Objective To analyze the effects of Dan Huang Ming Mu Recipe(DHMMR)(a pharmaceutical preparation from herbs and having the function of replenishing vital essence,removing heat,promoting blood circulation and excreting...Objective To analyze the effects of Dan Huang Ming Mu Recipe(DHMMR)(a pharmaceutical preparation from herbs and having the function of replenishing vital essence,removing heat,promoting blood circulation and excreting pathogenic water) on diabetic retinopathy(DR) after retinal laser photocoagulation through regulating the expression of vascular endothelial growth factor(VEGF) and pigment epithelium-derived factor(PEDF).Methods Forty male Brown Norway(BN) rats were randomly divided into blank group(group A,10 BN rats) and model group(30 BN rats).DR models were induced by 40 mg/kg streptozotocin(STZ) and the body weight and blood glucose of rats were monitored.After 12-week injection,fluorescein fundus angiography(FFA) and histopathological examination were detected to confirm the successful establishment of DR models.Subsequently,the right eyes of model group rats were conducted retinal laser photocoagulation with the left eyes having no retinal laser photocoagulation and rats of the model group were randomly divided into group B(the model control group),group C(the positive control group),and group D(the DHMMR group),with 10 rats in each group.Rats of the group A and the group B were given vehicle,and the group C were given calcium dobesilate suspension by gavage,and the group D were given DHMMR by gavage.After 4-week gavage,FFA was carried out to observe the fundus microvascular change.Then,the rats were sacrificed to do histopathological examinations and the serum levels of P-selectin,cAMP,cGMP,and the relative expression of the protein of VEGF and PEDF in retina were detected.Results After 12-week STZ injection,the blood glucose of the model group were pronouncedly higher than the blank group(P < 0.01).Fundus micro-hemangioma changes were observed in the rats of the model group through FFA,and the rats of the blank group did not see any changes.The pictures of HE staining showed that the retinal structure of the model group was more disordered than the blank group.After 4-week gavage,treatments with DHMMR showed dramatic reduction of serum levels of Pselectin,cAMP and cGMP compared with the group B(P < 0.01).FFA and histopathological examinations of DHMMR-treated rats revealed significantly suppression of retinal edema,fundus microvascular destruction and retinal destruction distinguishing from the group B.And the relative expression of VEGF protein of the group D and the group A was markedly lower than that of the group B(P < 0.01).The relative expression of PEDF protein of the group D and the group A was dramatically higher than that of the group B to the contrary(P < 0.01).Conclusions DHMMR demonstrates pronounced suppressive effects on the progression of DR after retinal laser photocoagulation,through down-regulating VEGF and upregulating PEDF.展开更多

The chronic atrophic gastritis (CAG)model in rats was made by pouring medicine which was a kind of mixed liquor including 2% sodium salicylate and 30% alcohol down the throat for 8 wk to stimulate rat gastric mucosa, combining with irregular fasting and compulsive sporting as pathogenic factors; 3.36, 4.80, and 6.24J/cm2doses of He-Ne laser were used, respectively for three different treatment groups, once a day for 20 d. The pH value of diluted gastric acid was determined by acidimeter,the histopathological changes such as the inflammatory degrees in gastric mucosa, the morphology and structure of parietal cells were observed, and the thickness of mucosa was measured by micrometer under optical microscope.RESULTS: In model control group, the secretion of gastric acid was little, pathologic morphological changes in gastric mucosa such as thinner mucous, atrophic glands, notable inflammatory infiltration were found. After 3.36 J/cm2 dose of He-Ne laser treatment for 20 d, the secretion of gastric acid was increased (P<0.05), the thickness of gastric mucosa was significantly thicker than that in model control group (P<0.01), the gastric mucosal inflammation cells were decreased (P<0.05). Morphology, structure and volume of the parietal cells all recuperated or were closed to normal.CONCLUSION: 3.36J/cm2 dose of He-Ne laser has a significant effect on CAG in rats.展开更多

METHODS: Sixty-four healthy male Wistar rats were ...AIM: To investigate the role of intestinal mucosal blood flow (IMBF) and motility in the damage of intestinal mucosal barrier in rats with traumatic brain injury. METHODS: Sixty-four healthy male Wistar rats were divided randomly into two groups: traumatic brain injury (TBI) group (n = 32), rats with traumatic brain injury; and control group (n = 32), rats with sham-operation. Each group was divided into four subgroups (n = 8) as 6, 12, 24 and 48 h after operation. Intestinal motility was measured by the propulsion ratio of a semi-solid colored marker (carbon-ink). IMBF was measured with the laser-Doppler technique. Endotoxin and D-xylose levels in plasma were measured to evaluate the change of intestinal mucosal barrier function following TBI. RESULTS: The level of endotoxin was significantly higher in TBI group than in the control group at each time point (0.382 ± 0.014 EU/mL vs 0.102 ± 0.007 EU/mL, 0.466 ± 0.018 EU/mL vs 0.114 ± 0.021 EU/mL, 0.478± 0.029 EU/mL vs 0.112 ±- 0.018 EU/mL and 0.412± 0.036 EU/mL vs 0.108 ±0.011 EU/mL, P 〈 0.05). D-xylose concentrations in plasma in TBI group were significantly higher than in the control group (6.68 ± 2.37 mmol/L vs 3.66 ±1.07 retool/L, 8.51 ± 2.69 mmol/L vs 3.15 + 0.95 mmol/L, 11.68 ±3.24 mmol/L vs 3.78 ± 1.12 mmol/L and 10.23 ± 2.83 mmol/L vs 3.34 ± 1.23 mmol/L, P 〈 0.05). The IMBF in TBI group was significantly lower than that in the control group (38.5 ± 2.8 PU vs 45.6 ± 4.6 PU, 25.2 ± 3.1 PU vs 48.2 ± 5.3 PU, 21.5 ± 2.7 PU vs 44.9 ± 2.8 PU, 29. 4 ± 3.8 PU vs 46.7 ± 3.2 PU) (P 〈 0.05). Significant decelerations of intestinal propulsion ratio in T8I groups were found compared with the control group (0.48% ± 0.06% vs 0.62%± 0.03%, 0.37% ±0.05% vs 0.64% ± 0.01%, 0.39% ± 0.07% vs 0.63% =1= 0.05% and 0.46% ± 0.03% vs 0.65% ± 0.02%) (P 〈 0.05). CONCLUSION: The intestinal mucosal permeability is increased obviously in TBI rats. Decrease of intestinal motility and IMBF occur early in TBI, both are important pathogenic factors for stress-related damage of the intestinal mucosal barrier in TBI.展开更多

The mice were exposed to γ- rays of ^60Co (whole body irradiation) in the dose of 7 Gy (the transitional clinical form of the acute radiation sickness). The dose rate at acute irradiation was 1.14 Gy/min, and at prolonged exposure, 0.027 Gy/min. Laser radiation in the dose l mJ/cm^2 was used to irradiate only the back of a mouse. First, the mice were exposed to γ-radiation, then to laser radiation. The time interval between two types of irradiation did not exceed 30 min. It was shown that the radiation protection of mice with laser radiation is possible at exposure to ionizing radiation in a wide dose interval and can reduce negative after-effects of both the acute and prolonged radiation exposure.展开更多

First, the mice were exposed to γ- radiation (whole body irradiation), then after 2 h or 24 h they were irradiated with laser radiation. The results of these studies have shown that the use of laser irradiation to reduce radiation damage in mice is effective 24 h after the exposure to 5 Gy ionizing radiation which leads to the bone-marrow clinical form of the ARS (Acute radiation sickness). In case of the lethal dose of ionizing radiation 7 Gy (the transitional clinical form of the ARS), the increase in life expectancy of mice is observed using laser radiation both 2 and 24 h after the exposure to γ- radiation, but the effectiveness of the laser used 2 h after the ionizing radiation is significantly more efficient.展开更多

Methods: 9 rats were randomly selected from 60 rats as normal con- trol group, and the other rats were used to replicate the animal model of chronic atrophic gastritis with comprehensive method, 5 rats were randomly inspected at the 8th week in the process of creating model for gastric mucosa pathological examination; when gastric mucosa shows CAG symptoms such as various degree of congestion, bleeding, atrophy, metaplasia, the model rats were randomly divided into model group, medicine group, acupuncture group and laser acupuncture group. The rats of medicine group were chron- ically administered with yanshenjianwei capsule and Kangfuxin Liquid; the groups of acupuncture and laser acupuncture selected Zusanli acupuncture as acupuncture points, after the treatment for 14 d, blood was removed, double antibody sandwich en- zyme linked immunosorbent assay (ELISA) was used to detect the levels of IL-2, IL-6 and TGF-β1 in serum. Results: Compared with normal group, the contents of IL-2, IL-6 and TGF-β1 in serum of model rats increased (P〈0.05 or P〈0.01). Medicine, acupuncture and laser acupuncture could reduce the contents of IL-2, IL-6 and TGF-β1, IL-2 in serum of the medicine group and laser acupuncture group was signifi- cantly decreased (P〈0.05); IL-6 in acupuncture group and laser acupuncture group were significantly decreased (P〈0.05 or P〈0.01);and the most significant is laser acupuncture group (P〈0.01). Medicine, acupuncture and laser acupuncture could significantly reduce TGF-β 1 content in serum (P〈0.05 or P〈0.01), and the particularly significant was laser acupuncture(P〈0.01). Conclusion: Laser acupuncture has the function of reducing the inflammatory response and adjusting immune function. It earl effectively reduce the ex- pression level of IL-2, IL-6 and TGF- β1 in serum of rats with chronic atrophic gastri- tis, and its effects are better than traditional acupuncture and medicine.展开更多
Objective: To observe the effect of acupuncture on blood oxygen concentration in the brain of rats with post-traumatic stress disorder (PTSD) based on functional near-infrared spectroscopy (fNIRS), thus to reveal the ...Objective: To observe the effect of acupuncture on blood oxygen concentration in the brain of rats with post-traumatic stress disorder (PTSD) based on functional near-infrared spectroscopy (fNIRS), thus to reveal the mechanisms of acupuncture in intervening the brain function of PTSD rats. Methods: Sixty Sprague-Dawley (SD) rats were randomly divided into a blank group, a model group, a grasping group, a paroxetine group and an acupuncture group, with 12 rats in each group. Except the blank group, rats in the other groups all received incarceration plus electric shock for 7 d to prepare the PTSD animal model. One hour before the stress model was established, rats in each group received the desig nated in tervention: rats in the blank group and the model group did not receive any intervention;rats in the grasping group received grasping and fixation;rats in the paroxetine group received paroxetine hydrochloride solution by intragastric administration;and rats in the acupuncture group received acupuncture. Six-day treatme nt was a course, with 2 courses of treatme nt con ducted for a total of 12 d. After the modeli ng, rats in each treatment group received intervention for 5 dz and the fNIRS system was used to collect and record the changes in the concentrations of oxygenated hemoglobin (HbO2), deoxygenated hemoglobin (d-Hb) and total hemoglobin (t-Hb) of the involved rat's brain regions, and also to assess the brain function. Results: Compared with the blank group, the con centrati on of HbO2 was significa ntly in creased, the concen trati on of d-Hb was significa ntly decreased, and the concen tration of t-Hb was sign ifica ntly in creased in the model group and the graspi ng group after the in tervention, and the differ en ces were statistically significant (all P<0.01). Compared with the model group, the concentrations of HbO2, d-Hb and t-Hb in the grasping group did not change significantly (all P>0.05). Compared with the grasping group, the concentration of HbC^was significantly decreased, the concentration of d-Hb was significantly in creased, and the concen trati on of t-Hb was sign ifica ntly decreased in the paroxeti ne group and the acup un cture group, and the differences were statistically sigrdficant (all P<0.05). There were no significant differences in the concentrations of HbO2, d-Hb and t-Hb between the paroxetine group and the acupuncture group (all P>0.05). Con elusion: Acup un cture can regulate the blood oxyge n conce ntrati on in the brain of PTSD model rats, which may be an important mechanism of acupuncture in intervening the brain function in PTSD rats.展开更多

Medicine, acupuncture and laser acupuncture could reduce the contents of IL-2, IL-6 and TGF-β1, IL-2 in serum of the medicine group and laser acupuncture group was signifi- cantly decreased (P〈0.05); IL-6 in acupuncture group and laser acupuncture group were significantly decreased (P〈0.05 or P〈0.01);and the most significant is laser acupuncture group (P〈0.01). Medicine, acupuncture and laser acupuncture could significantly reduce TGF-β 1 content in serum (P〈0.05 or P〈0.01), and the particularly significant was laser acupuncture(P〈0.01). Conclusion: Laser acupuncture has the function of reducing the inflammatory response and adjusting immune function. It earl effectively reduce the ex- pression level of IL-2, IL-6 and TGF- β1 in serum of rats with chronic atrophic gastri- tis, and its effects are better than traditional acupuncture and medicine.
文摘Objective: To observe the effect of acupuncture on blood oxygen concentration in the brain of rats with post-traumatic stress disorder (PTSD) based on functional near-infrared spectroscopy (fNIRS), thus to reveal the mechanisms of acupuncture in intervening the brain function of PTSD rats. Methods: Sixty Sprague-Dawley (SD) rats were randomly divided into a blank group, a model group, a grasping group, a paroxetine group and an acupuncture group, with 12 rats in each group. Except the blank group, rats in the other groups all received incarceration plus electric shock for 7 d to prepare the PTSD animal model. One hour before the stress model was established, rats in each group received the desig nated in tervention: rats in the blank group and the model group did not receive any intervention;rats in the grasping group received grasping and fixation;rats in the paroxetine group received paroxetine hydrochloride solution by intragastric administration;and rats in the acupuncture group received acupuncture. Six-day treatme nt was a course, with 2 courses of treatme nt con ducted for a total of 12 d. After the modeli ng, rats in each treatment group received intervention for 5 dz and the fNIRS system was used to collect and record the changes in the concentrations of oxygenated hemoglobin (HbO2), deoxygenated hemoglobin (d-Hb) and total hemoglobin (t-Hb) of the involved rat's brain regions, and also to assess the brain function. Results: Compared with the blank group, the con centrati on of HbO2 was significa ntly in creased, the concen trati on of d-Hb was significa ntly decreased, and the concen tration of t-Hb was sign ifica ntly in creased in the model group and the graspi ng group after the in tervention, and the differ en ces were statistically significant (all P<0.01). Compared with the model group, the concentrations of HbO2, d-Hb and t-Hb in the grasping group did not change significantly (all P>0.05). Compared with the grasping group, the concentration of HbC^was significantly decreased, the concentration of d-Hb was significantly in creased, and the concen trati on of t-Hb was sign ifica ntly decreased in the paroxeti ne group and the acup un cture group, and the differences were statistically sigrdficant (all P<0.05). There were no significant differences in the concentrations of HbO2, d-Hb and t-Hb between the paroxetine group and the acupuncture group (all P>0.05). Con elusion: Acup un cture can regulate the blood oxyge n conce ntrati on in the brain of PTSD model rats, which may be an important mechanism of acupuncture in intervening the brain function in PTSD rats.

文摘Nitric oxide(NO)and nitric oxide synthase(NOS)have an important role in pain signaling transmission in animal models.Low-level laser therapy(LLLT)is known to have an analgesic effect,but the mechanism is unclear.The aim of the study is to investigate the influence of LLLT on NO release and NOS synthesis in dorsal root ganglion(DRG)neurons,in order to find whether LLLI can ameliorate pain through modulating NO production at the cellular level.The results show that in stress conditions,the laser irradiation at 658 nm can modulate NO production in DRG neurons with soma diameter of about 20μm in a short time after illumination,and affect NOS synthesis in a dose-dependent manner.It is demonstrated that LLLT might treat pain by altering NO release directly and indirectly in DRG neurons.
