天麻姜制前后对小鼠宏观行为和脑组织形态结构的影响 被引量:7

目的研究姜天麻炮制前后对小鼠脑组织形态结构变化的影响,为深入研究天麻提供数据支撑。方法以昆明种小鼠SPF级为试验对象,将小鼠随机分为空白组、姜天麻组、天麻组和生姜组,每组10只。除空白组给予等量生理盐水外,其它组别给药浓度为0.... 目的研究姜天麻炮制前后对小鼠脑组织形态结构变化的影响,为深入研究天麻提供数据支撑。方法以昆明种小鼠SPF级为试验对象,将小鼠随机分为空白组、姜天麻组、天麻组和生姜组,每组10只。除空白组给予等量生理盐水外,其它组别给药浓度为0.1 mL/g,灌胃给药,每天1次,连续30 d,观察不同剂量组对小鼠宏观行为学和脑组织形态结构变化的影响。结果与空白组相比,对于宏观行为学考察,小鼠活动量的顺序依次为:天麻组<空白组<姜天麻组<生姜组;对于脑组织,姜天麻组脑神经元和小胶质细胞增多明显,并随给药时间延长,部分细胞有凋亡之势;生姜组小胶质细胞增多,随着给药时间延长,神经元细胞和小胶质细胞集结一起。结论天麻经姜制后,使小鼠脑组织结构发生了变化,这种变化与给药时间和剂量存在相关,提示天麻经姜制后,其对小鼠脑组织形态产生了影响。 展开更多

目的探讨姜天麻炮制前后对硝酸甘油诱导的大鼠偏头痛的影响。方法采用皮下注射硝酸甘油复制偏头痛大鼠模型,以酶联免疫吸附法(ELISA)测定血浆中内皮素(ET)、降钙素基因相关肽(CGRP),脑组织中5-羟色胺(5-HT)、多巴胺(DA)等含量;以生化法... 目的探讨姜天麻炮制前后对硝酸甘油诱导的大鼠偏头痛的影响。方法采用皮下注射硝酸甘油复制偏头痛大鼠模型,以酶联免疫吸附法(ELISA)测定血浆中内皮素(ET)、降钙素基因相关肽(CGRP),脑组织中5-羟色胺(5-HT)、多巴胺(DA)等含量;以生化法测定血浆中一氧化氮(NO)含量,对比各组别动物行为学及生化指标情况。结果与空白组比较,模型组挠头次数明显增多(P<0.01);血浆NO、CGRP含量明显升高(P<0.01),而ET、DA、5-HT含量均降低(P<0.01),说明模型复制成功。与模型组比较,正天丸对照组、生天麻组、姜天麻组在0~30、30~60 min两个时区的挠头次数明显降低(P<0.01);正天丸对照组中NO、CGRP含量明显降低(P<0.01),ET含量升高(P<0.05),DA、5-HT含量升高尤为明显(P<0.01);生天麻组中NO含量降低,而ET、DA含量升高,组间差异均具统计学意义(P<0.05),5-HT含量升高尤为明显(P<0.01),CGRP含量明显降低(P<0.01);姜天麻组中NO、CGRP含量均明显降低(P<0.01),ET含量升高(P<0.05),但5-HT、DA含量升高尤为明显(P<0.01),说明天麻经姜制后,对偏头痛的影响作用增大。结论天麻对硝酸甘油诱导大鼠偏头痛作用,可能是通过降低NO、CGRP和提高ET、DA、5-HT的含量,以改善偏头痛症状,且姜天麻较生品作用强,说明炮制后药效增强。本研究可为深入研究天麻提供基础。 展开更多

This product was identified as p-hydroxy benzyl alcohol on the basis of its 1H, 13C NMR and EI-MS spectral data. It could be inferred that the enzyme responsible for the biotransforma-tion reaction was a kind of extracellular and constitutive enzyme since the transformation reaction of the substrate could be carried out in cell free extracts of the fermentation broth of the Mucor spinosus. 展开更多

Chow were studied in Ganzi prefecture. The results showed that tree species had a very significant effect on asexual reproduction yield of wild Gastrodia elata Bl. f. glauca S. Chow. The drying rate and yield of Gastrodia elata Bl. f. glauca S. Chow cultured by close grained woods such as Quercus spinosa David ex Fr. was extremely significant higher than that cultured by coarse grained wood such as Betula platyphylla Suk. The dosage had a very significant effect on the yield of wild Gastrodia elata Bl. f. glauca S. Chow whereas it had no influence on the drying rate. 展开更多

Influences of Different Habitats on Asexual Propagation of Wild Gastrodia elata f. glauca being Domesticated in Ganzi

[Method] The research tri... [Objective] The influences of different habitats on asexual propagation of wild Geg(abbreviation for Gastrodia elata Bl. f. glauca S. Chow) being domesticated in Ganzi prefecture was studied. [Method] The research trial was carried out in the following three kinds of habitats at the same time: the plastic greenhouses, the birch forest of shady slope and the shrubbery of sunny slope, and the results were analyzed with group data average hypothesis test method. [Result] The overall tuber(the juvenile tuber, the immature tuber and the mature tuber) yield and the mature tuber yield were both very significantly higher in the plastic greenhouses than in the other two types of habitats, and respectively reached 9.52 and 6.70 kg/m2; the mature tuber drying rate was dramatically or very dramatically lower in the plastic greenhouses than in the other two types of habitats, and was 23.84%; the stabilities of the overall tuber yield, the mature tuber yield and the drying rate were all reduced in the order of greenhouses, birch forest and shrubbery; and the dry mature tuber yield of the plastic greenhouses was the highest in the three kinds of habitats, and was 1.60 kg/m2. [Conclusion] A good habitat had to be created or chosen in the domesticating cultivation of the wild Geg in Ganzi prefecture. The preferred choice was the plastic greenhouse, the alternative one was the deciduous broadleaved forest of shady slope. The imitating wild planting of the wild Geg should be done in the warm moist deciduous broad-leaved forest of shady slope. 展开更多

Gastrodia elata Bl.is a traditional Chinese herbal medicine with special pharmacological effects on headache and numbness.However,its wild sources are now in short supply because of over-excavation.A total of 14 endophytic fungi were isolated from Gastrodia elata Bl collected from Yinjing,in which 10 strains were identified,including Neonectria,Ascomycota,Fusarium,Oidiodendron and Dothiorella.All crude extracts showed some degrees of antioxidant activities by T-AOC method and DPPH methods,especially those from Oidiodendron GeR6 and Ascomycota sp.GeR2,which might be attributed due to their high phenolic contents.However,only Oidiodendron GeR6 contained gastrodin(39.2μg/g)as the host plant in the crude extracts.In conclusion,the notable activities of GeR6 indicated its medical use as a natural antioxidant agent,as well as substitutable use for Gastrodia elata Bl.in the future. 展开更多
