Results A new compound named herpetin was isolated, whose structure was determined to be 3-benzofuran methanol-2, 3-dihydm-2-(4- hydroxy-3-methoxyphenyl )-4-methoxy-6-[ tetrahydm-2-( 3-hydroxy-4-methoxyphenyl )-3-methanol ]-2- furanmethyl, showed significant inhibitory effects on HBV-DNA and the replication and expression of HBsAg and HBeAg. METHODS: From 42 IHS subjects and 21 controls, serum and biliary biochemical constituents, including total bilirubin (TBIL), direct bilirubin (DBIL), alanine aminotransferase (ALT), gamma-glutamyl transpeptidase (γ-GT), total bile acid (TBA), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) both in bile and serum, were assayed. The subjects with IHS were divided into a cholestasis group (n = 21) and a hepatitis group (n = 21). RESULTS: In the cholestasis group, serum TBIL, DBIL, ALT, γ-GT, TBA, IL-6 and TNF-α levels were higher than those in the control (P < 0.01); and also the biliary TBIL, DBIL, γ-GT and TBA levels were lower than those in the control, whereas biliary IL-6 and TNF-α levels were higher than those in the control (P < 0.01). In the cholestasis group, serum IL-6 and TNF-α levels were lower than those in bile (P < 0.01). In the hepatitis group, serum DBIL, ALT, γ-GT, TBA, IL-6 and TNF-α levels were higher than those in the control (P < 0.01 or 140.57 ± 70.32 vs 79.06 ± 35.25, P < 0.05), while biliary TBIL, DBIL, γ-GT and TBA levels were lower than those in the control (P < 0.01), and biliary IL-6 and TNF-α levels were higher than those in the control (P < 0.01). In the hepatitis group, serum IL-6 and TNF-α levels were also lower than those in bile (P < 0.01). Serum TBIL, DBIL, γ-GT, IL-6 and TNF-α levels in the cholestasis group were higher than those in the hepatitis group, while biliary IL-6 and TNF-α levels in the cholestasis group were higher than those in the hepatitisgroup. Biliary IL-6 and TNF-α were found to be more significantly increased than serum IL-6 and TNF-α in IHS (P < 0.01). The biliary IL-6 and TNF-α levels were positively correlated with serum DBIL, TBA and γ-GT levels in IHS subjects. CONCLUSION: Biliary biochemical constituents alter in coincidence with pathological changes in hepatocellular injury. Cholestasis is more serious in IHS patients of cholestasis subtype. Assay of biliary IL-6 and TNF-α levels can be specific and sensitive to determine the inflammatory status of impaired liver in IHS.展开更多

METHODS: Mice were administered the biologically active fraction of N J...AIM: To determine if the fraction of Nardostachysjata- mansi (N J) has the potential to ameliorate the severity of acute pancreatitis (AP). METHODS: Mice were administered the biologically active fraction of N J, i.e., the 4th fraction (N J4), intra- peritoneally, and then injected with the stable chole- cystokinin analogue cerulein hourly for 6 h. Six hours after the last cerulein injection, the pancreas, lung, and blood were harvested for morphological examination,measurement of cytokine expression, and examination of neutrophil infiltration. RESULTS: N J4 administration attenuated the sever- ity of AP and lung injury associated with AP. It also reduced cytokine production and neutrophil infiltration and resulted in the in vivo up-regulation of heine oxy- genase-1 (HO-1). Furthermore, NJ4 and its biologically active fraction, N J4-2 inhibited the cerulein-induced death of acinar cells by inducing HO-1 in isolated pan- creatic acinar cells. CONCLUSION: These results suggest that N J4 may be a candidate fraction offering protection in AP and N J4 might ameliorate the severity of pancreatitis by induc- ing HO-1 expression.展开更多

BRI1, a protein localized to the plasma membrane, functions as a BR receptor and it has been proposed that its kinase activity has an essential role in BR-regulated plant growth and development. Here we report the isolation and molecular characterization of a new allele of bril, bril-301, which shows moderate morphological phenotypes and a reduced response to BRs under normal growth conditions. Sequence analysis identified a two-base alteration from GG to AT, resulting in a conversion of 989G to 9891 in the BRI1 kinase domain. An in vitro assay of kinase activity showed that bril-301 has no detectable autophosphorylation activity or phosphorylation activity towards the BRI1 substrates TTL and BAK1. Furthermore, our results suggest that bril-301, even with extremely impaired kinase activity, still retains partial function in regulating plant growth and development, which raises the question of whether BRI1 kinase activity is essential for BR-mediated growth and development in higher plants.展开更多
In order to investigate the effects of microorganisms and their urease activities in macrophytic root zones on pollutant removal, four small-scale plots (SSPs) of vertical/reverse-vertical flow wetlands were set up to...In order to investigate the effects of microorganisms and their urease activities in macrophytic root zones on pollutant removal, four small-scale plots (SSPs) of vertical/reverse-vertical flow wetlands were set up to determine: a) the relationship between the abundance of microorganisms in the root zones and water purification efficiency; and b) the relationship between urease activities in the root zones and pollutant removal in a constructed wetland system. Total numbers of the microbial population (bacteria, fungi, and actinomyces) along with urease activities in the macrophytic root zones were determined. In addition, the relationships between microbial populations and urease activities as well as the wastewater purification efficiencies of total phosphorus (TP), total Kjeldahl nitrogen (TKN), biochemical oxygen demand in 5 days (BOD5), and chemical oxygen demand (COD) were also analyzed. The results showed that there was a highly significant positive correlation (r = 0.9772, P < 0.01) between the number of bacteria in the root zones and BOD5 removal efficiency and a significant negative correlation (r = -0.9092, P < 0.05) between the number of fungi and the removal efficiency of TKN. Meanwhile, there was a significant positive correlation (r -- 0.8830, P < 0.05) between urease activities in the root zones and the removal efficiency of TKN. Thus, during wastewater treatment in a constructed wetland system,microorganism and urease activities in the root zones were very important factors.展开更多
An inter-laboratory comparison of the AOAC mouse bioassay for paralytic shellfish poisoning (PSP) toxicity in shellfish was carried out among 25 Chinese laboratories to examine the overall performance for PSP testing ...An inter-laboratory comparison of the AOAC mouse bioassay for paralytic shellfish poisoning (PSP) toxicity in shellfish was carried out among 25 Chinese laboratories to examine the overall performance for PSP testing in China, and to analyze the main factors affecting the performance of this method. The toxic scallop Patinopecten yessoensis collected from coast of Bohai Sea, China, was used as a test sample in the comparison study. The results were reported and evaluated using robust statistical methods. The z scores showed that 80%, 8%, and 12% of laboratories reported satisfactory results, unsatisfactory results, and questionable results, respectively. This evaluation demonstrates that the PSP mouse bioassay is an appropriate method for screening and testing PSP toxicity in shellfish. However, it was found that the experience and skill of technicians, as well as the body weight and health status of mice being used significantly affected the accuracy of the method.展开更多

Methods: A constructed Tetl yeast expression system was used to investigate the immunological function of orally administered Tα1. Dried yeast containing three different concentration of Tα1 was fed to normal Balb/c mice and other Balb/c mice whose immunities were inhibited in advance by cyclophosphamide. Synthesized Tα1 peptide was used as positive control and dried yeast with empty plasmid was used as negative control. CD4^+ and CD8^+ levels were detected by flow cytometry assay. TNF-α, IFN-γ, IL-2, IL-6 and IL-10 levels were detected by liquid chip. Results: In normal Balb/c mice or immune inhibition Balb/c mice, CD8^+ levels were significantly increased. Especially in immune inhibition Balb/c mice, CD8^+ levels in synthesized Tα1 group (18.77%±4.72%), small dose group (13.48%±6.17%) and large dose group (22.74%±1.09%) were significantly higher than that in empty yeast control group (7.49%±2.14%). Conclusion: Orally administered Tα1 has its certain immunomodulatory function.展开更多

We utilized a unique culture system to analyze the expression patterns of gene, protein, and cell surface antigen, and the biological process of the related genes in erythroid and myeloid differentiation and switching...We utilized a unique culture system to analyze the expression patterns of gene, protein, and cell surface antigen, and the biological process of the related genes in erythroid and myeloid differentiation and switching of hematopoietic stem cells (HSCs) in response to cytokine alterations. Gene-specific fragments (266) identified from five populations of cytokine-stimulated HSCs were categorized into three groups: (1) expressed specifically in a single cell population; (2) expressed in two cell populations, and (3) expressed in three or more populations. Of 145 defined cDNAs, three (2%) were novel genes. Protein two-dimensional gel electrophoresis and flow cytometry analyses showed overlapped and distinguished protein expression profiles in the cell populations studied. Biological process mapping of mRNAs expressed in erythroid and myeloid lineages indicated that mRNAs shared by both lineages attended 'core processes,' whereas genes specifically expressed in either lineage alone were related to specific processes or cellular maturation. Data from this study support the hypothesis that committed HSCs (El4 or G14) cells can still be redirected to develop into myeloid or erythroid cells when erythropoietin (EPO) is replaced with granulocyte-colony stimulating factor (G-CSF) under erythroid-cultured condition or G-CSF with EPO in myeloid-cultured environment, respectively. Our results suggest that genes or proteins co-expressed in erythroid and myeloid lineages may be essential for the lineage maintenance and switching in hematopoiesis.展开更多

Yamaz.) from the mountain of Evota top in South Yakutia are shown. Extracts of these plants was analyzed by method of planar chromatography in thin layers using different specific detectors to exposure of some groups of biologically active matters. During the researches, it was established that polyphenol compounds of different degree of condensment prevail in rhododendron composition; catechines, flavonoids, coumarins, saponins, essential oils, phenol carboxylic acids and arbutin are present too. Due to detected features, all the researched species of rhododendrons are perspective for practical using in food industry and as adaptogenes for maintenance of people health in the conditions of north.展开更多

Purity of sesamin and sesamolin can reach 99.2% and 99.9 %, and they can be recovered by 99.9% and 99.4%, r...In this work, sesamin and sesamolin is separated and purified by SMB (simulated moving bed) chromatography. Purity of sesamin and sesamolin can reach 99.2% and 99.9 %, and they can be recovered by 99.9% and 99.4%, respectively. Concentrations of the extract and raffinate was monitored and periodical change of the concentration was observed on experiment. Effects of non-linear adsorption, dead volume, and temperature on the separation are reviewed by the triangle theory to explain the periodical change of concentration. It is found that dead volume has little effect, and temperature fluctuation significantly affects thc separation by SMB. The temperature seriously affects the robust operation of SMB due to the change of Henry's constant, which will change the selectivity and restrain the productivity. Although the productivity in this study is only 0.0416 g/L-hr, the cost of the stationary phase used in this study is much lower than that of packed in analytical column. By using this stationary phase, the authors are still able to separate the mixture of sesamin and sesmolin. This will economically attract attention for the application of SMB to purify bioactive compounds on developing botanical drugs.展开更多

Due to the importance of biological safety in drinking water quality and the disadvantages which exist in traditional methods of detecting typical microorganisms such as Cryptosporidium and Giardia, it is necessary to...Due to the importance of biological safety in drinking water quality and the disadvantages which exist in traditional methods of detecting typical microorganisms such as Cryptosporidium and Giardia, it is necessary to develop an alternative. Particle counts is a qualitative measurement of the amount of dissolved solids in water. The removal rate of particle counts was previously used as an indicator of the effectiveness of a biological activated carbon (BAC) filter in removing Cryptosporidium and Giardia. The particle counts in a BAC filter effluent over one operational period and the effects of BAC filter construction and operational parameters were investigated with a 10 m3/h pilot plant. The results indicated that the maximum particle count in backwash remnant water was as high as 1296 count/ml and it needed about 1.5 h to reduce from the maximum to less than 50 count/ml. During the standard filtration period, particle counts stay constant at less than 50 count/ml for 5 d except when influ- enced by sand filter backwash remnant water. The removal rates of particle counts in the BAC filter are related to characteristics of the carbon. For example, a columned carbon and a sand bed removed 33.3% and 8.5% of particles, respectively, while the particle counts in effluent from a cracked BAC filter was higher than that of the influent. There is no significant difference among particle removal rates with different filtration rates. High post-ozone dosage (>2 mg/L) plays an important role in particle count removal; when the dosage was 3 mg/L, the removal rates by carbon layers and sand beds decreased by 17.5% and increased by 9.5%, respectively, compared with a 2 mg/L dosage.展开更多

The results showed that there was a highly significant positive correlation (r = 0.9772, P < 0.01) between the number of bacteria in the root zones and BOD5 removal efficiency and a significant negative correlation (r = -0.9092, P < 0.05) between the number of fungi and the removal efficiency of TKN. Meanwhile, there was a significant positive correlation (r -- 0.8830, P < 0.05) between urease activities in the root zones and the removal efficiency of TKN. Thus, during wastewater treatment in a constructed wetland system,microorganism and urease activities in the root zones were very important factors.

文摘An inter-laboratory comparison of the AOAC mouse bioassay for paralytic shellfish poisoning (PSP) toxicity in shellfish was carried out among 25 Chinese laboratories to examine the overall performance for PSP testing in China, and to analyze the main factors affecting the performance of this method. The toxic scallop Patinopecten yessoensis collected from coast of Bohai Sea, China, was used as a test sample in the comparison study. The results were reported and evaluated using robust statistical methods. The z scores showed that 80%, 8%, and 12% of laboratories reported satisfactory results, unsatisfactory results, and questionable results, respectively. This evaluation demonstrates that the PSP mouse bioassay is an appropriate method for screening and testing PSP toxicity in shellfish. However, it was found that the experience and skill of technicians, as well as the body weight and health status of mice being used significantly affected the accuracy of the method.

文摘Objective: To investigate the immunological function of a yeast expression system for thymosin α1 (Tα1). Methods: A constructed Tetl yeast expression system was used to investigate the immunological function of orally administered Tα1. Dried yeast containing three different concentration of Tα1 was fed to normal Balb/c mice and other Balb/c mice whose immunities were inhibited in advance by cyclophosphamide. Synthesized Tα1 peptide was used as positive control and dried yeast with empty plasmid was used as negative control. CD4^+ and CD8^+ levels were detected by flow cytometry assay. TNF-α, IFN-γ, IL-2, IL-6 and IL-10 levels were detected by liquid chip. Results: In normal Balb/c mice or immune inhibition Balb/c mice, CD8^+ levels were significantly increased. Especially in immune inhibition Balb/c mice, CD8^+ levels in synthesized Tα1 group (18.77%±4.72%), small dose group (13.48%±6.17%) and large dose group (22.74%±1.09%) were significantly higher than that in empty yeast control group (7.49%±2.14%). Conclusion: Orally administered Tα1 has its certain immunomodulatory function.

文摘After comparing the operating status of other catalytic reforming units and evaluation of the side-cut stream tests, the refinery investigated the influence of the feedstock property, clay types, and operating regime of the clay tower and catalytic reforming unit on the service life of the clay. Test results had revealed that the low potential aromatic content of the reformer feed and high operating severity were the critical causes leading to high bromine number of the C8^+ aromatics feed for the PX unit and the shortened service life of clay. This article also puts forward the corresponding remedial measures.
文摘We utilized a unique culture system to analyze the expression patterns of gene, protein, and cell surface antigen, and the biological process of the related genes in erythroid and myeloid differentiation and switching of hematopoietic stem cells (HSCs) in response to cytokine alterations. Gene-specific fragments (266) identified from five populations of cytokine-stimulated HSCs were categorized into three groups: (1) expressed specifically in a single cell population; (2) expressed in two cell populations, and (3) expressed in three or more populations. Of 145 defined cDNAs, three (2%) were novel genes. Protein two-dimensional gel electrophoresis and flow cytometry analyses showed overlapped and distinguished protein expression profiles in the cell populations studied. Biological process mapping of mRNAs expressed in erythroid and myeloid lineages indicated that mRNAs shared by both lineages attended 'core processes,' whereas genes specifically expressed in either lineage alone were related to specific processes or cellular maturation. Data from this study support the hypothesis that committed HSCs (El4 or G14) cells can still be redirected to develop into myeloid or erythroid cells when erythropoietin (EPO) is replaced with granulocyte-colony stimulating factor (G-CSF) under erythroid-cultured condition or G-CSF with EPO in myeloid-cultured environment, respectively. Our results suggest that genes or proteins co-expressed in erythroid and myeloid lineages may be essential for the lineage maintenance and switching in hematopoiesis.
文摘The data on chemical compounds of three Rododendron species (Rh. adamsii Rehd., Rh. aureum Georgi. and Rh. lapponicum subsp, parvifolium (Adams) T. Yamaz.) from the mountain of Evota top in South Yakutia are shown. Extracts of these plants was analyzed by method of planar chromatography in thin layers using different specific detectors to exposure of some groups of biologically active matters. During the researches, it was established that polyphenol compounds of different degree of condensment prevail in rhododendron composition; catechines, flavonoids, coumarins, saponins, essential oils, phenol carboxylic acids and arbutin are present too. Due to detected features, all the researched species of rhododendrons are perspective for practical using in food industry and as adaptogenes for maintenance of people health in the conditions of north.
文摘In this work, sesamin and sesamolin is separated and purified by SMB (simulated moving bed) chromatography. Purity of sesamin and sesamolin can reach 99.2% and 99.9 %, and they can be recovered by 99.9% and 99.4%, respectively. Concentrations of the extract and raffinate was monitored and periodical change of the concentration was observed on experiment. Effects of non-linear adsorption, dead volume, and temperature on the separation are reviewed by the triangle theory to explain the periodical change of concentration. It is found that dead volume has little effect, and temperature fluctuation significantly affects thc separation by SMB. The temperature seriously affects the robust operation of SMB due to the change of Henry's constant, which will change the selectivity and restrain the productivity. Although the productivity in this study is only 0.0416 g/L-hr, the cost of the stationary phase used in this study is much lower than that of packed in analytical column. By using this stationary phase, the authors are still able to separate the mixture of sesamin and sesmolin. This will economically attract attention for the application of SMB to purify bioactive compounds on developing botanical drugs.

The aim of this work was to present bioactive compounds obtained from the silk fibre and isolated from the body of this insect, which may be used in medical and pharmacological applications. The most important are bioactive proteins. However, the juvenile stages of mulberry silkworm possess other very valuable active substances.

文摘Due to the importance of biological safety in drinking water quality and the disadvantages which exist in traditional methods of detecting typical microorganisms such as Cryptosporidium and Giardia, it is necessary to develop an alternative. Particle counts is a qualitative measurement of the amount of dissolved solids in water. The removal rate of particle counts was previously used as an indicator of the effectiveness of a biological activated carbon (BAC) filter in removing Cryptosporidium and Giardia. The particle counts in a BAC filter effluent over one operational period and the effects of BAC filter construction and operational parameters were investigated with a 10 m3/h pilot plant. The results indicated that the maximum particle count in backwash remnant water was as high as 1296 count/ml and it needed about 1.5 h to reduce from the maximum to less than 50 count/ml. During the standard filtration period, particle counts stay constant at less than 50 count/ml for 5 d except when influ- enced by sand filter backwash remnant water. The removal rates of particle counts in the BAC filter are related to characteristics of the carbon. For example, a columned carbon and a sand bed removed 33.3% and 8.5% of particles, respectively, while the particle counts in effluent from a cracked BAC filter was higher than that of the influent. There is no significant difference among particle removal rates with different filtration rates. High post-ozone dosage (>2 mg/L) plays an important role in particle count removal; when the dosage was 3 mg/L, the removal rates by carbon layers and sand beds decreased by 17.5% and increased by 9.5%, respectively, compared with a 2 mg/L dosage.
