characterized using cultivation-independent PCR-based denaturing gradient gel electrophoresis (DGGE). Groups resulting from unweighted pair-group method with arithmetic averages clustering of the DGGE profiles showed good consistency with the eco-environmental characteristics of the sea area they belonged to. Additionally, the clustering results based on DGGE fingerprinting and those based on morphological compositions were practically identical. The relationship of phytoplankton diversity to environmental factors was statistically analyzed. Temperature, dissolved inorganic nitrogen (DIN), and silicate-Si were found significantly related to the phytoplankton community composition. Canonical correspondence analysis (CCA) was performed to reveal the relationship between community composition and these three environmental factors. Generally, values of the ECS are clearly separated from those of the YS in the CCA biplot, due to mainly the effect of temperature and DIN.展开更多

LC3 was conducted. In the SDS-PAGE study, the relative molecular weights of the proteins were mainly in the range of 14 kDa-31 kDa and 43 kDa-66 kDa, and more proteins were detected between 14kDa and 31 kDa. With the improved protein preparation, the two-dimensional electrophoresis patterns indicated that the relative molecular weights of the proteins were between 14kDa and 100kDa, and most of them ranged from 14 kDa to 31 kDa. This was consistent with the result of the SDS-PAGE analysis. The isoelectric points were found to lie between 3.0 and 8.0, and most of them were in the range of 3.0-6.0. Better separation effect was acquired with pre-prepared immobilized gradient (IPG) strip (pH 3-5.6), and about 320 protein spots could be visualized on the 2-DE map by staining. Within pH 3-l0 and pH 3-5.6 strips, the protein samples of Alexandrium sp. LC3 could be separated well.展开更多

In the present study, the metabolic and phylogenetic profile of the bacterioplankton community in Guishan coastal water(Pearl River Estuary), South China Sea, at 12 sites(S1–S12) were explored by community-level physiological profiling(CLPP) with BIOLOG Eco-plate and denaturing gradient gel electrophoresis(DGGE). Our results showed that the core mariculture area(S6, S7 and S8) and the sites associating with human activity and sewage discharge(S11 and S12) had higher microbial metabolic capability and bacterial community diversity than others(S1–5, S9–10). Especially, the diversity index of S11 and S12 calculated from both CLPP and DGGE data(H >3.2) was higher than that of others as sewage discharge may increase water nitrogen and phosphorus nutrient. The bacterial community structure of S6, S8, S11 and S12 was greatly influenced by total phosphorous, salinity and total nitrogen. Based on DGGE fingerprinting, proteobacteria, especially γ- and α-proteobacteria, were found dominant at all sites. In conclusion, the aquaculture area and wharf had high microbial metabolic capability. The structure and composition of bacterial community were closely related to the level of phosphorus, salinity and nitrogen.展开更多

Sodium dodecyl ...The aim of this study was to compare chemical and biochemical properties of crude jellyfish protein extracted from salted sand jellyfish (Rhopilema hispidum) and white jellyfish (Lobonema smithii). Sodium dodecyl sulfate polyacylamide gel electrophoresis (SDS-PAGE) showed that the principal proteins of collagen standard types I and II were found among the protein constituents of desalted jellyfish collagen prepared in this study. The amino acid compositions of extracted collagen were typical collagen. The denaturation temperature (Td) of extracted collagen samples was 28-29 ℃. The solubility of all extracted collagens was found up to 4% NaCl. The results indicated that the commercial salting process could partially denature native jellyfish protein functionalities. However, the collagen could be extracted from desalted jellyfish for future use as a food ingredient.展开更多

Methods By sodium dodecyl sulphate polyacrylamide gel electrophoresis of ghostcorpuscles, we quantified the amount of protein by densitometric evaluation.Results The results showed that in β-thalassernia, the amount of spectrin, 4.5 protein and globin significantly increased compared with the controls (26.05 + 1.46, 21.69 + 1.86; 22.87 + 5.61, 12.99 +2.33; 15.23+3.31 and 4.97+2.73, respectively, P<0.05).Conclusion These data suggest that the erythrocyte membrane protein composition matched with globin in patients with β-thalassemia of the racial Li minority are different from the normal control. These factors increase rigidity but decrease deformability of the β-thalassemic red cell membrane, which may lead to hemolytic anemia.展开更多
