This study was undertaken to isolate and characterize the protease activity of human eye lens sample of mature and hyper mature cataract. Samples were collected just after surgery of the cataract lens and were stored ...This study was undertaken to isolate and characterize the protease activity of human eye lens sample of mature and hyper mature cataract. Samples were collected just after surgery of the cataract lens and were stored at -20 ℃. The total protein extract was isolated from 5 samples in each case (mature and hyper mature cataract) and clear supernatant obtained after centrifugation was used as an enzyme source. The optimum pH for the proteases of mature cataract was 7.5 while the proteases of hyper mature cataract were recorded for maximum activity at pH 5.5 and 7.5. The optimum temperature for both enzyme sources was 50 ℃. Effect of different metal ions such as potassium, lead, silver, zinc and borate was studied. In each case protease activity was increased. Reducing agent e.g. β mercaptoethanol also caused an increase in activity indicating the involvement of sulfhydryl groups. Protease activity was also located on agar plates.展开更多

NOS activity was first observed in the optic primordium and the lens placode at 5-somite...Expression of nitric oxide synthase (NOS) in the developing eye of zebrafish was studied by NADPH-diaphorase staining technique. NOS activity was first observed in the optic primordium and the lens placode at 5-somite stage, and remained basically unchanged up to the prim-5 stage. Upon hatching, NOS activity was nearly equally detected in the gangalion cell layer and the photoreceptor layer in the developing retina. However, it began declining in the inner plexiform layer and the inner nuclear layer at this stage. NOS activity disappeared in the lens although the anterior lens epithelium was strongly stained. Two days after hatching, NOS activity was still strong in the photoreceptor layer, but decreased markedly in the gangalion cell layer, the inner plexiform layer and the inner nuclear layer with the retinal patterning. These suggested that nitric oxide (NO), the product of NOS, is not only involved in the modulation of patterning and differentiation of the retinal cells but also in the regulation of proliferation, and differentiation of the lens fibrocytes.展开更多

Eye is the most important part on face for TCM doctors to diagnose patient. In this paper,we present several methods of eye quantification. We quantify eye movement,venation in white part of eye,tears in eye and the shape of upper eyelid,and then use cloud model to deduce volunteers' vitality condition by these measurements. We collect more than one thousand face images and dozens of face videos of patients',and some rules we mined with the highest support and interesting scores are right correspond to TCM diagnosis.展开更多
A fraction of cobra (Naja naja atra) venom has been discovered to have protective effect on rat cultural cerebellar granule neurons (CGNs) from apoptosis induced by removing serum and reducing the extracellular po...A fraction of cobra (Naja naja atra) venom has been discovered to have protective effect on rat cultural cerebellar granule neurons (CGNs) from apoptosis induced by removing serum and reducing the extracellular potassium concentration from 25 to 5 mM. This component has been purified and identified as secreted phospholipase A2 (cobra sPLA2). In order to study the relationship between the protection on CGNs and enzymatic activity of phospholipase A2, CGNs stained by Hoechst 33258 were quantified to determine survival rate under the fluorescence microscopy; the protective potencies on apoptosis of cultural CGNs were compared among cobra sPLA2, the cobra sPLA2 modified in carboxylate groups with water soluble carbodiimide and semicarbazide, and the heated cobra sPLA2 at 80℃ for 30 rain. The results showed that the CGN survival rate was unaffected significantly both in modified cobra sPLAz whose enzymatic activity of PLA2 had decreased by 80%, and in cobra sPLA2 adding 7, 7-Dimethyleicosadienoic acid, an inhibitor of sPLA2 at concentration of 10-fold IC50; contrary, the neuronal survival rate fell about 60% in heated cobra sPLA2, although its PLA2 activity only decreased by 10%. The protection on CGNs were also found in some of sPLA2s derived from venoms of bee, Naja naja mossambica, Crotalus atroxalso and Vipera Ammodytes Ammodytes but could not be found in other sPLA2s from bovine pancreas and Streptomyces violaceoruber. Above results suggest that the protection on CGNs of cobra sPLA2 is independent of its enzymatic activity.展开更多
