Expression vector p301-bG1 contains a Sw gene and a bialaphos resistance gene both driven by glyceraldehydes-3-phosphate dehydrogenase (GPD) gene promoter isolated from Lentinus edodes ( Berk.) Sing. Using p301-bG1, PEG-mediated transformation of protoplast of L. edodes was studied. Mixed with PEG-purified plasmid DNA, the protoplasts of L. edodes were treated with PEG solution and cultured on CYM regeneration plate containing 40 mug/mL bialaphos. Bialaphos-resistant and GUS-positive transformants were obtained using this transformation system. Although the transformation efficiency was relatively low, the protocols release large expenses on expensive instrument and restriction enzymes, providing a simple and economical method for mushroom breeding at the molecular level. 展开更多

In liquid-liquid solvent extraction processes, diluents have a strong influence on the extraction mechanism and efficiency. In this study, benzene, cyclohexane, trichloromethane, carbon tetrachloride, methyl isobutyl ketone (MIBK), butyl acetate, and 1-octanol were used as diluents in the extraction of oxalic acid by trialkylphos-phine oxide (TRPO). The effects of extractant concentration, initial concentration of oxalic acid and diluent type on the extraction equilibrium partition coefficient are analyzed. The sequence of the extraction ability by different diluents is MIBK > butyl acetate > cyclohexane=benzene > carbon tetrachloride > 1-octanol > trichloromethane. To understand the behavior of molybdenum and tungsten extracted by tri-alkyl phosphine oxide(TRPO)from peroxide solution,the extraction mechanism was studied by slope method and Raman and FTIR spectroscopy.The empirical formulas of molybdenum and tungsten extraction distribution ratio(D_(Mo)and D_(W))as functions of equilibrium pH,TRPO concentration and temperature were obtained by establishing mathematical models.Furthermore,the reliability of the empirical formula was verified in the H^(+)-W-Mo-H_(2)O_(2) solution.The results indicate that the calculated values of D_(Mo)or D_(W)were consistent with the experimental values.The apparent extraction equilibrium constants of molybdenum and tungsten wereK_(Mo)^(app)=8.51×10^(3)(0.74≤pH_(e)≤1.7),K_(Mo)^(app)=99.89×10^(3)(1.7<pH_(e)≤4.62)andK_(W)^(app)=2.65×10^(3)(0.92<pH_(e)<2.16)at 20°C,respectively.The main extraction complex of molybdenum or tungsten was[H_(2)(Mo or W)_(2)O_(3)(O_(2))_(4)(H_(2)O)_(2)]·2TRPO.These empirical formulas can be used to analyze and estimate the extraction and separation of Mo and W from low molybdenum and tungsten concentration solutions. 展开更多

Developing efficient,stable,and low-cost novel electron-cocatalysts is crucial for photocatalytic hydrogen evolution reaction.Herein,amorphous Ni-P alloy particles were successfully modified onto g-C3N4 to construct the Ni-P/g-C3N4 photocatalyst through a simple and green triethanolamine(TEOA)-mediated photodeposition method.It was found that the TEOA could serve as an excellent complexing agent to coordinate with Ni2+to form[Ni(TEOA)]^2+complex,which can promote the rapid and effective deposition of amorphous Ni-P alloy on the g-C3N4 surface.Photocatalytic tests suggest that the hydrogen-evolution performance of gC3N4 can be greatly promoted through integrating amorphous Ni-P alloy.Especially,the Ni-P/g-C3N4(5 wt%)exhibits the superior H2-generation activity(118.2μmol h^-1g^-1),which is almost 35.8 times that of bare g-C3N4.Furthermore,the amorphous Ni-P alloy cocatalyst can also serve as the general hydrogen-production cocatalyst to greatly enhance the photocatalytic performance of traditional semiconductor materials such as Ti O2 and Cd S.Based on the present results,the mechanism of the amorphous Ni-P alloy as the high-efficiency electron transfer medium was proposed for the boosted H2-generation rate.The present facile route may broaden the horizons for the efficient development of highly active cocatalysts in photocatalytic field. 展开更多
