Genetic Inheritance and Molecular Marker of Clubroot Resistance Genes in Brassica campestris ssp. chinensis

Objective] This study was conducted to investigate the genetic inheritance of clubroot resistance in Chinese non-heading cabbage (Brassica campestris ssp. chinensis). [Method] The clubroot resistance gene was introduced from a Brassica campestris ssp. pekinensis cultivar to non-heading Chinese cabbage, and the inheri-tance and molecular markers of clubroot resistance gene in parental lines, F1, F2 and BC1 of non-heading Chinese cabbage were studied through pathogen inoculation at seedling stage and ISSR-PCR. [Result] Clubroot resistance in non-heading Chi-nese cabbage was control ed by a single dominant gene. ISSR molecular markers with Bulk segregant analysis (BSA) found that primer-873 was linked to resistance gene, named CR-873, and the genetic distance between the marker and the resis-tance gene was 9.72 cM. [Conclusion] The results provide references for the molecular marker assisted breeding of non-heading Chinese cabbage. 展开更多

The adsorption state and catalytic properties of pepsin and acidic protease from microorganisms Asp. awamori and Asp. oryzae were studied in solid phase system (in presence of sorsilen, DEAE- and CM-cellulose). According to the results, adsorption capacity and catalytic activity of enzymes depend on the physical nature of surface groups of the solid phase. Changing the stability of enzymes in the system with solid phase is observed even the adsorption bond is less stable (in the case of DEAE- and CM-cellulose in acidic media). Injection to the medium ethanol, surfactants, sodium chloride and changing the temperature of the incubation medium could prevent the negative effects of the solid phases. When sorsilen is used as solid phase, pepsin and acidic protease from Asp. awamori suffer from high surface inactivation. Various surfactants influence adsorption state of enzymes differently. Non-ionic surfactants (Triton X-100) prevent adsorption and restore catalytic properties of enzymes. 展开更多

Protein malnutrition is quite common worldwide; its deficiency has adverse effects in all organs. Edible insect's traditional cultural food, commonly consumed in rural communities, now intake is also being incorporated in some countries. Grasshoppers comprise proteins, the building blocks of AA (amino acids), that are one of the five classes of complex biomolecules found in cells and tissues that play an important role in human metabolism and nutrition. This research was conducted to determine proximal composition of Sphenarium purpurascens Ch, grasshoppers, focused on proteins and AA. Sampling was performed during autumn, 2013, in Oaxaca State. Dry raw adult insects were ground and proceeded to macro molecules proximal analysis, protein by Kjeldahl AOAC (Association of Official Analytical Chemists) method and essential AA in a Beckman 6300 equipment. Obtained data were: total proteins 71.56%; lipids 8.14%; minerals 3.52%; fiber 7.48%; soluble carbohydrates 9.30%. Essential AA (mg/16 g-N): isoleucine 4.5; leucine 8.1; lysine 5.4; methionine + cysteine 5.1; phenylalanine + tyrosine 8.5; threonine 3.5; tryptophan 0.6; valine 5.5. Sphenarium purpurascens Ch, Grasshoppers represent a good source of proteins and essential AA that can grow almost everywhere, available mostly all year round and affordable to all social groups, thus being a good option to improve human health. 展开更多
