A laboratory experiment was set-up, with a semi-arid control soil and the same soil but artificially contaminated with diesel oil, to follow changes in the dominant species of the microbial community in the hydrocarbon-polluted soil via proteomics. Analysis of the proteins extracted from enriched cultures growing in Luria-Bertani (LB) media showed a change in the microbial community. The majority of the proteins were related to gIycolysis pathways, structural or protein synthesis. The results showed a relative increase in the complexity of the soil microbial community with hydrocarbon contamination, especially after 15 days of incubation. Species such as Ralstonia solanacearum, Synechococcus elongatus and different Clostridium sp. were adapted to contamination, not appearing in the control soil, although Bacillus sp. dominated the growing in LB in any of the treatments. We conclude that the identification of microbial species in soil extracts by culture-dependent proteomics is able to partially explain the changes in the diversity of the soil microbial community in hydrocarbon polluted semi-arid soils, but this information is much more limited than that provided by molecular methods.展开更多

METHODS: hALRcDNA clone was obtained from plasmid pGEM-T-hALR, and cDNA was subcloned into the prokatyotic expression vector pGEX-4T-2. The recombinant vector and pGEX-4T-2hALR were identified by enzyme digestion and DNA sequencing and transformed into E coli JM109. The positively selected clone was induced by the expression of GST-hALR fusion protein with IPTG, then the fusion protein was purified by glutathine s-transferase (GST) sepharose 4B affinity chromatography, cleaved by thrombin and the hALR monomer was obtained and detected by measuring H thymidine incorporation. RESULTS: The product of PCR from plasmid pGEM-T- hALR was examined by 1.5% sepharose electrophoresis. The specific strap was coincident with the theoretical one. The sequence was accurate and pGEX-4T-hALP digested by enzymes was coincident with the theoretical one. The sequence was accurate and the fragment was inserted in the positive direction. The recombinant vector was transformed into E coli JM109. SDS-PAGE proved that the induced expressive fusion protein showed a single band with a molecular weight of 41 kDa. The product was purified and cleaved. The molecular weights of GST and hALR were 26 kDa, 15 kDa respectively. The recombinant fusion protein accounted for 31% of the total soluble protein of bacterial lysate. HALR added to the culture medium of adult rat hepatocytes in primary culture and HepG2 cell line could significantly enhance the rate of DNA synthesis compared to the relevant control groups (P 〈 0.01).CONCLUSION: Purified hALR has the ability to stimulate DNA synthesis of adult rat hepatocytes in primary culture and HepG2 cells in vitro, and can provide evidence for its clinical application.展开更多

The test flasks containing the mineral medium, the test oil and the inoculum were placed in incubation together with flasks containing poisoned blanks for periods of 0 and 21 days, respectively. Flasks containing the reference materials in place of the test oil were run in parallel. At the end of the incubation period, the contents of the flasks were subjected to sonic vibration, and were acidified and extracted by using CCI4. The extracts were then analysed by infra-red (IR) spectrometer to measure the maximum absorption of the C-H stretch of CH2-CH3 band at wavelength of 2 930 cm^-1. The absorption values were used to calculate the residual oil contents of the poisoned and test flasks. Consequently the biodegradability of the test oil was calculated. The test results indicate that the differences in the biodegradability of test materials in different tests are within 5.5%, and consistent with the data described in Coordinating European Council (CEC) L-33- A-93. The biodegradability of lubricants can be evaluated by this method effectively.展开更多

Glomerular filtration rate(GFR) remains the ideal marker of kidney function. Unfortunately measuring GFR is time consuming and therefore GFR is usually estimated from equations that take into account endogenous filtration markers like serum creatinine(SCr) and cystatin C(Cys C). Other biomarkers such as albuminuria may precede kidney function decline and have demonstrated to have strong associationswith disease progression and outcomes. New potential biomarkers have arisen with the promise of detecting kidney damage prior to the currently used markers. The aim of this review is to discuss the utility of the GFR estimating equations and biomarkers in CKD and the different clinical settings where these should be applied. The CKD-Epidemiology Collaboration equation performs better than the modification of diet in renal disease equation, especially at GFR above 60 m L/min per 1.73 m2. Equations combining Cys C and SCr perform better than the equations using either Cys C or SCr alone and are recommended in situations where CKD needs to be confirmed. Combining creatinine, Cys C and urine albumin to creatinine ratio improves risk stratification for kidney disease progression and mortality. Kidney injury molecule and neutrophil gelatinase-associated lipocalin are considered reasonable biomarkers in urine and plasma to determine severity and prognosis of CKD.展开更多

In spite of high pressure of anthropogenic activities, the Rohtang Pass still harbours a variety of flowering plants with economic value, including various medicinal herbs. In order to observe species diversity in different landscape elements (LSEs),ground surveys were conducted in nine unique LSEs within the elevation range between 3624 m and 4332 m. Plant community structure in each LSE was studied using stratified random sampling where a total 56 quadrats of 1 m2 in size for herbs and 7 quadrats of 25 m2 for shrubs were made. Of the total 50 plant species belonging to 15 families recorded in different random quadrats,24 species were found of medicinal value. Maximum species richness (18) and value of Shannon diversity (H'=2.2648) were observed on northeast-facing slope in Picrorhiza kurrooa dominated LSE in moist area, followed by Rheum emodi LSE (species richness = 17 and H' = 2.4141) distributed on south-facing slope. Maximum values of species richness and Shannon diversity in Rheum emodi LSE were observed between 8 ~ 12 and 1.4797 ~ 2.1911,respectively. Rhododendron anthopogon dominated LSE on northwest-facing slope was found least diverse in terms of species richness where the Simpson index of dominance (D) was 0.4205. The species were equal in abundance in P. kurrooa LSE on east-facing slope and Pleurospermum candollii LSE on north-facing slope, showing the maximum similarity in terms of species distribution between the two LSEs. Low turnover of species across common LSEs gives an idea regarding their limited distribution. Five species of threatened category according to the IUCN criteria were observed in seven LSEs. The largest population of threatened medicinal plants was recorded on northeast and northwest-facing slopes where population density of Bergenia stracheyi (29.25 individuals/m2),Picrorhiza kurrooa (19.83 individuals/m2) and Rhododendron anthopogon (26.23 individuals/25m2) was highest. Aconitum heterophyllum was recorded for the lowest density (1 individual/m2). It was observed that the distribution of medicinal plants was very habitat-specific. Lower soil pH in most of the LSEs reflected its acidic nature. A significant positive correlation of Shannon diversity was found with soil organic matter, carbon content available and total nitrogen. Considering the continuous pressure of anthropogenic activities, this paper suggests effective ecotourism planning and plant conservation mechanisms to protect valuable plant bioresource at the Rohtang Pass.展开更多

In particular, it is extremely important that the oceans absorb massive amounts of carbon dioxide. This paper takes a pragmatic approach to analyzing the oceans with respect to the causes of discontinuities in oceanic variables of carbon dioxide sinks. We report on an application of chemical, physical and biological methods to analyze the changes of carbon dioxide in oceans. Based on the relationships among the oceans, land, atmosphere and sediment with respect to carbon dioxide, the foundation of carbon dioxide in shell-building and ocean acidification, the changes in carbon dioxide in the oceans and their impact on climate change, and so on, a vital conclusion can be drawn from this study. Specifically, under the condition that the oceans are not disturbed by external forces, the oceans are a large carbon dioxide sink. The result can also be inferred by the formula: C=A-B and G=E+F when the marine ecosystem can keep a natural balance and the amount of carbon dioxide emission is limited within the calrying capacity of the oceans.展开更多

The Hilbert-Huang transform (HHT) method, which is more effective to process non-stationary signal, is proposed to identify the characteristics of GI motility. We decompose the pressure activity data into intrinsic mode functions (IMFs), calculate the Hi/bert marginal spectrum and attain the peristalsis characteristics of GI tract. The IMFs represent the peristalses modes of GI tract activity embedded in the pressure data. The time-varying characteristic of the method suggests that the HHT is suitable to accommodate other non-stationary biomedical data analysis.展开更多

By applying this method, different types of perturbation solutions are obtained. Based on these approximate solutions, the analytical forms of soliton parameters, such as the velocity, the width and the initial position, are carried out and the effects of perturbation on solitons are analyzed at the same time. A numerical simulation of perturbed DNLSE finally verifies the results of the perturbation method.展开更多

After briefly introducing the main controversies arisen on biodiversity monetization and defining biodiversity banking schemes, discussions will be focused on evaluation methods mobilized in the context ofbiodiversity offset banking. Although there is currently no fixed framework assessing environment due resolutely to the specific nature of biodiversity, evaluation methods are analyzed through a review of the academic and empirical literature. This step allows to select few methods justifying these choices and presenting their pros and cons by keeping the aim to contribute to the debates. Thus, ecological assessments (service-to-service and resource-to-resource) and economic valuations (value-to-value and value-to-cost) carried out in biodiversity banking schemes will be distinguished. Finally, this paper will emphasize the inherent differences of the two evaluating forms and their specificities on the one hand, and highlight their opportunities and risks from methodological perspective on the other hand.展开更多

Integrating both epidemiological and molecular approaches, the authors studied: (a) the frequency of the HH main mutations; (b) the association between mutations and HH cases. The majority of patients with HH are homozygous for the C282Y mutation of the HFE gene. A second mutation (H63D) is more widely distributed and its connection with HH isn't clear, but a low penetrance is attributed to this variant. The population-based study consists of three steps: (1) determination of iron biochemical parameters, (2) genetic test, and (3) sequencing of HFE gene and bioinformatics studies. A case report is presented in a 41-year-old male (genotype: H63D/wt) with biochemical and clinical evidences of HH, in absence of secondary iron overload factors. In the cohort of studied patients (150M:62F), there are 18 homozygous patients; H63D/H63D genotype is found in 11 cases. In the heterozygous group, H63D/wt is the predominant genotype (61/68 subjects). All the H63D/wt residents in the same village (Mont.) show altered biochemical parameter levels. In our case study, a substitution localized into the HFE promoter (nt225A 〉 C) is found. Results show that the H63D genotype is responsible for most cases of HH. The peculiar clinical manifestation found in Mont. suggests a founder effect. In our case, the iron overload is related to a presence of an undetected mutation, critical for the transcriptional regulation of the HFE gene.展开更多

The involvement of metabolic enzymes in the resistance strain of cotton aphid to the neonicotinoids was determined by the biochemical biomarkers and the resistance mechanism was determined as CaE. In another study, three different bioassay experiments were designed for detecting the susceptibility of cotton aphid to imidacloprid and thiametoxam and the effect of these two insecticides on the enzymatic activity of cotton aphid was assessed in the adult aphids treated with three different bioassay methods using a modified version of the FAO dip test, residue bioassay procedure and starvation method. Our findings suggested that the type of bioassay methods is very important when aphids' populations assess for the resistance against the neonicotinod insecticides. It has shown the starvation method is the most reliable method compared with other methods.展开更多

The reasons of a primary arterial hypertension occurrence, and also some accompanying it physical and physiological phenomena are considered. The surgical method of treatment of the primary arterial hypertension, connected with denervation of renal arteries is substantiated.展开更多

文摘Pressure activity data as an important index of gastrointestinal (GI) motility can be obtained from the wireless radiotelemetry capsule. The time-varying characteristic of the method suggests that the HHT is suitable to accommodate other non-stationary biomedical data analysis.

文摘We extend Lou's direct perturbation method for solving the nonlinear Schrodinger equation to the case of the derivative nonlinear Schrodinger equation (DNLSE). By applying this method, different types of perturbation solutions are obtained. Based on these approximate solutions, the analytical forms of soliton parameters, such as the velocity, the width and the initial position, are carried out and the effects of perturbation on solitons are analyzed at the same time. A numerical simulation of perturbed DNLSE finally verifies the results of the perturbation method.

文摘A series of novel fluorine-containing acrylates 6a-6g were synthesized via the condensation of ethyl cyanoacetate and trifluoroacetic anhydride, followed by chloridization and the coupling reaction with amines. These new compounds exhibited some biological activity as preliminary bioassay indicated. A plausible reaction mechanism was outlined and discussed.
文摘The assessment of ecological impacts represents a key element of biodiversity offsetting success. Finally, this paper will emphasize the inherent differences of the two evaluating forms and their specificities on the one hand, and highlight their opportunities and risks from methodological perspective on the other hand.
文摘The present study aims to investigate the genotype-phenotype correlation of the hereditary hemochromatosis (HH), a genetic disorder of iron metabolism, in Matera province (Basilicata, Italy). In our case, the iron overload is related to a presence of an undetected mutation, critical for the transcriptional regulation of the HFE gene.
文摘The educational project prepared for students of High School 15-17 years old with the subject of Biodiversity in Minoan Period and Nowadays. It was an interdisciplinary approach of ecological and cultural views of biodiversity. Special educational material was prepared with photos and texts of archaeological excavations and field sampling which was used in real and virtual environment, in t~ discovery teaching of Natural Sciences and History subjects. Students, in a scientific research project with constructive teachings, followed the procedures by which scientists gather, analyze, synthesize, process and evaluate information of organisms of Crete since 4000 years. Crete is located between the natural limits of three continents, having the unique privilege to host permanently or in migration, distinctive species of these three continents. After having the research questions, teachers collected the educational material, organized the work groups, implemented the didactic intervention in the classroom, had the field sampling of plants, recognized the synchronous local and scientific names of the plants, constructed the botany book, photographed the collected organisms and all the concentrated material was delivered to the team members.
文摘In this study, the susceptibility of three populations of cotton aphid, Aph& gossypii Glover (Hemiptera:Aphididae) was assayed to imidacloprid (35SC) and thiametoxam (50WG). It has shown the starvation method is the most reliable method compared with other methods.
文摘It is shown that flow of a blood in a passive elastic tube essentially unstable process accompanying with a flutter of a stream. Biophysical principles of neuroreflex regulation of an arterial bloodstream in cardiovascular system are considered. It is shown that this regulation has dual-purpose character: reduction of the hydraulic resistance and maintenance stable no flutter stream of blood. The reasons of a primary arterial hypertension occurrence, and also some accompanying it physical and physiological phenomena are considered. The surgical method of treatment of the primary arterial hypertension, connected with denervation of renal arteries is substantiated.
