目的了解宁夏地区黄牛博尔纳病病毒(Borna disease virus,BDV)的自然感染情况,对检测到的BDVp24基因阳性扩增片段进行核苷酸序列比对,探讨其与国际标准病毒株及其分离株的同源性。方法采用荧光定量巢式逆转录酶聚合酶链反应(FQ-nRT-PCR... 目的了解宁夏地区黄牛博尔纳病病毒(Borna disease virus,BDV)的自然感染情况,对检测到的BDVp24基因阳性扩增片段进行核苷酸序列比对,探讨其与国际标准病毒株及其分离株的同源性。方法采用荧光定量巢式逆转录酶聚合酶链反应(FQ-nRT-PCR)检测宁夏地区205例健康牛外周血单个核细胞(PB-MCs)BDVp24基因片段,并对其中的阳性扩增产物进行基因序列测定,然后应用BLAST软件以及DNAsist5.0软件对测序结果分析。结果205例中3例阳性,阳性率为1.5%;该BDVp24片段的核苷酸序列与StrainH3915同源性最高达97.67%,与标准病毒株H1766同源性为96.51%,与StrainV/FR的同源性为95.35%,且它们编码的氨基酸相同。结论宁夏地区黄牛中存在动物源性BDV的自然感染,该BDVp24核苷酸序列与标准病毒株具有高度同源性。 展开更多

The biological features of most foamy viruses(FVs) are poorly understood, including bovine foamy virus(BFV). BFV strain 3026(BFV3026) was isolated from the peripheral blood mononuclear cells of an infected cow in Zhangjiakou, China. A full-length genomic clone of BFV3026 was obtained from BFV3026-infected cells, and it exhibited more than 99% amino acid(AA) homology to another BFV strain isolated in the USA. Upon transfection into fetal canine thymus cells, the full-length BFV3026 clone produced viral structural and auxiliary proteins, typical cytopathic effects, and virus particles. These results demonstrate that the full-length BFV3026 clone is fully infectious and can be used in further BFV3026 research. 展开更多
