In this work, we calculated the average probability of residue-residue contacts in 470 globular proteins and analyzed the distribution of contacts in the different interval of residues using Contacts of Structural Units (CSU) and Structural Classification (SCOP) software. It was found that the relationship between the average probability PL and the residue distance L for four structural classes of proteins could be expressed as lgPL=a+b×L, where a and b are coefficients. We also discussed the connection between two aspects of proteins which have equal array residue number and found that the distribution probability was stable (or un- stable) if the proteins had the same (or different) compact (for example synthase) in the same structural class.展开更多
The growing interest in skeletal muscle regeneration is associated with the opening of new therapeutic strategies for muscle injury after trauma, as well as several muscular degenerative pathologies, including dystrop...The growing interest in skeletal muscle regeneration is associated with the opening of new therapeutic strategies for muscle injury after trauma, as well as several muscular degenerative pathologies, including dystrophies, muscu- lar atrophy, and cachexia. Studies focused on the ability of extracellular factors to promote myogenesis are therefore highly promising. We now report that an adipocyte-derived factor, globular adiponectin (gAd), is able to induce mus- cle gene expression and cell differentiation, gAd, besides its well-known ability to regulate several metabolic func- tions in muscle, including glucose uptake and consumption and fatty acid catabolism, is able to block cell cycle entry of myoblasts, to induce the expression of specific skeletal muscle markers such as myosin heavy chain or caveolin-3, as well as to provoke cell fusion into multinucleated syncytia and, finally, muscle fibre formation, gAd exerts its pro- differentiative activity through redox-dependent activation of p38, Akt and 5'-AMP-activated protein kinase path- ways. Interestingly, differentiating myoblasts are autocrine for adiponectin, and the mimicking of pro-inflammatory settings or exposure to oxidative stress strongly increases the production of the hormone from differentiating cells. These data suggest a novel function of adiponectin, directly coordinating the myogenic differentiation program and serving an autocrine function during skeletal myogenesis.展开更多

Methods: Eighty-seven PTC patients after ...Objective: The aim of this study was to compare the effect of 131I therapy of different clinical stages in postopera- tive patients with papillary thyroid carcinoma (PTC). Methods: Eighty-seven PTC patients after surgery ablated with high doses of 131I from 2004 to 2010 were retrospectively reviewed. The efficacy of 131I therapy was assessed by three diagnostics that serum thyroglobulin (Tg) was normal or significantly reduced, 131I whole body scan (131I-WBS) was negative or the metas- tases shrank or the number of them decreased and new metastases was not found in cervical ultrasound examination. The x2 test was used to analyze 3 factors which might affect the therapeutic efficacy of 131I in patients of different clinical period, including different surgical ways (total or subtotal thyroidectomy along with half or double sides neck lymph node dissection), age (〈 45 years and 〉 45 years) and ablative 131I dose. Results: Of 87 patients, the effective rate of 46 patients I stage was 89.13% (41); the effective rate of 22 cases III stage was 77.27% (17); the effective rate of 19 cases IV stage was 36.84% (7). The corresponding intra-groups statistical difference of 3 stages was significant by x2 test (x2 = 1.72, 19.03, 6.87; P 〉 0.25, P 〈 0.005, P 〈 0.01). The effective rate was 91.67% (44) in 48 cases undergoing total thyroidectomy; the effective rate was 53.85% (21) in 39 patients undergoing subtotal thyroidectomy. There was a significant difference between the two groups above by x2 test (x2 = 16.291; P 〈 0.005). Conclusion: The efficacy of 131I ablation of stage I and stage III in postoperative PTC patients was almost alike, while the efficacy of stage IV descended markedly. The results was mainly determined by residual thyroid tissue size because of different surgical modus.展开更多

Methods: One thousand and two normal cases, 95 thyroid cancer patients and 243 nodular goiter patients were selected to measure serum β2-MG levels using double-antibody sandwich enzyme-linked immunosorbent assay (ELISA). Results: The positive rate of 7.78% in normal population (78/1002) and 31.57% in thyroid cancer patients (30/95). There were significant differences between the normal population and thyroid cancer patients (X2 = 55.352; P = 0.000). The positive rate of 7.81% in nodular goiter patients (19/243) and there were no significant differences between the normal population and nodular goiter patients (X2 = 0.0004; P = 0.986), but significant differences between nodular goiter patients and thyroid cancer patients (x2 = 31.106; P = 0.000). Meanwhile, the significant difference of the positive rate existed in between the various pathological types of thyroid cancer (X2 = 10.015; P = 0.007), anaplastic thyroid cancer patients with the highest positive rate and The significant difference was found between the positive lymph node metastasis groups and negative lymph node metastasis groups (x2 = 4.441; P = 0.035), the presence of distant metastasis group and absence of distant metastasis group (X2 = 9.795; P = 0.002). Statistically significant increased ratio of the nucleus and the cytoplasm height and the nucleolus and the nucleus height is characteristically for cancer cells compared to normal cells. AFM method allows determining the viral changes in squamous cells by detecting the perinuclear area of enlightenment. AFM allows objectifying immunocytochemistry data by quantifying the height of immune complexes. Possibilities of quantitative immunocytochemistry with AFM are shown in the model of Her2/neu oncoprotein expression in breast cancer and thyroglobulin in papillary thyroid cancer.展开更多
