Detection and Analysis of Antibodies of PRRSV and CSFV

Objective] The passing rates of porcine reproductive and respiratory syn-drome virus (PRRSV) vaccine and classical swine fever virus (CSFV) vaccine were investigated in this paper. The PRRSV vaccine was injected in 14-d-old piglets and the CSFV vaccine was injected in 25-d-old piglets. [Method] The antibody expres-sion amounts were detected with the PRRSV antibody ELlSA detection kit and the CSFV antibody ELlSA detection kit. [Result] The antibody positive rates were 6.7%, 26.7%, 85% and 100% 3, 7, 14 and 30 d after the vaccination of PRRSV vaccine. While for the CSFV vaccine, 13%, 40%, 58.3% and 83.3% of the immune individu-als were antibody positive 3, 7, 14 and 30 d after the vaccination. [Conclusion] The PRRSV vaccine showed the lowest antibody tilter 3 d after the inoculation, but had a great vaccination effect 30 d after the inoculation. The piglets were not immune enough against viral invasion 3 d after the inoculation of CSFV vaccine. Although the antibody positive rate of PRRSV and CSFV vaccines al reached the required level by the Ministry of Agriculture (70%) 30 d after the inoculation, the vaccination of CSFV vaccine could not achieve the ideal effect of 100%. 展开更多

Effect of Eliminated the Breeding Pigs with Unqualified CSFV Antibody on Sows' Reproductive Performance and Pigs' Growth Performance

[Objective] The aim was to explore the effect of eliminated the breeding pigs with unqualified CSFV antibody on sow's reproductive performance and pig's growth performance so as to provide a scientific basis for the eliminating of the breeding pigs with unqualified CSFV antibody in large-scale pig farms.[Method] Serum samples from three scale pig farms were collected and tested with HerdChek CSFV-ELISA kit,then provided a certain scientific basis for scale pig farm to eliminate the unqualified(antibody blocking rate<50%)breeders after booster immunization.[Result ] After eliminating the breeding pigs with unqualified CSFV antibody,the average litter number of piglet in three pig farms(A,B,C)were increased by 0.29(P<0.05),0.40(P<0.01),0.39(P<0.01)respectively;the average number of survival piglets were increased by 0.54(P<0.01),0.35(P<0.01),0.62(P<0.01)respectively;the average litter number of weaned piglets were increased by 0.65(P<0.01),0.71(P<0.01),0.81(P<0.01)respectively.The difference in weight gain of piglet at 30-60 d of age was extremely significant(P<0.01),but inconspicuousness for the swine pregnancy rate,the survival rate of weaned piglet and piglet at 30-60 d of age.[Conclusion] The eliminating the breeding pigs with unqualified CSFV antibody can significantly improve the performance of breeder and piglets. 展开更多

Recently, a novel porcine circovirus-like virus P1 with a circular DNA genome of 0.648 kb was identified. P1 antigen was detected both in vitro and in vivo by synthetic peptide-derived polyclonal antibody-based immunochemistry. The designed peptides were synthesized by solid-phase technique, purified by high performance liquid chromatography, coupled to Keyhole limpet hemocyanin, and injected into rabbits to prepare polyclonal antibody. The emergence of positive cells revealed that synthetic peptide could elicit antibodies against P1 and viral protein could be synthesized. The polyclonal peptide antibodies described here was successfully applied to immunochemical staining and proved helpful in diagnosing P1. 展开更多
