A full-length cDNA encoding fructose-6-phosphate, 2-kinase/fructose-2,6-bisphosphatase from maize (Zea mays L.) was cloned by the methods of reverse transcription-polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends (RACE), and designated as mF2KP. The encoded protein is composed of two regions. Its COOH-terminal region is catalytic region and homologous to the enzymes from other eukaryotes; and its NH 2-terminal region is common and special region only in plant. A truncated fragment of mF2KP covering integrated catalytic region was expressed in Escherichia coli. The fusion protein had the activities of fructose-6-phosphate, 2-kinase as well as fructose-2,6-bisphosphatase. Northern blot showed that the transcript level of mF2KP in seedlings initiated from strong-vigor seeds is lower than that from weak-vigor seeds. 展开更多

To investigate the variation law of pericarp tenderness in growth progress of super sweet corn kernel, the values of pericarp tenderness of 10 super sweet corn inbreeds were measured during kernel growth, and the variations under differ- ent conditions were analyzed. The results showed that there existed gradient differ- ences in pericarp tenderness among the 10 materials, of which PE10 had the best pericarp tenderness, T105 took medial place, and $33205 performed worst in peri- carp tenderness. Pericarp tenderness values of these 3 inbreeds increased curvedly from 12 to 24 days after pollination, in the spring (Wuhan, Hubei) and winter (Ling- shui, Hainan) of 2014. Moreover, the average pericarp tenderness at different time points presented the same decreasing order of $33205, T105, PE10, which was not altered by enviroment. With the growth of kernel, for one material, the difference of pericarp tenderness under different environments presented a law of increment, re- duction, uniformity. As for optimum-picking time, there was four days difference be- tween the spring in Wuhan and winter in Lingshui. However, there was no obvious difference in pericarp tenderness on the optimum picking time, which indicated that growing environment could affect the variation ratio of pericarp tenderness, but it still depended on the nature of materials. 展开更多

Control Effect of Bio-guide Wasp Virus Missile on Ostrinia furnacalis and Its Benefits for Maize Production in Nanchong City

In order to promote "green plant protection", reduce the administration dosage and residue of pesticides and control cost of environment pollution, and improve maize production, quality and agricultural ecological environment safety. In this study, the control effect and benefit of bio-guide wasp virus missile(BGWVM) on Ostrinia furnacalis in Nanchong City was investigated by using insect light traps and combined method of field investigation and experimental demonstration. According to the results, applying 30-37 pairs of BGWVM per hectare exhibited an average control efficiency of 78.0%, and the control cost was 139.50-172.50 CNY/hm2, which was reduced by 142.50-168.00 CNY/hm^2 compared with conventional chemical control. Average maize yield in BGWVM demonstration plot was 817.8 kg/hm2 higher and improved by 11.0% compared with blank control plot. The output value was improved by 1 799.16 CNY/hm^2 and the revenue increased by 1 941.66-1 967.16CNY/hm2. During 2003-2014, the cumulative demonstration and popularization area of BGWVM technique in Nanchong City had reached 24 507.33 hm^2, and the cumulative yield increment reached 20.042 1 million kg. BGWVM application improved total output value by 44.092 6 million CNY and saved control cost by 3.492 3-4.117 2 million CNY, thus increasing the revenue by 47.584 9-48.209 8 million CNY. BGWVM application reduced 551.41 t of 5% dimehypo GG or 12.87 t of 90%monosultap WP. In addition, BGWVM technique is simple to use, labor-saving and safe for human, animals, environment and crops, with significant economic, ecological and social benefits and without any side effect, which has become an ideal substitute for controlling O. furnacalis and reducing chemical pesticides in organic agriculture industry, modern agriculture industry, production of green and pollution-free agricultural products and IBM prevention and control demonstration gardens. 展开更多

The results showed that 6% tebuconazole FS was effective to control maize head smut, and the control effect ranged from 73.4% to 91.1%. In addition, it also improved the maize yield by 36.7%-44.1%, compared with the blank control. The optimal dose of 6% tebuconazole FS for seed dressing was 166.5 g/100 kg to control head smut on maize. 展开更多

A pot experiment was conducted to study the effects of intercropping with Echinochloacrusgali, Polygonumorientale, Conyzacanadensis and Senecioscan-dens on growth and zinc uptake of fruit maize seedlings. The results showed that intercropping with E. crusgali_and C. canadensis increased root, stem and sheath, leaf and aboveground part biomasses of fruit maize seedlings, increased above-ground part biomasses of fruit maize seedlings by 58.89% and 8.76%, respectively;but intercropping with P. orientale and S. scandens decreased the biomass of fruit maize seedlings. Compared with monoculture, intercropping with E. crusgali_and C. canadensis also enhanced the chlorophyl and carotenoid contents of fruit maize seedlings, and intercropping with P. orientale_and S. scandens decreased them. On zinc uptake, intercropping with E. crusgali_and C. canadensis_enhanced the zinc contents in roots, stems and sheaths, leaves and aboveground part of fruit maize seedlings, enhanced by 1.24% and 3.89% in aboveground part; intercropping with P. orientale_and S. scandens decreased zinc contents in fruit maize seedlings. There-fore, intercropping with E. crusgali_and C. canadensis could promote the growth and zinc uptake of fruit maize seedlings. 展开更多

In this study, the result sh... Chitinases play an important role in regulating plant growth and development, especially defending themselves from fungal pathogens. It is important to do the biological analyses in crops. In this study, the result showed that the chitinases were distributed into the whole genome in rice, and nearly the whole genome in maize expect for Chromosome 9. The clustering results showed that one out of three chitinases from maize and rice belonged to new groups, which were separated from those in the conformed Classes I-VII. The identification of most amino acid sequences was very low among the chitinases from rice and/or maize. It was inferred that the chitinases with different functions were relatively stable during plant evolution. The relationship of chitinases expression between leaf blade and anther was positively significant in maize, but not significant in rice. Additionally, the ratio of chitinases with up- or down-regulated expression in sensitive maize under Fusarium moniliforme inoculation was different from that in sensitive rice under Magnaporthe grisea inoculation. It might be result from different tissues infected by different fungi. The number of chitinases from resistant maize was less than that from sensitive maize, which inferred that the resistant pathways on F. moniliforme should be not only chitin induced Pathogen-associated molecular PTI (patterns-triggered immunity) pathway, but also might include other PTI pathways that improve tolerance to F. moniliforme. The analysis of expression pattern of chitinases from maize and rice under fungi inoculation will be contributed into further research on the defense mechanism of fungi in crops. 展开更多

Control Effect of 80% Nicosulfuron·Atrazine Water-dispersible Granules on Weeds in Spring Maize Field

[Objective] This study aimed to investigate the control effect of different concentrations of 80% nicosulfuron·atrazine water-dispersible granules on weeds in spring maize field. [Method] Maize field was sprayed with 300, 375, 450, 750 g/hm280% nicosulfuron·atrazine water-dispersible granules respectively, with 40 g/L nicosulfuron suspending concentrate and 38% atrazine suspending concentrate as control agents. Artificial weeding and control(CK) plots were set. [Result] Fresh weight control efficiency of 375-750 g/hm280% nicosulfuron·atrazine water-dispersible granules was significantly higher than that of 40 g/L nicosulfuron suspending concentrate and 38% atrazine suspending concentrate; no obvious phytotoxicity symptoms were observed after application of 300-450 g/hm280% nicosulfuron·atrazine waterdispersible granules; 750 g/hm280% nicosulfuron·atrazine water-dispersible granules posed certain impact on the growth of maize seedlings. Compared with control plots, various doses of 80% nicosulfuron·atrazine water-dispersible granules significantly improved the yield of maize. [Conclusion] In the present study, 375-450 g/hm280% nicosulfuron·atrazine water-dispersible granules exhibited high control effect on weeds in maize field and were safe for the growth of maize seedlings. 展开更多
