Increased iron content of the substantia nigra (SN) has been found in PD patients, and divalent metal transporter 1 (DMT1) has been shown to be up-regulated in the SN of both MPTP-induced PD models and PD patients. However, the mechanisms underlying DMT1 up-regulation are largely unknown. In the present study, we observed that in the SN of 6-hydroxydopamine (6-OHDA)-induced PD rats, DMT1 with the iron responsive element (IRE, DMTI+IRE), but not DMT1 without IRE (DMTI-IRE), was up- regulated, suggesting that increased DMTI+IRE expression might account for nigral iron accumulation in PD rats. This possibility was further assessed in an in vitro study using 6-OHDA-treated and DMTl+IRE-over-expressing MES23.5 cells. In 6-OHDA-treated MES23.5 cells, increased iron regulatory protein (IRP) 1 and IRP2 expression was observed, while silencing of IRPs dramatically diminished 6-OHDA-indueed DMTI+IRE up-regulation. Pre- treatment with N-acetyl-L-cysteine fully suppressed IRPs up-regulation by inhibition of 6-OHDA-indueed oxidative stress. Increased DMTI+IRE expression resulted in increased iron influx by MES23.5 cells. Our data provide direct evidence that DMTI+IRE up-regulation can account for IRE/IRP-dependent 6-OHDA-induced iron accumulation initiated by 6-OHDA-induced intracellular oxidative stress and that increased levels of intracellular iron result in ag- gravated oxidative stress. The results of this study provide novel evidence supporting the use of anti-oxidants in the treatment of PD, with the goal of inhibiting iron accumulation by regulation of DMT1 expression.展开更多

In this manuscript authors evaluated the importance of turning the wastes and of inoculating microorganisms and worms in order to unprove the composting process at domestic scale. Four treatments (Control without turning- C-, Control with Turning- CT- inoculated Mountain Microorganism with turning- MM- and Worms without turning- W-) were tested in a random experiment with tour replicates. Sixteen composting bins were fed with 300 kg of organic wastes from a local street market. The process of compostnig lasted 13 weeks after which all the composts were sifted and submitted to a range of chemical, physical and biological analysis. According to the results MM slightly increased the initial temperature and enzymatic activiry. This investigation outcome don't provide of sufficient grounds for a precise recommendation about worms inoculation in compost bins. A decrease between 75% and 80% in the fresh weight of the organic waste was found in the first thirteen weeks after starting the composting process in all the treatments. The quality level of the compost was acceptable, with very low heavy metal content. Turn over helps to keep the product hygienic especially after the inoculation with MM microbial starts. In conclusion and according to the results of this investigation, the use of 320 L compost bins for the organic waste management is strongly advisable.展开更多

The Ancient Greek Philosophers never showed the two gods together. Nevertheless, in the main myths related to the netherworld such as those of Orpheus or Persephone, the two gods are in a continuous fight which has determined the human condition until nowadays. The author wishes in this paper to examine how the Orpheus' myth related to the duality Eros and Thanatos is treated in Theo Angelopoulos' world and especially how it appears in Ulysses' Gaze (1995) and Trilogy: The Weeping Meadow (2004) and Dust of Time (2008). In this last Angelopoulos' Trilogy, we will search the unfolds of the love story of two young people started in Odessa in 1918 and finished by the death of the woman at the end of the century, under the lens of the duality Eros and Thanatos as literal and metaphorical qualities. The historical and personal adventures of the two characters represent a daedal itinerary from East to West, which offers multiple levels of reading of the filmic texts. A comparative approach with myth, literature, and historical research will demonstrate the richness of artistic expression and a profound relationship of Angelopoulos' thought with the formulaic myths he introduces in his filmic work.展开更多
