Previously, we have reported a prenylated isoflavonoid, isoangustone A(IAA) from licorice(Glycyrrhiza uralensis), which induces apoptosis in colorectal cancer cells by disrupting mitochondrial functions. In the present study, we compared a group of flavonoids from licorice with IAA for their anti-proliferation activities and effects on intracellular signaling. The results indicated that the isoprenyl groups on the A and B rings, the hydroxyl groups at the ortho position of isoprenyl on A ring and the conjugated plane of C ring might contribute to the anti-cancer activity of prenylated flavonoids. Based on the above structure-activity relationship, we further identified four prenylated flavonoids with similar anti-cancer activities from licorice. Taken together, our present study established a preliminary structure-activity relationship of anti-cancer prenylated flavonoids, and our data provided important leading compounds from licorice, which deserved further research and development.展开更多

The mobile phase consisted of (A) acetonitrile and (B) 0.026% aqueous H3PO4 (V/V) using a gradient elution of 20%-25% A at 0-20 min, 25%-33% A at 20-30 min, 33%-50% A at 30-55 min, 50%-60% A at 55-65 min, and 60% A between 65 min and 80 min, and peaks were detected at 280 rim. The fourteen compounds were assigned by HPLC-Orbitrap MS methods. The regression coefficient for the linear equations for the 14 compounds ranged between 0.9998 and 1. The limits of detection and quantification lay in the range of 0.032-2.461 and 0.154-8.202 μg·mL^-1, respectively. The relative recovery rates for the 14 compounds were in the range of 93.90%-106.73% with RSDs being less than 5%. Coefficient variations for intra-day and inter-day precisions were in the range of 0.27%-2.38% and 0.31%-3.51%, respectively. In summary, the validated method was applied to the simultaneous determination of the 14 components in 29 different licorice samples and was proven to be suitable for quality evaluation of licorices and their active fractions.展开更多

A total of 18 phenolic compounds were isolated by silica gel and Sephadex LH-20 chromatography, together with preparative HPLC methods. By analysis of the MS and NMR spectroscopic data and comparison with those in literature, these 18 compounds were identified as apigenin (1), galangin (2), luteolin (3), kempherol (4), quercetin (5), hispidulin (6), 6-methoxykaempferol (7), eupafolin (8), 3-methylquercetin (9), ermanin (10), 5,7,4'-trihydroxy-3,6-dimethoxyflavonone (11), bracteoside (12), 7-O-(β-D-glucopyranosyl)-galactin (13), isochlorogenic acid B (14), isochlorogenic acid C (15), 4-hydroxybenzoic acid (16), 5-pentadecylbenzene-l,3-diol (17) and scopoletin (18). Among them, compounds 1-13 were identified as flavonoids, Compounds 2 and 6-17 were isolated from both the nlant and the Matricaria genus for the first time.展开更多
