以荧光高分子为相分离载体和指示物的免疫分析新方法

以6-甲氧基-β-(5-乙烯基-2-奎宁环基)-4-喹啉甲醇(奎尼丁,Qd)为单体合成了一种pH敏感荧光高分子聚N-异丙基丙烯酰胺(NIP)-奎尼丁-N,N,-二甲基氨丙基甲基丙烯酰胺(DMAPM)[P(NIP-DMAPM-Qd)]。采用共聚法将布氏杆菌抗原(BrAg)与P(NIP-DMAP... 以6-甲氧基-β-(5-乙烯基-2-奎宁环基)-4-喹啉甲醇(奎尼丁,Qd)为单体合成了一种pH敏感荧光高分子聚N-异丙基丙烯酰胺(NIP)-奎尼丁-N,N,-二甲基氨丙基甲基丙烯酰胺(DMAPM)[P(NIP-DMAPM-Qd)]。采用共聚法将布氏杆菌抗原(BrAg)与P(NIP-DMAPM-Qd)连接,制备了P(NIP-DMAPM-Qd)-BrAg连接物。加入待测布氏杆菌抗体(BrAb),免疫反应后,调节至pH7.6,使高分子相变,从而将体系中的免疫反应复合物P(NIP-DMAPM-Qd-BrAg)-BrAb和未反应连接物P(NIP-DMAPM-Qd)-BrAg与非特异性组分以及未反应的待测BrAb分离.利用蛋白A对抗体的亲和性,捕获P(NIP-DMAPM-Qd-BrAg)-BrAb,从而将其与P(NIP-DMAPM-Qd-BrAg)分开,通过测定P(NIP-DMAPM-Qd-BrAg)-BrAb的荧光来定量BrAb。该方法通过高分子相变分离和蛋白A捕获双重分离作用,基本消除了非特异性组分以及未反应的特异性免疫成分等的干扰;以高分子自身荧光为检测信号源,无须另外的标记物,大大提高了免疫分析的灵敏度和简便性。以布氏杆菌抗体(BrAb)为检测对象,测得线性范围为0~168ng/L,抗体检出限为1.2ng/L,结果令人满意。 展开更多

The feasibility of a new method for separating arsenic from arsenic-antimony-bearing dusts using Cu S was put forward,in which Sb was transformed into Sb2O4 and Sb2S3 that stayed in the roasted calcine while As was volatilized in the form of As4O6.The factors such as roasting temperature and Cu S addition amount were studied using XRD,EPMA and SEM-EDS.Cu S has an active effect on the separation of arsenic due to the destruction of(Sb,As)2 O3 structures in the original dust and the simultaneous release of As in the form of As4O6.At a roasting temperature of 400°C and Cu S addition amount of 130%,the volatilization rates of arsenic and antimony reach 97.80 wt.%and 8.29 wt.%,respectively.Further,the high As volatile matter can be used to prepare ferric arsenate after it is oxidized,with this treatment rendering the vapor harmlessness. 展开更多

In a study carried out in pilot plants with distillation testsystems there was found a good agreement between the experimental separation results and thoseobtained by computing with a theorectical model; the latter is based on the assumption of phaseequilibrium between the vapour formed on an infinitely small element of area in a liquid film of anygiven concentric periphery of the vertically arranged evaporator.
