Chemical communication plays an important role in survival and reproductive success in mammalian species. In the present study, we examined the ontogenetic pattern of behavioral responses of male giant pandas (Ailuropoda melanoleuca ) to urine odors of conspecific individuals. Our data showed that exposure to the urine of adult females induced a significant increase in sniffing and environmental sniffing/licking behaviors, but a decrease in biting behavior, in males. Males of different ages displayed specific behaviors to female urine odors. Adult males spent more time licking than juvenile and sub-adult males. Further, sub-adult and adult males displayed high levels of environmental sniffing/licking, which was absent in the juvenile males. Juvenile males displayed scent rubbing behavior significantly more frequently than sub-adult and adult males, and also spent more time showing biting behavior than sub-adult males. Finally, juvenile and sub-aduh males showed no difference in response to female and male urine odors. Together, these data suggest that chemosensory cues from conspecific urines induce age-specific responses in male giant pandas. 展开更多

Several studies have characterized the cellular and molecular mechanisms of hepatocyte injury caused by the retention of hydrophobic bile acids (BAs) in cholestatic diseases. BAs may disrupt cell membranes through their detergent action on lipid components and can promote the generation of reactive oxygen species that, in turn, oxidatively modify lipids, proteins, and nucleic acids, and eventually cause hepatocyte necrosis and apoptosis. Several pathways are involved in triggering hepatocyte apoptosis. Toxic BAs can activate hepatocyte death receptors directly and induce oxidative damage, thereby causing mitochondrial dysfunction, and induce endoplasmic reticulum stress. When these compounds are taken up and accumulate inside biliary cells, they can also cause apoptosis. Regarding extrahepatic tissues, the accumulation of BAs in the systemic circulation may contribute to endothelial injury in the kidney and lungs. In gastrointestinal cells, BAs may behave as cancer promoters through an indirect mechanism involving oxidative stress and DNA damage, as well as acting as selection agents for apoptosis-resistant cells. The accumulation of BAs may have also deleterious effects on placental and fetal cells. However, other BAs, such as ursodeoxycholic acid, have been shown to modulate BA-induced injury in hepatocytes. The major beneficial effects of treatment with ursodeoxycholic acid are protection against cytotoxicity due to more toxic BAs; the stimulation of hepatobiliary secretion; antioxidant activity, due in part to an enhancement in glutathione levels; and the inhibition of liver cell apoptosis. AIM: To investigate the ability of ursodeoxycholic acid (UDCA) to scavenge superoxide anion (O2-).METHODS: We assessed the ability of UDCA to scavenge (O2-) generated by xanthine-xanthine oxidase (X-XO) in a cell-free system and its effect on the rate of O2--induced ascorbic acid (AA) oxidation in hepatic post-mitochondrial supernatants.RESULTS: UDCA at a concentration as high as 1 mmol/Ldid not impair the ability of the X-XO system to generate O2-, but could scavenge O2- at concentrations of 0.5 and 1 mmol/L, and decrease the rate of AA oxidation at a concentration of 100 μmol/L.CONCLUSION: UDCA can scavenge O2-, an action that may be beneficial to patients with primary biliary cirrhosis. 展开更多

From 1989-2009, 1,430 bear management report forms collected at Brooks Camp, KNP (Katmai National Park), were placed in 1 of 9 categories representing bear-human interactions of concern. Using non-parametrical statistics the categories identified dominance interactions, fish-related dominance interactions, food obtained, food-related incidents, fish stolen, property damage, and use of deterrents either decreasing or exhibiting no significant trends over time. Aggressive behavior by bears towards park staff in bear management situations increased. An elevated walkway and platform at Brooks Falls and an electric fence at the campground marked significant decrease in bear-human interactions in these areas. Changes in fishing regulations in 1998 marked significant decreases in number of fish stolen. Changes in minimum distance regulations in 2003 marked no change in number of dominance interactions or bluff charges. Efforts to facilitate visitor traffic in areas around the floating bridge have marked significant increases in number of bear-human interactions. Management and policy efforts have been successful in minimizing bear-human interactions. Habituation of bears to human activity has likely contributed to both minimizing general interactions and increased aggressive bear behavior towards management staff. This study provides examples of successful adaptive management tactics for bears and humans in a visitor oriented setting. 展开更多

It not only gives a chance for the audiences in western countries to enjoy the traditional Chinese culture, but also for the Chinese to enjoy this visual feast. This article will illustrate the implicit and explicit Chinese elements in Kung fu Panda, in hope of finding out the extensive use of Chinese elements in today's Hollywood movies. According to the collection of media data concluded, the film production team undertook detailed investigation and studies in the traditional Chinese culture so that today's Hollywood movies can successfully interpret Chinese elements. Thus, the international cultural communications are found hiding behind the widely used of Chinese elements. With the analysis of this article, Chinese reader can know further about how important the diffusion of Chinese traditional culture is. Extant giant pandas are divided into Sichuan and Qinling subspecies.The giant panda has many speciesspecific characteristics,including comparatively small organs for body size,small genitalia of male individuals,and low reproduction.Here,we report the most contiguous,high-quality chromosomelevel genomes of two extant giant panda subspecies to date,with the first genome assembly of the Qinling subspecies.Compared with the previously assembled giant panda genomes based on short reads,our two assembled genomes increased contiguity over 200-fold at the contig level.Additional sequencing of 25 individuals dated the divergence of the Sichuan and Qinling subspecies into two distinct clusters from 10,000 to 12,000 years ago.Comparative genomic analyses identified the loss of regulatory elements in the dachshund family transcription factor 2(DACH2)gene and specific changes in the synaptotagmin 6(SYT6)gene,which may be responsible for the reduced fertility of the giant panda.Positive selection analysis between the two subspecies indicated that the reproduction-associated IQ motif containing D(IQCD)gene may at least partly explain the different reproduction rates of the two subspecies.Furthermore,several genes in the Hippo pathway exhibited signs of rapid evolution with giant panda-specific variants and divergent regulatory elements,which may contribute to the reduced inner organ sizes of the giant panda. 展开更多

Predicting the Impact of Climate Change on Vulnerable Species in Gandaki River Basin,Central Himalayas

Gandaki River Basin(GRB)is an important part of the central Himalayan region,which provides habitat for numerous wild species.However,climatic changes are making the habitat in this basin more vulnerable.This paper aims to assess the potential impacts of climate change on the spatial distributions of habitat changes for two vulnerable species,Himalayan black bear(Ursus thibetanus laniger)and common leopard(Panthera pardus fusca),using the maximum entropy(Max Ent)species distribution model.Species occurrence locations were used along with several bioclimatic and topographic variables(elevation,slope and aspect)to fit the model and predict the potential distributions(current and future)of the species.The results show that the highly suitable area of Himalayan black bear within the GRB currently encompasses around 1642 km^(2)(5.01%area of the basin),which is predicted to increase by 51 km^(2) in the future(2050).Similarly,the habitat of common leopard is estimated as 3999 km^(2)(12.19%of the GRB area),which is likely to increase to 4806 km^(2) in 2050.Spatially,the habitat of Himalayan black bear is predicted to increase in the eastern part(Baseri,Tatopani and north from Bhainse)and to decrease in the eastern(Somdang,Chhekampar),western(Burtibang and Bobang)and northern(Sangboche,Manang,Chhekampar)parts of the study area.Similarly,the habitat of common leopard is projected to decrease particularly in the eastern,western and southern parts of the basin,although it is estimated to be extended in the southeastern(Bhainse),western(Harichaur and northern Sandhikhark)and north-western(Sangboche)parts of the basin.To determine the habitat impact,the environmental variables such as elevation,Bio 15(precipitation seasonality)and Bio 16(precipitation of wettest quarter)highly contribute to habitat change of Himalayan black bear;while Bio 13(precipitation of wettest month)and Bio 15 are the main contributors for common leopard.Overall,this study predicted that the suitable habitat areas of both species are likely to be impacted by climate change at different altitudes in the future,and these are the areas that need more attention in order to protect these species. 展开更多

Understanding present patterns of genetic diversity is critical in order to design effective conservation and manage- ment strategies for endangered species. Tangjiahe Nature Reserve (NR) is one of the most important national reserves for giant pandas Ailuropoda melanoleuca in China. Previous studies have shown that giant pandas in Tangjiahe NR may be threatened by population decline and fragmentation. Here we used 10 microsatellite DNA markers to assess the genetic variability in the Tang- jiahe population. The results indicate a low level of genetic differentiation between the Hongshihe and Motianling subpopulations in the reserve. Assignment tests using the Bayesian clustering method in STRUCTURE identified one genetic cluster from 42 in- dividuals of the two subpopulations. All individuals from the same subpopulation were assigned to one cluster. This indicates high gene flow between subpopulations. F statistic analyses revealed a low Fzs-value of 0.024 in the total population and implies a randomly mating population in Tangjiahe NR. Additionally, our data show a high level of genetic diversity for the Tangjiahe population. Mean allele number (A), Allelic richness (AR) and mean expected heterozygosity (HE) for the Tangjiahe population was 5.9, 5.173 and 0.703, respectively. However there are still losses during the process of moving along corridors. In this study, a mathematical model with Allee effect is carried out to describe the diffusion of giant pandas between n patches. Some criteria are obtained to keep the system persisting. It is proved that the system has a unique positive w-periodic solution which is globally asymptotically stable. The ecological meanings of these findings are discussed following the results. And some numerical simulations in the Qinling Mountain giant panda nature reservation area are also presented in the end. 展开更多
