METHODS:Pancreatic acini were prepared from male Wistar rats. Isolated acinar cells were suspended in Eagle's MEM solution. After adding drugs, the incubation was performed at 37 degrees for a set period of time. Amylase of supernatant was assayed using starch-iodide reaction. Isolated acinar single cell was incubated with Fura-2/AM at 37 degrees, then cells were washed and resuspended in fresh solution and attached to the chamber. Cytoplasm [Ca2+]i of a single cell was expressed by fluorescence ratio F340/F380 recorded in a Nikon PI Ca2+ measurement system. RESULTS: Rate course of amylase secretion stimulated by SA(I) in rat pancreatic acini appeared in bell-like shape. The peak amplitude increased depended on SA(I) concentration. The maximum rate responded to 1 x 10(-5)mol/L SA(I) was 13.1-fold of basal and the rate decreased to basal level at 30 min. CCK-8 receptor antagonist Bt(2)-cGMP markedly inhibited amylase secretion stimulated by SA(I) and the dose-effect relationship was similar to that by CCK-8. [Ca2+]i in a single acinar cell rose to the peak at 5 min after adding 5 x 10(-6)mol/L SA(I) and was 5.1-fold of basal level. In addition, there was a secondary increase after the initial peak. GDP could inhibit both the rate of amylase secretion and rising of [Ca2+]i stimulated by SA(I) in a single pancreatic acinar cell. CONCLUSION: SA(I) is highly efficient in promoting the secretion of enzymes synthesized in rat pancreatic acini and raising intracellular [Ca2+]i. Signaling transduction pathway of SA(I) involves activating special membrane receptor and increase in cytoplasm [Ca2+]i sequentially.展开更多

Methods:Cultured human laryngeal cancer cellline Hep-2 and human vascular endothelial cells in vitro, cells got into the period of exponential phase of growth, was diviced into 3 groups:group I (control group), group II (DDP group), group III (Rg3 group). Added to the Hep-2 cells Rg3 and DDP, made Rg3 final concentration was 300μg/mL, and DDP was 3μg/mL. 48 h later, specimens from sample to be done immunocytochemistry, and the protein of VEGF in Hep-2 cells to be detected. Col ecting Hep-2 cells supernatant, some was used to measure the protein level of VEGF in Hep-2 cells supernatant by ELISA. Some was used to culture HVEC. 24 h later, cellgrowth inhibition rate of human vascular endothelial was determined by MTT. Results:The protein level of VEGF was evi-dently higher in group I compared to group II and group III, it was not only in Hep-2 cells, but also in supernatant of Hep-2 cells. There was no significantly dif erent between group II and group III. MTT results showed that, the human vascular endothelial cellgrowth inhibition rate of group I was significantly lower than that of group II and group III (P〈0.05). At the same time the HVEC growth inhibition rate of group II was significantly lower than that of group III (P〈0.05). Conclusion:The inhibition to tumor angiogenesis of Rg3 is stronger than traditional chemotherapy drug cisplatin. It worke by reducing the biological ef ects of secreted VEGF, But the ef ecting worke by reducing the activity of secreted VEGF itself or af ecting endothelial function of VEGF receptor or some other ways to be further studied.展开更多

The main reason is the error judgment of the intermediate layer suspended between the aqueous and organic layer during the liquid–liquid extraction process. This study shows that the intermediate layer consists of lots of medium- to long-chain carboxylic salts for their special physical and chemical properties. An improved saponification extraction method is also developed and the results show that the carboxylic salts distributed in the intermediate layer could be obtained completely, which greatly enhances the authenticity and accuracy of fatty acid analysis. Additionally, the possible reasons of formation of the intermediate layer are also discussed.展开更多

It is widely planted for windbreaks and soil erosion control and largely recommended like food of cattle since it can provide a source of fodder, protein and metabolic energy. Its flowers are incredibly attractive for pollinating insects and thus, form a source of pollen and nectar for honey. This tree was introduced in Europe in 1700 and was introduced by the colonists into Algeria in 1949. In order to valorize the natural substances of this species, the evaluation of the composition of its fruits (pods and seeds separately) in polyphenols, condensed tannins and hydrolysable tannins, which considered as anti-nutritional substances has been done. The obtained results showed that the fruits of Gleditsia triacanthos L. contain very low values in anti-nutritional factors with (0.13%-0.03%) of condensed tannins, (0.78%-0.45%) of hydrolysable tannins and (0.44%-0.16%) of total phenols for pods and seeds, respectively. It arises that these fruits can be of a great interest for many industries especially for feedstock.展开更多

It has been widely used in China for non-small cell lung cancer (NSCLC). However, there are only a few clinical trials to study the effectiveness of Rg3 on NSCLC, and almost them are small- samples, so we performed a meta-analysis on the results of the studies we collected in order to investigate the effectiveness of Rg3 on NSCLC. METHODS A meta-analysis was conducted in all the selected randomized controlled trials evaluating the effectiveness of Rg3 on NSCLC patients. All on-line databases regarding Rg3 from 1950 to 2011 were searched. Supplemental hand searching of the references of retrieved articles was performed. RESULTS Six trials met the inclusion criteria. Four of them compared chemotherapy plus Rg3 with chemotherapy alone, and the other 2 compared chemotherapy plus Rg3 with chemotherapy plus placebo. These trials are homogeneous. Two of the trials report overall survival, but the data are not suitable for a meta-analysis. After meta-analysis was conducted in the included studies comparing the effects of chemotherapy plus Rg3 with that of chemotherapy alone or chemotherapy plus placebo, it was suggested that chemotherapy plus Rg3 increased the response rate [odds ratio: 2.64 (95% Ch 1.70-4.11), fixed effects model] and disease control rate [odds ratio: 3.34 (95% CI: 1.92-5.81); fixed effects model] of the patients at stage II-IV, especially for the patients at stage Ill-IV. CONCLUSION Meta-analysis of the available evidence suggests that Rg3 plus chemotherapy improves the response rate of NSCLC patients, and well-designed RCTs with large sample size are needed.展开更多

This technology can effectively separate the starch and cellulose from Dioscorea Zingiberensis, thus the output of diosgenin is raised by 20%. Meanwhile using the suspension after separation, the synergy of enzymes, which are under the best condition for enzymolysis is studied, and an enzymolysis model, which uses two enzymes at the same time, is established. This model provides a theoretical basis for finding new enzyme resources. What's more orthogonal experiments show that by using this model, either the output of diosgenin can be increased by 50.58% or the amount of acid used can be reduced by 83% which means less pollution.展开更多

The RP-HPLC assay was performed on a reversed-phase C18 column with a gradient elution. The mobile phase consisted of acetonitrile and water containing 0.1% (v/v) phosphoric acid. The flow rate was 1.0 mL/min and the detection wavelength was set at 210 nm. The six marker constituents were separated well with good linearity (r2〉0.9996), precision, stability and repeatability. The overall recoveries were in the range of 99.00%-104.52%. Cluster analysis was employed to analyze 15 batches of samples. The result indicated this method provide an efficient way to perform quality control as well as a scientific rationale for the Geo-authentication of I. pubescens.展开更多

Methods The trial was a double-blind, placebo-controlled, parallel- group design. The eligible patients were randomized into 2 treatment groups: sarsasapogenin group (sarsasapogenin plus risperidone for 8 weeks, n = 41) and placebo group (risperidone only for 8 weeks, n = 39). At the baseline, as well as at weeks 2, 4 and 8 of treatment, the therapeutic response was measured by using scales including Positive and Nega- tive Symptoms Scale (PANSS), Wechsler Memory Scale (WMS), modified Chinese Wechsler Adult Intelligence Scale (mWAIS), Clinical Global Impression (CGI) and Brief Psychiatry Rating Scale (BPRS). The study period for each subject was 8 weeks and duration of overall trial was 2 years. Results Patients treated with sarsasapogenin plus risperidone demonstrated no statistically significant differences in changes in PANSS, WMS or mWAIS score at the end-point of the trial compared with patients treated with placebo plus risperidone. The incidence of treatment-emergent adverse events in patients treated with sarsasapogenin was not different from that observed in placebo group. Conclusion Sarsasapogenin did not augment the efficacy of risperidone in treating negative symptoms dominated schizophrenia. Sarsasapogenin at a dosage of 200 mg per day added to a flexible dosage of risperidone at 2-4 mg per day is safe and well tolerated by patients with negative symptoms dominated schizophrenia.展开更多

The aim ofpresent study is to investigate the protective effect ofPanax notoginseng saponins (PNS) against chronic ethanol-induced hepaticsteatosis in vivo. Mice were pair-fed a modified Lieber-DeCarli liquid diet containing alcohol or isocaloric maltose dextrin ascontrol diet with or without PNS (200 mg/kg, BW) for 8 weeks. Animals supplemented with PNS were protected against hepaticlipid accumulation induced by chronic ethanol exposure. Accordingly, PNS could significantly decrease the elevation of plasmatriglyceride, plasma enzyme activities, i.e. alanine aminotransferase (ALT) and aspartate aminotransferase (AST), and hepaticTNF-ct and IL-6 levels which were induced by chronic alcohol exposure. In addition, PNS markedly reduced the lipolysis ofwhite adipose tissue (WAT) that stimulated by alcohol feeding through the inhibiting protein expression of phosphorylation ofhormone-sensitive lipase (p-HSL), rather than total HSL. Furthermore, alcohol exposure also enhanced fatty acid uptake capacityin liver by elevated hepatic CD36 expression, which could attenuated by PNS treatment. These results demonstrate that PNSsupplementation protects against chronic ethanol-induced hepatic steatosis, which is associated with ameliorating dysfunctionallipid metabolism of WAT and the reduced inflammatory cytokines. Our findings suggested that PNS might be potential to bedeveloped as an effective agent for the treatment of chronic alcoholic steatosis.展开更多

文摘With the rapid increase of water contamination, membrane separation technology and their corresponding molecular weight cut-off(MWCO) evaluation method become more necessary. In this study,Panax notoginseng saponins was used as a new standard marker to determinate ultrafiltration(UF) membrane MWCOs,series of Millipore membranes were selected as control group to analyze and calculate the relationship between retention rate and MWCOs with exponential or logarithmic equation.A new and convenient method was provided for determining the membrane MWCO by modeling analysis retention rate with MWCOs,and the regression coefficients >0.990.The feasibility and practicability of established method was verified by different manufactures' membrane and dextrans. In the detection progress,as the main ingredient of Panax notoginseng saponins,Notoginsenoside R_1,Ginsenoside Rg_1,Ginsenoside Rb_1 and Ginsenoside Rd with different surface activity, the MWCO range of UF membranes can be divided into two zones mainly due to the retention rate difference among Notoginsenoside R_1,Ginsenoside Rg_1,Ginsenoside Rb_1 and Ginsenoside Rd.Zone 1,1000-10000;and Zone II,10000-100000.Thus,the new method would be helpful to improve the applicability of UF membrane in separation technology.

文摘AIM: This investigation was to reveal the characteristics and mechanism of enzyme secretion and increase in [Ca2+]i stimulated by saikosaponin(I) (SA(I)) in rat pancreatic acini. Signaling transduction pathway of SA(I) involves activating special membrane receptor and increase in cytoplasm [Ca2+]i sequentially.

文摘The application of waste alkali liquids as a substitute of sodium hydroxide for the saponification to improve the collection performance of fatty acids was investigated by saponification reaction test and flotation test. The results of the saponification reaction test indicated that the optimal conditions for the saponification were stirring rate of 55 r/min,initial temperature of 40℃ and stirring time of 45 min.Meanwhile, the laboratory scale and industrial scale flotation experiments showed that the fatty acid salt synthesized by wastewater achieved an index comparable to fatty acid sodium synthesized by sodium hydroxide. As a consequence,it was feasible to replace sodium hydroxide with the wastewater from zeolite production for fatty acid saponification. The cross-border utilization of waste alkali liquids not only reduced environmental pollution, but also produced excellent economic benefits.
文摘Objective:The aim of this study was to investigate the af ecting of Rg3 to secreted VEGF of human laryngeal carcinoma Hep-2 cells and its mechanism of inhibition to tumor angiogenesis. It worke by reducing the biological ef ects of secreted VEGF, But the ef ecting worke by reducing the activity of secreted VEGF itself or af ecting endothelial function of VEGF receptor or some other ways to be further studied.

文摘The conventional saponification method could result in lower recoveries and artificial changes of longchain fatty acids. Additionally, the possible reasons of formation of the intermediate layer are also discussed.
文摘Honey locust (Gleditsia triacanthos L.), also called sweet-locust or thorny-locust, is a moderately fast growing tree. It arises that these fruits can be of a great interest for many industries especially for feedstock.
文摘OBJECTIVE Ginsenoside Rg3 (Rg3) has shown anti-tumor effects on various tumor cells. CONCLUSION Meta-analysis of the available evidence suggests that Rg3 plus chemotherapy improves the response rate of NSCLC patients, and well-designed RCTs with large sample size are needed.

文摘Based on a through study of the chemical compositions of Dioscorea Zingiberensis, a new separation technology is presented in this paper. What's more orthogonal experiments show that by using this model, either the output of diosgenin can be increased by 50.58% or the amount of acid used can be reduced by 83% which means less pollution.

文摘A reversed-phase high performance liquid chromatography (RP-HPLC) method with diode array detector (DAD) has been established to simultaneously determine six bioactive compounds in the roots ofllex pubescens, namely tortoside A, (+)-syringaresinol, ilexsaponin B3, ilexsaponin A1, ilexsaponin B1, ilexgenin A. The result indicated this method provide an efficient way to perform quality control as well as a scientific rationale for the Geo-authentication of I. pubescens.

文摘Objective To identify whether sarsasapogenin, a sapogenin from the Chinese medicinal herb Anemarrhena Asphodeloides Bunge, would augment the efficacy of risperidone and significantly improve cognitive functions in patients with negative symptoms dominated schizophrenia. Sarsasapogenin at a dosage of 200 mg per day added to a flexible dosage of risperidone at 2-4 mg per day is safe and well tolerated by patients with negative symptoms dominated schizophrenia.

文摘Chronic alcohol consumption induces hepatic steatosis, the early stage of alcoholic liver disease (ALD). Our findings suggested that PNS might be potential to bedeveloped as an effective agent for the treatment of chronic alcoholic steatosis.
