目的:观察解毒灌肠方灌肠对中晚期原发性肝细胞癌经导管动脉栓塞术(TAE)后综合征的防治作用。方法:将53例拟接受TAE的中晚期原发性肝细胞癌患者随机分为治疗组(28例)和对照组(25例)。对照组于TAE后接受西医常规治疗;治疗组在对照组治疗... 目的:观察解毒灌肠方灌肠对中晚期原发性肝细胞癌经导管动脉栓塞术(TAE)后综合征的防治作用。方法:将53例拟接受TAE的中晚期原发性肝细胞癌患者随机分为治疗组(28例)和对照组(25例)。对照组于TAE后接受西医常规治疗;治疗组在对照组治疗的基础上,于TAE前3h至术后第7天加用解毒灌肠方中药灌肠,每日1次。于术后第8日对2组患者发热、肝区疼痛、腹胀症状进行评分,并做统计学比较;比较术前与术后第8日肝功能指标、卡氏(KPS)评分改变情况。结果:TAE后第8日,治疗组发热、肝区疼痛、腹胀症状评分均明显低于对照组(P<0.05)。2组患者术后第8日谷草转氨酶(AST)、谷丙转氨酶(ALT)、总胆红素(TBIL)均明显高于术前(P<0.05),但治疗组上述指标明显低于对照组(P<0.05)。治疗组术后第8日KPS评分明显高于术前(P<0.05)和对照组术后(P<0.01)。结论:解毒灌肠方灌肠对原发性肝细胞癌TAE后综合征具有良好的防治作用。 展开更多

Plants synthesize the osmoprotectant glycine betaine (GB) via choline→betaine aldehyde→glycine be- taine[1]. Two enzymes are involved in the pathway choline monooxygenase (CMO) and betaine aldehyde dehydrogenase (BADH). A full length CMO cDNA (1,643bp) was cloned from Amaranthus tricolor. The open reading frame encoded a 442-amino acid polypeptide, which showed 69% identity with CMOs in Spina- cia oleracea L. and Beta vulgaris L. DNA gel blot analysis indicated the presence of one copy of CMO gene in the A. tricolor genome. The expressions of CMO and BADH proteins in A.tricolor leaves significantly increased under salinization, drought and heat stress (42℃), as determined by immunoblot analysis, but did not respond to cold stress (4℃), or exogenous ABA application. The increase of GB content in leaves was parallel to CMO and BADH contents. 展开更多

Genome-wide and candidate gene association studies have identified several variants that predispose indi- viduals to developing nonalcoholic fatty liver disease (NAFLD). However, the gene that has been consis- tently involved in the genetic susceptibility of NAFLD in humans is patatin-like phospholipase domain contain- ing 3 (PNPLA3, also known as adiponutrin). A nonsyn- onymous single nucleotide polymorphism in PNPLA3 (rs738409 C/G, a coding variant that encodes an amino acid substitution I148M) is significantly associated with fatty liver and histological disease severity, not only in adults but also in children. Nevertheless, how PNPLA3 influences the biology of fatty liver disease is still an open question. A recent article describes new aspects about PNPLA3 gene/protein function and suggests that the I148M variant promotes hepatic lipid synthesis due to a gain of function. We revise here the published data about the role of the I148M variant in lipogen- esis/lipolysis, and suggest putative areas of future research. For instance we explored in silico whether the rs738409 C or G alleles have the ability to modify miRNA binding sites and miRNA gene regulation, and we found that prediction of PNPLA3 target miRNAs shows two miRNAs potentially interacting in the 3' UTR region (hsa-miR-769-3p and hsa-miR-516a-3p). In addition, interesting unanswered questions remain to be explored. For example, PNPLA3 lies between two CCCTC-binding factor-bound sites that could be tested for insulator activity, and an intronic histone 3 lysine 4 trimethylation peak predicts an enhancer element, cor- roborated by the DNase I hypersensitivity site peak. Finally, an interaction between PNPLA3 and glycerol- 3-phosphate acyltransferase 2 is suggested by data miming. 展开更多

One possible application for the excess of glycerol, which is an exceeding byproduct in biodiesel industry, was used as carbon and energy sources for bioproducts synthesis. This work aims to evaluate biosurfactant production from glycerol by Pseudomonas aeruginosa EQ 109 isolated from crude oil-contaminated soil. Factorial design 2^3 was utilized to optimize the amount of biosurfactant produced, by using pH (A), initial biomass concentration (B), and initial glycerol concentration (C) as independent factors. The experiments were carried out in flasks containing 100 mL of mineral medium. Biosurfactant production was monitored by increase of the emulsification of aviation kerosene (E24) and surface tension reduction (STr). The results have shown that, at pH = 7.0, in order to increase E24, variables as B and C are the most influential, leading to a maximum value of E24 = 79%, as well as for an increase of GC (GCmax = 49%). STR was the variable with the best correlation factor for the proposed linear model (R2=0.96) and its maximum value was 48%. Xfwas not significant to the model, although it was influenced by pH and C, with C = 40g/L (Xfmax = 4.56 g/L). 展开更多

The effect of monoethylphosphate (MEP, commercial available or synthesized) together with IL-2 on the selective proliferation of human γ~δ T cells in Vitro from peripheral blood mononuclear cells (PBMC) of healthy donors and of cancer patients was investigated. The γ~δ T cells were stimulated by MEP to proliferate in a dose-dependent manner. The effect of synthesized MEP was 10 times greater than that of commercial MEP. When the PBMCs of healthy donors were cultured for 25 d in the medium containing different concentrations of MEP, the total cell number increased about 1000-3000 fold; and the ratio of γ~δ T cells reached to 70-80%. The selective expansion of γ~δ T cells depended on the synergic action of MEP and IL-2. The bulk cultured γ~δ T cells exhibited obvious cytotoxic activities against allogenic tumor cell lines (SQ-5,K562 alld Daudi) and autologous tumor cells. The culture system described here not only offers a simple method for obtaining a large number of γ~δ T cells which may become a new effector in the adoptive immunotherapy, but also provides a useful model for the further studies of the structure and function of γ~δ T cells in vitro. 展开更多

Methods: Thirty-two cases of simple obesity patients were enrolled into this study. Acupoints of the Stomach Meridian and Spleen Meridian as Zhongwan (中脘 CV 12), Liangmen (梁门 ST 21), Tianshu (天枢 ST 25), Guayuan (关元 CV 4), etc. were punctured, once daily in the first 5 days, and once every other day afterwards, with 10 sessions being a therapeutic course. Before treatment and after 3 courses of treatment, the body weight, waistline, weight index, seam cholesterol (CH), triglyceride and aggregation index of red blood cell (RBC) were detected. Results: After acupuncture treatment, all the indexes of body weight, waistline, weight index, serum CH, triglyceride and aggregation index of RBC decreased significantly in comparison with those of pre-treatment(P< 0.05). Conclusion: Acupuncture can apparently improve cellular hemorheology, reduce body weight, serum cholesterol and TG levels in simple obesity patients. 展开更多

The aim of this study was to assess the effect of long-term in vitro sub-culturing on the varietal degeneration of three sweet potato varieties, namely, Monate, Mokone and Ndou which were sub-cultured for 32, 23 and 12 generations, respectively. Each generation was cultured in a media which is made from 4.43 g/L Murashige and Skoog (MS), 30 g/L sucrose and 2 g/L gelrite, respectively, and grown under 16 h light and 8 h dark photoperiod for 30 d. For each generation, 45 plantlets were acclimatized for two months in a glasshouse. Data on in vitro growth performance and 11 morphological characteristics during acclimatization were recorded. Early root and shoot formation was observed after the 27th and 21st sub-cultured generations of Monate and Mokone, respectively. During acclimatization, plantlets from the same variety showed differences in morphological traits such as leaf colour, abaxial leaf pigmentation, vine pigmentation, petiole pigmentation, leaf wrinkling and flowering. However, the rate of these morphological differences is random and irrespective to increase in sub-culturing. Therefore, to understand the genetic base of these morphological variability, two plantlets from each variety were subjected to genetic analysis by using five simple sequence repeat (SSR) primers (IB-242, IB-318, IB-255F, 1B-248 and IB-255). Although SSR loci IB-255F and IB-318 could distinguish between the three varieties, there were no allelic polymorphisms detected in plantlets from the same varieties. Therefore, long-term sub-culturing do not leads to quality degeneration in the three sweet potato varieties. 展开更多

Among the numerous inland river basins in the arid northwest China,Shiyang River basin is known for its most serious water shortage that constrains its social and economic developments and for some of the worst ecological and environmental deterioration in China.The research on the value of water resources used for ecosystem is the basis for reasonable allocation of water resources between users of ecosystem and economic system.In this paper the concept of dynamic value for the ecosystem services is proposed.A modified coefficient for the ecosystem service value per unit area is proposed according to the cover degrees of biomes combined with expert consultation.Based on the developmental stage coefficient and scarcity of ecological resources,a dynamic evaluation method is proposed.The theoretical formula and simple calculation formula of the sharing benefits coefficient and benefits per unit ecological water utilization are proposed.The result shows that the benefit of unit ecological water utilization in the lower reach was greater than that in the upper reach in the Shiyang River basin. 展开更多

Glycerol mono laurate(GML)has been widely used as an effective antibacterial emulsifier in the food in?dustry.A total of 360 44-week-old Hy-Line brown laying hens were randomly distributed into four groups each with six replicates of 15 birds,and fed with com-soybean-meal-based diets supplemented with 0,0.15,0.30,and 0.45 g/kg GML,respectively.Our results showed that 0.15,0.30,and 0.45 g/kg GML treatments significantly decreased feed conversion ratios(FCRs)by 2.65%,7.08%,and 3.54%,respectively,and significantly increased the laying rates and average egg weights.For egg quality,GML drastically in creased albume n height and Haugh units,and enhanced yolk color.Notably,GML increased the concentrations of polyunsaturated and monounsaturated fatty acids and reduced the concentration of total saturated fatty acids in the yolk.The albumen composition was also significantly modified,with an increase of 1.02%in total protein content,and increased 8ntents of His(4.55%)and Glu(2.02%)under the 0.30 g/kg GML treatment.Additionally,GML treatments had positive effects on the lipid metabolism of laying hens,including lowering the serum triglyceride and total cholesterol levels and reducing fat deposit!on in abdominal adipose tissue.Intestinal morphology was also improved by GML treatment,with increased villus length and villus height to crypt depth ratio.Our data demonstrated that GML supplementation of laying hens could have beneficial effects on both their productivity and physiological properties,which indicates the potential application of GML as a functional feed additive and gives us a new in sight into this traditional food additive. 展开更多

The intestinal permeability of two flavonoid compounds liquiritin (LQ) and isoliquiritin (ILQ) was investigated using the Caco-2 cell monolayer model. In order to evaluate the permeability and predict the absorption mechanism of the two compounds, the study on bidirectional permeability from the apical (AP) side to the basolateral (BL) side as well as from the BL side to the AP side was carried out. The determination was performed by HPLC-UV method. And the permeability parameters, especially the apparent permeability coefficients (Papp), were then calculated. The Papp values of LQ and ILQ are (5.40±0.16)× 10^-7 and (8.69±0.15)× 10^-7 cm/s, respectively. The results of time- and concentration-dependent transport experiments indicate that both LQ and ILQ are poor absorbed mainly through passive diffusion. 展开更多

The 1-O-monoalkyl glycerol ethers (MAGEs) were initially viewed as the biomarkers for sulfate-reducing bacteria (SRB) me- diating anaerobic oxidation of methane in the marine environments. However, limited information is known about their distri- bution in terrestrial and other aquatic settings including soils, fresh water lakes, and cave sediments, which may obscure our understanding of their biological sources. Here we found that MAGEs were ubiquitous but differed obviously in distributional pattern among those environments. The surface soils are dominated generally by iC15:0-MAGE, followed by nCI6:o-MAGE whereas the lake sediments show the opposite, resulting in significantly higher iC15:0/nC16:0 ratios in soils than in lake sedi- ments. The cave deposits are characterized by considerably higher proportions of branched MAGEs than the former two envi- ronments. The logarithm of iC15:0/aC15:0 ratio shows a significant negative correlation with soil pH, likely reflecting an adapta- tion of microbial cell membrane to change in the ambient proton concentration. The MAGE profiles in cultured bacteria cannot fully explain the MAGE distribution in all the samples analyzed. Therefore, MAGEs in soil, lake sediments, and cave deposits likely have additional biological source(s) other than SRB and cultured MAGE-producing bacteria. The difference in MAGE pattern among environments is likely to be attributed to change in microbial communities. 展开更多

When exposed to biotic or abiotic stress conditions, plants produce ethylene from its immediate precursor 1-aminocyclopropane-1- carboxylate (ACC), leading to retarded root growth and senescence. Many plant growth-promoting rhizobacteria contain the enzyme ACC deaminase and this enzyme can cleave ACC to form a-ketobutyrate and ammonium, thereby lowering levels of ethylene. The aim of this study was to isolate and characterize ACC deaminase-producing bacteria from the rhizosphere of salt-stressed canola (Brassica napus L.). Out of 105 random bacterial isolates, 15 were able to utilize ACC as the sole source of nitrogen. These 15 isolates were also positive for indole acetic acid (IAA) production. Phylogenetic analysis based on partial 16S rDNA sequences showed that all isolates belonged to fluorescent Pseudomonas spp. In the canola rhizosphere investigated in this study, Pseudomonas fluorescens was the dominant ACC deaminase-producing species. Cluster analysis based on BOX-AIR-based repetitive extragenic palindromic-polymerase chain reaction (BOX-PCR) patterns suggested a high degree of genetic variability in ACC deaminase-producing P. fluorescens strains. Because glycidyl(Gly) contains an epoxy and an active hydroxyl group, the Gly unit is difficult to introduce into certain polymeric chains in a controlled manner and usually yields hyperbranched polyglycidyl. Alternatively, the monomer 1-ethoxyethyl glycidyl ether(EEGE), derived from Gly and ethyl vinyl ether, has shown potential for application in polymer chemistry, and homopolymerization of this monomer directly produces linear poly(1-ethoxyethyl glycidyl ether) and further yields linear polyglycidyl. In this review, the initiation system of the EEGE monomer is first discussed in terms of chain transfer to monomers in ring-opening polymerization of epoxides with substituent groups. Then, random copolymerization of EEGE with other epoxides is considered. In addition, because the EEGE units on polymers can be transferred to Gly units and further used to construct copolymers with complicated architectures, the applications of EEGE monomers to block, graft, and hyperbranched copolymers are reviewed. Finally, the synthesis of main chain and terminal functional polyethers by transforming the hydroxyl groups at the polymer end or on the main chain into certain functional groups are also discussed. Chemistry based on EEGE has been proved to be an efficient, versatile route to constructing copolymers containing Gly units and ultimately yielding the target properties and applications. 展开更多
