Full-length cDNA Cloning and Tissues Expression Analysis of Ferritin Gene from Acipenser sinensis 被引量:2

According to the conserved sequence of the ferritin gene, a homologous sequence was obtained from the EST database through a BLAST search against the GenBank database. This sequence was amplified with the method of RT... According to the conserved sequence of the ferritin gene, a homologous sequence was obtained from the EST database through a BLAST search against the GenBank database. This sequence was amplified with the method of RT-PCR, false sequencing was corrected, and full length eDNA of the ferritin subunit from the Chinese sturgeon was obtained. After being submitted to the GenBank database, the sequence accession number EU348782 was assigned. With the length of 896 bp, this eDNA includes entire coding regions of 53 lbp, which encodes 176 amino acids (aa). The molecular weight was predicted to be 20339.9Mr and the theoretical isoelectric point 5.66. It shares 82.9% protein sequence homology with the ferritin of the Atlantic salmon. This gene is expressed in many organs of the Chinese sturgeon, for example, the liver, pancreas, muscle, brain, heart and gastric mucosa. The highest expression level was found in the pancreas and the heart, while the muscular tissue showed the lowest. Homology modeling was used to predict the 3-D structure of the protein, which included 5 alpha helices and 10 turns. The ferritin protein structure could be overlapped and showed high similarity with that of human, flog and bacteria. It was revealed that this kind of ferritin was highly conserved in structure and function. 展开更多

[Objective] The aim of this study was to realize efficient expression of the porcine reproductive and respiratory syndrome virus (PRRSV) ORF7 gene in genetic engineering bacteria and analYze the immunological activity of the recombinant protein after purification. [ Method] The constructed recombinant expression vector pET-ORF7 was transformed into Escherichia co1BL21 (DE3) and induced by IPTG under the optimal condition. After analysis of SDS-PAGE and Western Blot, the expression products were purified by Ni-NTA His · Bind Resin chrom- atographic column under denaturing condition and renatured by gradient dialysis. Subsequently, the immunological activity of the renatured recombinant protein was detected by Westem Blot and indirect ELISA. [ Result] The recombinant plasmid pET-ORF7 expressed in E. coli successfully, and the fusion protein was in the form of inclusion body. By SDS-PAGE detection, the molecular weight of the expression protein was approximate 33 kD, according with the expectation. Analysis by Bandscan software showed that the expressed fusion protein was about 50% of total bacterial protein of BL21 (DE3). Wastem Blot and indirect ELISA detection showed that the renatured protein could react with PRRSV positive serum specifically, indicating its good immunological activity. [ Conclusion] This study lays a foundation for the preparation of PRRSV monoclonal antibody and diagnostic kit. 展开更多

A Dendroclimatic Study of Relic Picea meyeri in Inner Mongolian Steppe 被引量:6

Dendroclimatic methods were used to investigate the fundamental relationships between the temperature variables and the growth of climatically relic Picea meyeri Rehd. et Wils. on sandy land of Inner Mongolian grasslands in the Xilin River Basin. The annual mean temperature and accumulated temperature (above 5 ℃, 10 ℃) respectively showed no significant correlation with the spruce growth relative to summer mean temperature, which displayed strong coherence with tree growth ( P <0.01). The mean temperature in May showed significantly negative correlation with spruce growth ( P <0.05). Furthermore, it was revealed that the negative influence of May temperature was due to monthly mean maximum temperature ( T max ) ( P <0.01) other than monthly mean minimum temperature ( T min ) values ( P <0.1), which indicated that the use of T min and T max separately can allow much more insights on the temperate influence. In addition, extreme maximum temperature in May and June might impose the most detrimental influence on tree growth in semi_arid Inner Mongolian grassland. The analysis of the recorded meteorological data demonstrated that the increases in temperature were synchronous with slight decreases in precipitation in the growing season, which suggested that P. meyeri may reduce growth due to temperature_induced drought. On the other hand, the analysis for decadal periods explored that trees appeared to reduce the sensitivity to the warming, and consequently increased sensitivity to rainfall. This may serve as a baseline for more accurate predictions of the potential impacts of altered climate regimes on tree growth. 展开更多

Photochemical Efficiency of PSⅡ and Membrane Lipid Peroxidation in Leaves of indica and japonica Rice (Oryza sativa) Under Chilling Temperature and Strong Light Stress Conditions 被引量:18

Relationships between fluorescence parameters and membrane lipid peroxidation in leaves of indica and japonica rice (Oryza sativa L.) during later growth stage were studied under chilling temperature and strong light stress conditions. Results showed that D1 protein contents of PSⅡ in photosynthetic apparatus dropped, the generation of antheraxanthin (A) and zeaxanthin (Z) of xanthophyll cycle were inhibited partly, PSⅡ photochemical efficiency (F v/F m)and non-photochemical quenching (q N) were also decreased obviously. In addition, endogenous active oxygen scavenger—superoxide dismutase (SOD) reduced, superoxide anion radical (O -· 2) and malondialdehyde (MDA) accumulated, as a result, photooxidation of leaves occurred under chilling temperature and strong light stress conditions. Obvious differences in the changes of the above mentioned physiological parameters between indica and japonica rice were observed. Experiments in leaves treated with inhibitors under chilling temperature and strong light conditions showed that indica rice was more sensitive to chilling temperature with strong light and subjected to photooxidation more than japonica rice. Notable positive correlation between D1 protein contents and F v/F m or (A+Z)/(A+Z+V), and a marked negative correlation between F v/F m and MDA contents were obtained by regression analysis in indica and japonica rice during chilling temperature and strong light conditions. According to the facts mentioned above, it was inferred that PSⅡ photochemical efficiency(F v/F m) was the key index to forecast for the prediction of photooxidation under stress circumstances and the physiological basis were the synthetic capacity of D1 protein and the protection of xanthophyll cycle. 展开更多

This species was thought to be intermediate between P. bungeana Zuec. ex Endl. of subgen. Strobus and P. yunnanensis Franch. of subgen. Pinus by the original author. We made an effort to address the question of the phylogenetic relationship of this peculiar and important species in the context of the major groups of the genus Pinus by using sequences of the rbcL, matK,genes, rpl20-rps18 spacer, trnV intron of the chloroplast genome and the nuclear ribosomal ITS region. The results of the separated analysis and the combined analysis of the four cpDNA sequences and ITS sequence indicated that P. squamata was a stable member of subsect. Gerardianae and P. gerardiana Wall. was the closest species of P. squamata phylogenetically. As a result, the causes of the distribution pattern of subsect. Gerardianae were also discussed. 展开更多

[Objective]The α subunit gene of phycobiliprotein from Spirulina maxima was studied in order to provide a basis for the subsequent study of phycobiliprotein.[Method] Amino acids composition,signal peptides,hydrophobicity/hydrophilicity and trails-membrane topological structure of α subunit gene of phycobiliprotein from Spirulina maxima which registered in GenBank(GenBank AF441177) were analyzed and predicted by the tools of bioinformatic analysis.Meanwhile,phylogenetic tree was constructed based on α subunit gene of phycobiliprotein from Spirulina maxima,and its molecular evolution was also analyzed.[Result]The phycobiliprotein was rich in amino acids,which not only contained 18 kinds of essential amino acids,but also contained some non-essential amino acids like glycine,aspartic acid,etc.;Analysis on signal peptides and trails-membrane topological structure showed that the phycobiliprotein belonged to intracellular protein;Analysis on hydrophobicity/hydrophilicity showed that the phycobiliprotein belonged to hydrophilic protein;Phylogenetic analysis showed that the phycobiliprotein had a high homology with Arthrospira,which reached 99%-100%.[Conclusion]The study provided a certain reference for studying the relationship and interaction between α subunit and β subunit. 展开更多

Expression of Survivin in HL-60 Cells Treated with Chemotherapeutic Drugs

Objective: To explore the relationship between survivin and drug resistance, and the changes of the survivin expression in HL-60 cells treated with three kinds of chemotherapeutic drugs. Methods: HL- 60 cells were treated with appropriate concentration of daunomycin (DNR), mitoxantrone (MIT) or arsenic trioxide (As2O3). The expression of survivin mRNA and protein on the first or third day was detected by RT-PCR and Western blot respectively. Results: The expression of survivin mRNA was decreased on the first day by 10% in DNR-treated group, 40% in MIT-treated group (P〈0.01) and 25% in As2O3-treated group (P〈0.01) respectively. On the third day, the expression of survivin mRNA in DNR- and MIT-treated group was up-regulated to 120% (P〈0.05) and 165% (P〈0.01) respectively as compared with that on the first day, but down-regulated to 68% in As2O3-treated group (P〈0.01). As compared with control group, the expression of survivin protein in DNR- or MIT-treated group was increased by 14% or 11% on the third day respectively, but it was decreased by 18% in As2O3-treated group. Conclusion: In DNR- and MIT-treated group, the expression of surivin was decreased at first and then increased obviously, which may be one of the causes for resistance to chemotherapy against leukemia. Different from other two drugs, As2O3 may play an important role in restoring chemotherapy sensitivity. 展开更多

The experiments w... The vacuum aluminothermic reduction of the mixture of calcined magnesite and calcined dolomite was studied. An isothermal reduction method satisfying the vacuum aluminothermic reduction was proposed. The experiments were carried out at 4 Pa. The results indicate that the reduction rate is increased with increasing temperature, content of aluminum and pellet forming pressure. The XRD patterns of pellets at different reduction stages confirm that the reduction process can be roughly classified into three stages:the formation of MgAl2O4, and Ca12Al14O33 phases;the phase transformation from MgAl2O4 and C12A7 to CaAl2O4;the formation of CaAl4O7 phase. The experimental data were divided into three parts according to the kinetic models. The apparent activation energies of the three parts were determined to be 98.2, 133.0 and 223.3 kJ/mol, respectively. 展开更多

Objective: To observe whether there is evidence for vascular channel formation by osteosarcoma cellsin vitro and to illustrate mechanism of vasculogenic mimicry in osteosarcoma.Methods: Osteosarcoma cell lines (U-2OS) were tested for their ability to form tubular networks in three-dimensional culture containing type I collagen. The structures of the tubular networks were observed under a phase contrast microscope and an electron microscope.Results: Observation under light microscopy and electron microscopy showed that high aggressive osteosarcoma cells line (U-2OS) formed networks containing channels when grown in three-dimensional culture containing type I collagen in the absence of endothelial cells or fibroblasts.Conclusion: These observations strongly suggest that aggressive osteosarcoma cells may generate vascular channels that facilitate tumor perfusion independent of tumor angiogenesis and have the ability of vasculogenic mimicry. Pharmacokinetics of Ivermectin in Salvelinus leucomaenis Following Two Ways of Administration

[Objective] This study was aimed to study the pharmacokinetics of iver- mectin in Salvelinus leucomaenis following a single oral administration or intraperi- toneal injection. [Method] After a single oral administration or intraperitoneal injection of ivermectin with a dosage of 0.3 mg/kg body weight, samples were taken at dif- ferent time points and determined by high-performance liquid chromatography-ultravi- olet detection (HPLC-UV). The pharmacokinetic parameters were calculated by 3p97 software. [Result] The concentration-time relations of ivermectin in plasma, muscle, liver and kidney were well described by one-department open model with first-order absorption. After oral administration, the pharmacokinetic parameters in plasma were as follows: Tmax=4.503 h, Cmax=0.252mg/L, t1/2ka=0.476 h, t1/2ka=331.160 h, AUC=121.524 (mg/L) .h. After intraperitoneal injection, the pharmacokinetic parameters in plasma were as follows: Tmax=2.751 h, Cmax=0.230 mg/L, t1/2ka=0.306 h, t1/2ka=153.868 h, AUC= 51.689 (mg/L) .h. [Conclusion] There were differences in the pharmacokinetics of ivermectin in Salvelinus leucomaenis between two different administration routes, and intraperitoneal injection led to more rapid absorption than oral administration. 展开更多

Freecalcium ([Ca^(2+])i) was determined by Fura-2... To investigate apoptosis in mouse leukemia cell (WEHI-3) induced by Econazoleand its mechanism. Methods: Apoptosis induced by Econazole was examined by flow cytometry. Freecalcium ([Ca^(2+])i) was determined by Fura-2 fluorescein load technique. The protein was isolatedfrom endoplasmic reticulum of WEHI-3 cells, and then the expression of caspase-12 and caspase-7 wasdetected by Western blot. Results: WEHI-3 cells exhibited typical change of apoptosis when they weretreated by Econazole. [Ca^(2+)]i was significantly higher in Econazole-treated group than incontrol group. The expression of caspase-12 and caspase-7 was increased with the increase ofEconazole concentration. Conclusion: Caspase-12 may play a key role in WEHI-3 apoptosis induced byEconazole. 展开更多

The experiment of rearing Chouioia cunea with Tussah (Silkworm) pupa were carried out by 搕hree-cut method?and inoculating method. The results showed that three-cut method is effective way for breed-ing Chouioia cunea, with a parasitical rate of 95%. The biological characteristics and the life cycle of Chouioia cunea were ob-served and described and more hosts of Chouioia cunea were found. 揟hree-cut method?as a new technique of rearing Chouioia cunea has been put into practice. 展开更多
