为探究光唇鱼不饱和脂肪酸合成关键酶脂肪酸去饱和酶2(fads2)对鱼类不饱和脂肪酸合成的调控,首先对fads2基因克隆,得到克隆基因序列后对fads2基因的结构进行了生物信息学分析,并且在不同温度梯度下饲养光唇鱼,使用实时荧光定量(qRT-PCR... 为探究光唇鱼不饱和脂肪酸合成关键酶脂肪酸去饱和酶2(fads2)对鱼类不饱和脂肪酸合成的调控,首先对fads2基因克隆,得到克隆基因序列后对fads2基因的结构进行了生物信息学分析,并且在不同温度梯度下饲养光唇鱼,使用实时荧光定量(qRT-PCR)测定不同温度下fads2基因的表达量差异,对比不同温度下fads2基因的表达量。生物信息学分析该基因为亲水性蛋白,结构稳定,含有与脂肪酸合成有关的Cyt-b5结构域。从fads2的定量表达结果来看,在28℃条件下fads2表达量显著上调(P<0.05),研究表明,fads2基因的表达量与温度有关。研究结果有助于深入了解光唇鱼fads2基因的结构和功能,为进一步的研究鱼类脂质代谢提供理论基础,从而选育出高不饱和脂肪酸合成较高的优良光唇鱼品种。 展开更多

An approximately 800 bp cDNA ( Lhcb 2) encoding light_harvesting chlorophyll a/b_binding protein complex (type Ⅱ) was cloned from the seedling of pea ( Pisum sativum L.) with RT_PCR method. Southern blotting using special probe demonstrated that there existed one copy of Lhcb 2 in pea genome. RT_PCR and Northern blotting revealed the expression of Lhcb 2 which was regulated by light in a time_dependent expression manner. The Lhcb 2 gene didn't express untill 2 h after irradiated with white light. Low temperature (4 ℃) also affected the Lhcb 2 gene by decreasing half of its expression under 25 ℃. 展开更多

In this study, we examined the effect of elevated temperature on the expression patterns of genes, i.e., nacrein, irr, n16, n19, and hsp70 in the pearl oyster Pinctada fucata. The experiment was carried out at 4 temperatures, i.e., 20℃(ambient, control), 24, 28℃, and 32℃. The expression levels of target genes in P. fucata were assayed at 0, 6, 24, 48, and 96 h via real-time polymerase chain reaction. Results showed that the expression levels of nacrein and irr had no significant variations among different time points below 28℃, but significantly increased over time at 32℃. The expression levels of n16 and n19 did not change markedly at 20℃. The former increased significantly at 6 h and 24 h while the latter substantially decreased during 6–96 h at 24, 28 and 32℃. Among different temperatures, the level of n16 was significantly lower at 20℃ than at other temperatures during 6–96 h, and the level of n19 significantly varied among different temperatures at 48 h and 96 h. The expression level of hsp70 was significantly higher at 32℃ than at 20, 24 and 28℃ at 24 h. These results demonstrated that elevated temperature impacted the physiological processes of P. fucata and potentially influenced its adaptability to thermal stress. 展开更多

It is also well known that, in scrotal mammals, spermatogenesis depends greatly on the maintenance of comparatively low temperature in the scrotum. To investigate whether the expression of cdc2 and Cyclin B1 in spermatogenic cells during spermatogenesis is actually a temperature dependent event, in situ hybridization, Western blotting and immunohistochemistry analysis were used to study the expression of cdc2 and Cyclin B1 in normal and cryptorchid testis. Results showed that the abdominal temperature had no significant influence on the transcription of cdc2 and Cyclin B1 in the spermatogonia and pachytene/diplotene primary spermatocytes, but it blocked the translation of them. Due to the deficiency of p34cdc2 and Cyclin B1, the spermatogonia and pachytene/diplotene primary spermatocytes were unable to form MPF, hence, they couldn’t undergo karyokinesis. The development of primary spermatocytes was arrested at the G2 to M phase transition. We also found that testosterone could regulate the Cyclin B1 expression in spermatogenic cells. Muscular injection of testosterone could recover spermatogenesis in the unilateral scrotal testis which was influenced by the contralateral cryptorchid testis, but it could not salvage the spermatogenesis block in the cryptorchid testis. 展开更多

Based on the form of the n-dimensional generic power-law potential, the state equation and the heat capacity, the analytical expressions of the Joule-Thomson coefficient (3TC) for an ideal Bose gas are derived in n-dimensional potential. The effect of the spatial dimension and the external potential on the JTC are discussed, respectively. These results show that: (i) For the free ideal Bose gas, when n/s ≤ 2 (n is the spatial dimension, s is the momentum index in the relation between the energy and the momentum), and T → Tc (Tc is the critical temperature), the JTC can obviously improve by means of changing the throttle valve's shape and decreasing the spatial dimension of gases. (ii) For the inhomogeneous external potential, the discriminant △= [1 - y∏^ni=1(kT/εi)^1/tiГ(1/ti+1)] (k is the Boltzmann Constant, T is the thermodynamic temperature, ε is the external field's energy), is obtained. The potential makes the JTC increase when △ 〉 0, on the contrary, it makes the JTC decrease when A 〈△. In this study, we investigate pairwise non-classical correlations measured using a one-way quantum deficit as well as quantum coherence in the XY spin-1/2 chain in a transverse magnetic field for both zero and finite temperatures. The analytical and numerical results of our investigations are presented. In the case when the temperature is zero, it is shown that the one-way quantum deficit can characterize quantum phase transitions as well as quantum coherence. We find that these measures have a clear critical point at λ = 1. When λ 1, the one-way quantum deficit has an analytical expression that coincides with the relative entropy of coherence. We also study an XX model and an Ising chain at the finite temperatures. 展开更多

Heat transport phenomenon of two-dimensional magnetohydrodynamie Casson fluid flow by employing Cattaneo-Christov heat diffusion theory is described in this work. The term of heat absorption/generation is incorporated in the mathematical modeling of present flow problem. The governing mathematical expressions are solved for velocity and temperature profiles using RKF 45 method along with shooting technique. The importance of arising nonlinear quantities namely velocity, temperature, skin-friction and temperature gradient are elaborated via plots. It is explored that the Casson parameter retarded the liquid velocity while it enhances the fluid temperature. Fhrther, we noted that temperature and thickness of temperature boundary layer are weaker in case of Cattaneo-Christov heat diffusion model when matched with the profiles obtained for Fourier's theory of heat flux. 展开更多
