目的观察重组结核杆菌热休克蛋白10(CPN10)对成骨细胞(OB)-外周血单个核细胞(PBMs)共培养体系中破骨细胞生成及相关基因表达的影响。方法建立培养上清相通但二者互相不接触的成骨细胞一单个核细胞共育模型。实验分对照组和CPN10(10μg/... 目的观察重组结核杆菌热休克蛋白10(CPN10)对成骨细胞(OB)-外周血单个核细胞(PBMs)共培养体系中破骨细胞生成及相关基因表达的影响。方法建立培养上清相通但二者互相不接触的成骨细胞一单个核细胞共育模型。实验分对照组和CPN10(10μg/ml)处理组。主要观察指标:①采用TRAP染色及扫描电镜检测破骨细胞生成及小牛骨磨片吸收陷窝,②应用Realtime PCR检测与破骨细胞生成相关基因NFATc1、c-Fos、RANKL、OPG的基因表达。结果两组细胞均有TRAP阳性多核破骨细胞生成,并在小牛骨磨片上形成吸收陷窝;但对照组所获TRAP阳性多核细胞数目、吸收陷窝数目及面积均显著小于CPN10组。Realtime PCR检测结果显示CPN10组与对照组相比NFATc1、c-Fos、RANKL、OPG相对浓度分别为7.410±1.738、8.844±1.981、22.4272±2.058、2.445±0.2517(P<0.05),对照组各基因表达均显著低于CPN10组。结论 CPN10在成骨细胞-单个核细胞(OB-PBMs)共培养体系中可促进OC的生成及骨吸收,CPN10通过对成骨细胞的作用,致其分泌的OPG/RANKL比例失调,并上调破骨细胞相关基因NFATc1、c-Fos、RANKL、OPG的基因表达。 展开更多

Callus Induction and Plant Regeneration from Mature Seeds of Perennial Ryegrass 被引量:15

Objective The aim was to explore callus induction and plant regeneration of perennial ryegrass, as well as provide the foundation for transgenic research on perennial ryegrass.[ Methed] Mature seeds of perennial ryegrass were used as explants to study the effects of different hormone compositions on callus induction, proliferation and plant differentiation. Result The result showed that the induction rate achieved its highest on 2,4-D of 8 mg/L combining with 6-BA of 0.025 mg/L, which was up to 56.42%. Callus were differentiated after two to three generations, the highest differentiation rate 34.14% was achieved in the medium contained MS medium with 6-BA of 2 mg/L, and the differentiation rate was obviously affected by the callus condition after proliferation. The root inducing medium, containing 0.5 mg/L NAA and MS medium with half of macroelement, gained 98% root inducing rate. Conclusien A high frequency genetic regeneration system was established. 展开更多

A full habituated cell line C_ 20hi was screened from 2,4_D dependent line (C_ 20D) of Catharanthus roseus (L.) G. Don. The investigation involved the cell growth, ajmalicine production and enzyme activity related to indole alkaloid biosynthesis in both cell lines. These results indicated that C_ 20hi cells grew faster than C_ 20D cells, and average ajmalicine content in C_ 20hi cells was 18.4 times more than that in C_ 20D when cultured in the production medium. In the growth medium, average ajmalicine content in C_ 20hi cells was 31.9 times more than that in C_ 20D cells, while the cell growth has no obvious difference. The comparison of enzyme activities in C_ 20hi and C_ 20D cells indicated that tryptophan decarboxylase (TDC), strictosidine synthase (SSS) and geraniol_10_dehydrogenase (G10H) activities have no close relation to ajmalicine accumulation, although the activities of these enzymes were higher when cells were cultured in the production medium than in the growth medium. The C_ 20hi cells are relatively stable in five years of culture. 展开更多

In this study, we compared the transcriptomes of Nannochloropsis oceanica cultured in f/2 medium prepared with seawater and freshwater, respectively, aiming to understand the acclimation mechanism of this alga to freshwater. Differentially expressed genes were mainly assigned to the degradation of cell components, ion transportation, and ribosomal biogenesis. These find- ings indicate that the algal cells degrade its components (mainly amino acids and fatty acids) to yield excessive energy (ATP) to maintain cellular ion (mainly K+ and Ca〉) homeostasis, while the depletion of amino acids and ATP, and the reduction of ribosomes attenuate the protein translation and finally slow down the cell growth. 展开更多

In this work, sucrose utilizing microbes from soil were screened to evaluate their ability for accumulation of biopolymer of polyhydroxyalkanoate (PHA). Among 72 isolates were transferred to mineral salt medium (MSM), 33 strains can be grown on sucrose agar medium. However, only one strain showed a strong black color for Sudan Black and gave positive result for Nile blue A. Identification by 16S rDNA nucleotide sequence homology of the isolate showed very closely to Hydrogenophaga sp. (99% identify). To consider PHA production, the isolate was grown in the medium containing sucrose as a sole carbon under controlled conditions of 35 ℃ and at pH 7. Maximum dry cell weight (DCW) and PHA production were obtained at 3.61 g/L and 2.41 g/L after 36 and 42 h batch fermentation. Sucrose uptake measured in term of total organic carbon (TOC) showed at 14.73 g within 48 h. The highest PHA was 68.15% (gPHA/gDCW) giving maximum PHA yield (YP/s) of 0.17 (gPHA/gs ) and a productivity of 0.057 gPHA/L.h. This highlights the potential of microbial resources in soil environment and may be an exploitable application for the industrial production of PHA. 展开更多

Cordyceps ophioglossoides is a valuable traditional medicinal material.We have found that intracellular polysaccharide(IPS) is the major biologically active ingredient in Cordyceps ophioglossoides.This study is the first time to optimize the yield of IPS from Cordyceps ophioglossoides.The optimal medium for IPS production consists of glucose 54.50 g·L·1,yeast powder 25.50 g·L·1,NaH2PO4 0.4 g·L·1 and K2HPO4 0.4 g·L·1.The suggested culture conditions are 24 ℃,initial pH 4.5 with a rotary speed of 120 r·min·1 for 168 h.The yield of IPS is 737.93 mg·L·1,which is 50% higher than the yield under the conditions prior to optimization.The anti-oxidative activities of IPS in Cordyceps ophioglossoides L2 are also characterized using various in vitro assay.The anti-oxidative activity may explain the reason why IPS from Cordyceps ophioglossoides can be used to fight against neurodegenerative dis-eases and menopausal symptoms. 展开更多

The bacteria of Zymomonas mobilis has b... Glutaminase is used industrially to enhance flavor and aroma and to enrich foodstuffs nutritionally. It also has potential for pharmaceutical application as anti-leukemia agent. The bacteria of Zymomonas mobilis has been studied for ethanol production, however, there is no study regarding glutaminase production. The aim of the present study was to establish the influencing factors for the growth of Z mobilis and its relationship with glutaminase production using statistical tools. Analysis of variance in blocks was carried out in a complete block design and the Tukey test demonstrated the importance of the components of the culture medium, absence of agitation and fermentation time. Minimum culture medium was used in the optimization varying the glucose concentrations (10, 30, and 50 g/L), glutamine (0, 0.5 and 1g/L) and culture time (18, 24 and 30 hours). Maximum production was obtained at 8.86 U/L glutaminase. Optimized conditions were used in the growth kinetics, where typical exponential growth was observed. Glutaminase production was shown to be related to biomass production. 展开更多
