In yeast, the deadenylation reaction is predominantly mediated by CCR4 and CCR4- associated factor 1 (CAF1), two components of the well-characterised protein complex named CCR4-NOT. We report here that AtCAF1a and AtCAF1b, putative Arabidopsis homologs of the yeast CAF1 gene, partially complement the growth defect of the yeast call mutant in the presence of caffeine or at high temperatures. The expression of At-CAF1a and AtCAFlb is induced by multiple stress-related hormones and stimuli. Both AtCAF1a and AtCAFlb show deadenylation activity in vitro and point mutations in the predicted active sites disrupt this activity. T-DNA insertion mutants disrupting the expression of AtCAF1a and/or AtCAF1b are defective in deadenylation of stress-related mRNAs, indicating that the two AtCAF1 proteins are involved in regulated mRNA deadenylation in vivo. Interestingly, the single and double mutants of AtCAF1a and AtCAFlb show reduced expression of pathogenesis-related (PR) genes PR1 and PR2 and are more susceptible to Pseudomonas syringae pv tomato DC3000 (Pst DC3000) infection, whereas transgenic plants over-expressing AtCAFla show elevated expression of PR1 and PR2 and increased resis-tance to the same pathogen. To investigate the influence of sodium to potassium (Na/IO ratios on the growth performance and physiological response of the Pacific white shrimp (Litopenaeus vananmei), various concentrations of KC1 were added to...To investigate the influence of sodium to potassium (Na/IO ratios on the growth performance and physiological response of the Pacific white shrimp (Litopenaeus vananmei), various concentrations of KC1 were added to low-salinity well water (salinity 4) in an 8-week culture trial. Six treatments with NWK ratios of 60:1, 42:1, 33:1, 23:1, 17:1, and 14:1 were replicated in triplicate. The highest weight-gain rate (3 506±48)% and survival rate (89.38±0.88)% was observed in well water with Na/K ratios of 23:1 and 42:1, respectively, while the feed conversion ratio (1.02~0.01), oxygen consumption, and ammonia-N excretion rate was the lowest in the medium with a Na/K ratio of 23:1. Gill Na+-K+-ATPase activity, as an indicator of osmoregulation, peaked in the treatment where the Na/K ratio was 17:1. The total hemocyte count, respiratory burst, and immune-related enzyme activities (ALP, LSZ, PO, and SOD) ofL. vananmei were affected significantly by Na/K ratios (P〈0.05). After challenged with Vibrio harveyi, the cumulative mortality of shrimp reared in a Na/K ratio of 23:1 (30±14.14)% was significantly lower than the control (75~7.07)%. In conclusion, the addition of K+ to low-salinity well water in L. vannamei cultures is feasible. Na/K ratios ranging from 23:1 to 33:1 might improve survival and growth. Immunity and disease resistance are also closely related to the Na/K ratio of the low-salinity well water. The findings may contribute to the development of more efficient K^+ remediation strategies for L. vananmei culture in low-salinity well water.展开更多
The Cross River Estuary,Nigeria,is an important shrimping area for artisanal fishermen of the coastal communities.The multi-species Macrobrachium fishery is exploited with three main gears,namely beach seine,push net ...The Cross River Estuary,Nigeria,is an important shrimping area for artisanal fishermen of the coastal communities.The multi-species Macrobrachium fishery is exploited with three main gears,namely beach seine,push net and trap.Studies on species composition of this fishery recorded thirteen shrimp species,one swimming crab (Callinectes amnicola) and two fish species (Eleotris sp.and Pellonula leonensis).The shrimp species identified included Macrobrachium macrobrachion (83.39% and 55.69% by number and weight,respectively),M.vollenhovenii (9.66% and 37.18%),M.equidens (3.8% and 2.87%),juveniles-sub-adults of Penaeus notialis (1.11% and 1.3%),M.dux,M.felicinum,Palaemonetes africanus,Palaemon maculatus,Palaemon elegans,Desmocaris sp.,Leander sp.,Nematopalaemon hastatus and Alpheus pontederiae.While the selectivity index for trap was 0.25,beach seine and push net had a lower index of 0.063.The results present the first comprehensive and representative report for the Estuary shrimp fishery and will assist in the management of the biodiversity of this ecosystem.展开更多

While components of the polycomb repressive complex2 (PRC2) are evolutionarily conserved and their functions are extensively studied in plants, PRCI differs considerably between animals and plants, and its functions in plants are as yet not well described. Previous studies have identified the Arabidopsis AtRINGla and AtRINGlb as homologues of the animal PRC1 subunit RING1. Here, we show that the Atringla Atringlb double mutant exhibits derepression of embryonic traits during vegetative growth. Accordingly, several key regulatory genes involved in embryogenesis and stem cell activity are ectopically expressed in the mutant. Furthermore, we show that the mutant phenotypes and increased expression of regulatory genes are enhanced by the PRC2 mutant c/f. Finally, we show that three homologues of the animal PRCl-subunit ring-finger protein BMI1, AtBMIIa, AtBMIlb and AtBMIlc, can bind with AtRINGla or AtRINGIb, and in addition, AtBMIlc can bind with LHP1. The Atbmila Atbmilb double mutant shows derepression of embryonic traits similar to that of the Atringla Atringlb double mutant. The present study was conducted to evaluate the feasibility of replacing fish meal(FM) with bioactive peptides(BPs) in diet of white shrimp(Litopenaeus vannamei). The changes in growth performance, body composition, n...The present study was conducted to evaluate the feasibility of replacing fish meal(FM) with bioactive peptides(BPs) in diet of white shrimp(Litopenaeus vannamei). The changes in growth performance, body composition, non-specific immunity, and water quality were examined after the shrimp were fed four diets, in which 0%(control), 33.3%, 66.7% and 100% of FM was replaced by BPs, respectively. The groups were designated as Con, 1/3BPs, 2/3BPs, and 3/3BPs. A total of 720 shrimp with an initial body weight of 1.46 ± 0.78 g were fed the experimental diets for 56 days. The results revealed that: 1) the weight gain rate(WGR) in 1/3BPs, 2/3BPs, and 3/3BPs was significantly higher than that in Con(P < 0.05), while no significant difference was found on survival rate and feed conversion ratio(FCR); 2) the whole-body crude protein(CP) and crude lipids(CL) were significantly different among groups, while there was no significant difference between crude ash and phosphorus contents; 3) the levels of acid phosphatase(ACP), lysozyme(LZM), superoxide dismutase(SOD), phenol oxidase(PO) and bactericidal activity increased significantly with the inclusion of BPs; 4) in terms of water quality, no significant difference was found in p H and dissolved oxygen among diets during the whole experimental period. Moreover, even though nitrite and ammonium levels tended to increase with time, there was no significant difference among groups. The results indicated that BPs is an applicable alternative of protein source, which can substitute FM in the diets of L. vannamei; it is able to effectively promote growth performance and improve immunity. BFT (Biofloc Technology) has been currently applied in Cantho University to a number of targeted species from fresh-to marine and saline water species in the Mekong Delta, e.g. striped catfish (Pangasianodon hypoph...BFT (Biofloc Technology) has been currently applied in Cantho University to a number of targeted species from fresh-to marine and saline water species in the Mekong Delta, e.g. striped catfish (Pangasianodon hypophthalmus), white leg shrimp (Litopenaeus vannamei), tiger shrimp (Penaeus monodon) and Artemia Vinhchau strain (Artemia franciscana) for both in the lab-scales or production scales. The best growth and survival rate of striped catfish was obtained in 6%o. For white leg shrimp, the results indicated: (1) C (Carbon) source e.g. rice-flour or molasses supplementary based on the feed provided to promote survival, growth and shrimp biomass harvested; (2) it was fed less than 20% as usual but showed similar to those in the control; (3) combination of rice-flour and molasses at a ratio of 70:30 by weight of C and N (Nitrogen) in a ration of 15:1 enhanced shrimp survival and growth; (4) in intensive culture, shrimp could be against the infection of disease and end up with higher survival, growth in earthen ponds. For tiger shrimp, a set up for larvae till post larvae at different ratios of C and N and C:N of 30 PLI5 displayed with better survival rate (49.73 ± 7.07%) and production (74,596 ± 10.608 PL/m3). Artemiafranciscana was set up at C:N = 10:1 and salinities from 35, 60, 80 and 100 ppt. After two weeks, there were no significant difference among treatment and the control (without biofloc) in term of survival and growth. Moreover, total embryos per female was not significant different with the control and even the number of embryos as cysts tended to be higher. BFT displayed its advantages when applied on culture system of different targeted species and there is no doubt that it could help to sustain aquaculture and save environment in the Mekong Delta in near future.展开更多
