翼扇涵体构型无人机风切变下的响应特性

为了分析风切变影响下的响应特性,首先根据翼扇涵体无人直升机的构型特点,建立了该型直升机在风切变场中的非线性飞行动力学模型,依据美国联邦航空局的统计数据,给出了典型的风切变模型,最后研究分析了翼扇涵体构型无人直升机在不同维数风切变场中的响应特性,以及弹性支承件对该构型直升机抗风特性的影响。研究结果表明:翼扇涵体构型无人直升机受高维数的风切变影响较大;以提高操纵性为目的弹性支承件降低了该构型无人直升机的抗风能力,因此在对弹性支承件的刚度进行设计时,需要综合考虑其对操纵性与抗风特性的影响。

Clinical Diagnosis Technique of Goat Pox Disease

[Objective] The aim was to provide theoretical basis for effective prevention of goat pox disease.[Method] 5 cases of infected goats were diagnosed for goat pox with microbiology examination.The poxes on their skin,rumen,reticulum,omasum,abomasum and submandibular lymph nodes,bronchial lymph nodes,lung and spleen were macroscopically and microscopically observed with pathanatomical and histopathological technique.[Result] Poxes on skin mainly showed ashen hemisphere state and gave prominence to the surface of skin; some cases had hemorrhage in the poxes and showed dark purplish red.Poxes on gastric mucosa showed ashen.Cytoplasmic inclusion body could be all observed in epithelial cells of the poxes and macrphages of lymph node,lung and spleen.[Conclusion] Poxes on skin,lung and the surface of gastric mucosa as well as cytoplasmic inclusion body in the epithelial cells of pox and the macrphages of lymphoid organs were the especial pathochanges of goat pox,which could be taken as the proof of goat pox＇s clinic diagnisis.

Baculovirus per os Infectivity Factors Are Involved in HearNPV ODVs Infection of HzAM1 Cells in vitro

Baculoviruses produce two viral phenotypes, the budded virus （BV） and the occlusion-derived virus （ODV）. ODVs are released from occlusion bodies in the midgut where they initiate a primary infection. Due to the lack of an in vitro system, the molecular mechanism of ODV infection is still unclear. Here we present data demonstrating that Helicoverpa armigera nucleopolyhedrovirus （HearNPV） ODV infected cultured Hz-AM1 cells in a pH dependent manner. The optimal pH for ODV infection was 8.5, which is same to that in the microvilli of midgut epithelial cells, the ODV native infection sites. Antibodies neutralization analysis indicated that four HearNPV oral infection essential genes p74, pif-l, pif-2 and pif-3 are also essential for HearNPV ODV infection in vitro. Thus, HearNPV-HzAM1 system can be used to analyze the mechanism of ODV entry.

The expression and antigenicity identification of recombinant rat TGF-β1 in bacteria

In order to study structure-function details of TGF-beta1, the recombinant mature form of rat TGF-beta1 was expressed in bacteria. Synthesis of the 112 amino-acid carboxyl-terminal part of TGF-beta1 (amino acid 279-390) was controlled by an inducible gene expression system based on bacteriophage T7 RNA polymerase. This system allowed an active and selective synthesis of recombinant TGF-beta1. The molecular weight of expressed TGF-alpha1 monomer determined on SDS-polyacrylamide gel under reducing conditions was about 13 kD. Serial detergent washes combined with a single gel-filtration purification step were sufficient to purify the expression product to homogeneity. Amino-terminal sequencing revealed that the N-terminal of the recombinant protein was identical to the published data. In Western blot analysis the recombinant polypeptide showed excellent antigenicity against polyclonal TGF-beta1 antibody. The mature recombinant rat TGF-beta1 expressed in this study provides a useful tool for future detailed structural and functional studies.

Optimization of the Purification Methods for Recovery of Recombinant Growth Hormone from Paralichthys olivaceus

This study aimed to optimize the purification of recombinant growth hormone from Paralichthys olivaceus. Recombinant flounder growth hormone （r-fGH） was expressed by Escherichia coli in form of inclusion body or as soluble protein under different inducing conditions. The inclusion body was renatured using two recovery methods, i.e., dilution and dialysis. Thereafter, the refolded protein was purified by Glutathione Sepharase 4B affinity chromatography and r-fGH was obtained by cleavage of thrombin. For soluble products, r-fGH was directly purified from the lysates by Glutathione Sepharase 4B affinity chromatography. ELISA-receptor assay demonstrated that despite its low receptor binding activity, the r-fGH purified from refolded inclusion body had a higher yield （2.605 mg L^-1） than that from soluble protein （1.964 mg L^-l）. Of the tested recovery methods, addition of renaturing buffer （pH 8.5） into denatured inclusion body yielded the best recovery rate （17.9%）. This work provided an optimized purification method for high recovery of r-fGH, thus contributing to the application of r-fGH to aquaculture.

Numerical Study on Influence of Key Parameters of Aerodynamic Characteristics of Shaftless Ducted Rotor

Shaftless ducted rotor(SDR)is a new type of ducted rotor system designed with ducted-rotor-motor integration,which is quite different from traditional ducted rotor(DR)in aerodynamic characteristics.The sliding mesh based on unstructured grid is used to simulate the aerodynamic characteristics of SDR and DR.Then,the effects of five key parameters,namely,the rotor disk height,the number of blades,the spread angle of the duct,the central hole radius and the ducted lip radius on the aerodynamic characteristics of the SDR are investigated.It is found that the same-sized SDR produces a larger total lift than the DR in hovering,but the lift proportion of its duct is reduced.In the forward flight,a large low-speed region is generated behind the SDR duct,and the reflux vortex in blade root above the advancing blade has the trend for inward diffusion.The rotor disk height has similar effects on SDR and DR.Increasing the number of blades can effectively increase the total lift of SDR,which also increases the lift proportion of duct.Increasing the spread angle of the duct will lead to the rotor lift coefficient decrease,reducing the central hole radius can increase the total lift,but the component lift coefficient decreases.Appropriately increasing the ducted lip radius can increase the total lift,which begins to decrease after reaching a certain value.

Characterization of rtSHSp13 gene encoding a development protein involved in vesicular traffic in spermiogenesis

A cDNA, designated as rtSH3p13, was isolated from a rat testis cDNA library. It consists of 1463 bp nuclear acids,which encodes a protein of 312 amino acids and was assigned the GenBank accession number AF227439. The deduced rtSH3p13 protein is a truncated isoform of SH3p13 as a result of mRNA alternative splicing. It is mainly expressed in the rat testis, detected in spermatids at the steps 8-19 of spermiogenesis, and found around the acrosome. During postnatal development, rtSH3p13 appears on day 18 and reaches maximum on day 60. Further experimental results suggested that rtSH3p13 forms a complex with activated epidermal growth factor receptor (EGFR) and interacts with synaptojanin I. Surprisingly, similar to SH3 domain, the V region of rtSH3p13 also inhibits endocytosis in CHO cells.Our results reveal a link between an rtSH3p13-synaptojanin-clathrin complex-mediated formation of pits and the process of spermiogenesis.
