目的通过观察左金丸对溃疡性结肠炎(UC)小鼠的影响,从miR-21→STAT3/PI3K信号通路研究左金丸治疗溃疡性结肠炎的作用机制。方法将60只BALB/c小鼠随机分为空白组、模型组、美沙拉嗪组(MS,0.3 g/kg)、左金丸组(6 g/kg),用葡聚糖硫酸钠(DSS... 目的通过观察左金丸对溃疡性结肠炎(UC)小鼠的影响,从miR-21→STAT3/PI3K信号通路研究左金丸治疗溃疡性结肠炎的作用机制。方法将60只BALB/c小鼠随机分为空白组、模型组、美沙拉嗪组(MS,0.3 g/kg)、左金丸组(6 g/kg),用葡聚糖硫酸钠(DSS)构建急性UC小鼠模型,造模第二天开始灌胃给药,持续7 d,观察小鼠一般情况。采用HE染色法观察小鼠结肠组织形态,Western blot法检测小鼠结肠部位的STAT3、PI3K、AKT蛋白表达;运用qrt-PCR法检测小鼠结肠部位的miR-21基因表达。结果左金丸溶液和MS溶液对UC模型小鼠有明显治疗作用,且左金丸组小鼠效果更为显著,通过对STAT3、PI3K、AKt、Mir-21的抑制,减轻对结肠组织的损伤。结论左金丸对UC小鼠免疫调节有效,且miR-21应为左金丸干预UC小鼠的关键环节。 展开更多

The aniline degradation by persulfate activated with ferrous ion (Fe2+ ) was investigated in batch reactor at ambient temperature. The aniline degradation by persulfate activated with ferrous ion (Fe2+ ) was investigated in batch reactor at ambient temperature. The experimental factors in aqueous solutions including persulfate concentration, Fe2+ concentration, pH and ionic strength level were discussed. It is demonstrated that, aniline degradation rate increases with increasing persulfate concentration, but much more ferrous ion inhibits the aniline degradation. When the aniline concentration is 0.10 mmol/L, the maximum aniline degradation occurs at the S2O82- to Fe2+ molar ratio of 250/5 at pH 7.0. In the pH range of 5.0-8.5, increasing pH causes higher aniline degradation. What's more, the increase of ionic strength in solution causes inhibiting in the reaction. Produced intermediates during the oxidation process were identified using gas chromatography-mass spectrometry (GC-MS) technology. And degradation pathways of aniline were also tentatively proposed. 展开更多

For this problem, we are given two potential uniform machines to process a sequence of independent jobs. Machines need to be activated before starting to process, and each machine activated incurs a fixed machine activation cost. No machines are initially activated, and when a job is revealed, the algorithm has the option to activate new machines. The objective is to minimize the sum of the makespan and the machine activation cost. We design optimal online algorithms with competitive ratio of (2s+1)/(s+1) for every s≥1. 展开更多

Electrophoretic display(EPD) technology has become one of the main supporting pillars of the electronic paper display industry.Despite its benefits,the EPD technology suffers from several disadvantages such as non-fixed threshold voltage value for gray scale display.In addition,the display has to repeatedly refresh between white and black states to eliminate ghost image when it needs to update a new image.The traditional driving waveform for the EPD includes four stages: erasing the original image,resetting to black state,clearing to white state,and writing a new image.A flicker can be found when transferring between two adjacent stages.A new driving waveform based on the improvement of activation pattern is proposed to weaken the ghost image and reduce the flicker.Experimental results show that the proposed driving waveform could weaken the ghost image effectively and reduce the number of flickers by 50%.Compared with the traditional driving waveform,the driving waveform of this work has a better performance. 展开更多

Properties and organic activation of ocean clay in the East Pacific

In order to make use of ocean clay reasonably and excavate its potential applications the properties of ocean clay in Chinese investigating area of the East Pacific have been tested with the chemical analysis, XRFS, ICP/MAS, granulometry and SSA, etc. The results show that the clay sediments of this area are rich in clay minerals, fine granularity, bigger SSA, bigger water absorbability and lower ion changing ability etc. To improve the adaptability of ocean clay and macromolecule materials, using the dry milling method with silane, aluminate, stearic acid coupling agents etc the properties of ocean clay are improved. The effect has been evaluated by testing the water absorbing of the modified ocean clay and their dispersing in paraffin liquid. The result is that KH550 is better than any other active reagents, and the best activation technics is that 1.5% (volume) of KH550 quantity, 20min of milling time. The DTA, XRD, SEM and IR analyses and SSA testing show that the organic activation can not change the mineral structure but can densify the structure of clay, and activation mechanism is chemical covered mainly and chemical combined secondly. 展开更多

This paper presents the construction of a pneumatic active suspension system for a one-wheel car model using fuzzy reasoning and a disturbance observer. The one-wheel car model can be approximately described as a nonlinear two degrees of freedom system subject to excitation from a road profile. The active control is composed of fuzzy and disturbance controls, and the active control force is constructed by actuating a pneumatic actuator. A phase lead-lag compensator is inserted to counter the performance degradation due to the delay of the pneumatic actuator. The experimental result indicates that the proposed active suspension improves much the vibration suppression of the car model. 展开更多

ERK1/2 contributes negative regulation to STAT3 activity in HSS-transfected HepG2 cells 被引量:3

Signal transducer and activator of transcription 3 (STAT3) is a recently characterized transcription factor which is essential to liver regeneration. We have previously reported that hepatic stimulator substance (HSS), a novel growthpromoting substance, phosphorylated the epidermal growth factor (EGF) receptors and activated downstream RasMAP kinase (extracellular signal-regulated kinases, ERK1/2) cascade. However, whether HSS signal is related to STAT3pathway remains unclear. The present study is aiming to explore the regulatory effect of activation of ERK1/2 evoked by HSS on STAT3 phosphorylation and STAT3 signaling. Human hepatoma cell line HepG2 was stably transfected with HSS cDNA and HSS expression was measured by Northern blot. The results showed that the transfection of HSS into HepG2 resulted in remarkable increase in cellular proliferation as compared with the non-transfected cells, and it was further proved that the cellular proliferation in the HSS-transfected cells was related to ERK1/2 activation. Treatment of the cells with 50 μM of PD98059, an ERK1/2 specific upstream inhibitor, resulted in ERK1/2 inactivation completely.Inhibition of ERK1/2 allowed the tyrosine of STAT3 to be phosphorylated in a dose-dependent manner to PD98059.Furthermore, transient transfection of STAT3 mutant (STAT3S727A) into HSS-bearing cells could remarkably reverse the inhibitory effect of ERK1/2 on STAT3 phosphorylation. Based upon these results, it is concluded that ERK1/2negatively modulates STAT3 phosphorylation and this function is dependent on residual serine-727 (S727) of STAT3. 展开更多
