Methods Ion-exchange resin-drug mefformin hydrochloride complexes were prepared as core materials, and followed by coating using ethylcellulose (EC) by the emulsion solvent diffusion technique. The release rate of mefformin from the microcapsules was highly dependent on the encapsulating formulation, thus being used as an index for formulation screening. Orthogonal experiments were performed to optimize the coating formulation. Results The final chosen formulation for coating of mefformin microcapsules were as follows: ( 1 ) the ratio of EC (20cps) to EC (45cps) was 50:50; (2) the ratio of plasticizer to coating materials was 20% ;and (3) the ratio of resin-mefformin complexes to coating materials was 5 : 1. The chromatographic processes were c...An effective cation-exchange chromatographic method for lysozyme isolation from chicken egg white is presented, using supermacroporous cryogel grafted with sulfo functional groups. The chromatographic processes were carried out by one-step and sequential elution, respectively. Sodium phosphate buffer (pH 7.8) containing different concentrations of NaC1 is used as elution agent. The corresponding breakthrough characteristics and elution behaviors in the cryogel bed were investigated and analyzed. Purity of lysozyme in the elution effluent was assayed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The maximum purity of the obtained lysozyme was about 96%, and the cryogel is demonstrated as a potential separation medium for purification of high-purit lysozyme from chicken egg white.展开更多
The purification of lactic acid based on the esterification of raw lactic acid from fermentation broth and then the catalytic distillation hydrolysis of methyl lactate simultaneously to achieve pure lactic acid is rep...The purification of lactic acid based on the esterification of raw lactic acid from fermentation broth and then the catalytic distillation hydrolysis of methyl lactate simultaneously to achieve pure lactic acid is reported. The esterification kinetics of lactic acid with methanol catalyzed by strong-acid cation-exchange resins (Amberlyst-15,D001, D002, NKC, 002) was studied under the condition that simulates the real catalytic environment. Experimental results were correlated by a Langmuir-Hinselwood model and the nonideality of the solution was taken into account by using activities calculated by the universal quasichemical functional group activity coefficient (UNIFAC) method.A good agreement between the model and the experimental data was achieved. Continuous purification experiments were conducted to find the optimum column configuration and operation condition for the system. The effects of various parameters, e.g. the length of different section of the column, feed rate and ratio of reactants, packing material and catalyst type, were studied. This novel system shows good separation results in lab scale, and is potential for industrial application.展开更多

An analysis of equilibrium isotherm models showed that the best fit model was the Langmuir–Freundlich. The kinetics of the ion exchange process have been investigated and four kinetic models have been tested namely: Ritchie model, pseudo-second order model, pseudo-first order model and the Elovich model. Atomic absorption spectrum and X-ray diffraction analysis were carried out to investigate the effects of Ag^+ , Cu^2+ and Zn^2+ on antimicrobial abilities of natural porous minerals, and the effect of preparation method on ion exchange capacity of antimicrobial material, respectively. The results show that for the ion exchange capacity, clay mineral is higher than fibrous mineral, i.e. both zeolite and montmorillonite are higher; the antimicrobial ability of material with Ag^+ is the bests the exchange capacities of materials with Cu^2+ or Zn^2+ are all higher, but the antimicrobial ability of Cu^2+ is better than that of Zn^2+ .展开更多

With 16S rRNA gene sequencing, we identified the strain as Tamlana sp., and t...We isolated a bacterial strain (HC4) that is able to degrade k-carrageenan from a live specimen of the red alga Hyalosiphonia caespitosa. With 16S rRNA gene sequencing, we identified the strain as Tamlana sp., and then purified an extracellular K-carrageenase from a culture of Tamlana sp. HC4 by ammonium sulfate precipitation, Sephadex G-200 gel filtration chromatography, and DE-cellulose 52 anion-exchange chromatography. The purified enzyme yields a single band on SDS-PAGE with a molecular mass of 66.4 kDa. The optimal pH and temperature for κ-carrageenase activity are at 8.0 and 30~C, respectively. The enzyme is stable over the range ofpH 7.2-8.6 below 45℃. The enzyme activity is strongly inhibited by Zn2+ and Cu2+ at 1 mmol/L. The enzyme-catalyzed reaction follows Michaelis-Menten kinetics with the Michaelis constant (Kin) at 7.63 mg/ml. Analysis of the degradation products of the κ-carrageenase by ESI-MS and 13C-NMR spectroscopy indicates that the enzyme degrades κ-carrageenan down to the level ofκ-neocarrabiose sulfate.展开更多
Soil and tissue analyses are usually used in identifying potassium(K) deficiencies and predicting K fertilizer requirements of crops.The critical levels of both soil and fresh leaf tissue at seventh leaf stage were de...Soil and tissue analyses are usually used in identifying potassium(K) deficiencies and predicting K fertilizer requirements of crops.The critical levels of both soil and fresh leaf tissue at seventh leaf stage were developed and assessed for canola,chickpea and sunflower grown on two Saskatchewan,soils,with six rates of K fertilizer supply, in a growth chamber experiment.The available potassium in soils was determined by two methods:1)resin strip extraction, and 2) NH4OAC extraction. The potassium in fresh leaves was determined at seventh leaf stage by a simple procedure using a common garlic press and injector to extract the plant sap and testing the sap with a handheld ion selection electrode meter. The results shoWed significant relationships between the resin strip extractable K and NH4OAC extractable K, and between the plant uptake of total K and the supply of available K in the soils determined by the two methods. Good relationships were also found between the potassium in fresh leaves and the plant uptake of total K for canola, chickpea and sunflower.The resin strip extraction for K was calibrated using common NH4OAC extraction, and recommended for routine analyses because of its simplicity and sensitivity.展开更多

The synthesis of zinc doped bentonite nanocomposite was accomplished by p...Nanocomposites of zinc/bentonite clay were synthesized for use as an antibacterial material by a quick and simple alkaline ion exchange method. The synthesis of zinc doped bentonite nanocomposite was accomplished by placing bentonite in a melting bath of ZnSO4 for 10, 20, 40, 60 and 90 rain. The complexes were characterized by XRD, SEM and DRS. XRD analyses and SEM observations confirmed the diffusion of zinc to the clay surfaces. Antibacterial activity tests against Escherichia coli showed that bentonite did not present any antibacterial properties, but after alkaline ion exchange treatment, inhibition was noted. The highest antibacterial activity was observed with ZnO/bentonite composite alkaline ion exchange for 60 and 90 rain. Interestingly, the leaching test indicated that ZnO/bentonite did not present any risk for drinking water treatment.展开更多

In this study, purification of recombinant DsbA was investigated by examining four important factors with Box-Behnken design method, a statistic-based design of experiments. The optimal operation conditions were obtained by adopting the effectiveness coefficient method on the multi-objective problem, which takes the protein recovery, purification efficiency and throughput of ion-exchange chromatography into account. After the optimization, protein recovery of 96.8% and purity higher than 95% DsbA was achieved, and the productivity was (377.9±1.7) mg soluble DsbA per liter broth. The purified protein was identified by peptide mass fingerprinting matching the record of gil2624856, a mutant of DsbA. The DsbA was preliminarily applied to the refolding of denatured lysozyme in vitro.展开更多

Comparing with the corresponding relationships derived from damage mechanics, we find the weakness factor has the same significance as the damage factor. We simulate the wave field using a staggered-grid pseudospectral method to show the influence of the weakness factor qualitatively. Applying the analytical solution of plane waves, we give the velocity and attenuation coefficient of three body waves, which are affected by the wave frequency and the weakness factor of saturated discrete media. Our results show that velocity decreases with increasing weakness factor, the attenuation coefficient increases with an increase in the weakness factor, and that the influence of weakness depends on the mode of the body waves.展开更多
