We investigated the complete mitochondrial genome(mitogenome) of Argyreus hyperbius.The 151 56 bp long genome harbored the gene content(13 protein coding genes,22 tRNA genes,2 rRNA genes and an A+T-rich region) a...We investigated the complete mitochondrial genome(mitogenome) of Argyreus hyperbius.The 151 56 bp long genome harbored the gene content(13 protein coding genes,22 tRNA genes,2 rRNA genes and an A+T-rich region) and the gene arrangement was identical to all known lepidopteran mitogenomes.Mitogenome sequence nucleotide organization and codon usage analyses showed that the genome had a strong A+T bias,accounting for A+T content of 80.8%,with a small negative AT skew(?0.019).Eleven intergenic spacers totaling 96 bp,and 14 overlapping regions totaling 34 bp were scattered throughout the whole genome.As has been observed in other lepidopteran species,12 of the 13 protein-coding genes(PCGs) were initiated by ATN codons,while the COI gene was tentatively designated by the CGA codon.A total of 11 PCGs harbored the complete termination codon TAA,while the COI and COII genes ended at a single T residue.All of the 22 tRNA genes showed typical clover structures except that the tRNASer(AGN) lacks the dihydrouridine(DHU) stem which is replaced by a simple loop.The intergenic spacer sequence between the tRNASer(AGN) and ND1 also contained the ATACTAA motif,which is conserved in all other lepidopterans as well.Additionally,the 349 bp A+T-rich region was not comprised of large tandem repetitive sequences,but harbored a few structures common to other lepidopteran insects,such as the motif ATAGA followed by a 20 bp poly-T stretch,a microsatellite-like(AT)9 element preceded by the ATTTA motif,and a 5 bp poly-A site present immediately upstream of tRNAMet.The mitochondrial genomic sequence features found in this study not only contribute to genetic diversity information of the group,but also are useful in future studies of the endangered nymphalid butterfly in population genetic dynamics,species conservation,phylogeography and evolution.展开更多

Using reciprocal cross, self-cross, and backcross, w...To fully understand the tomato ovary locule formation of inheritance, two varieties, ovary multi-locule (MLK1) and few-locule (FL1), were used in these studies. Using reciprocal cross, self-cross, and backcross, we have constructed various groups to seek the genetic law and mechanisms of tomato locule formation, for modifying the tomato breeding theory. The parental and the resulting FI and F2 generations have been used to examine the heredity of the locule number. The results showed that few-locule was incompletely dominant. The data from backcross demonstrated a significant effect on the locule number, whereas, the reciprocal cross showed an insignificant effect. Not surprisingly, the locule number was controlled by nuclear genomes, not by exogenous substances. The model that inherited the locule number belonged to the additive-dominant model: additive effects played a very important role and were partially dominant. In addition, the results revealed that the locule number in tomato was mainly controlled by a single gene, whereas, it was modulated by a number of other genes. Finally, the general inheritability and narrow inheritability of the locule number were 69.44% and 52.98%, respectively.展开更多

Fifteen taxa, of which eleven were ingroups representing ten of the eleven families, were selected to produce a phylogenetic inference of nepomorphan superfamilial or familial relationships via parsimony and Bayesian techniques. Our results supported the monophylies of Nepomorpha, Naucoroidea (Aphelocheiridae + Naucoridae), Nepoidea (Belostomatidae + Nepidae), Ochteroidea (Ochteridae + Gelastocoridae) and Pieoidea (Pleidae + Helotrephidae); the most basal lineage of Ochteroidea; Notonectoidea contained Notonectidae only and formed a new sister relationship with (Pleoidea + Naucoroidea); and the sister relationship of (Nepoidea + Corixoidea). The Hox genes are suitable molecular markers to resolve phylogenetic relationships at superfamily or family levels in Heteroptera.展开更多

JF437925) was determined as a circular DNA molecule of 15 242 bp, with common genes of 13 putative proteins, 2 rRNAs, and 22 tRNAs and of the same gene arrangement as in other sequenced lepidopterans. All protein-coding genes had the typical start codon ATN, except for the COI's using CGA as its start codon as previously demonstrated in other lepidopteran species. The comparison of the nucleotide sequences of the A. ilia mitogenome with ten other Nymphalidae species showed nearly identical gene orientation and arrangement, with only a few alterations in non-coding fragments. The nucleotide composition and codon frequency all fell into the range estimated for the order Lepidoptera. The A. ilia mitochondrial genome had the canonical set of 22 tRNA genes folded in the typical cloverleaf structure, with an unique exception of tRNAs^r (AGN). The mitochondrial genes from A. ilia were overlapped in a total of 33 bp at 9 locations, as well as interleaved with a total of 155 bp intergenic spacers, spread over 12 regions with the size ranging from 1 to 49 bp. Furthermore, the spacer between ND6 and Cyt b harbored a microsatellite-like repeat (TA)23 not found in other completely sequenced nymphalid genomes. The 403 bp AT-rich region harbored two conserved motifs (ATAGA, ATTTA), a 21 bp polyT stretch, a 10 bp poly-A region, along with two microsatellite-like repeats ( (TA)10 and (TA)7), as detected in other nymphalid butterflies.展开更多

[Method] The representative heat shock protein gene Bmhsp24.3 was screened by bioinfor- matic analysis method, and carried out real-time PCR expression analysis. [Result] The target gene Bmhsp24.3 expressed in different B. mori materials, but the expres- sion level in different materials significantly varied. The relative expression level of the gene had different degrees of changes under different rearing conditions. With the increase of rearing temperature, the gene expression was upregulated. The ma- terials with better healthiness had remarkable increase in expression of target gene, while the materials with poorer healthiness had less increase in expression of target gene. The expression difference of target gene Bmhsp24.3 was exactly consistent with the healthiness of breeds. [Conclusion] The healthiness of materials had rela- tionship with expression of target gene Bmhsp24.3. the higher the expression of tar- get gene Bmhsp24.3 was, the better the healthiness of materials was; conversely, the lower the expression of target gene Bmhsp24.3 was, the poorer the healthiness of materials was.展开更多
The phylogenetic relationships of four species,Bufo melanostictus,Hyla chinensis,Rana limnocharis and Rana guentheri,which belong to three differrent families of Anura,were detected with RAPD technique.The genomic DNA...The phylogenetic relationships of four species,Bufo melanostictus,Hyla chinensis,Rana limnocharis and Rana guentheri,which belong to three differrent families of Anura,were detected with RAPD technique.The genomic DNA of each species was amplified with 19 random primers.16 primers given clear amplified bands were used for analysis and the genetic distances between four species were calculated.The results show that RAPD bands obtained by all 16 primers evinced different degree polymorphisms.The genetic distance between R.limnocharis and R.guentheri is the nearest,that between B.melanostictus and H.chinensis is the second nearest,and that between B.melanostictus and R.guentheri is the furthest.The different distances also indicated that the relationship between Bufoidae and Hylidae is closer than that between Bufonidae and Ranidae at genomic DNA level.In concordant with the conclusion of the morphology,chromosomal and mitochondrial DNA studies,our results provide a new evidence of the systematic evolution of the three families of Anura at DNA molecular level.展开更多

The Expo 2010 Shanghai is taken as an example, and a structural equation model is employed to analyze the dynamic mechanism of trip mode choice behavior and the effectiveness of the transportation management measures at different stages. Based on the difference between the objective-oriented stated preference (SP) survey results and the objectives, together with the feedback from the previous stage survey, some adjustments on the transportation management measures are made in the next stage of the planning process until the objectives are eventually achieved. The results indicate that the adjustments on transportation management measures at different stages can effectively raise the transit share to 88.6%. Nonlocal visitors are inclined to choose nonstop modes of transportation and the companion attributes have the most significant effects on the trip mode choices of visitors. The research method is proved to be an effective way to support the decision making process of transportation management measures during planned special events in the future.展开更多

Thirdly, based on the theory of plates and shells, the simplified model is calculated and the theoretical calculation formula to determine the safe thickness of cave roof under pile tip can be obtained when the edges of the cave roof are simply supported. In order to resolve the coordination and optimization of the power network planning effectively, on the basis of introducing the concept of power intelligence center (PIC), the key factor power flow, line investment a...In order to resolve the coordination and optimization of the power network planning effectively, on the basis of introducing the concept of power intelligence center (PIC), the key factor power flow, line investment and load that impact generation sector, transmission sector and dispatching center in PIC were analyzed and a multi-objective coordination optimal model for new power intelligence center (NPIC) was established. To ensure the reliability and coordination of power grid and reduce investment cost, two aspects were optimized. The evolutionary algorithm was introduced to solve optimal power flow problem and the fitness function was improved to ensure the minimum cost of power generation. The gray particle swarm optimization (GPSO) algorithm was used to forecast load accurately, which can ensure the network with high reliability. On this basis, the multi-objective coordination optimal model which was more practical and in line with the need of the electricity market was proposed, then the coordination model was effectively solved through the improved particle swarm optimization algorithm, and the corresponding algorithm was obtained. The optimization of IEEE30 node system shows that the evolutionary algorithm can effectively solve the problem of optimal power flow. The average load forecasting of GPSO is 26.97 MW, which has an error of 0.34 MW compared with the actual load. The algorithm has higher forecasting accuracy. The multi-objective coordination optimal model for NPIC can effectively process the coordination and optimization problem of power network.展开更多

[ Result ]Allium macrostemon Bunge in Laizhou City introduced in this study was tdploid; its somatic chromosome number was 24 and karyotype formula was K(2n) =3x=24m(2SAT) + 1Bs, thus the karyotype belongs to 1A type. One of the chromosome No. 3 contained satellite, and chromosome deletion may be existed in one of the chromosome No. 5. In addition, B chromosome was observed in some cells.[ Conclusion] This introduction of triploid A. macrostemon found in China was the first time.展开更多

Phylogenetic analysis based on ge- netic distance from related gene sequences of 10 flatfish showed that this method was ideal to explore the rela- tionship between species, genera and families. Phylogenetic trees set-up is based on neighbor-joining, maximum parsimony and maximum likelihood methods that accords to the general rule of Pleuronectiformes evolution. But they also resulted in some confusion. Unlike data from morphological characters, P. olivaceus clustered with K. bicoloratus, but P. cinnamomeus did not cluster with P. olivaceus, which is worth further studying.展开更多

Methods: To obtain HBVE, the...Objective: The aim of the study was to observe the transfection efficacy of hepatitis B virus envelope (HBVE) and evaluate its ability as a gene transfer vector for liver cancer cells. Methods: To obtain HBVE, the supematant fluid of HepG 2.2.15 cells was mixed with a PEG8000 solution for concentration and was inactivated by β-propiolactone. The acquired HBVE was used to pack plRES2-EGFP to test its package ability. Then, we examined its quantity and quality with ELISA, PCR, SDS-PAGE and electron microscopy. The plRES2-EGFP was packed with HBVE and obtained the product HBVE-GFP. The plRES2-EGFP was packed with liposome and obtained the product liposome-GFP. HBVE-GFP and liposome-GFP were used to transfer HepG 2 cells to study the transfection efficiency. HBVE-GFP was used to transfer HepG 2, A549, HeLa and FB cells to study the targeting ability. The green fluorescent protein (GFP) expression was observed under a fluorescent microscope. The rate of GFP positive cells was determined by flow cytometry. Results: 1. The acquired HBVE could retain the surface protein HBsAg + pre S1 + pre S2 and had no virus DNA. It had good package ability for plRES2-EGFP. 2. Transfection efficiency: The GFP could be observed in both the liposome group and HBVE group under the fluorescent microscope. But the HBVE group had a higher fluorescent intensity than liposome group. The transfection rate of liposome group was 49.97% + 2.37% while the HBVE group was 70.65% + 3.15% and the fluorescent intensity of the HBVE group was 3-4 times (P = 0.000) for liposome group with the determination of flow cytometry. 3. Targeting ability: The GFP could be observed in the four groups under the fluorescent microscope. The HepG 2 group had the highest fluorescent intensity among the four groups. The transfection rate of HepG 2 group was 71.35% + 0.03% which was highly expressed than other groups (P = 0.000) and the fluorescent intensity of the HepG 2 group was 2-3 times (P = 0.000) for the other 3 groups with the determination of flow cytometry. Conclusion: HBVE can be constructed successfully with the methods of PEG8000 and β-propiolactone from the supernatant fluid of HepG 22.15 cells. The HBVE can be a candidate gene transfer vector for liver cancer cells.展开更多

[Method] The representative heat shock protein gene Bmhsp24.3 was screened by bioinfor- matic analysis method, and carried out real-time PCR expression analysis. [Result] The target gene Bmhsp24.3 expressed in different B. mori materials, but the expres- sion level in different materials significantly varied. The relative expression level of the gene had different degrees of changes under different rearing conditions. With the increase of rearing temperature, the gene expression was upregulated. The ma- terials with better healthiness had remarkable increase in expression of target gene, while the materials with poorer healthiness had less increase in expression of target gene. The expression difference of target gene Bmhsp24.3 was exactly consistent with the healthiness of breeds. [Conclusion] The healthiness of materials had rela- tionship with expression of target gene Bmhsp24.3. the higher the expression of tar- get gene Bmhsp24.3 was, the better the healthiness of materials was; conversely, the lower the expression of target gene Bmhsp24.3 was, the poorer the healthiness of materials was.
文摘The phylogenetic relationships of four species,Bufo melanostictus,Hyla chinensis,Rana limnocharis and Rana guentheri,which belong to three differrent families of Anura,were detected with RAPD technique.The genomic DNA of each species was amplified with 19 random primers.16 primers given clear amplified bands were used for analysis and the genetic distances between four species were calculated.The results show that RAPD bands obtained by all 16 primers evinced different degree polymorphisms.The genetic distance between R.limnocharis and R.guentheri is the nearest,that between B.melanostictus and H.chinensis is the second nearest,and that between B.melanostictus and R.guentheri is the furthest.The different distances also indicated that the relationship between Bufoidae and Hylidae is closer than that between Bufonidae and Ranidae at genomic DNA level.In concordant with the conclusion of the morphology,chromosomal and mitochondrial DNA studies,our results provide a new evidence of the systematic evolution of the three families of Anura at DNA molecular level.
文摘In this paper, an improved radial basis function networks named hidden neuron modifiable radial basis function (HNMRBF) networks is proposed for target classification, and evolutionary programming (EP) is used as a learning algorithm to determine and modify the hidden neuron of HNMRBF nets. The result of passive sonar target classification shows that HNMRBF nets can effectively solve the problem of traditional neural networks, i. e. learning new target patterns on line will cause forgetting of the old patterns.

文摘This paper aims to provide a decision-making method for the transportation management strategies in guiding the transformation of trip mode choice during planned special events. The Expo 2010 Shanghai is taken as an example, and a structural equation model is employed to analyze the dynamic mechanism of trip mode choice behavior and the effectiveness of the transportation management measures at different stages. Based on the difference between the objective-oriented stated preference (SP) survey results and the objectives, together with the feedback from the previous stage survey, some adjustments on the transportation management measures are made in the next stage of the planning process until the objectives are eventually achieved. The results indicate that the adjustments on transportation management measures at different stages can effectively raise the transit share to 88.6%. Nonlocal visitors are inclined to choose nonstop modes of transportation and the companion attributes have the most significant effects on the trip mode choices of visitors. The research method is proved to be an effective way to support the decision making process of transportation management measures during planned special events in the future.

文摘Cave roofs are used to support pile foundation in many engineering projects. Accurate stability analysis method of cave roof under pile tip is important in order to ensure the safety of the pile foundation structure. Firstly the mechanical model to analysis the stability of cave roof under pile tip is founded aiming to solve the problems that the simplified mechanical model has. Secondly, the boundary of cave roof is simply supposed to be supported according to the integrity of the rock mass in the boundary of cave roof. Thirdly, based on the theory of plates and shells, the simplified model is calculated and the theoretical calculation formula to determine the safe thickness of cave roof under pile tip can be obtained when the edges of the cave roof are simply supported. In the end, the analysis of the practical engineering project proves the feasibility and the rationality of the method which can be a new method to calculate the safe thickness of cave roof under pile tip.
文摘The TGF-β superfamily members have important roles in controlling patterning and tissue formation in both invertebrates and vertebrates. Two types of signal transducers, receptors and Smads, mediate the signaling to regulate expression of their target genes. Despite of the relatively simple signal transduction pathway, many modulators have been found to contribute to a tight regulation of this pathway in a variety of mechanisms. This article reviews the negative regulation of TGF-β signaling with focus on its roles in vertebrate development.

文摘In order to resolve the coordination and optimization of the power network planning effectively, on the basis of introducing the concept of power intelligence center (PIC), the key factor power flow, line investment and load that impact generation sector, transmission sector and dispatching center in PIC were analyzed and a multi-objective coordination optimal model for new power intelligence center (NPIC) was established. To ensure the reliability and coordination of power grid and reduce investment cost, two aspects were optimized. The evolutionary algorithm was introduced to solve optimal power flow problem and the fitness function was improved to ensure the minimum cost of power generation. The gray particle swarm optimization (GPSO) algorithm was used to forecast load accurately, which can ensure the network with high reliability. On this basis, the multi-objective coordination optimal model which was more practical and in line with the need of the electricity market was proposed, then the coordination model was effectively solved through the improved particle swarm optimization algorithm, and the corresponding algorithm was obtained. The optimization of IEEE30 node system shows that the evolutionary algorithm can effectively solve the problem of optimal power flow. The average load forecasting of GPSO is 26.97 MW, which has an error of 0.34 MW compared with the actual load. The algorithm has higher forecasting accuracy. The multi-objective coordination optimal model for NPIC can effectively process the coordination and optimization problem of power network.

文摘[Objective]The study was to analyze the karyotype of Allium macrostemon Bunge in Laizhou City of Shandong Province.[ Method] The root tip of A. macrostemon was pretreated with 8-hydroxyquinoline solution, fixed, dissociated and stained for preparing the glass slide to reveal the chromosome number via the microscopic examination; the sparse cells with good chromosome morphology were photographed under microscope. [ Result ]Allium macrostemon Bunge in Laizhou City introduced in this study was tdploid; its somatic chromosome number was 24 and karyotype formula was K(2n) =3x=24m(2SAT) + 1Bs, thus the karyotype belongs to 1A type. One of the chromosome No. 3 contained satellite, and chromosome deletion may be existed in one of the chromosome No. 5. In addition, B chromosome was observed in some cells.[ Conclusion] This introduction of triploid A. macrostemon found in China was the first time.

文摘A 605 bp section of mitochondrial 16S rRNA gene from Paralichthys olivaceus, Pseudorhombus cinnamomeus, Psetta maxima and Kareius bicoloratus, which represent 3 families of Order Pleuronectiformes was amplified by PCR and sequenced to show the molecular systematics of Pleuronectiformes for comparison with related gene sequences of other 6 flatfish downloaded from GenBank. Phylogenetic analysis based on ge- netic distance from related gene sequences of 10 flatfish showed that this method was ideal to explore the rela- tionship between species, genera and families. Phylogenetic trees set-up is based on neighbor-joining, maximum parsimony and maximum likelihood methods that accords to the general rule of Pleuronectiformes evolution. But they also resulted in some confusion. Unlike data from morphological characters, P. olivaceus clustered with K. bicoloratus, but P. cinnamomeus did not cluster with P. olivaceus, which is worth further studying.

文摘Objective: The aim of the study was to observe the transfection efficacy of hepatitis B virus envelope (HBVE) and evaluate its ability as a gene transfer vector for liver cancer cells. Methods: To obtain HBVE, the supematant fluid of HepG 2.2.15 cells was mixed with a PEG8000 solution for concentration and was inactivated by β-propiolactone. The acquired HBVE was used to pack plRES2-EGFP to test its package ability. Then, we examined its quantity and quality with ELISA, PCR, SDS-PAGE and electron microscopy. The plRES2-EGFP was packed with HBVE and obtained the product HBVE-GFP. The plRES2-EGFP was packed with liposome and obtained the product liposome-GFP. HBVE-GFP and liposome-GFP were used to transfer HepG 2 cells to study the transfection efficiency. HBVE-GFP was used to transfer HepG 2, A549, HeLa and FB cells to study the targeting ability. The green fluorescent protein (GFP) expression was observed under a fluorescent microscope. The rate of GFP positive cells was determined by flow cytometry. Results: 1. The acquired HBVE could retain the surface protein HBsAg + pre S1 + pre S2 and had no virus DNA. It had good package ability for plRES2-EGFP. 2. Transfection efficiency: The GFP could be observed in both the liposome group and HBVE group under the fluorescent microscope. But the HBVE group had a higher fluorescent intensity than liposome group. The transfection rate of liposome group was 49.97% + 2.37% while the HBVE group was 70.65% + 3.15% and the fluorescent intensity of the HBVE group was 3-4 times (P = 0.000) for liposome group with the determination of flow cytometry. 3. Targeting ability: The GFP could be observed in the four groups under the fluorescent microscope. The HepG 2 group had the highest fluorescent intensity among the four groups. The transfection rate of HepG 2 group was 71.35% + 0.03% which was highly expressed than other groups (P = 0.000) and the fluorescent intensity of the HepG 2 group was 2-3 times (P = 0.000) for the other 3 groups with the determination of flow cytometry. Conclusion: HBVE can be constructed successfully with the methods of PEG8000 and β-propiolactone from the supernatant fluid of HepG 22.15 cells. The HBVE can be a candidate gene transfer vector for liver cancer cells.
