Effects of two kinds of transgenic poplar on protective enzymes system in the midgut of larvae of American white moth 被引量:10

The leaves of Bt (Bacillus thuringiensis) transgenic poplar (Populus nigra L.) and CpTI (Cowpea trypsin inhibitor) transgenic poplar ((P. tomentosa×P. bolleana)×P. Tomentosa) were taken to feed the 4th-5th-instar larvae of American white moth (Hyphantria cunea (Drury)) for determination of the activities of the protective enzyme system inside larvae’s body. The physiological and biochemical effects of the transgenic poplars on the larvae were studied. The results showed that the two kinds of transgenic poplars had similar effects on the protective enzyme system in the midgut of larvae. The activities of superoxide dismutase, catalase, and peroxidase in midgut of the larvae increased gradually, reached the highest value at a certain time, and then decreased suddenly. For the larvae that were fed with the leaves of Bt transgenic poplar, the peak value of superoxide dismutase and catalase presented at the time of 24-h feeding, while the peak of peroxidase took place at the time of 12-h feeding. The activities of these protective enzymes for the larvae that were fed with leaves of CpTI transgenic poplar peaked 12 h later than that of those fed with leaves of Bt transgenic poplar. The comparison of activities of the protective enzymes was also carried out between the larvae with different levels of intoxication. It was found that the activities of protective enzyme of the seriously intoxicant larvae were higher than that of the lightly intoxicant larvae. This difference was more obvious in the group treated with CpTI transgenic poplar. 展开更多

Larch (Larix olgensis), Manchurian ash (Fraxinus mandshurica), Korean pine (Pinus koraiensis) and White birch (Betula platyphylla) are the major planting species in northeast China. The samples of forest litters were collected from the stands of the above 4 species in Laoyeling and Jianlagou experiment stations of Maorshan Exp. Forest Farm (45?2-45?0N, 127?0-127?8E), Northeast Forestry University, in early October 2002. Quantitative analysis and qualitative analysis were carried out on the organic acids existing in freshly fallen litters (L layer) and hemi-decomposed litters (F layer) of the four forest species by using Gas Chromatogram system. A wide variety of organic acids were identified, including oxalic, malonic, fumaric, succinic, maleic, malic, citric, C16:0, C18:0, C18:1, C18:2, C18:3 and C20:0 acids. In respect of L litters of all samples, the oxalic acid content (over 30 mg/g) was the highest of the seven low-molecular-weight organic acids identified, while the content of oleic or linoleic (above 40 mg/g) was found to be highest among the six high aliphatic acids identified. As to F litters, oxalic acid content was also the highest, followed by linoleic and oleic. For the same tree species or the same forest, the kinds and contents of organic acids in L litters were more abundant than that in F litters. 展开更多

The essential oil wa... To analyze the constituents of essential oil from Chinese eaglewood [resinous wood of Aquilaria sinensis (Lour.) Gilg] and its anti-methicillin-resistant Staphylococcus aureus (MRSA) activity. The essential oil was extracted by water-steam distillation and analyzed by GC/MS method. The relative contents of the compounds were determined by normalization. The compounds were characterized by NIST05 and WILEY275L database matching and comparison of their MS spectra with those of literature data. Antibacterial activity of the oil was assayed by the filter paper disc agar diffusion method. The oil showed significant antibacterial activity against MRSA. Sixty-six chromatographic peaks were detected, among them thirty compounds comprising 59.80% of the total essential oil were characterized. Twenty-six compounds comprising 54.26% of the oil were identified as sesquiterpenes. β-Agarofuran (8.96%), kusunol (7.82%), (-)-jinkoh-eremol (5.04%), agarospirol (4.53%), baimuxifuranic acid (4.09%) were the major sesquiterpenes. Four nor-sesquiterpenes and some other sesquiterpenes, such as 10-epi-γ-eudesmol, α-agarofuran, epi-ligulyl oxide, etc. were detected in Chinese eaglewood oil for the first time. This is the first report about anti-MRSA activity of Chinese eaglewood oil from A. sinensis. 展开更多

There were 8 gliadin and 3 high_molecular_weight (HMW)_glutenin patterns in 14 Yunnan hulled wheat ( Triticum aestivum ssp. yunnanese King) accessions, 9 gliadin and 4 HMW_glutenin patterns in 9 Tibetan weedrace ( T. aestivum ssp. tibetanum Shao ) accessions, and 9 gliadin and 5 HMW_glutenin patterns in 9 Xinjiang rice wheat ( T. petropavlovskyi Udacz. et Migusch.) accessions. One accession (i.e. Daomai 2) carried new subunits 2.1+10.1 encoded by Glu_D1. Among the three Chinese endemic wheat groups, a total of 10, 14 and 11 alleles at Gli_1 locus; 11, 14 and 12 alleles at Gli_2 locus; and 5, 6 and 8 alleles at Glu_1 locus were identified, respectively. Among Yunnan hulled wheat, Tibetan weedrace and Xinjiang rice wheat, the Nei's genetic variation indexes were 0.3798, 0.5625 and 0.5693, respectively. Objective: To investigate the association between the mutations in lipoprotein lipase gene and hypertriglyceridemia (HTG). Methods: The lipoprotein lipase (LPL) gene was screened for mutations in 386 Chinese subjects with (108 cases in the HTG group) or without HTG (278 cases in the control group), by single-strand conformation polymorphism (SSCP) analysis and DNA sequencing. Results: One novel silent mutation L103L, one missense mutation P207L, three splicing mutations Int3/3' -ass/C(-6)→T, and the common S447X polymorphism has been identified in the whole coding region and exon-intron junctions of the LPL gene were examined. Heterozygous P207L found in the HTG group was the first case reported in Asia and subsequently another P207L heterozygote was found in the proband's family, all of which suggested that P207L was one of the causes of familial combined hyperlipidemia, but was not so prevalent as that in French Canadian. Int3/3'-ass/C(-6)→T was found in both groups in the present study although it was regarded as a pathogenic variant to HTG earlier on. Moreover about the beneficial polymorphism S447X, there was also some supportive evidence that the levels of triglycerides (TG) in S447X carriers were significantly lower than noncarders in the subjects without HTG. Conclusions: The association between the LPL variants and HTG is quite complicated and versatile, genotyping of LPL in a larger-scale screening should be necessary and justifiable. 展开更多

[Method] Chinese cabbages were used as experimental materials and irrigated with control water, 50% reclaimed water and 100% reclaimed water, and then the number of endophytic bacteria in Chinese cab- bages was measured. [Result] Using 50% reclaimed water, the fresh weight of Chi- nese cabbage was improved by 68.94%; however, with the deepening internalization of exogenous microorganism internalization, the total number of endophytic bacteria and coliform flock in Chinese cabbages treated with 50% reclaimed water was sig- nificantly higher than the other two treatment groups (P〈0.05). [Conclusion] Re- claimed water enhances both the weight of Chinese cabbage and the number of endophytic bacteria, which is unsuitable for irrigation of edible crops such as Chi- nese cabbage.
