Objective:The biological characteristics of rabbit oral mucosal epithelial cells(OMECs) were studied and the suitable procedure for the culture of OMECs was explored.Methods:Cultured OMECs by different surface disinfection,by different Dispase Ⅱ preparation,in different medium,in different calciumion concentration of K-SFM medium.Results:Microbial contamination of cell culture could significantly reduce with 2% iodine solution used for sterilization of oral cavity before cell culture.The OMECs digested with Dispase Ⅱin K-SFM could attach to the culture flask quickly,the OMECs digested with Dispase Ⅱ in PBS rare attached to the culture flask.The proliferations of OMECs cultured in 0 or 0.09 mmol/L calcium concentrations of K-SFM medium showed not statistically significant.the OMECs grew fast with serum,but it was easy to differentiate into fibroblast-like cells.Conclusion:OMECs could grow well and had the typical oral mucosal epithelial cell morphology by the procedure that described in this experiments. 展开更多

Objective:Bovine endometritis is one of the most common reproductive disorders in cattle.The aim of this study was to investigate the anti-inflammation potential of punicalagin in lipopolysaccharide(LPS)-induced bovine endometrial epithelial cells(bE ECs)and to uncover the underlying mechanisms.Methods:bE ECs were stimulated with different concentrations(1,10,30,50,and 100μg/ml)of LPS for 3,6,9,12,and 18 h.MTT assay was used to assess cell viability and to identify the conditions for inflammatory injury and effective concentrations of punicalagin.Quantitative real-time polymerase chain reaction(q RT-PCR)was used to assess gene expression of pro-inflammatory cytokines.Western blotting was used to assess levels of inflammation-related proteins.Results:Treatment of b EECs with 30μg/ml LPS for 12 h induced cell injury and reduced cell viability.Punicalagin(5,10,or 20μg/ml)pretreatment significantly decreased LPS-induced productions of interleukin(IL)-1β,IL-6,IL-8,and tumor necrosis factor-α(TNF-α)in bE ECs.Molecular research showed that punicalagin inhibited the activation of the upstream mediator nuclear factor-κB(NF-κB)by suppressing the production of inhibitorκBα(IκBα)and phosphorylation of p65.Results also indicated that punicalagin can suppress the phosphorylation of mitogen-activated protein kinases(MAPKs)including p38,c-Jun N-terminal kinase(JNK),and extracellular signal-regulated kinase(ERK).Conclusions:Punicalagin may attenuate LPS-induced inflammatory injury and provide a potential option for the treatment of dairy cows with Escherichia coli endometritis. 展开更多

Relation between eardrum flap area and healing outcome of traumatic eardrum perforation

Objective: To retrospectively study the clinical effects of eardrum flap area on the healing outcome following traumatic perforation.Methods: Totally 291 traumatic eardrum perforations with in-/everted edges were included in this study. They were randomly divided into three groups and received conservative treatment, epidermal growth factor (EGF) via Gelfoam patching, or edge-approximation plus Gelfoam patching respectively. Patients in each group were further divided into two subgroups according to the eardrum flap area ≤ 1/2 or 〉1/2 of the perforation size. The healing rate and mean closure time after tympanic membrane perforation were evaluated at three months.Results: Of the total 291 participants, 281 were included in the final statistical analysis. The area of curled edge did not affect the healing outcome significantly in any groups (P〉0.05). The healing rate varied slightly: 90.7% vs 92.3% in spontaneous healing group, 98.2% vs 97.4% in EGF via Gelfoam patching group, and 96.5% vs 100% in edge-approximation plus Gelfoam patching group. In addition, in all groups the area of curled edge did not affect the mean closure time significantly (P〉0.05). The closure time was (32.3±2.4) d vs (30.6±3.1) d in sponaneous healing group, (13.4±2.5) d vs (13.1± 1.9) d in EGF via Gelfoam patching group, and (11.9±3.1) d vs (12.2±2.1) d in edge-approximation plus Gelfoam patching group.Conclusion: The eardrum flap area of traumatic eardrum perforation does not significantly affect the clinical outcomes. 展开更多
