OBJECTIVE To report results of radiation therapy treatment of 30 B-cell lymphoma patients with an initial cutaneous presentation according to the new classification by the WHO/EORTC. METHODS Thirty patients with cutan...OBJECTIVE To report results of radiation therapy treatment of 30 B-cell lymphoma patients with an initial cutaneous presentation according to the new classification by the WHO/EORTC. METHODS Thirty patients with cutaneous B-cell lymphoma (CBCL) were treated by cutaneous irradiation based on the number and location of the lesions and the stage of their tumor. Treatment was conducted using a Satume Clinac. RESULTS A complete response (CR) from the treatment for our series was 86%. The length of complete remission ranged from 4 to 301 months. Three patients (11%) developed a partial response (PR). One patient was progressive. Disease-free survival(DFS) at 10 years was 87%. Three patiens died [One PCMZL two PCLBCL leg type (29%)]. Radiotherapy was generally well tolerated. CONCLUSION According to the WHO/EORTC classification, the survivor results were good for PCMZL and PCFCL. The PCLBCL leg type had a poor prognosis. Localized field irradiation is an effective treatment for some localized forms of primary cutaneous B-cell lymphoma, and this mode of therapy can produce prolonged remissions.The patients with wide-spread skin involvement are usually candidates for extended field irradiation and/or chemotherapy. For advanced stages of cutaneous B-cell lymphoma, where chemotherapy is the treatment of choice, a degree of palliation can be achieved using local field irradation.展开更多

These patients were 41 females and 16 males, with an average age of 49 yea...AIM: TO investigate colonic endocrine cells in lympho- cytic colitis (LC) patients. METHODS: Fifty-seven patients with LC were in- cluded. These patients were 41 females and 16 males, with an average age of 49 years (range 19-84 years). Twenty-seven subjects that underwent colonoscopy with biopsies were used as controls. These subjects underwent colonoscopy because of gastrointestinal bleeding or health worries, where the source of bleed- ing was identified as haemorrhoids or angiodysplasia. They were 19 females and 8 males with an average age of 49 years (range 18-67 years). Biopsies from the right and left colon were obtained from both patients and controls during colonoscopy. Biopsies were fixed in 4% buffered paraformaldehyde, embedded in paraffin and cut into 5 μm-thick sections. The sections immunostained by the avidin-biotin-complex method for se- rotonin, peptide YY (PYY), pancreatic polypeptide (PP) enteroglucagon and somatostatin cells. The cell densi- ties were quantified by computerised image analysis using Olympus software. RESULTS: The colon of both the patient and the control subjects were macroscopically normal. Histo- pathological examination of colon biopsies from con- trols revealed normal histology. All patients fulfilled the diagnosis criteria required for of LC: an increase in intraepithelial lymphocytes (〉 20 lymphocytes/100 epithelial cells) and surface epithelial damage with increased lamina propria plasma cells and absent or minimal crypt architectural distribution. In the colon of both patients and control subjects, serotonin-, PYY-, PP-, enteroglucagon- and somatostatin-immunoreac- tive cells were primarily located in the upper part of the crypts of Lieberk0hn. These cells were basket- or flask-shaped. There was no statistically significant dif- ference between the right and left colon in controls with regards to the densities of serotonin- and PYY- immunoreactive cells (P = 0.9 and 0.1, respectively). Serotonin cell density in the right colon in controls was 28.9 ± 1.8 and in LC patients 41.6±2.6 (P = 0.008). In the left colon, the corresponding figures were 28.5± 1.9 and 42.4± 2.9, respectively (P = 0.009). PYY cell density in the right colon of the controls was 10.1 ± 1 and of LC patients 41 ± 4 (P = 0.00006). In the left colon, PYY cell density in controls was 6.6± 1.2 and in LC patients 53.3 ± 4.6 (P = 0.00007). CONCLUSION: The change in serotonin cells could be caused by an interaction between immune cells and serotonin cells, and that of PYY density might be sec- ondary.展开更多
AIM:To investigate the association between human papilloma virus(HPV) infection and colorectal cancer.METHODS:Sixty-nine patients with pathologically confirmed primary colorectal cancer including 6 stage Ⅰ,24 stage ...AIM:To investigate the association between human papilloma virus(HPV) infection and colorectal cancer.METHODS:Sixty-nine patients with pathologically confirmed primary colorectal cancer including 6 stage Ⅰ,24 stage Ⅱ,21 stage Ⅲ,and 18 stage Ⅳ patients were enrolled in this study to investigate whether HPV 16 could be involved in colorectal tumorigenesis.Nested-polymerase chain reaction(nested-PCR) was used to detect HPV16 DNA in colorectal tumor tissues and further confirmed by in situ hybridization(ISH).In addition,immunohistochemistry analysis was performed to examine the E6 oncoprotein in colorectal tumors.To verify whether E6 could inactivate the p53 transcriptional function,the levels of p21 and Mdm2 mRNA expression were evaluated by real-time reverse transcription(RT)-PCR.RESULTS:Of the 69 colorectal tumors,HPV16 DNA was detected in 11(16%) by nested-PCR,and HPV16 DNA was present in 8 of the 11(73%) tumors which was confirmed by ISH.The presence of HPV16 DNA in colorectal tumors was not associated with patients' clinical parameters including age,gender,smoking status,tumor site;however,HPV16 infection was more common in stage Ⅰ patients than in late-stages patients(Ⅱ,Ⅲ and Ⅳ).We next asked whether HPV16 infection could be linked with colorectal cancer development.Immunohistochemical data indicated that 8 of the 11 HPV16 DNA-positive tumors had E6 oncoprotein expression.Moreover,we also observed that the adjacent normal tissues including endothelial cells,lymphocytes,fibroblasts,and gland cells in E6-positive tumors had E6 oncoprotein expression.In addition,3 of the 4(75%) E6-positive tumors carrying p53 wild-type had negative immunostaining,but one tumor had less p53 immunostaining.We further examined whether E6-positive and/or p53 mutated tumors reduce p53 transcriptional activity.Real-time RT-PCR analysis indicated that p21 and mdm2 mRNA expression levels in E6/p53-wildtype tumors were significantly lower than in their adjacent normal tissues;as expected,E6-positive/p53-mutated tumors had lower p21 and mdm2 mRNA expression levels compared with their adjacent normal tissues.These results clearly indicate that the E6 oncoprotein expressed in p53 wildtype tumors may reduce p21 and mdm2 expression via p53 inactivation.CONCLUSION:These results suggest that HPV16 infection may be involved in a subset of colorectal cancer,and we suggest that the transmission of HPV to the colon and rectum might occur through peripheral blood lymphocytes.展开更多
Purpose: to evaluate the in vitro influence of Copaiferalangsdorffii essential oil on the antinecrotic activity in macrophages, and on the antiapoptotic activity in macrophages and lymphocytes of Swiss mice. Methods...Purpose: to evaluate the in vitro influence of Copaiferalangsdorffii essential oil on the antinecrotic activity in macrophages, and on the antiapoptotic activity in macrophages and lymphocytes of Swiss mice. Methods: lymphocytes and macrophages were obtained from ten 60-day-old male mice weighing 42-45 g, and then cultured in 24-well plates. Cells were treated with concentrations of 2%, 5%, 10% and 20% of oil from Copaiferalangsdorffii before or after induction of apoptosis with ultraviolet light. We used positive and negative controls. Hoechst staining was performed, and then images of cells under fluorescence microscope were captured and digitalized. We used Image J_1.46o to quantify the fluorescence intensity/brightness of condensation of the chromatin ofapoptotic cells. We used one-way ANOVA with Bonferroni's and Tukey's multiple comparison tests. Results are expressed as the means of relative densities after adjustment and 95% confidence intervals; P 〈 0.05 was considered statistically significant. Results: oil from Copaiferalangsdorffii at different concentrations has decreased necrosis and apoptosis of both lymphocytes and macrophages from mice, when compared to C+ (all P 〈 0.05). Discussion: Copaiferalangsdorffii is typically Brazilian and found in the Amazon region, with anti-inflammatory, antioxidant and antineoplastic effects due to its oil compounds, such as xyloglucans and kaurenoic acid. Its potential therapeutical usage was demonstrated in a national study. The present study showed that its oil has significant in vitro antiapoptotic and antinecrotic activity and it should be tested experimentally in vivo.展开更多
Accidentally or occupationally exposures to ionizing radiation generally involving the partial-body exposures and this may pose significant health hazards that are indicated by chromosome aberration (CA) induction. ...Accidentally or occupationally exposures to ionizing radiation generally involving the partial-body exposures and this may pose significant health hazards that are indicated by chromosome aberration (CA) induction. In this experiment, the quantification of the frequencies of CA was carried out based on cytogenetic analyses of peripheral blood samples obtained from 4 healthy volunteers as a result of simulation of partial-body exposures. The percentages of mixtures of blood samples irradiated in vitro with 2 Gy of gamma rays were 10, 25, 50, 75 and 100.0%. Lymphocytes were cultured and first-division metaphase cells were collected after culture times of 48 h and then harvested with standard procedures. The results showed that frequencies of unstable CA were depended on the percentage/portion of irradiated blood. All frequencies of observed CA was lower than that of calculated from 100% exposed blood, except in one case, indicating a phenomena of"dilution" of unirradiated into irradiated lymphocytes though there could be a bystander effects taken place. The increasing in frequency for 25-100% portions was also comparable with other similar experiments. The quantification of CAs in lymphocytes is an important methodology of dose assessment for partial-body exposure to ionizing radiation, however, the scenario of exposure should be determined.展开更多
