[Method] The enzymatic digestion method, scraping method, serial filtration and differential velocity adherent technique were used to isolate BESC and BEGEC, then immunocytochemical method and TRYPAN-Blue assay were used to determine the purity and survival rate of isolated cells. [Result] The BESC and BEGEC were successfully isolated and cultured while immunocytochemical method and cell count method demonstrated that the purity was over 90%. The result of TRYPAN-Blue assay shown that survival rate of BESC and BEGEC was 91% and 78% respectively. [Conclusion] The enzymatic digestion method, scraping method, serial filtration and differential velocity adherent technique could isolate BESC and BEGEC with high purity.展开更多

We present two cases of PCI associated with massive peritoneal free air and/or retroperitoneal air that mimicked perforated peritonitis. These cases highlight the clinical importance of PCI that mimics perforated peritonitis, which requires emergency surgery. Preoperative imaging modalities and diagnostic laparoscopy are useful to make an accurate diagnosis.展开更多
AIM: To explore the effects of ultrasound exposure combined with microbubble contrast agent (SonoVue) on the permeability of the cellular membrane and on the expression of plasrnid DNA encoding enhanced green fluor...AIM: To explore the effects of ultrasound exposure combined with microbubble contrast agent (SonoVue) on the permeability of the cellular membrane and on the expression of plasrnid DNA encoding enhanced green fluorescent protein (pEGFP) transfer into human umbilical vein endothelial cells (HUVECs). METHODS: HUVECs with fluorescein isothiocyanatedextran (FD500) and HUVECs with pEGFP were exposed to continuous wave (1.9 MHz, 80.0 mW/cm^2) for 5 min, with or without a SonoVue. The percentage of FD500 taken by the HUVECs and the transient expression rate of pEGFP in the HUVECs were examined by fluorescence microscopy and flow cytornetry, respectively. RESULTS: The percentage of FDS00-positive HUVECs in the group of ultrasound exposure combined with SonoVue was significantly higher than that of the group of ultrasound exposure alone (24.0%± 5.5% vs 66.6% ± 4.1%, P 〈 0.001). Compared with the group of ultrasound exposure alone, the transfection expression rate of pEGFP in HUVECs was markedly increased with the addition of SonoVue (16.1% ± 1.9% vs 1.5% ± 0.2%, P 〈 0.001). No statistical significant difference was observed in the HUVECs survival rates between the ultrasound group with and without the addition of SonoVue (94.1% ± 2.3% vs 91.1% ± 4.1% ). CONCLUSION: The cell membrane permeability of HUVECs and the transfection efficiency of pEGFP into HUVECs exposed to ultrasound are significantly increased after addition of an ultrasound contrast agent without obvious damage to the survival of HUVECs. This non- invasive gene transfer method may be a useful tool for clinical gene therapy of hepatic tumors.展开更多

In recent decades the application of stomatal frequency data that are relied...Leaf cuticle analysis has long been a powerful tool for fossil plant identification, systematics, and palaeoclimatological recon- struction. In recent decades the application of stomatal frequency data that are relied on precise calculation of stomata on plant fossil cuticles to reconstruct ancient atmospheric CO2 concentration made the preparation of cuticular membrane with sufficient size a critical technique in palaeoclimatological research. However, for plants with originally thin and fragile cuticles, e.g., most deciduous plants, conventional techniques sometimes fail to obtain cuticular membranes with sufficient size, or sometimes unable to recover any. This has largely hampered the usage of fossil cuticle analysis in palaeobotanical and palaeo- climatological research. Here, we describe a new method using clear nail polish as a medium to "strengthen" the originally thin and fragile cuticles prior to maceration procedures. We demonstrate the method by using middle Eocene Metasequoia fossils that were notorious for the difficulty of recovering large-sized clean cuticular membranes due to their thin and fragile nature. Metasequoia, with well-documented and widely-distributed fossil records since the Late Cretaceous and with a living repre- sentative, 114. glyptostroboides, as a comparative reference, bas been widely used as a model genus for the study of evolution of plants, palaeoclimatological reconstruction, and plant adaptation to climate changes. But its deciduous habit produces thin cuticles and makes the preparation of clean cuticular membranes a tedious process. The new method successfully allows us to recover its delicate cuticular membranes with sufficient sizes for SEM observation and stomatal frequency analysis.展开更多
