应用免疫组织化学方法研究甲苯四氢吡啶(MPTP)对单胺氧化酶B的作用。单胺氧化酶B被认为可使MPTP转化为甲苯吡啶(MPP+ ),而MPP+ 是损毁黑质致密部神经元的细胞毒性物质。在猫腹腔注射MPTP导致选择性的抑制中... 应用免疫组织化学方法研究甲苯四氢吡啶(MPTP)对单胺氧化酶B的作用。单胺氧化酶B被认为可使MPTP转化为甲苯吡啶(MPP+ ),而MPP+ 是损毁黑质致密部神经元的细胞毒性物质。在猫腹腔注射MPTP导致选择性的抑制中缝核神经元内的单胺氧化酶B免疫反应,分布显示中缝吻侧完全被抑制,而尾侧仅存少量免疫染色非常弱的神经元。但是MPTP对下丘脑及各脑区星状胶质细胞内的单胺氧化酶B无任何作用。这些作用伴随黑质多巴胺能神经元的丢失,但中缝核的细胞数量和形态以及5-羟色胺(5-HT)免疫组织化学反应与对照组相同。根据上述结果我们认为,MPTP抑制中缝核内单胺氧化酶B,但不破坏5-HT能神经元,并且中缝核内单胺氧化酶B在MPTP导致黑质损伤的过程中不起关键作用。 展开更多

Neuroprotective Effects of Modafinil on MPTP Mouse Model of Parkinson′s Disease

Aim To observe the neuroprotective effects of modafinil on the Parkinson'sdisease ( PD ) model induced by 1-methyl-4-phenyl-1, 2,3, 6-tetrahydropyridine (MPTP ). Methods Themodel of PD was induced by intraperitoneal injection of MPTP into C57BL/6J mice for 4 d. Modafinil(ip, 50 or 100 mg·kg^(-1)·d^(-1)) was administered following MPTP for 4 d and for another 10 dconsecatirely. The effects of modafinil on the locomotor activity, and the incubation, maintenanceperiod and grade of the tremor, the duration of the climbing rod of mouse, and the distribution ofpositive cells of ty-rosine hydroxylase (TH) and Nissl bodies in the striatum and substantia nigra(SN) were observed. The contents of dopam-ine (DA) , noradrenaline (NA) and 5-hydroxytryptamine(5-HT) in the striatum were determined. Results Modafinil (50 and 100 mg·kg^(-1)) significantlyprevented the locomotor, the tremor and climbing rod defect behavior in a dose-dependent manner (P <0.05 and P < 0.01, n = 10), prevented the decrease in the number of TH-positive cells and Nisslbodies (P<0.05, n=10), and reduced the decrease of DA, NA, and 5-HT in the striatum (P < 0.05, n =10) induced by MPTP. Conclusion Modafinil improves the behavioral deficits and prevents themonoaminergic neuron lesion in seriously impaired MPTP mouse model. 展开更多

The title compound (C20H15ClN4O, Mr = 362.81) was synthesized by the reaction of ethyl 2-cyano-3-(2-chlorophenyl)-1-acylate with 5-amino-3-methyl-1-phenylpyrazole under microwave irradiation and its structure was determined by single-crystal X-ray diffraction. The crystal is of monoclinic, space group P21/n with a = 8.8192(19), b = 15.976(3), c = 13.094(3) ? = 103.074(4), V = 1797.1(7) ?, Z = 4, Dc = 1.341 g/cm3, = 0.229 mm-1, F(000) = 752, R = 0.0632 and wR = 0.1278. X-ray analysis reveals that the pyridine ring adopts an envelope conformation. The dihedral angle between the pyrazole and pyridine planes is 2.19, and inter- molecular hydrogen bonds are also observed in the molecule. 展开更多

Objective To study the apoptotic effects of 1 methyl 4 phenyl 1,2,3,6 tetrahydropyridine (MPTP) on the nigral dopaminergic neurons of mice and 1 methyl 4 phenylpyridium ion (MPP +) on pheochromocytoma (PC12) cells, as well as the antagonism of Eldepryl against MPTP's apoptotic effect Methods Three groups of C 57 BL mice were treated with MPTP, Eldepryl plus MPTP and normal saline, respectively, for 7 days before performing TUNEL (terminal deoxyneucleotidyl transferase mediated dUTP x nick end labeling) and FACS (fluorescence activated cell sorting) analyses of neuronal apoptosis in the substantia nigra The same tests were employed in cell culture to examine apoptosis in PC12 cells treated with MPP +, MPTP or PBS Results Intraperitoneal administration of MPTP 30?mg/kg could induce nigral apoptosis, and oral use of Eldepryl prior to MPTP treatment could completely prevent the nigral apoptosis caused by MPTP MPP +, an intermediate metabolite of MPTP, could lead to the apoptosis of PC12 cells, whereas MPTP itself had no such effect on PC12 cells Conclusions The experiment indicated that the neurotoxin, MPTP, might cause the death of nigral neurons through a mechanism of apoptosis and this effect might be mediated by its bioactive intermediate metabolite MPP + Eldepryl could protect the neurotoxicity from MPTP 展开更多

As a widely used traditional Chinese medicine (TCM), Swertia punicea Hemsl has exhibited effects on anti-hepatitis B virus (HBV), liver protection, hypoglycemic activity and cholecystitis. In this study, we confirmed that xanthone extract from Swertia punicea Hemsl (XSPH) improved the motor deficit, increased the levels of striatal dopamine (DA) and homovanilic acid (HVA), and alleviated the loss of tyrosine hydroxylase (TH)-positive neurons located in substantia nigra pars compacta (SNpc) in MPTP-induced mouse model of Parkinson's disease (PD). In conclusion, the present results indicated that XSPH offered neuroprotective effects against the neurotoxicity of MPTP and it might be a potential treatment for PD. 展开更多
