We therefore expected impairments in saccade perf...Neurodegeneration in Parkinson’s disease dementia (PDD) and dementia with Lew y bodies (DLB) affect cortical and subcortical networks involved in saccade gene ration. We therefore expected impairments in saccade performance in both disorde rs. In order to improve the pathophysiological understanding and to investigate the usefulness of saccades for differential diagnosis, saccades were tested in a ge-and education-matched patients with PDD (n=20) and DLB (n=20), Alzheimer’s disease (n=22) and Parkinson’s disease (n=24), and controls (n=24). Reflexive (gap, overlap) and complex saccades (prediction, decision and antisaccade) were tested with electrooculography. PDD and DLB patients had similar impairment in a ll tasks (P > 0.05, not signifi cant). Compared with controls, they were impaire d in both reflexive saccade execution (gap and overlap latencies, P < 0.0001; ga ins, P < 0.004) and complex saccade performance (target prediction, P < 0.0001; error decisions, P < 0.003; error antisaccades: P < 0.0001). Patients with Alzhe imer’s disease were only impaired in complex saccade performance (Alzheimer’s disease versus controls, target prediction P < 0.001, error decisions P < 0.0001 , error antisaccades P < 0.0001), but not reflexive saccade execution (for all, P >0.05). Patients with Parkinson’s disease had, compared with controls, similar complex saccade performance (for all, P > 0.05) and only minimal impairment in reflexive tasks, i.e. hypometric gain in the gap task (P=0.04). Impaired saccade execution in re flexive tasks allowed discrimination between DLB versus Alzheimer’s disease (se nsitivity ≥60%, specificity ≥77%)-and between PDD versus Parkinson’s disea se (sensitivity ≥60%, specificity ≥88%) when ±1.5 standard deviations was u sed for group discrimination. We conclude that impairments in reflexive saccades may be helpful for differential diagnosis and are minimal when either cortical (Alzheimer’s disease) or nigrostriatal neurodegeneration (Parkinson’s disease) exists solely; however, they become prominent with combined cortical and subcor tical neurodegeneration in PDD and DLB. The similarities in saccade performance in PDD and DLB underline the overlap between these conditions and underscore dif ferences from Alzheimer’s disease and Parkinson’s disease.展开更多

[Result] The multiplex RT-PCR amplification, agarose gel electrophoresis and sequencing results showed that the self-designed primers are successful for detecting AIV. In mid-June, the wilting rate of L. lancifolium in seriously-infected field was even up to 50%-70%. In some fields, the shoots of L. lancifolium even al wilted. The pathogen was isolated from the infected leaves of L. lancifolium. Its pathogenici-ty, spore morphology, 18S rDNA sequence and biological characteristics were stud-ied. The results showed the isolated pathogen was Alternaria alternate. The lethal temperature of mycelial growth was 55 ℃. The optimum pH was 6-7. Among the tested carbon sources and nitrogen sources, the optimum carbon source was mal-tose, and the optimum nitrogen source was yeast extract.展开更多

[Method] Twenty-five C. gloeosporioides strains were isolated and identified from the diseased samples...[Objective] This study aimed to explore the biological characteristics of Col etotrichum gloeosporioides in pears. [Method] Twenty-five C. gloeosporioides strains were isolated and identified from the diseased samples. Their pathogenicity was identified by inoculating the surface of punctured pears with fungal discs. The effects of different temperatures, pH values, carbon sources and nitrogen sources on the growth of C. gloeosporioides mycelia were explored by incubating fungal discs on the center of plates. [Result] Among the twenty-five C. gloeosporioides strains, three had strong pathogenicity, and eighteen had intermediate pathogenicity, and four strains had weak pathogenicity. Those highly-pathogenic strains had darker colonies, with dense mycelia, whereas those lowly-pathogenic ones had white colonies, with sparse mycelia. Those with fast-growing colonies showed strong pathogenicity, while those with slowly-growing colonies displayed weak pathogenicity. There was no relationship between conidia yield and pathogenicity. The optimum temperature for the growth of C. gloeosporioides mycelia was 25-30 ℃, and the optimum pH was 5.0-7.0. C. gloeosporioides could make use of various carbon sources (monosaccharide and disaccharide), inorganic and organic nitrogen sources, and the optimal carbon source and nitrogen source were sucrose and beef extract, respectively. [Conclusion] Our study benefits further understanding of C. gloeospori-oides and helps to control pear anthracnose more effectively.展开更多

METHODS The expression of VEGF in 128 gastric carcinomas was investigated by immunohistochemical staining with a polyclonal antibody against VEGF. Cor- relations between the expression of VEGF and various clinicopathologic factors and prognosis were studied. RESULTS The VEGF-rich expression rate was 64.1% in gastric carcinoma. VEGF-rich expression rate of patients with stage Ⅲ and stage Ⅳ disease was greater than that of patients with stage f disease (P <0.05). Significant differences of expression rate ex- isted with respect to growth pattern,serosal invasion and lymph node metastasis. The VEGF-rich rate was much higher in tumors with expanding growth pattern (71.8%) or serosal invasion (73.5%) than in those with infiltrative growth pattern (52.0%) or non-serosal invasion (53.3%) (P<0.025,respectively),and it was also significantly higher in patients with lymph node metastases (75.0%) than in those without such metastases (50.0%) (P<0.05). In addition,postop- erative survey of 86 patients who had been followed up for at least 5 years demonstrated that the 5-year sur- vival rate of patients with VEGF-rich tumors was signifi- cantly lower than that of patients with VEGF-poor tu- mors (P<0.05). CONCLUSIONS The expression of VEGF may be as- sociated with the invasion and metastasis and may also be a useful prognostic indicator of gastric carcinoma.展开更多

[Method]The hemolytic bacteria were isolated from the hepatopancreas of Babylonia suffered from proboscis edema with blood agar plate. The dominant bacterial community in the ill Babylonia was identified by 16 S r DNA sequence analysis, and the bacterial cultural and biochemical characteristics and pathogenicities were studied. [Result]The Shewanella bacteria, including Shewanella algae and Shewanella abalone, are the dominant bacterial community in Babylonia suffered from proboscis edema.The colony characteristics of Shewanella algae in nutrient agar medium, TCBS agar medium and CHROMagar vibrio colored medium were similar to those of Shewanella abalone. Shewanella algae possessed β-hemolysis and Shewanella abalone possessed α-hemolysis in the blood agar plate. The biochemical reaction of Shewanella algae and Shewanella abalone was all of non-fermentation type. The results of artificial infection test showed that half lethal dose(LD50) of the test strains of Shewanella algae was 10-5.50/0.1 ml. The test strains of Shewanella algae have strong toxicity, and could cause mice and chickens to die of sepsis with mortality of100%. The mortality of Babylonia infected with Shewanella algae was 10%; while the survived Babylonia lost the ability of moving and intaking for a long time, but they were not suffered from proboscis edema. There was no death in mice or chicks infected with Shewanella abalone, but their livers and spleens were slightly hyperemic and swelling. There was also no death in Babylonia infected with Shewanella abalone, but their intaking and moving ability was lost for a short time.[Conclusion] Although Shewanella algae and Shewanella abalone were the dominant bacteria in Babylonia suffered from proboscis edema, they were not the main pathogenic bacteria for proboscis edema. Shewanella algae had strong pathogenicity to mice, chicks and Babylonia, while Shewanella abalone showed no marked pathogenicity to those experimental animals in this study.展开更多

The efficient artificial propagation method was to feed the host larvae with virus suspension,and the average mortality of the insects was 65.0%. The death peak of the pests appeared 4-8 d after virus infection. The high temperature, high humidity and poor light could help the virus infection and propagation. Filed control efficacy of Chenzhou strain was 86.6% in laboratory, which was better than of another commercial strain. The corrected control efficacy of this strain was 88.4% the field, which was higher than that of avermectin pesticide significantly. It was detected that the occlusion body(OB) concentration of the initial virus' s stock solution was 1.03×1011OBs/ml,and it was a strong SpliNPV strain, as it showed an excellent efficacy to control the pest Spodoptera litura, and thus there will be a good prospect of application and development of this SpliNPV strain.展开更多

After...OBJECTIVE To establish a murine uterine cervical cancer cell line and to define its biological characters. METHODS Transplanted tumor tissue was used for in vitro primary culture of U14 cervical carcinoma cells. After 20 passages, we examined its morphology, chromosomes, tumorigenicity and produced a growth curve. CK was detected by immunohistochemistry, the cell cycle determined by flow cytometry and the metastatic potential assessed in 615 and C57BL/c mice. We also transfected the cells with the pEGFP-N1 plasmid. RESULTS A newly established murine cell line was passaged 50 times over a period of 10 months. The cells grow as a partially suspended culture, and are immunohistochemically CK(+). The cell line is characterized by a hypotetraploid karyotype, a chromosomal number of 64-68 and a doubling time of 21.8 h. Exponential growth occurs by the third and forth day of culture. Cell cycle analysis showed G1 34%, G2 26%, and 40% in the S phase. The tumorigenicity was 100% upon implantation. No mycoplasma contamination was detected. A monoclonal continuous U14-GFP cell strain which was 100% GFP (+) was also produced. CONCLUSION We successfully established a new murine cervical U14 carcinoma cell line and an U14-GFP monoclonal strain. These cell lines are ideal for combined in vivo and in vitro tumor research.展开更多

And 17 fungicides were sprayed to evaluate the efficacy and effect of the fungicides tested in the field trials on the rice characters, [Result] The results showed that epoxicona- zole, difenoconazole, propiconazole and procloraz exhibited high inhibitory activity against mycelial growth of Ustilaginoidea virens with the ECso values 0.04, 0.07, 0.12 and 0.11 pg/ml, respectively. The results of field trials showed that the efficacy of Wen- quning, and fungicides such as difenoconazole, prochloraz, propiconazole, epoxi- conazole and their mixtures in controlling rice false smut were all 70% or more. [Conclusion] The 17 tested fungicides behaved efficacy in controlling rice false smut and did not cause drug injury on leaves and grains of rice plants, sprayed when flag leaves of rice fully expanded.展开更多

Akin to other functional gastrointestinal disorders, research has demonstrated an association betwe...The pathogenesis of Functional Dyspepsia (FD) remains unclear, appears diverse and is thus inadequately understood. Akin to other functional gastrointestinal disorders, research has demonstrated an association between this common diagnosis and psychosocial factors and psychiatric morbidity. Conceptualising the relevance of these factors within the syndrome of FD requires application of the biopsychosocial model of disease. Using this paradigm, dysregulation of the reciprocal communication between the brain and the gut is central to symptom generation, interpretation and exacerbation. Appreciation and understanding of the neurobiological correlates of various psychological states is also relevant. The view that psychosocial factors exert their influence in FD predominantly through motivation of health care seeking also persists. This appears too one-dimensional an assertion in light of the evidence available supporting a more intdnsic aetiological link. Evolving understanding of pathogenic mechanisms and the heterogeneous nature of the syndrome will facilitate effective management. Co-morbid psychiatric illness warrants treatment with conventional therapies. Acknowledging the relevance of psychosocial variables in FD, the degree of which is subject to vadation, has implications for assessment and management. Available evidence suggests psychological therapies may benefit FD patients particularly those with chronic symptoms. The rationale for use of psychotropic medications in FD is apparent but the evidence base to support the use of antidepressant pharmacotherapy is to date limited.展开更多

We aimed to systematically review the literature to determine which hygiene-related factors are associated with the development of IBD. Publications identified from a broad based MEDLINE and Current Contents search between 1966 and 2007 on key terms relevant to the 'hygiene hypothesis' and IBD including H pylori exposure, helminths, cold chain hypothesis, measles infection and vaccination, antibiotic use, breastfeeding, family size, sibship, urban upbringing, day care attendance and domestic hygiene were reviewed. The literature suggests that the hygiene hypothesis and its association with decreased microbial exposure in childhood probably plays an important role in the development of IBD, although the strength of the supporting data for each of the factors varies considerably. The most promising factors that may potentially be associated with development of IBD include H pylori exposure, helminths, breastfeeding and sibship. However, the vast majority of studies in this area are plagued by serious methodological shortcomings, particularly the reliance on retrospective recall of information making it difficult to truly ascertain the importance of a 'hygiene hypothesis' in IBD. The 'hygiene hypothesis' in IBD is an important area of research that may give clues to the aetiology of this disease. Directions for future research are recommended.展开更多

Clinically significant is the early onset of pleural effusion, which heralds a poor outcome of acute pancreatitis. The role of circulating trypsin, phospholipase A2, platelet activating factor, release of free fatty acids, chemoattractants such as tumor necrsosis factor (TNF)- alpha, interleukin (IL)-1, IL-6, IL-8, fMet-leu-phe (a bacterial wall product), nitric oxide, substance P, and macrophage inhlbitor factor is currently studied. The hope is that future management of acute pancreatitis with a better understanding of the pathogenesis of lung injury will be directed against the production of noxious cytokines.展开更多

Estrogens are not only essential for the female reproductive system, but they also control fundamental functions in other tissues including the cardiovascular system, bone, brain and liver. Recently, estrogens have been shown to target the biliary tree, where they modulate the proliferative and secretory activities of cholangiocytes, the epithelial cells lining bile ducts. By acting on both estrogen receptors (ER-α) and (ER-β) subtypes, and by activating either genomic or non-genomic pathways, estrogens play a key role in the complex loop of growth factors and cytokines, which modulates the proliferative response of cholangiocytes to damage. Specifically, estrogens activate intracellular signalling cascades JERK1/2 (extracellular regulated kinases 1/2, PI3-kinase/AKT (phosphatidylinositol-3' kinase/AKT)] typical of growth factors such as insulin like growth factor (IGF1), nerve growth factor (NGF) and vascular endothelial growth factor (VEGF), thus potentiating their action. In addition, estrogens stimulate the secretion of different growth factors in proliferating cholangiocytes. This review specifically deals with the recent advances related to the role and mechanisms by which estrogens modulate cholangiocyte functions in normal and pathological conditions.展开更多

CD is currently seen as a predominantly T-lym-phocyte-driven disease characterized by the presence of a c...Many advances have been made in the understanding of Crohn’s disease (CD) pathogenesis during the last decade. CD is currently seen as a predominantly T-lym-phocyte-driven disease characterized by the presence of a complex cocktail of interacting cytokines, chemokines and other mediators produced by a variety of cell types. Prevailing theories of CD pathogenesis suggest that patients’ T-lymphocytes are inappropriately activated in the setting of an immune imbalance, which is itself caused by an unfortunate confluence of genetic and en- vironmental factors. The T-cell response then leads to the chronic inflammation characteristic for the disease. Various environmental factors may play a role in the development of CD, but microbes are most consistently implied. This theory is based on epidemiological, clinico- pathological, genetic and experimental evidence. Despite the abundance of arguments for the implication of bac-teria in the aetiopathogenesis of CD, the precise role of bacteria in this disease still remains elusive. Three not necessarily mutually exclusive theories have been pro- posed: (1) an unidentified persistent pathogen; (2) an abnormally permeable mucosal barrier leading to exces-sive bacterial translocation; and (3) a breakdown in the balance between putative "protective" versus "harmful" intestinal bacteria ("dysbiosis"). At present, one cannot exclude with certainty any of these three proposed hy-potheses; they may all apply to CD to a certain extent.展开更多

Patients with Alzhe imer’s disease were only impaired in complex saccade performance (Alzheimer’s disease versus controls, target prediction P < 0.001, error decisions P < 0.0001 , error antisaccades P < 0.0001), but not reflexive saccade execution (for all, P >0.05). Patients with Parkinson’s disease had, compared with controls, similar complex saccade performance (for all, P > 0.05) and only minimal impairment in reflexive tasks, i.e. hypometric gain in the gap task (P=0.04). Impaired saccade execution in re flexive tasks allowed discrimination between DLB versus Alzheimer’s disease (se nsitivity ≥60%, specificity ≥77%)-and between PDD versus Parkinson’s disea se (sensitivity ≥60%, specificity ≥88%) when ±1.5 standard deviations was u sed for group discrimination. We conclude that impairments in reflexive saccades may be helpful for differential diagnosis and are minimal when either cortical (Alzheimer’s disease) or nigrostriatal neurodegeneration (Parkinson’s disease) exists solely; however, they become prominent with combined cortical and subcor tical neurodegeneration in PDD and DLB. The similarities in saccade performance in PDD and DLB underline the overlap between these conditions and underscore dif ferences from Alzheimer’s disease and Parkinson’s disease.

文摘With the progress of aging,the incidence of vascular calcification(VC)gradually increases,which is correlated with cardiovascular events and all-cause death,aggravating global clinical burden.Over the past several decades,accumulating approaches targeting the underlying pathogenesis of VC have provided some possibilities for the treatment of VC.Unfortunately,none of the current interventions have achieved clinical effectiveness on reversing or curing VC.The purpose of this review is to make a summary of novel perspectives on the interventions of VC and provide reference for clinical decision-making.

文摘[Objective] The research aimed to isolate and identify the anthracnose pathogen of Cassava and study its biological charcteristics.[Method] Two isolates of anthracnose (CCGHN01 and CCGHN03) in Cassava were isolated from the diseased leaves collected from Hainan of China.They were identified through the morphological observation of conidia and ITS sequence analysis.And the morphological characteristics were studied.[Result] The morphological observation of conidia and ITS sequence analysis indicated that the two isolates were Colletotrichum gloeosporioides.The morphological characteristics study results showed that optimum medium for the growth of two strains was PSA,the optimum temperatures were 26 ℃ and 30 ℃ respectively,the optimum pH was 8.0,the optimum light conditions were alternative light and dark and complete darkness respectively.For the conidia germination of two strains,the optimum temperatures were 28 ℃ and 30 ℃ respectively and the lethal temperature was 55 ℃ remaining 10 min.[Conclusion] The research laid the foundation for further control of anthracnose in Cassava.

文摘[Objective] The research aimed to design primers that are suitable for detecting H5 and H7 subtypes of avian influenza virus (AIV) ; [Method] DNAStar was used to analyze the homology of the sequences of H5 and H7 subtypes of AIV accessed in GenBank, and design primers( by Primer Premier 5.0) on high homologous region of these sequences, and then amplified by RT-PCR. [Result] The multiplex RT-PCR amplification, agarose gel electrophoresis and sequencing results showed that the self-designed primers are successful for detecting AIV. [Conclusion] It is feasible to rapidly diagnose AIV through this method.

文摘With the increasing planting area of Lilium lancifolium, the leaf fungal dis-ease of L. lancifolium is becoming more and more serious. In June and July of 2014, the excessive rainfal leads to the serious outbreak of leaf disease of L. lanci-folium. In mid-June, the wilting rate of L. lancifolium in seriously-infected field was even up to 50%-70%. In some fields, the shoots of L. lancifolium even al wilted. The pathogen was isolated from the infected leaves of L. lancifolium. Its pathogenici-ty, spore morphology, 18S rDNA sequence and biological characteristics were stud-ied. The results showed the isolated pathogen was Alternaria alternate. The lethal temperature of mycelial growth was 55 ℃. The optimum pH was 6-7. Among the tested carbon sources and nitrogen sources, the optimum carbon source was mal-tose, and the optimum nitrogen source was yeast extract.

文摘Objective The ventral part of the medial prefrontal cortex(mPFC)plays an important role in initiation and control of voluntary movement,mood and cognition.However,after the degeneration of the nigrostriatal pathway,the neuronal activity of the ventral mPFC and the role of serotonin1A(5-hydroxytryptamine,5-HT1A)receptors in the firing of the neurons are still unknown.The present study is to investigate the change of neuronal activity in the ventral mPFC and the effect of systemic administration of the selective 5-HT1Areceptor antagonist WAY-100635 on the activity of the neurons in normal and 6-hydroxydopamine(6-OHDA)-lesioned rats.Methods Single unit responses were recorded extracellularly with glass microelectrodes from ventral mPFC neurons in normal rats and 6-OHDA unilaterally lesiond rats in vivo.Results 6-OHDA lesion of the substantia nigra pars compacta(SNc)significantly increased the firing rate with no change in the firing pattern of neurons of the ventral mPFC in rats.Systemic administration of WAY-100635(0.1 mg/kg,i.v.)did not change the mean firing rate and firing pattern of ventral mPFC neurons in normal rats.In contrast,WAY-100635 signifi- cantly decreased the mean firing rate of the neurons in rats with 6-OHDA lesion of the SNc.Conclusion These data suggest that the degeneration of the nigrostriatal pathway results in an increase of neuronal activity of ventral mPFC and dysfunction of 5-HT1Areceptor.

文摘[Objective] This study aimed to explore the biological characteristics of Col etotrichum gloeosporioides in pears. [Method] Twenty-five C. gloeosporioides strains were isolated and identified from the diseased samples. Their pathogenicity was identified by inoculating the surface of punctured pears with fungal discs. The effects of different temperatures, pH values, carbon sources and nitrogen sources on the growth of C. gloeosporioides mycelia were explored by incubating fungal discs on the center of plates. [Result] Among the twenty-five C. gloeosporioides strains, three had strong pathogenicity, and eighteen had intermediate pathogenicity, and four strains had weak pathogenicity. Those highly-pathogenic strains had darker colonies, with dense mycelia, whereas those lowly-pathogenic ones had white colonies, with sparse mycelia. Those with fast-growing colonies showed strong pathogenicity, while those with slowly-growing colonies displayed weak pathogenicity. There was no relationship between conidia yield and pathogenicity. The optimum temperature for the growth of C. gloeosporioides mycelia was 25-30 ℃, and the optimum pH was 5.0-7.0. C. gloeosporioides could make use of various carbon sources (monosaccharide and disaccharide), inorganic and organic nitrogen sources, and the optimal carbon source and nitrogen source were sucrose and beef extract, respectively. [Conclusion] Our study benefits further understanding of C. gloeospori-oides and helps to control pear anthracnose more effectively.
文摘AIMS To investigate the clinical significance of vas- cular endothelial growth factor (VEGF) expression in gastric carcinoma. METHODS The expression of VEGF in 128 gastric carcinomas was investigated by immunohistochemical staining with a polyclonal antibody against VEGF. Cor- relations between the expression of VEGF and various clinicopathologic factors and prognosis were studied. RESULTS The VEGF-rich expression rate was 64.1% in gastric carcinoma. VEGF-rich expression rate of patients with stage Ⅲ and stage Ⅳ disease was greater than that of patients with stage f disease (P <0.05). Significant differences of expression rate ex- isted with respect to growth pattern,serosal invasion and lymph node metastasis. The VEGF-rich rate was much higher in tumors with expanding growth pattern (71.8%) or serosal invasion (73.5%) than in those with infiltrative growth pattern (52.0%) or non-serosal invasion (53.3%) (P<0.025,respectively),and it was also significantly higher in patients with lymph node metastases (75.0%) than in those without such metastases (50.0%) (P<0.05). In addition,postop- erative survey of 86 patients who had been followed up for at least 5 years demonstrated that the 5-year sur- vival rate of patients with VEGF-rich tumors was signifi- cantly lower than that of patients with VEGF-poor tu- mors (P<0.05). CONCLUSIONS The expression of VEGF may be as- sociated with the invasion and metastasis and may also be a useful prognostic indicator of gastric carcinoma.

文摘[Objective] The biological characteristics and pathogenicities of Shewanella algae and Shewanella abalone from Babylonia were studied in this paper. [Method]The hemolytic bacteria were isolated from the hepatopancreas of Babylonia suffered from proboscis edema with blood agar plate. The dominant bacterial community in the ill Babylonia was identified by 16 S r DNA sequence analysis, and the bacterial cultural and biochemical characteristics and pathogenicities were studied. [Result]The Shewanella bacteria, including Shewanella algae and Shewanella abalone, are the dominant bacterial community in Babylonia suffered from proboscis edema.The colony characteristics of Shewanella algae in nutrient agar medium, TCBS agar medium and CHROMagar vibrio colored medium were similar to those of Shewanella abalone. Shewanella algae possessed β-hemolysis and Shewanella abalone possessed α-hemolysis in the blood agar plate. The biochemical reaction of Shewanella algae and Shewanella abalone was all of non-fermentation type. The results of artificial infection test showed that half lethal dose(LD50) of the test strains of Shewanella algae was 10-5.50/0.1 ml. The test strains of Shewanella algae have strong toxicity, and could cause mice and chickens to die of sepsis with mortality of100%. The mortality of Babylonia infected with Shewanella algae was 10%; while the survived Babylonia lost the ability of moving and intaking for a long time, but they were not suffered from proboscis edema. There was no death in mice or chicks infected with Shewanella abalone, but their livers and spleens were slightly hyperemic and swelling. There was also no death in Babylonia infected with Shewanella abalone, but their intaking and moving ability was lost for a short time.[Conclusion] Although Shewanella algae and Shewanella abalone were the dominant bacteria in Babylonia suffered from proboscis edema, they were not the main pathogenic bacteria for proboscis edema. Shewanella algae had strong pathogenicity to mice, chicks and Babylonia, while Shewanella abalone showed no marked pathogenicity to those experimental animals in this study.

文摘The laboratory bioassay and field control efficacy of Spodoptera litura nucleopolyhedrovirus(Spli NPV) Chenzhou strain were preliminarily examined. The efficient artificial propagation method was to feed the host larvae with virus suspension,and the average mortality of the insects was 65.0%. The death peak of the pests appeared 4-8 d after virus infection. The high temperature, high humidity and poor light could help the virus infection and propagation. Filed control efficacy of Chenzhou strain was 86.6% in laboratory, which was better than of another commercial strain. The corrected control efficacy of this strain was 88.4% the field, which was higher than that of avermectin pesticide significantly. It was detected that the occlusion body(OB) concentration of the initial virus' s stock solution was 1.03×1011OBs/ml,and it was a strong SpliNPV strain, as it showed an excellent efficacy to control the pest Spodoptera litura, and thus there will be a good prospect of application and development of this SpliNPV strain.
文摘OBJECTIVE To establish a murine uterine cervical cancer cell line and to define its biological characters. METHODS Transplanted tumor tissue was used for in vitro primary culture of U14 cervical carcinoma cells. After 20 passages, we examined its morphology, chromosomes, tumorigenicity and produced a growth curve. CK was detected by immunohistochemistry, the cell cycle determined by flow cytometry and the metastatic potential assessed in 615 and C57BL/c mice. We also transfected the cells with the pEGFP-N1 plasmid. RESULTS A newly established murine cell line was passaged 50 times over a period of 10 months. The cells grow as a partially suspended culture, and are immunohistochemically CK(+). The cell line is characterized by a hypotetraploid karyotype, a chromosomal number of 64-68 and a doubling time of 21.8 h. Exponential growth occurs by the third and forth day of culture. Cell cycle analysis showed G1 34%, G2 26%, and 40% in the S phase. The tumorigenicity was 100% upon implantation. No mycoplasma contamination was detected. A monoclonal continuous U14-GFP cell strain which was 100% GFP (+) was also produced. CONCLUSION We successfully established a new murine cervical U14 carcinoma cell line and an U14-GFP monoclonal strain. These cell lines are ideal for combined in vivo and in vitro tumor research.

文摘[Objective] This study aims to screen for the high effective fungicides which could significantly decrease the disease incidence and disease index of rice false smut. [Method] The inhibitory activities of the fungicide against mycelial growth of Ustilaginoidea virens were measured to in vitro evaluate the ECho values. And 17 fungicides were sprayed to evaluate the efficacy and effect of the fungicides tested in the field trials on the rice characters, [Result] The results showed that epoxicona- zole, difenoconazole, propiconazole and procloraz exhibited high inhibitory activity against mycelial growth of Ustilaginoidea virens with the ECso values 0.04, 0.07, 0.12 and 0.11 pg/ml, respectively. The results of field trials showed that the efficacy of Wen- quning, and fungicides such as difenoconazole, prochloraz, propiconazole, epoxi- conazole and their mixtures in controlling rice false smut were all 70% or more. [Conclusion] The 17 tested fungicides behaved efficacy in controlling rice false smut and did not cause drug injury on leaves and grains of rice plants, sprayed when flag leaves of rice fully expanded.

文摘The family of bile acids includes a group of molecular species of acidic steroids with very peculiar physical-chemical and biological characteristics.They are synthesized by the liver from cholesterol through several complementary pathways that are controlled by mechanisms involving finetuning by the levels of certain bile acid species.Although their best-known role is their participation in the digestion and absorption of fat,they also play an important role in several other physiological processes.Thus,genetic abnormalities accounting for alterations in their synthesis,biotransformation and/or transport may result in severe alterations,even leading to lethal situations for which the sole therapeutic option may be liver transplantation.Moreover,the increased levels of bile acids reached during cholestatic liver diseases are known to induce oxidative stress and apoptosis,resulting in damage to the liver parenchyma and,eventually,extrahepatic tissues.When this occurs during pregnancy,the outcome of gestation may be challenged.In contrast,the physical-chemical and biological properties of these compounds have been used as the bases for the development of drugs and as pharmaceutical tools for the delivery of active agents.
文摘The pathogenesis of Functional Dyspepsia (FD) remains unclear, appears diverse and is thus inadequately understood. Akin to other functional gastrointestinal disorders, research has demonstrated an association between this common diagnosis and psychosocial factors and psychiatric morbidity. Conceptualising the relevance of these factors within the syndrome of FD requires application of the biopsychosocial model of disease. Using this paradigm, dysregulation of the reciprocal communication between the brain and the gut is central to symptom generation, interpretation and exacerbation. Appreciation and understanding of the neurobiological correlates of various psychological states is also relevant. The view that psychosocial factors exert their influence in FD predominantly through motivation of health care seeking also persists. This appears too one-dimensional an assertion in light of the evidence available supporting a more intdnsic aetiological link. Evolving understanding of pathogenic mechanisms and the heterogeneous nature of the syndrome will facilitate effective management. Co-morbid psychiatric illness warrants treatment with conventional therapies. Acknowledging the relevance of psychosocial variables in FD, the degree of which is subject to vadation, has implications for assessment and management. Available evidence suggests psychological therapies may benefit FD patients particularly those with chronic symptoms. The rationale for use of psychotropic medications in FD is apparent but the evidence base to support the use of antidepressant pharmacotherapy is to date limited.
文摘The hygiene hypothesis is thought to be a significant contributor to the growing incidence of inflammatory bowel disease (IBD) around the world, although the evidence for specific factors that underlie the hygiene hypothesis in IBD is unclear. We aimed to systematically review the literature to determine which hygiene-related factors are associated with the development of IBD. Publications identified from a broad based MEDLINE and Current Contents search between 1966 and 2007 on key terms relevant to the 'hygiene hypothesis' and IBD including H pylori exposure, helminths, cold chain hypothesis, measles infection and vaccination, antibiotic use, breastfeeding, family size, sibship, urban upbringing, day care attendance and domestic hygiene were reviewed. The literature suggests that the hygiene hypothesis and its association with decreased microbial exposure in childhood probably plays an important role in the development of IBD, although the strength of the supporting data for each of the factors varies considerably. The most promising factors that may potentially be associated with development of IBD include H pylori exposure, helminths, breastfeeding and sibship. However, the vast majority of studies in this area are plagued by serious methodological shortcomings, particularly the reliance on retrospective recall of information making it difficult to truly ascertain the importance of a 'hygiene hypothesis' in IBD. The 'hygiene hypothesis' in IBD is an important area of research that may give clues to the aetiology of this disease. Directions for future research are recommended.
文摘Acute pancreatitis in its severe form is complicated by multiple organ system dysfunction, most importantly by pulmonary complications which include hypoxia, acute respiratory distress syndrome, atelectasis, and pleural effusion. The pathogenesis of some of the above complications is attributed to the production of noxious cytokines. Clinically significant is the early onset of pleural effusion, which heralds a poor outcome of acute pancreatitis. The role of circulating trypsin, phospholipase A2, platelet activating factor, release of free fatty acids, chemoattractants such as tumor necrsosis factor (TNF)- alpha, interleukin (IL)-1, IL-6, IL-8, fMet-leu-phe (a bacterial wall product), nitric oxide, substance P, and macrophage inhlbitor factor is currently studied. The hope is that future management of acute pancreatitis with a better understanding of the pathogenesis of lung injury will be directed against the production of noxious cytokines.

文摘The scientific framework concerning estrogen effects on different tissues has expanded enormously during the last decades, when estrogen receptor (ER) subtypes were identified. Estrogens are not only essential for the female reproductive system, but they also control fundamental functions in other tissues including the cardiovascular system, bone, brain and liver. Recently, estrogens have been shown to target the biliary tree, where they modulate the proliferative and secretory activities of cholangiocytes, the epithelial cells lining bile ducts. By acting on both estrogen receptors (ER-α) and (ER-β) subtypes, and by activating either genomic or non-genomic pathways, estrogens play a key role in the complex loop of growth factors and cytokines, which modulates the proliferative response of cholangiocytes to damage. Specifically, estrogens activate intracellular signalling cascades JERK1/2 (extracellular regulated kinases 1/2, PI3-kinase/AKT (phosphatidylinositol-3' kinase/AKT)] typical of growth factors such as insulin like growth factor (IGF1), nerve growth factor (NGF) and vascular endothelial growth factor (VEGF), thus potentiating their action. In addition, estrogens stimulate the secretion of different growth factors in proliferating cholangiocytes. This review specifically deals with the recent advances related to the role and mechanisms by which estrogens modulate cholangiocyte functions in normal and pathological conditions.
文摘Many advances have been made in the understanding of Crohn’s disease (CD) pathogenesis during the last decade. CD is currently seen as a predominantly T-lym-phocyte-driven disease characterized by the presence of a complex cocktail of interacting cytokines, chemokines and other mediators produced by a variety of cell types. Prevailing theories of CD pathogenesis suggest that patients’ T-lymphocytes are inappropriately activated in the setting of an immune imbalance, which is itself caused by an unfortunate confluence of genetic and en- vironmental factors. The T-cell response then leads to the chronic inflammation characteristic for the disease. Various environmental factors may play a role in the development of CD, but microbes are most consistently implied. This theory is based on epidemiological, clinico- pathological, genetic and experimental evidence. Despite the abundance of arguments for the implication of bac-teria in the aetiopathogenesis of CD, the precise role of bacteria in this disease still remains elusive. Three not necessarily mutually exclusive theories have been pro- posed: (1) an unidentified persistent pathogen; (2) an abnormally permeable mucosal barrier leading to exces-sive bacterial translocation; and (3) a breakdown in the balance between putative "protective" versus "harmful" intestinal bacteria ("dysbiosis"). At present, one cannot exclude with certainty any of these three proposed hy-potheses; they may all apply to CD to a certain extent.
