[Methed] Bio-safety disposal and recovering processing of excrements in a duck farm were researched based on technology of configuration, detection, digestion, EM and poultry breeding. [Result] The integrated application technology is quite simple and the cost is not high. During breeding period, excellent organic fertilizers and high-protein forages could be obtained without any antibiotics. Furthermore, secondary wastes and pollution would not occur. In addition, ammonia was lower in excrements processed with earthworm and air pollution was reduced; the produced humus provided organic fertilizers and improved barren soils. [Conclusion] The research provides references of multi-technology integration for related industries.展开更多

METHODS: Five methods of extracting bacterial DNA, including Tris-EDTA buffer, chelex-100, ultrapure water, 2% sodium dodecyl sulfate and 10% Triton-100 with and without sonication, were compared with the commercial fecal DNA extraction kit method, which is considered as the gold standard for DNA extraction. The comparison was based on the yield and purity of DNA and the indexes of the structure and property of micro-organisms that were reflected by ERIC-PCR. RESULTS: The yield and purity of DNA obtained by the chelex method was similar to that obtained with the fecal DNA kit. The ERIC-PCR results obtained for the DNA extracted by the chelex method and those obtained for DNA extracted with the fecal DNA kit were basically the same.CONCLUSION: The chelex method is recommended for ERIC-PCR experiments in view of its simplicity and cost- effectiveness; and it is suitable for extracting total DNA from intestinal micro-organisms, particularly for handling a large number of samples.展开更多

We present a new edge recognition technology based on the normalized vertical derivative of the total horizontal derivative which has the functions of both edge detection and enhancement techniques. First, we calculate the total horizontal derivative (THDR) of the potential-field data and then compute the n-order vertical derivative (VDRn) of the THDR. For the n-order vertical derivative, the peak value of total horizontal derivative (PTHDR) is obtained using a threshold value greater than 0. This PTHDR can be used for edge detection. Second, the PTHDR value is divided by the total horizontal derivative and normalized by the maximum value. Finally, we used different kinds of numerical models to verify the effectiveness and reliability of the new edge recognition technology.展开更多
Culturable thermophilic microorganisms were enriched from samples collected from Lau Basin hydrothermal vents in artificial seawater medium at 45 ℃ and pH 7.0. Microbial diversities of the enriched communities were d...Culturable thermophilic microorganisms were enriched from samples collected from Lau Basin hydrothermal vents in artificial seawater medium at 45 ℃ and pH 7.0. Microbial diversities of the enriched communities were defined by performing a restriction fragment length polymorphism (RFLP) analysis of 16S rRNA gene sequences with enzymes MspI and Hin6 I. A total of 14 phylotypes have been detected by the RFLP patterns identified for 16S rRNA clone libraries of the enrichment. Analysis of sequences showed that at least four bacterial divisions presented in the clones libraries. The phyla Proteobacteria and Firmicutes were the most dominant groups. The majority of the sequences included in this analysis affiliated with Gamma Proteobacteria (71%) and Bacillus (23%). Scanning electron micrographs revealed that there were abundant rod and coceoidal forms encased in sulphur and sodium chloride precipitate. These results revealed that there were a diversity of moderate thermophilic bacterial populations thrived in Lau Basin hydrothermal vents that were previously not detected by either molecular retrieval or strain purification techniques.展开更多

The evolutionary relationships of S. sclerotiorum and other fungi in atom gene were studied. Results showed that the atom gene from of S. sclerotiorum has a single open reading frame of 4 773 bp and does not include any introns. The derived amino acid sequence consists of 1 590 residues, and it is homologous to all fungal AROM proteins studied so far. The theoretical isoelectric point (pl) and molecular weight (Mw) is 6.5 and 172.66 kD, respectively GC percentage of the arom gene is 44.94. According to the results of searching from CDD and Prosite database, AROM protein of S. sclerotiorum contains five conserve domains: 3-dehydroquinate synthase domain, 3-dehydroquinate dehydratase (3-dehydroquinase) domain, shikimate 5-dehydrogenase domain, shikimate kinase domain, and -enolpyruvylshikimate-3-phosphate synthase (EPSP sythase) domain, and four motifs: two EPSP synthase signatures, dehydroquinase class I active site, shikimate kinase signature. According to the PIR Site Rule PIRSR000514-1, four functionally important amino acid residues are found by alignment. Putative TATA box and CAAT box locate separately in -23 and -77 loci in 5' un-translated region, and two loci found in downstream atom gene are likely polyadenylation signals. Leaf edges show chlorosis starting from the t...Experimental trail conducted in Jasenica, Bosnia and Herzegovina have showed apricot (Prunus persica L) leaf injures especially on the edges in the middle of the season. Leaf edges show chlorosis starting from the tip due to lack of Chlorophyll. Affected leaves start falling and renew leaves masse as respond to stress conditions which also delay fruits maturation. Technological development in detecting the stress of high plants trough the leaf fluorescence and Chlorophyll concentration have led to the introduction of instruments which utilize fluorescence as a basis for determining stress level. The paper was aimed to show whether the productivity values calculated from Chlorophyll fluorescence Fv/Fm were comparable to those produced by spectrophotometric method in detecting Chlorophyll a concentration and SPAD values detected by Chlorophyll meter (SPAD-502). Measurements of nitrogen content were also provided in order to obtain accuracy of SPAD meter since Chlorophyll molecules contain nitrogen in their structure. To test the relationship between these three technique regression relationship was obtained. Regression coefficient between Fv/Fm values and Chlorophyll a concentration mg g-t was high. Also, strong regression coefficient was observed between Chlorophyll a and SPAD values indicating a good accuracy of this device which was also confirmed by good regression between nitrogen content and SPAD values. Fluorometer measurements on injured leaves samples of apricot have also reviled the Fv/Fm values below 0.83 which is in according with numerous authors who considered a line for indicating stress factors.展开更多
