Mangrove-associated fungi ...This review describes the present hot research areas of mangrove-associated fungi, including its biodiversity, ecological roles, novel metabolites productions and biotechnological potential. Mangrove-associated fungi were divided into saprophytic, parasitic and true symbiotic fungi based on its ecological roles. Saprophytic fungi are fundamental to decomposition and energy flow of mangrove, additionally, their potential toxicity also exists. Pathogenic fungi have significant effects on mangrove survival, growth, and fitness. Endophytic fungi, the most prolific source of diverse bioactive compounds found among that of mangrove-associated fungi, are found in most species of mangroves. Although a significant number of reports focused on the antimicrobial, insecticidal and other bioactive metabolites as well as many novel enzymes isolatcd from mangrove-associated fungi, and many of those metabolites from endophytic fungi are suspected to be of significant to mangrove, only few studies have provided convincing evidence for symbiotic producers in mangrove. Hence, this paper discusses the present progress of molecular methods used to correlate the ecological roles of endophytic fungi with their bioactive metabolites;, meanwhile, the potential of using metabolic engineering and post-genomic approaches to isolate more novel enzymes and bioactive compounds and to make their possible commercial application was also discussed.展开更多

The emergence of drug resistant strains has directed to the identification of pros...Seaweeds are known to exhibit various antimicrobial properties, since it harbours an enormous range of indigenous bioactive compounds. The emergence of drug resistant strains has directed to the identification of prospective metabolites from seaweed and its endophytes, thereby exploiting the properties in resisting bacterial diseases. The current study was aimed to assess the antimicrobial activity of extracts obtained from Ulva reticulate, for which metabolites of Ulva reticulata and its endophytes were extracted and assessed against human pathogens like Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Salmonella typhi, and Bacillus subtilis. It was observed that the hexane extract of isolate VITDSJ2 was effective against all the tested pathogens but a significant inhibition was observed for Staphylococcus aureus and Escherichia coli. Further, Gas chromatography coupled with Mass spectroscopy (GC-MS) revealed the existence of phenol, 3, 5-bis (1, 1-dimethylethyl) in the crude hexane extract which is well-known to possess antibacterial activity. The effective isolate VITDSJ2 was identified to be the closest neighbour of Pseudomonas smtzeri by phenotypic and genotypic methods. The crude extracts of the seaweed Ulva reticulata was also screened for antibacterial activity and the hexane extract was effective in showing inhibition against all the tested pathogens. The compound in the crude extract of Ulva reticulata was identified as hentriacontane using GC-MS. The extracts obtained from dichloromethane did not show significant activity in comparison with the hexane extracts. Hence the metabolites of Ulva reticulata and the bacterial secondary metabolites of the endophytes could be used in the treatment of bacterial infections.展开更多

In such systems, solar thermal energy can be used, on one hand, directly to charge the buffer storage and, on the other hand, as heat source for the evaporator oftbe HP (heat pump). In this work systems, in which solar heat is only used directly (parallel operation of solar and HP), systems using the collectors also as a heat source for the HP are analyzed and compared to conventional air HP systems. With a combined parallel solar thermal HP system, the system performance compared to a conventional HP system can be significantly increased. Unglazed selectively coated collectors as source for the HP have the advantage that the collector can be used as an air heat exchanger. If solar radiation is available and the collector is used as source for the HP, higher temperatures at the evaporator of the HP can be achieved than with a conventional air HP system.展开更多

[Method] The bark of J. regia was washed with running water and placed on medium to let the fungi grown; the fungal colonies were studied via morphological observation and anti-microbial activity. [Result] An endophytic fungus showing broad-scope, highly active anti-microbial activity was screened out. This fungus showed anti-microbial activity seven Gram-positive and four Gram-negative bacterial indicators, six skin pathogenic fungi and seven pathogenic fungi to different extent. Morphological observation on the ITS sequence showed that this fungus is consistent with Paecilomyces lilacinus in genus Paecilomyces bainier(with homology of over 99%). Accordingly this fungus was designated as Paecilomyces lilacinus JY-1024. [Conclusion] This study for the first time reported the isolation of P. lilacinus from J. regia and its in vitro antimicrobial activity to several pathogens.展开更多

Second experiment conducted in large pots to investigate the effects of the two bacterial isolates on some morphological, productive and physiological characters of sesame plants against same fungi under normal conditions. Results showed very low germination and slowly seedlings growth in Pythium, Fusarium and Alternaria treatments respectively, but adding bacterial vaccine of Pseudomonas putida2 and Pseudomonas fluorescens3 as a Biocide to fungi treatments, increased the germination percent and seedlings growth of sesame crop significantly, 20 days after planting compared with control treatment. In the second experiment, control treatment scored germination by 52%, while vaccines of Pseudornonas putida2 and Pseudomonas fluorescence3 treatments increased germination percentage in Fusarium, Pythium and Alternaria treatments significantly in average more than 71%. Both isolates increased significantly Leaf number per plant, leaf area per plant, height of plant, branches number per plant, total dry weight of shoot per plant and chlorophyll content compared with other and control treatments. Similar significant effect for both isolates was recorded in seeds number per pod per plant, total weight of 1,000 seeds per plant and pods number per plant and percentage of N, P, K in total dry weight of shoot per plant and oil percentage in seeds per plant compared with other and control treatments. P. putida2 increased oil percentage in seeds by 43.3, 48.0 and 45.0% respectively while with P. fluorescens3 increased to 42.7, 44.0 and 43.7% respectively compared with control treatment (27.7%). In general Pseudomonas putida2 increased most of growth characters much higher than Pseudomonas fluorescens3, it may related to siderophores compound and genetic factors.展开更多

Potato dextrose agar and potato dextrose broth were used for fungi purification and fermentation. Fungal metabolites were extracted with methanol (1 : 1 v/v) for 24 h before vacuum evaporated to yield the crude extracts. Antimicrobial activity test was done according to the Kirby-Bauer test. The inhibition zone around each disc was considered to measure antibacterial activity of endophytic fungi. Only three endophytic fungi showed inhibition activity on S. aureus ATCC 9144 and two endophytic fungi showed inhibition on E. coil ATCC 8739. The results suggested that the extracts have a high potential for enteric diseases treatment caused by foodbome pathogen.展开更多
