Objective:The causal relationship between eczema and autoimmune diseases has not been previously reported.This study aims to evaluate the causal relationship between eczema and autoimmune diseases.Methods:The two‐sam...Objective:The causal relationship between eczema and autoimmune diseases has not been previously reported.This study aims to evaluate the causal relationship between eczema and autoimmune diseases.Methods:The two‐sample Mendelian randomization(MR)method was used to assess the causal effect of eczema on autoimmune diseases.Summary data from the Genome-Wide Association Study Catalog(GWAS)were obtained from the Integrative Epidemiology Unit(IEU)database.For eczema and autoimmune diseases,genetic instrument variants(GIVs)were identified according to the significant difference(P<5×10−8).Causal effect estimates were generated using the inverse‐variance weighted(IVW)method.MR Egger,maximum likelihood,MR-PRESSO,and MR-RAPS methods were used for alternative analyses.Sensitivity tests,including heterogeneity,horizontal pleiotropy,and leave-one-out analyses,were performed.Finally,reverse causality was assessed.Results:Genetic susceptibility to eczema was associated with an increased risk of Crohn’s disease(OR=1.444,95%CI 1.199 to 1.738,P<0.001)and ulcerative colitis(OR=1.002,95%CI 1.001 to 1.003,P=0.002).However,no causal relationship was found for the other 6 autoimmune diseases,including systemic lupus erythematosus(SLE)(OR=0.932,P=0.401),bullous pemphigoid(BP)(OR=1.191,P=0.642),vitiligo(OR=1.000,P=0.327),multiple sclerosis(MS)(OR=1.000,P=0.965),ankylosing spondylitis(AS)(OR=1.001,P=0.121),rheumatoid arthritis(RA)(OR=1.000,P=0.460).Additionally,no reverse causal relationship was found between autoimmune diseases and eczema.Conclusion:Eczema is associated with an increased risk of Crohn’s disease and ulcerative colitis.No causal relationship is found between eczema and SLE,MS,AS,RA,BP,or vitiligo.展开更多

Frozen sect...Sixteen patients with osteoarthritis (13 knees and 3 hips), 3 patients with rheumatoid arthritis (RA) and 4 cadaver were studied for evidence of immune complex in the destroyed articular cartilage tissues. Frozen sections of the articular cartilage from arthroplasty were stained with fluoresceinated antibodies to human immunoglobulins IgG, IgA, IgM and complement C3. The results showed: 1. There were immune complexes linear deposited in the surface of the irregular articular cartilage tissues and on some chondro- cytes remained in most patients with osteoarthritis (14/16). The patterns of immune complexes are IgA, complement C3, IgG and IgM, their percentage is 81. 25%, 75%, 75% and 50% respectively. 2. In all of 3 patients with RA, the surfaces of articular tissues were seen with patchy diffusely positive areas for IgA, IgG, IgM (excepting negative in I case) and complement C3. 3. There were no immune complexes deposited in the surfaces of 4 cases of normal articular tissues. The presence of immune complexes in the cartilages suggested that an autoimmune reaction participated in the pathological process of osteoarthritis and that the autoimmunity may be responsible for the continuous degeneration of the osteoarthritis.展开更多

Methods HPVs in CA tissues from 60 patients were detected by immunofluorescence and PCR, respectively. Different subtypes of HPVs were also identified with restriction fragment length polymorphism (RFLP) . Resuits The positive detective rates of immunofluorescence and PCR were 56. 67 % (34/60) and 96. 67 % (58/ 60), respectively ( P 〈 0. 01 ). RFLP results showed HPV6 and HPV11 were the main subtypes in the detected virus, which accounted for 98.28%. Conclusion The sensibility of PCR is superior to that of immunofluorescence.展开更多
The study was performed to determine whether the srum concentrations of IL (interleukin)-6 are elevated in patients with RA (rheumatoid arthritis) and to investigate the relationship between IL-6 levels and iron s...The study was performed to determine whether the srum concentrations of IL (interleukin)-6 are elevated in patients with RA (rheumatoid arthritis) and to investigate the relationship between IL-6 levels and iron status in RA patients. 95 serum samples were obtained, 70 of them from patients with RA who had visited the department of Rheumatology at Al-Sadder medical city in Najaf governorate (Iraq) and 25 age and sex-matched healthy controls. The authors assessed the clinical parameters of the disease, including ESR (erythrocyte sedimentation rate), CRP (C-reactive protein), and RF (rheumatoid factor). Serum levels of iron and TIBC (total iron binding capacity) were measured spectrophotometrically, while TS% (transferrin saturation percentage) and transferrin concentration were calculated mathematically. Serum concentrations of IL-6 (interleukin-6) and ferritin were measured using an ELISA (enzyme-linked immunosorbent assay). The results of serum concentration of IL-6 (interleukin-6) and ferritin were significantly elevated (P 〈 0.0001) in patients with RA compared to those of healthy controls. On the other hand, serum concentrations of iron, TIBC (total iron binding capacity), TS% (transferrin saturation percentage) and transferrin concentration were significantly decreased in patients with RA compared with those of healthy controls. These findings suggest that anemia is the most frequent observations in patients with RA and mostly associative with increasing level of interleukin-6.展开更多
This study was undertaken to investigate the regulatory effect of Resveratrol (Res) on the proliferation and apop- tosis of synoviocytes of patients with rheumatoid arthritis (RA), as the proliferation of synoviocytes...This study was undertaken to investigate the regulatory effect of Resveratrol (Res) on the proliferation and apop- tosis of synoviocytes of patients with rheumatoid arthritis (RA), as the proliferation of synoviocytes of patients was deter- mined by MTT chromatometry and the apoptosis of these cells was assayed with TUNEL flow cytometry. It was found in this experiment that the degree of cell proliferation of the Res-treated group with dosages of 50-400 μM was significantly reduced in comparison with that of the control group, but percentage of the apoptotic cells demonstrated with TUNEL labeling was el- evated under treatment with Res at the same dosages in a concentration-dependent manner. The difference between the Res- treated group and the control group was quite significant ( P < 0.01). It is concluded that Res shows a potent anti-prolifera- tive effect on synoviocytes of patients with RA with induction of cell apoptosis, and it is likely a valuable candidate for the chemotherapy and management of patients with RA.展开更多

Methods 100 RA cases were enrolled from January 2012 to February 2013 into this study. HIV screening was conducted with ECLIA detecting both HIV-1 p24 antigen, HIV-1 and HIV-2 antibodies, with ELISA and colloidal gold method detecting HIV-1 and HIV-2 antibodies. The samples producing positive results were submitted to the Center for Disease Control for confirmation using Western blotting method. The antibody titers of rheumatoid factors(RF) including RF-IgG, RF-IgM, RF-IgA, and CCP-IgG were analyzed by ELISA. Results The HIV positive-rate determined by ECLIA was significantly higher than that by ELISA and colloidal gold method(P<0.01). The false-positive rate of HIV screening was associated with antibody titers of RF-IgG, RF-IgM, RF-IgA, and CCP-IgG in RA(P<0.01). Conclusion Immunological factors, including RF and anti-CCP antibody, may influence the screening of HIV by ECLIA, producing false-positive result.展开更多
