目的:观察黄皮叶乙醇提取物对非酒精性脂肪肝大鼠肝组织ATP、ADP含量的影响,旨在探讨黄皮叶醇提物对肝组织能量代谢的干预作用,为中药防治非酒精性脂肪肝提供实验依据。方法:采用灌饲高脂肪乳剂方法制备非酒精性脂肪肝大鼠模型,用黄皮... 目的:观察黄皮叶乙醇提取物对非酒精性脂肪肝大鼠肝组织ATP、ADP含量的影响,旨在探讨黄皮叶醇提物对肝组织能量代谢的干预作用,为中药防治非酒精性脂肪肝提供实验依据。方法:采用灌饲高脂肪乳剂方法制备非酒精性脂肪肝大鼠模型,用黄皮叶乙醇提取物灌胃给药4周,高效液相色谱法(HPLC)测定观察黄皮叶醇提取对肝组织ATP、ADP含量的影响。结果:ATP回归方程Y=0.138X+0.413,在1.0~100.0μg/ml范围内线性关系良好;ADP回归方程Y=0.283X+1.177,在5.0~160.0μg/ml范围内线性关系良好;与对照组比较,非酒精性脂肪肝模型组大鼠肝组织ATP、ADP含量明显降低(P<0.05);与模型组比较,黄皮叶250~1000mg/kg灌胃给药能提升NAFLD大鼠肝组织ATP、ADP的含量,其中低剂量组(250mg/kg)的ADP含量显著升高(P<0.05),中、高剂量组(500 mg/kg、1000mg/kg)的ATP、ADP含量均显著升高(P<0.05)。结论:本研究建立的检测ATP、ADP含量的HPLC法快速、简便、准确,可为生物组织中能量物质的评价提供依据。黄皮叶醇提取物可提高NAFLD大鼠肝组织ATP、ADP的含量,有效改善NAFLD后肝组织能量代谢障碍,从而保护肝功能。 展开更多

Effects of Ca^(2+)concentrations on accumulations of mineral elements in the components of Pteroceltis tatarinowii 被引量:6

The bark of Pteroce/tis tatarinowii is a raw material for manufacturing XuanPaper. The effects of Ca^(2+) concentrations on the accumulation of mineral elements in the bark,leaf and root of Pteroceltis tatarinowii were studied under controlled conditions. The types ofHoagland nutrient solution with three Ca^(2+) concentrations levels (200, 400 and 600 μg·g^(-1))and a control (without Ca^(2+)) were designed to culture Pteroceltis tatarinowii. After 6 months,contents of seven mineral elements including Ca, K, Mg, Mn, Zn, Cu and Na in the root, leaf and barkwere analyzed. The results indicated that Ca accumulations content in the root, leaf and bark hadpositively relation with Ca^(2+) concentrations (200, 400, 600 μg · g^(-1)), and the order of theCa content in the three components was root>leaf>bark. Ca content in the root treated with 600 μg·g^(-1) Ca^(2+) concentrations was 5.5 times as high as that of the control, and about 1.4 times ashigh as that of the root treated in 200 and 400 μg/g Ca^(2+) concentrations respectively. On thecontrary, K and Mg contents in the root, leaf and bark were negatively related to Ca^(2+)concentrations, especially in the bark, and their accumulation trend followed the order ofleaf>root>bark. K content in the bark treated with 600 μg ·g^(-1) Ca^(2+) concentrations was 39.3%of that of the control, and was 79.0% and 91.8% of that of the bark treated with 200 μg ·g^(-1)and 400 μg ·g^(-1) Ca^(2+) concentrations respectively; Mg content in the bark treated with 600μg ·g^(-1) Ca^(2+) concentrations was 23.4% of that of the control, and was 27.1% and 35.4% ofthat of the bark treated with 200 and 400 μg·g^(-1) Ca^(2+) concentrations respectively. Comparedwith the control, the general tendency of Mn, Zn and Cu content decreased with increasing of Ca^(2+)concentrations and their contents were in the order: root>leaf>bark. Based on the results of thisstudy, the experiment has been useful for providing academic bases in improving the bark quality ofPteroceltis tatarinowii on non-limestone soil. 展开更多

For finding valuable systematic characters, leaf epidermis of 77 taxa, representing 12 sections of the genus Clematis and three related genera in the Ranunculaceae, were examined mainly by means of light microscopy (LM), and partially by scanning electron microscopy (SEM). It was shown that the leaf epidermal cells were usually irregular or polygonal in shape. The patterns of anticlinal walls were straight, arched or undulate. The stomatal apparatus is anomocytic and exists in abaxial epidermis of all species, and in the adaxial epidermis of some species. Under SEM observation, the leaf epidermis was often striated, seldom nearly smooth, and often with flakes attached. Evidence from leaf epidermis serves as a criterion for distinguishing the subsections in sect. Meclatis (Spach) Tamura and in sect. Fruticella Tamura. The results also support that there are several separate evolutionary processes in the genus Clematis . 展开更多

Objective The ventral part of the medial prefrontal cortex(mPFC)plays an important role in initiation and control of voluntary movement,mood and cognition.However,after the degeneration of the nigrostriatal pathway,the neuronal activity of the ventral mPFC and the role of serotonin1A(5-hydroxytryptamine,5-HT1A)receptors in the firing of the neurons are still unknown.The present study is to investigate the change of neuronal activity in the ventral mPFC and the effect of systemic administration of the selective 5-HT1Areceptor antagonist WAY-100635 on the activity of the neurons in normal and 6-hydroxydopamine(6-OHDA)-lesioned rats.Methods Single unit responses were recorded extracellularly with glass microelectrodes from ventral mPFC neurons in normal rats and 6-OHDA unilaterally lesiond rats in vivo.Results 6-OHDA lesion of the substantia nigra pars compacta(SNc)significantly increased the firing rate with no change in the firing pattern of neurons of the ventral mPFC in rats.Systemic administration of WAY-100635(0.1 mg/kg,i.v.)did not change the mean firing rate and firing pattern of ventral mPFC neurons in normal rats.In contrast,WAY-100635 signifi- cantly decreased the mean firing rate of the neurons in rats with 6-OHDA lesion of the SNc.Conclusion These data suggest that the degeneration of the nigrostriatal pathway results in an increase of neuronal activity of ventral mPFC and dysfunction of 5-HT1Areceptor. 展开更多

The leaf epidermis of Japanese honeysuckle (Lonicera japonica Thunb.) and Wild Honeysuckle (Lonicera confusa) in the genus of Flos Lonicerae were mainly observed by scanning electron microscopes (SEM) to study the characteristics of stomata, trichomes and dermal cell, etc.. The results showed that stoma exists only on the lower epidermis and its distribution is irregular, and leaf epidermis consist of epidermis cells, stoma complexes and bushy trichomes including glandular hair and non-glandular hair. On the upper epidermis, anticlinal wall caves in sinuous groove to countercheck the transpiration. Evidences from leaf morphological structures serve as another proof on drought-resistant mechanisms. Some strumaes distributing regularly are hypothesized as oxalic calcium on the lower epidermis under laser scanning confocal microscopy (LSCM) with Fluo-3/AM, which can increase their endurance to drought stress. Therefore, the above characteristics of Flos Lonicerae can reduce the loss of water and make Japanese honeysuckle and Wild Honeysuckle adapt to the droughty environment at Karst area in southwest China. However, there is some difference of the two species. From the SEM (Scanning Electron Microscopy) result, it is shown that on the upper epidermis, some glandular hair regularly present along the midrib of Japanese honeysuckle, but Wild Honeysuckle has no glandular hair on the upper epidermis, which can verify the relationships of Flos Lonicerae species and provide the significance for classification of Flos Lonicerae. 展开更多

Fluorescence spectra of native purple bacterial reaction center (RC) and bacterial pheophytin (Bphe) replaced RCs were obtained from 600 nm to 900 nm with a selective excitation at 597 nm. With the help of measuring the fluorescence from bacterial chlorophyll, bacterial pheophytin and plant pheophytin, the corresponding components in the RCs are classified for fluorescence emission. Results showed that pheophytin substitution influences the composition of fluorescence spectra. Therefore, four, three and two components were obtained from fluorescence spectra of native RC, Bphe B_replaced RC and Bphe A,B _replaced RC, respectively. Fluorescence components are well correlated to the binding of plant pheophytin. The decay of excited state of primary electron donor P in different RCs was also studied by measuring the fluorescence decay at 686.4, 674.1 and 681.1 nm, respectively. The decaying kinetics changed in different RCs, indicating that pheophytin replacement influenced the energy transduction and primary photochemical reaction in purple bacterial reaction centers. 展开更多

[Objective] The aim of this study was to investigate the effects of salt stress on cell expansion in Arabidopsis thaliana rosette leaves.[Method] Arabidopsis seedlings were treated by sodium chloride at the concentration of 0,100 or 150 mmol/L. At the 7th and 14th d of treatment,with nail enamel printing mark method and computer software,the leaf blades area and abaxial epidermal pavement cells area was measured and compared using statistical analysis in Excel. [Result] The growth of Arabidopsis rosette leaves was inhibited under salt stress. Leaves treated for 7 or 14 d expanded less compared with controls. The salt-mediated decrease in leaf expansion is associated with a decrease in abaxial pavement cell expansion. [Conclusion] The decreased leaf and epidermal cell expansion under salt stress is the most important characteristic of plant physiological response to salt stress. 展开更多

These leaf cushions are very similar to Lepidodendron lepidophloides Yao both in size and morphology. They probably belong to the same species in different preservative condition. Comparisons on morphology and anatomy of L. lepidophloides and lepidodendralean leaf cushion genera, such as Lepidodendron, Lepidophloios, Sigillaria and Cathaysiodendron revealed that L. lepidophloides possesses mixed features of all these genera and it can not be assigned into any of them without doubt. It probably represents a new genus. Because the anatomical features of the other parts of the stem as well as the fertile organs related to these leaf cushions are unknown, the specific name L. lepidophloides is adopted. We leave this taxonomic problem open till the relationship between these leaf cushions and the stem and fertile organs become clear. This is the first report on the detailed anatomical features of the Cathaysian lepidodendralean leaf cushions. 展开更多

The orbitofrontal cortex (OFC) is particularly important for the neural representation of reward value. Previous studies indicated that electroencephalogram (EEG) activity in the OFC was involved in drug administration and withdrawal. The present study investigated EEG activity in the OFC in rats during the development of food reward and craving. Two environments were used separately for control and food-related EEG recordings. In the food-related environment rats were first trained to eat chocolate peanuts; then they either had no access to this food, but could see and smell it (craving trials), or had free access to this food (reward trials). The EEG in the left OFC was recorded during these trials. We showed that, in the food-related environment the EEG activity peaking in the delta band (2-4 Hz) was significantly correlated with the stimulus, increasing during food reward and decreasing during food craving when compared with that in the control environment. Our data suggests that EEG activity in the OFC can be altered by food reward; moreover, delta rhythm in this region could be used as an index monitoring changed signal underlying this reward. 展开更多
