Bioinformatics w...Affymetrix U133A oligonucleotide microarrays were used to study the differences of gene expressions between high (H) metastatic ovarian cancer cell line, HO-8910PM, and normal ovarian tissues (C). Bioinformatics was used to identify their chromosomal localizations. A total of 1,237 genes were found to have a difference in expression levels more than eight times. Among them 597 were upregulated [Signal Log Ratio (SLR) ≥3], and 640 genes were downregulated (SLR≤-3). Except one gene, whose location was unknown, all these genes were randomly distributed on all the chromosomes. However, chromosome 1 contained the most differentially expressed genes (115 genes, 9.3%), followed by chromosome 2 (94 genes, 7.6%), chromosome 12 (88 genes, 7.1%), chromosome 11 (76 genes, 6.1%), chromosomes X (71 genes, 5.7%), and chromosomes l7 (69 genes, 5.6%). These genes were localized on short-arm of chromosome (q), which had 805 (65.1%) genes, and the short arms of No.13, 14, 15, 21, and 22 chromosomes were the only parts of the chromosomes where the differentially expressed genes were localized. Functional classification showed that most of the genes (306 genes, 24.7%) belonged to the enzymes and their regulator groups. The subsequent group was the nucleic acid binding genes (144 genes, 11.6%). The rest of the top two groups were signal transduction genes (137 genes, 11.1%) and proteins binding genes (116 genes, 9.4%). These comprised 56.8% of all the differentially expressed genes. There were also 207 genes whose functions were unknown (16.7 %). Therefore it was concluded that differentially expressed genes in high metastatic ovarian cancer cell were supposed to be randomly distributed across the genome, but the majority were found on chromosomes 1, 2, 12, 11, 17, and X. Abnormality in four groups of genes, including in enzyme and its regulator, nucleic acid binding, signal transduction and protein binding associated genes, might play important roles in ovarian cancer metastasis. [Method]The yield components and yield per plant of two cross combination of double-low hybrid rapeseed (B02, D04) and Shuza 6 were correlatively analyzed and compared, besides, the path analysis was also carried on to them. [Result] Among B02, D04 and Shuza No.6, effective pod number per plant and seeds per silique, seeds per pod and 1 000-grain weight were all negative correlation. In high yield hybrid, pod number per plant, seeds per pod had more impaction on yield per plant than 1 000-grain weight and the difference was at 0.01 significant level. In the control variety Shuza No.6, the impactions of pod number per plant and seeds per pod on yield per plant were bigger than that of 1 000-grain weight on yield per plant, however, the difference was not significant. [Conclusion] The improvement of effective pod number per plant was an important aim of breeding work of double low rapeseed breeding in Sichuan ecological region.展开更多
The contents of the soil microbial biomass nitrogen (SMBN) in the soils sampled from the Loess Plateau of China were determined using chloroform fumigation aerobic incubation method (CFAIM), chloroform fumigation anae...The contents of the soil microbial biomass nitrogen (SMBN) in the soils sampled from the Loess Plateau of China were determined using chloroform fumigation aerobic incubation method (CFAIM), chloroform fumigation anaerobic incubation method (CFANIM) and chloroform fumigation-extraction method (CFEM).The N taken up by ryegrass on the soils was determined after a glasshouse pot experiment. The flushes of nitrogen (FN) of the soils obtained by the CFAIM and CFANIM were higher than that by the CFEM, and there were significantly positive correlations between the FN obtained by the 3 methods. The N extracted from the fumigated soils by the CFAIM, CFANIM and CFEM were significantly positively correlated with the N uptake by ryegrass. The FN obtained by the 3 methods was also closely positively correlated with the plant N uptake. The contributions of the SMBN and mineral N and mineralized N during the incubation period to plant N uptake were evaluated with the multiple regression method. The results showed that the N contained in the soil microbial biomass might play a noticeable role in the N supply of the soils to the plant.展开更多
Compound 209 is a newly synthesized dithiocarbamate derivative with antiproliferation activity in vitro, however, its antitumor effect in vivo and the underlying mechanisms have yet to be identified. We explored the a...Compound 209 is a newly synthesized dithiocarbamate derivative with antiproliferation activity in vitro, however, its antitumor effect in vivo and the underlying mechanisms have yet to be identified. We explored the antitumor effect of compound 209 and the possible mechanisms for its inhibition of the growth of HT-29 xenograff tumor and proliferation of HT-29 cells. Cell proliferation was evaluated with SRB assay in vitro. The results showed that compound 209 had significant antiproliferation activity on HT-29 cells. Furthermore, the xenograff HT-29 nude mouse model was used to study the antitumor effect of compound 209 in vivo. We found that compound 209 significantly inhibited tumor growth and did not cause loss of body weight or leukocytopenia. Analysis by flow cytometry indicated that compound 209 arrested HT-29 cell cycle in G~ phase. Western blotting analysis suggested that compound 209 increased the expression of p27, cyclin E, CDK2, cyclin D1 and CDK4. These results demonstrated the antitumor effect of compound 209 and its potential use as an anticancer drug.展开更多
