Objective:To examine the level of expression of anti skin aging gene of Ganoderma lucidum polysacchayides and clarify its mechanism with anti aging of this ancient Chinese medicine.Methods:HacaT cell of keratinocytes ...Objective:To examine the level of expression of anti skin aging gene of Ganoderma lucidum polysacchayides and clarify its mechanism with anti aging of this ancient Chinese medicine.Methods:HacaT cell of keratinocytes lines were cultured and treated with the polysaccharides.The total RNA was extracted with Trizol reagent and cDNA was synthesized by reverse thanscription.The obtained cDNAs were then fluorescently labeled with cy3 and cy5 respectively and hybridized with gene expressing pedigree cDNA chip.The images were scanned and analyzed with special software.The scan data were analyzed with software and checked by real time PCR.Results:Among total 18 346 human genes,the expression of 103 ones was up-regulated and 378 ones down-regulated.It was demonstrated evidently that Ganoderma lucidum polysaccharides affected the expression of genes of anti skin aging.Two ways are anastomotic.Conclusion:it is concluded by analysis of function of these up-regulation and down-regulation genes that Ganoderma lucidum polysaccharides may play an important role in boosting cell growth and against skin aging.It shows that the results of gene array reliable by real time PCR.展开更多

PSC was characterized as a type I collagen, and its imino acid content w...Characteristics and antioxidant activities of pepsin-soluble collagen (PSC) from yellow goosefish (Lophius litulon) skins were investigated. PSC was characterized as a type I collagen, and its imino acid content was 193 residues/1 000 residues. PSC's denaturation temperature was -17.56℃ and Fourier transform infrared spectra confirmed the presence of triple helices. Solubility analysis showed good solubility at acidic pH (1-6) or low NaCl concentrations (≤2%). PSC showed scavenging activity against hydroxyl radicals and superoxide anions in a concentration-dependent manner. Furthermore, PSC could protect D-galactose-induced skin aging by significantly controlling malondialdehyde formation and improving the activity of superoxide dismutase, glutathione peroxidase, catalase, glutathione, and hydroxyproline. PSC may be a promising antioxidant in appropriate applications.展开更多

This study investig...Objectives: The burn repair associated with diabetes complications showed considerable changes in the healing process and the development of alternative therapies to favor the repair is important. This study investigated the efficacy of the Casearia sylvestris in the burn repair in diabetic and non-diabetic rats. Methods: The animals were divided into four groups (n = 20): (C) non-diabetics treated with carbopol gel; (G) non-diabetic treated with C. sylvestris extract in carbopol gel; (DM-C) diabetics treated with carbopol gel; (DM-G) diabetics treated with C. sylvestris extract in carbopol gel. Burn was induced using a metal plate (2 cm diameter/120 °C/20 sec). Samples were collected on the 3rd, 7th, 14th and 21st days after the injury for histomorphometric analysis (inflammatory infiltrate, fibroblasts, blood vessels and collagen), MPO (Myeloperoxidase) and NAG (N-Acetilglicosaminidase) dosage, hydroxyproline, glycosaminoglycans, TGF-β1, VEGF, collagen I and III. Key findings: C. sylvestris decreased the inflammatory process during the experimental period in diabetic and non-diabetic animals which has been demonstrated by MPO, NAG quantification, and TGF-β1 expression. C. sylvestris also increased blood vessels and decreased VEGF expression during the studied period in non-diabetic animals. Fibroplasia and collagenase were increased in all experimental periods in the DM-G group by means of de hydroxyproline quantification, collagen I, collagen III and glycosaminoglycans. Conclusions: C. sylvestris application can modulate favorably the time of inflammation and collagenous process in the repair of burn injuries in diabetic rats.展开更多
OBJECTIVE:To reveal the effects of Guasha(scraping therapy) on the histomorphology of scraped skins and on the expressions of calcitonin gene-related peptide(CGRP) and substance P(SP).METHODS:50 rats,as experimental s...OBJECTIVE:To reveal the effects of Guasha(scraping therapy) on the histomorphology of scraped skins and on the expressions of calcitonin gene-related peptide(CGRP) and substance P(SP).METHODS:50 rats,as experimental subjects,were randomly divided into 5 groups according to different observation time points.Dorsal setae were shaved for the exposure of skin on both sides of the spine.Even reinforcing and reducing method was applied to the rats in Guasha groups on the site equivalent to bladder meridian of human body on one side of the spine,and the skin was scraped from top to bottom until rash of measles occurred.The skin tissues with rash of measles were taken down after perfusion.The corresponding sites of rats in group A were also taken down as control.The tissues were made to sections and used for immunofluorescence histochemical staining and HE staining of antibodies such as SP and CGRP.RESULTS:After Guasha,there were significant differences in appearance,hair follicle and blood vessel on local skin scraped on the back of rats when compared with control;In different time points,the differences reduced.There was no significant difference in expression of CGRP and SP when compared local skin scraped on the back of rats in different time points.CONCLUSION:Guasha didn't significantly change the morphology of nerve fibers inside local skin,and the histomorphology of hair follicle,blood vessel and etc.However,after Guasha they basically returned to normal level within five days.展开更多

Methods: The subacute senile model of mouse was made by subcutaneous injection of D-galactose into the neck, then they were randomly allocated into an acupuncture group, a moxibustion group, a model group, and a blank group, and measured the content of malondialdehyde (MDA), activity of glutathione peroxidase (GSH-Px), and content of hydroxyproline (Hyp) in the skin tissue. Results: Acupuncture and moxibustion at Zusanli (ST 36) could decrease the content of MDA, enhance the activity of GSH-Px, and increase the content of Hyp in the skin tissue of senile mice. Conclusion: Acupuncture and moxibustion at Zusanli (ST 36) could delay the senescence progress of mice's skin.展开更多

Saline control was the group injected by saline. HMG model were created by injection of estrogen and progestogen. Moxibustion group was also injected of estrogen and progesto-gen with moxibustion at the same time. The Changes of nipple diameter and height were measured.The rats' skin temperature was recorded by an infrared thermal camera at the nipples, mammary areas,Danzhong(CV 17) and Ganshu(BL 18). Pathological changes of mammary gland in rats were also observed under light microscope.RESULTS: The diameter and height of the nipples in model group were prominently bigger and higher than that in control group(P < 0.01). The diameter and height in moxibustion group were prominently smaller and lower than that in model group(P < 0.01), and there was no significant difference between moxibustion group and control group.Compared with control group, skin temperature of the nipples, mammary area, and acupoints Danzhong(CV 17) and Ganshu(BL 18) decreased prominently in model group(P < 0.01-0.05). Compared with model group, skin temperature of that in moxibustion group increased prominently(P <0.05).CONCLUSION: Treatment with moxibustion can effectively decrease the HMG rats' nipple diameter and height, and increase the skin temperature in HMG model rats at the nipples, mammary areas,Danzhong(CV 17) and Ganshu(BL 18). This study convinces the therapeutic effect of moxibustion on mammary gland hyperplasia.展开更多

Methods: The 18-month-old senile Wistar rats were randomly divided into three groups, a blank group, a surrounding needling group, a surrounding needling plus Zusanli (ST 36) group, comparing with the 8-month-old young rats. The tests included the content of dissoluble hydroxyproline (Hyp) and the activity of superoxide dismutase (SOD) in the skin of rats' abdomen. The transmission electron microscope was used for the morphological observation of the fibroblasts and the arrangement of the collagen fiber. Results: The dermal organelles in the fibroblasts of the rats decreased with the cellular structures. Meanwhile, the spaces among the collagen fibers extended and the aging bridging increased. The surrounding needling improved the activity of the fibroblasts. The dissoluble Hyp content in the surrounding needling group was much higher than the rats in the blank group, but the SOD activity didn't change significantly. However, the surrounding needling plus Zusanli (ST 36) enhanced the SOD activity. Conclusion: Acupuncture may improve the activity of the fibroblasts in the skin to increase the dissoluble collagen content, resulting in the improvement of an aged appearance. Surrounding needling plus needling Zusanli (ST 36) could enhance the SOD activity.展开更多
