To clarify which factors may influence pathological tumor response and affect clinical outcomes in patients with locally advanced rectal carcinoma treated with neo-adjuvant chemoradiotherapy and surgery.METHODSTumor regression grade (TRG) according to the Dworak system and yTNM stage were assessed and correlated with pre-treatment clinico-pathological variables in 215 clinically locally advanced (cTNM stage II and III) rectal carcinomas. Prognostic value of all pathological and clinical factors on disease free survival (DFS) and cancer specific survival (CSS) was analyzed by Kaplan Meier and Cox-regression analyses.RESULTScN+ status, mucinous histotype or poor differentiation in the pre-treatment biopsy were significantly associated with lower pathological response (low Dworak grade and TNM remaining unchanged/upstaging). Cases showing acellular mucin pools in surgical specimens all had unremarkable clinical courses with no deaths or recurrences during follow-up. Dworak grade had prognostic significance for DFS and CSS. However, compared to the 5-tiered system, a simplified two-tiered grading system, in which grades 0, 1 and 2 were grouped as absent/partial regression and grades 3 and 4 were grouped as total/subtotal regression, was more reproducible and prognostically informative. The two-tiered Dworak system, yN stage, craniocaudal extension of the tumor and radial margin status were significant independent prognostic variables.CONCLUSIONOur data suggest that caution should be applied in using a conservative approach in rectal carcinomas with cN+ status, extensive/lower involvement of the rectum and mucinous histotype or poor differentiation. Although Dworak TRG is prognostically significant, a simplified two-tiered system could be preferable. Finally, cases with acellular mucin pools should be carefully evaluated to definitely exclude residual mucinous carcinoma. 展开更多

Isolated pancreatic granulocyte sarcoma is extremely rare. We report a very unusual pancreatic granulocytic sarcoma in a patient without acute myeloid leukemia. The patient presented with acute epigastric pain because of splenic infarction due to a mass consisting of myeloblasts in the pancreatic tail. The patients underwent splenectomy and distal pancreatectomy. Pathology and immunohistochemistry suggested a GS. Despite local surgery, an isolated tumor recurred 2 mo after operation and the patient died 3 mo after removal of the tumor. Only 7 reported cases of pancreatic GS were identified in the literature and the mass was located in the pancreatic head. This is the first report of GS in the pancreatic tail with splenic infarction. 展开更多

Some well-preserved conifer leafy shoots and female cones from the Lower Cretaceous Changcai For- mation near Fudong of Helong, eastern Jilin, Northeast China are studied. Based on gross morphological and cuticular study, a new species, Elatides helongensis Sun et Zhao ( sp. nov. ) is described systematically. The new species is characterized by persistent, linear or slightly falcate leaves with obtusely acute apex, attached helically on the shoots. Female cones of the new species are terminal and oval, composed of persistent helically- arranged rhomboidal scales and erect seeds. One erect seed is growing on each scale. Leaf cuticles are hyposto- matic. Monocylic stomata are ellipse, composed of 2 sunken guard ceils and 4-8 subsidiary cells. Moreover, the cuticles of a young female cone of Pityostrobus yingchengensis Yang are described for the first time. 展开更多

The objective of this study was characterizing the gelatin from the chicken feet by FTIR (Fourier transform infrared) spectroscopy. Two gelatin samples were prepared for the experiment which were extracted of 200 g of chicken feet by a thermal bath of 4% acetyl acid solution at 60 ℃. A commercial gelatin was used to compare the results. FTIR spectra of gelatin samples were recorded using a Nicolet iS5 FTIR spectrometer equipped with an ATR/iD3 with argon horizontal cell in which the spectra were varied in the range of 400-4,000 cm1 at 16 ~C. The results showed that FTIR spectra of the chicken feet gelatin had the most with the vibration peak at wave numbers of 1,652.01 cm~ to the amide I, of 1,539.87 cm1 to the amide II, of 1,241.29 cm~ to the amide III, of 2,923.72 cm-~ to the amide B and of 3,399.56 cm~ to the amide A. The collagen composition of the chicken feet gelatins was twotimes larger than the commercial gelatin and the vibration bands to beef gelatin have been different from chicken feet gelatins. This demonstrates that chicken feet gelatins are very good nutritional quality when compared to the commercial gelatin. 展开更多

It showed that the cortical cytoskeleton was composed of non-ciliature structure, ciliature structure including adoral zone of membranelles (AZM), undulating membranes (UM), frontal-ventral-transverse cirri (FVTC), caudal cirri (CC), dorsal kineties, as well as base-associated microtubules. The silverline system is composed of longitudinal and transverse microtubules, concave structure appeared on the cell dorsal side, the base of DK containing rosette-like skeleton structure. All these suggested that the non-ciliature structure, ciliature structure of Euplotes be quite different from that of other species of ciliate, the silverline-system be true pare of cortical microtubute cytoskeleton in Euplotes, rosette structure be part of the basecytoskeleton. 展开更多

Antibody-based fusion proteins are the next generation of antibody therapies for cancer and other diseases.CD20 antigen,which is overexpressed on cell membranes in nearly 95% of cases of B-cell Non-Hodgkin's Lymphoma,is an attractive target for the therapy of B-lymphoid malignancies.Lidamycin (LDM) is a potent enediyne-containing antitumor antibiotic that now has entered phase II clinical trials.In this study,we prepared an engineered fusion protein,scFv-LDP,consisting of an anti-CD20 scFv fragment and the apoprotein LDP of LDM using DNA recombination.After purification and refolding,scFv-LDP was found to bind specifically to CD20-positive lymphoma cells using ELISA and indirect immunofluorescent cytochemical staining assays.The energized fusion protein scFv-LDP-AE was obtained using molecular reconstitution of the active chromophore AE of LDM and scFv-LDP.MTT assay revealed potent cytotoxicity of scFv-LDP-AE to CD20-positive Raji and Daudi cells,with IC 50 values of 1.21×10-11 and 6.24×10-11 mol L-1,respectively.An in vivo subcutaneous xenograft model of CD20-positive B cell lymphoma in BALB/c (nu/nu) mice was also utilized.Drugs were given intravenously on day 14 and 21 after tumor transplantation.In terms of maximal tolerated doses,scFv-LDP-AE at 0.3 mg kg-1 suppressed tumor growth by 79.3%,and LDM at 0.05 mg kg-1 by 68.6% (P<0.05).Results suggested scFv-LDP-AE could be a potential candidate for tumor-targeting therapy. 展开更多

Regulatory Action of Charred Gossamer Urocteae on the Functions of Mouse Oral Fibroblasts

Objective: To explore the influence of charred Gossamer urocteae (CGU) on the functions of primary cultured mouse oral fibroblasts and reveal its mechanism in wound healing. Methods: CGU was extracted with different solvents and ethanol extract (EE), ethyl acetate fraction (EF), n-butanol fraction (BF) and aqueous fraction (AF) were obtained. The effects of different fractions on the proliferation, matrix metaUoproteinase-2,9 (MMP-2,9) activities, synthesis of collagen and tissue inhibitor of metalloproteinase 1 (TIMP-1) in the mouse oral fibroblasts were determined by MTT, gelatin zymography, chloramine-T method, and enzyme-linked immunosorbent assay (ELISA) respectively. Results: EE, EF and BF at high concentrations could significantly inhibit proliferation of fibroblasts (P〈0.05 or P〈0.01), and at low concentrations EF and BF could promote proliferation of fibroblasts, and BF and AF could significantly inhibit collagen synthesis (P〈0.05 or P〈0.01). EE, EF and AF at high concentrations could significantly increase the MMP-9 activity, and BF and AF could significantly inhibit synthesis of TIMP-1. Conclusion: CGU at high concentrations can inhibit the proliferations of fibroblasts and synthesis of collagen, and in healing of wound, CGU at high concentrations possibly has the functions of anti-fibrosis and anti-scar, and the mechanism to promote degradation of collagen is possibly related to the increase in MMP-9 activity and the inhibition of TIMP-1 synthesis. 展开更多
