Cholestasis results in a buildup of bile acids in serum and in hepatocytes.Early studies into the mechanisms of cholestatic liver injury strongly implicated bile acidinduced apoptosis as the major cause of hepatocellu...Cholestasis results in a buildup of bile acids in serum and in hepatocytes.Early studies into the mechanisms of cholestatic liver injury strongly implicated bile acidinduced apoptosis as the major cause of hepatocellular injury.Recent work has focused both on the role of bile acids in cell signaling as well as the role of sterile inflammation in the pathophysiology.Advances in modern analytical methodology have allowed for more accurate measuring of bile acid concentrations in serum,liver,and bile to very low levels of detection.Interestingly,toxic bile acid levels are seemingly far lower than previously hypothesized.The initial hypothesis has been based largely upon the exposure of μmol/L concentrations of toxic bile acids and bile salts to primary hepatocytes in cell culture,the possibility that in vivo bile acid concentrations may be far lower than the observed in vitro toxicity has far reaching implications in the mechanism of injury.This review will focus on both how different bile acids and different bile acid concentrations can affect hepatocytes during cholestasis,and additionally provide insight into how these data support recent hypotheses that cholestatic liver injury may not occur through direct bile acid-induced apoptosis,but may involve largely inflammatory cell-mediated liver cell necrosis.展开更多
The contribution of the genetic make-up to an individual’s capacity has long been recognized in modern pharmacology as a crucial factor leading to therapy ineffciency and toxicity, negatively impacting the economic b...The contribution of the genetic make-up to an individual’s capacity has long been recognized in modern pharmacology as a crucial factor leading to therapy ineffciency and toxicity, negatively impacting the economic burden of healthcare and restricting the monitoring of diseases. In practical terms, and in order for drug prescription to be improved toward meeting the personalized medicine concept in drug delivery, the maximum clinical outcome for most, if not all, patients must be achieved, i.e. , pharmacotyping. Such a direction although promising and of high expectation from the society, it is however hardly to be afforded for healthcare worldwide. To overcome any existed hurdles, this means that practical clinical utility of personalized medicine decisions have to be documented and validated in the clinical setting. The latter implies for drug delivery the effcient implementation of previously gained in vivo pharmacology experience with pharmacogenomics knowledge. As an approach to work faster and in a more productive way, the elaboration of advanced physiologically based phar-macokinetics models is discussed. And in better clarifying this topic, the example of tamoxifen is thoroughly presented. Overall, pharmacotyping represents a major challenge in modern therapeutics for which pharmacologists need to work in successfully fulflling this task.展开更多

This paper discusses one of the modern methods of time series analysis--a method of determini...The article presents the results of recent investigations into Holter monitoring of ECG, using non-linear analysis methods. This paper discusses one of the modern methods of time series analysis--a method of deterministic chaos theory. It involves the transition from study of the characteristics of the signal to the investigation of metric (and probabilistic) properties of the reconstructed attractor of the signal. It is shown that one of the most precise characteristics of the functional state of biological systems is the dynamical trend of correlation dimension and entropy of the reconstructed attractor. On the basis of this it is suggested that a complex programming apparatus be created for calculating these characteristics on line. A similar programming product is being created now with the support of RFBR. The first results of the working program, its adjustment, and further development, are also considered in the article.展开更多

The composition, microstructure, magnetic property, adsorption and photocatalytic activity of the prepared samples were characterized by means of modem analytical techniques. The results indicated that ZT0.6F1.4CNTs composites not only held the original special structure and excellent adsorption properties of CNTs, but also had suitable magnetic property and excellent photocatalytic activity. The removal rate of the samples on Rhodamine B (RhB) depended on the adsorption of CNTs and the photocatalytic degradation of ZTo.6F1.4 in the composites. The maximum adsorption amount (qm) of ZT0.6F1.4/CNTs with the mass ratios of ZT0.6F1.4 to CNTs (mz/c)=l was up to 17.153 mg g-t for RhB, its adsorption behavior was in accord with Langmuir model, and its photocatalytic degradation activity on RhB had a positive correlation with the content of ZT0.6F1.4 in the sample. The experimental results indicate that the total removal rate of composite with rnz/c=l on RhB was more than 95% and the composite had good decontamination capability on industrial dye wastewater. In addition, the samples can be recovered conveniently, activated easily and had good performance for recycling.展开更多
