AIM:To construct a recombinant strain which expresses adhesin AlpA of Helicobacter pylori (H pylori) and to study the immunogenicity of adhesin AlpA. METHODS: Gene Ab, which was amplified from H pylori chromosomal DNA...AIM:To construct a recombinant strain which expresses adhesin AlpA of Helicobacter pylori (H pylori) and to study the immunogenicity of adhesin AlpA. METHODS: Gene Ab, which was amplified from H pylori chromosomal DNA by PCR technique, was sequenced and the biological information was analyzed, and inserted into the Nco I and Not I restriction fragments of the expression vector pET-22b(+) using T4 DNA ligase. The resulting plasmid pET-AlpA was transformed into competent E.coli BL21(DE3) cells using ampicillin resistance for selection. Recombinant strains were incubated in 5 mL LB with 100 μg/mL ampicillin overnight at 37 ℃. Sonication of BL21(DE3)pET-22b(+)/AlpA was analyzed by Western blot to detect AlpA immunogenicity. RESULTS: The gene encoding AlpA protein was amplified by PCR with chromosomal DNA of H pylori Sydney strain (SS1) as templates. It revealed that AlpA DNA fragment amplified by PCR had approximately 1 500 nucleotides, compatible with the previous reports. The recombinant plasmid pET-22b(+)/AB was successfully constructed. DNA sequencing showed one open reading frame with the length of 588 bp. It encoded seven conservative regions that showed good antigenicity and hydrophobicity by Parker and Welling method. Furthermore, INTERNET EXPASY, NNPREDICT and ISREC predicted that it was a porin-like structure consisting of β-pleated sheets that were embedded in the outer membrane. BLAST analyzed 836 767 protein sequences and found that the similar sequences were all belonging to H pylori OMP sequences. SDS-PAGE and scan analysis showed that the molecular weight of AB was 22.5 ku and recombinant protein amounted to 29% of the total bacterial protein, among which dissolved expression amounted to 21.9% of sonicated supernatant. The rAB purity amounted to 96% through affinity chromatography. Western blot analysis of rAB confirmed that it could be specially recognized by serum form rabbit immunized with AlpA and H pylori infected. CONCLUSION: Adhesin AlpA recombinant protein may be a potential vaccine for control and treatment of H pylori infection.展开更多

Unlike the previous methods where the coding rate ...A new neural network based method for solving the problem of congestion control arising at the user network interface (UNI) of ATM networks is proposed in this paper. Unlike the previous methods where the coding rate for all traffic sources as controller output signals is tuned in a body, the proposed method adjusts the coding rate for only a part of the traffic sources while the remainder sources send the cells in the previous coding rate in case of occurrence of congestion. The controller output signals include the source coding rate and the percentage of the sources that send cells at the corresponding coding rate. The control methods not only minimize the cell loss rate but also guarantee the quality of information (such as voice sources) fed into the multiplexer buffer. Simulations with 150 ADPCM voice sources fed into the multiplexer buffer showed that the proposed methods have advantage over the previous methods in the aspect of the performance indices such as cell loss rate (CLR) and voice quality.展开更多
Advanced intelligent or "smart" meters are being deployed in Asia. A result of deployment of smart meters, with associated equipment, is the electric power industry faced with new and changing threats, vulnerabiliti...Advanced intelligent or "smart" meters are being deployed in Asia. A result of deployment of smart meters, with associated equipment, is the electric power industry faced with new and changing threats, vulnerabilities and re-evaluate traditional approaches to cyber security. Protection against emerging cyber-security threats targeting smart meter infrastructures will increase risk to both the utility and customer if not addressed within initial rollouts. This paper will discuss the issues in SMI (smart meter infrastructures) deployments that pertain to cyber security. It will cover topics such as the threats to operations, infrastructure, network and people and organization and their associated risks. SMI deployments include not only the smart meter, but also the interfaces for home energy management systems as well as communication interfaces back to the utility. Utilities must recognize and anticipate the new threat landscape that can attack and compromise the meter and the associated field network collectors. They must also include threats to the WAN (wide-area-network) backhaul networks, smart meter headends, MDMS (meter data management systems) and their interfaces to CIS (customer information systems) and billing and OMS (outage management systems). Lessons learned from SMI implementations from North America, Europe and recently, Japan, will be discussed. How white-box and black-box testing techniques are applied to determine the threat impact to the SMI. Finally, organizational change risk will be discussed and how utilities have responded to re-organizing and developing a security governance structure for the SMI and other smart grid applications.展开更多

This microgrid has photovoltaic panels as microsources, energy storage, and a flexible AC distribution interfacing system that...The results of the implementation of an actual microgrid in the Netherlands are presented. This microgrid has photovoltaic panels as microsources, energy storage, and a flexible AC distribution interfacing system that can operate connected to the public grid or autonomously where it regulates the site's voltage and frequency. In this paper, the potential of the microgrid in improving power quality issues of the site, specifically harmonic distortions, is demonstrated. Results show that flexible AC distribution interfacing system devices were able to compensate voltage harmonics when the microgrid was operating connected to the public grid and when operating autonomously. Other tests such as short-circuit, synchronization and blackstart were also conducted. The improvement in power quality and positive results of the other tests demonstrate that a self-supporting, reliable and efficient operation of the microgrid can be achieved.展开更多
This article presents our experimental studies to unravel the dynamic photovoltaic processes occurring at donor:acceptor(D:A)and electrode:active layer(E:A)interfaces under device-operating conditions by using two uni...This article presents our experimental studies to unravel the dynamic photovoltaic processes occurring at donor:acceptor(D:A)and electrode:active layer(E:A)interfaces under device-operating conditions by using two unique magneto-optical measurements,namely photo-induced capacitance and magnetic field effect measurement.First,we have found that a higher surface polarization of dielectric thin film can decrease the surface charge accumulation at E:A interface.The photo-induced capacitance results indicate that dielectric thin film plays a crucial role in the charge collection in generating photocurrent in organic solar cells.Second,our experimental results from magnetic field effect show that the binding energies of charge transfer(CT)states at D:A interface can be evaluated by using the critical bias required to completely dissociate the CT states.This is the first experimental demonstration that the binding energies of CT states can be measured under deviceoperating conditions.Furthermore,we use our measurement of magnetic field effect to investigate the most popular organic photovoltaic solar cells,organometal halide perovskite photovoltaic devices.The results of magneto-photoluminescence show that the photogenerated electrons and holes are inevitably recombined into electron–hole pairs through a spin-dependent process in the perovskites.Therefore,using spin polarizations can present a new design to control the photovoltaic loss in perovskites-based photovoltaic devices.Also,we found that introducing D:A interface can largely affect the bulk charge dissociation and recombination in perovskite solar cells.This indicates that the interfacial and bulk photovoltaic processes are internally coupled in developing photovoltaic actions in perovskite devices.Clearly,these magneto-optical measurements show a great potential to unravel the deeper photovoltaic processes occurring at D:A and E:A interfaces in both organic bulk-heterojunction and perovskite solar cells under device-operating conditions.展开更多
