中华猕猴桃是原产于中国的本土果树,野生种质资源丰富,系统评价中华猕猴桃野生种质对细菌性溃疡病的抗性,筛选优异抗病种质,可为猕猴桃抗病品种选育提供理论依据.以高感中华猕猴桃‘红阳’、耐病美味猕猴桃‘徐香’品种为对照,利用猕猴... 中华猕猴桃是原产于中国的本土果树,野生种质资源丰富,系统评价中华猕猴桃野生种质对细菌性溃疡病的抗性,筛选优异抗病种质,可为猕猴桃抗病品种选育提供理论依据.以高感中华猕猴桃‘红阳’、耐病美味猕猴桃‘徐香’品种为对照,利用猕猴桃溃疡病菌(Pseudomonas syringae pv.actinidiae)人工接种离体枝条的方法,对国家猕猴桃种质资源圃内收集自7个省份的129份中华猕猴桃野生种质进行抗性评价.基于枝条上的病斑长度将种质划分为高抗至高感7个等级,发现中华猕猴桃野生种质间存在明显抗性分化,其中含高抗种质1份、中抗种质1份、低抗种质3份、耐病种质5份、低感种质25份、中感种质15份及高感种质79份,抗病种质比例仅为3.87%.在供试的猕猴桃种质中共筛选得到5份野生中华猕猴桃抗病种质,其中‘四川B2585’为四倍体,‘湖北3021’‘江西B2755’‘湖南B2243’及‘湖南B2244’为二倍体.研究结果证实,对中华猕猴桃野生种质进行抗病性评价可直接筛选获得高抗种质,用作抗病育种的亲本材料,或作为材料进行抗病机理研究及抗病基因挖掘验证,为中华猕猴桃高抗种质的开发创制奠定基础. 展开更多

柑橘链格孢褐斑病(Alternaria brown spot,ABS)是新近在我国局部地区新发现的病害,主要为害宽皮柑橘及某些杂交种柑橘,引起落叶、落果和枯梢,不仅影响产量,还影响果实的销售,造成严重损失,并且病害呈逐渐扩散和加重的趋势。药剂防治是... 柑橘链格孢褐斑病(Alternaria brown spot,ABS)是新近在我国局部地区新发现的病害,主要为害宽皮柑橘及某些杂交种柑橘,引起落叶、落果和枯梢,不仅影响产量,还影响果实的销售,造成严重损失,并且病害呈逐渐扩散和加重的趋势。药剂防治是目前防治ABS的最有效措施,然而目前尚无药剂被登记用于该病害的防治。本研究以来自我国各褐斑病发生区的54个代表菌株构成一个群体,应用刃天青(resazurin,RZ)显色法测定病菌群体对新烟酰胺类杀菌剂啶酰菌胺、氟酰胺和噻氟酰胺,以及吡啶类杀菌剂氟啶胺的敏感性。结果表明,该病菌种群孢子萌发对啶酰菌胺、氟酰胺、噻氟酰胺和氟啶胺而言都是野生的敏感群体,尚未发现抗药性菌株,平均EC50值分别为0.467 7μg/mL、0.010 3μg/mL、0.061 9μg/mL和0.010 0μg/mL。广东和浙江菌株对啶酰菌胺较不敏感,重庆菌株对噻氟酰胺更敏感,而对氟酰胺和氟啶胺的敏感性则与菌株的地理来源无关。柑橘褐斑病菌种群对这4种杀菌剂敏感性的确定,可为选择药剂防治ABS提供依据,并为这些药剂推广后的抗药性监测提供基础研究。 展开更多

A novel method for on-site determination of trace iron was developed using membrane preconcentration and spectrophotometric detection. Fe(II)-ferrozine complex was reacted with cetyltrimethylammonium bromide (CTAB) to form a Fe(II)-ferrozine-CTAB paired compound, which was collected on a membrane by filtration under vacuum. The membrane was immersed in 2 mL of ethanol-nitric acid and the absorbance of the solution measured for quantitative analysis. Various factors affecting the iron collection and determination were investigated. With different sample preconcentration volumes, the range of determination was broadened to 0.5-120 ~tg/L. The detection limit of this method reached 0.19 ktg/L and the recoveries were between 97.2 and 109% when the concentration enrichment was about 45. The relative standard deviation (n = 7) was 1.9% for samples containing 10 ~g/L Fe. Twelve seawater samples were analyzed on-site using the proposed method, and two were also analyzed in inductively coupled plasma mass spectrometry. No significant difference was shown between the two methods by the Student's t-test. The method has also been used on-site for iron enrichment experiments with phytoplankton and concluded to be simple, accurate and inexpensive. 展开更多

Levulinic acid(LA) has been identified as a promising green,biomass derived platform chemical.Response surface analysis(RSA) with a four-factor-five-level central composite design(CCD) was applied to optimize the hydrolysis conditions for the conversion of bamboo(Phyllostachys Praecox f.preveynalis) shoot shell(BSS) to LA catalyzed with ionic liquid [C4mim]HSO4.The effects of four main reaction parameters including temperature,time,C[C4mim]HSO4(initial [C4mim]HSO4 concentration) and XBSS(initial BSS intake) on the hydrolysis reaction for yield of LA were analyzed.A quadratic equation model for yield of LA was established and fitted to the data with an R2 of 0.9868,and effects of main factors and their corresponding relationships were obtained with RSA.Model validation and results of CCD showed good correspondence between actual and predicted values.The analysis of variance(ANOVA) of the results indicated that the yield of LA in the range studied was significantly(P&lt;0.05) affected by the four factors.The optimized reaction conditions were as follows:temperature of 145 ℃,time of 103.8 min,C[C4mim]HSO4 of 0.9 mol.L-1 and XBSS of 2.04%(by mass),respectively.A high yield [(71±0.41)%(by mol),triplicate experiment] was obtained at the optimum conditions of temperature of 145 ℃,time of 104 min,C[C4mim]HSO4 of 0.9 mol.L-1 and XBSS of 2%(by mass),which obtained from the real experiments,concurred with the model prediction [73.8%(by mol) based on available C6 sugars in BSS or 17.9%(by mass) based on the mass of BSS],indicating that the model was adequate for the hydrolysis process. 展开更多

Two silkworm strains viz, B20 A (high cocoon shell ratio) and C.Nichi (low cocoon shell ratio) were sib mated for 10 generations to determine the homozygosis. Both bulked segregant analysis(BSA) and near isogenic lines (NIL) studies were done to identify the RFLP markers closely linked to cocoon shell parameters. Three hundred and fifty two random clones were identified as the low copy number sequence and used for identification of Restriction Fragment Length Polymorphic (RFLP) marker linked to cocoon weight and cocoon shell character. In the bulk segregant analysis, DNA from the parents (B20 A, C.Nichi), F 1 and F 2 progeny of high shell ratio (HSR) and low shell ratio (LSR) were screened for hybridization with the random clones. Polymorphic banding pattern achieved through southern hybridization with different probes indicated the probable correlation of polymorphism with high and low cocoon shell character which are possible landmarks in identifying the putative marker(s) for the cocoon shell character. Out of the 100 probes tried with parents, F 1, F 2 and their bulks, 10 probes were found to be closely linked to cocoon shell characters. 展开更多

The authors numerically investigated the characteristics of surface plasmons excited on a thin metal grating placed in planer or conical mounting. After formulating the problem, the solution method, Yasuura's method (a modal expansion approach with least-squares boundary matching) was described. Although the grating is periodic in one direction, coupling between TE and TM waves Occurs because arbitrary incidence is assumed. This requires the employment of both TE and TM vector modal functions in the analysis. Numerical computations showed: (l) the excitation of surface plasmons with total or partial absorption of incident light; (2) the resonance character of the coefficient of an evanescent order that couples the plasmon surface wave; (3) the field profile and Poynting's vector. The plasmons excited on the surfaces of a thin metal grating are classified into three types: SISP, SRSP, and LRSP, different from each other in the feature of field profile and energy flow. In addition, the eigenvalue of a plasmon mode was obtained by solving a sequence of diffraction problems with complex-valued angles of incidence and using the quasi-Newton algorithm to predict the real angle of incidence at which the absorption occurs. 展开更多

A SLon full-scale continuous centrifugal concentrator was used to reconcentrate hematite from a high gradient magnetic separation concentrate to study the effect of impact angle, concentrate mass and drum rotation speed on the impact energy of turbulent water sprays for continuous centrifugal concentration, under conditions of feed volume flow rate around 9 m3/h, feed solid concentration of 25%-35% and reciprocating velocity of water sprays at 0.05 m/s. The results indicate that a minimal critical impact energy is required in the water sprays for achieving continuous concentration of the concentrator; an unfitted impact angle reduces the impact efficiency, and the highest impact efficiency of 0.6416 is found at the mpact angle of 60°; the increase in concentrate mass leads to an increase in impact energy, and the highest impact efficiency is maintained when the concentrate mass varies in the range of 0.44-0.59 kg/s; when the concentrate mass and the pressure of water sprays are kept at around 0.45 kg/s and in the range of 0.4-0.6 MPa respectively, the impact energy increases proportionally with the increase of drum rotation speed. 展开更多

A plasma-assisted method was employed to prepare Ni/γ-All2O3 catalyst for carbon dioxide reforming of methane reaction. The novel catalyst possessed higher activity and better coke-suppression performance than those of the conventional calcination catalyst. To achieve the same CH4 conversion, the conventional catalyst needed higher reaction temperature, about 50 ℃ higher than that of the N2 plasma-treated catalyst. After the evaluation test, the deactivation rate of the novel catalyst was 1.7%, compared with 15.2% for the conventional catalyst. Different from the characterization results of the calcined catalyst, a smaller average pore diameter and a higher specific surface area were obtained for the plasma-treated catalyst. The variations of the reduction peak temperatures and areas indicated that the catalyst reducibility was promoted by plasma assistance. The dispersion of nickel was also remarkably improved, which was helpful for controlling the ensemble size of metal atoms on the catalyst surface. The modification effect of plasma- assisted preparation on the surface property of alumina supported catalyst was speculated to account for the concentration increase of absorbed CO2. An enhancement of CO2 adsorption was propitious to the inhibition of carbon formation. The coke amount deposited on plasma treated catalyst was much smaller than that on the conventional catalyst. 展开更多

The 185/333 gene family involved in the immune response of sea urchin.One 185/333 c DNA was isolated from Strongylocentrotus intermedius,and named as Si185/333-1.Its full-length c DNA was 1246 bp in length with a 906 bp open reading frame encoding a protein of 301 aa.The molecular weight of the deduced protein was approximately 33.1 k D with an estimated PI of p H 6.26.Si185/333-1 had high identities(70%–86%) to most of Sp185/333.An extraordinary identity of 92% was found between Si185/333-1 and Sp185/333 C5 alpha(ABR22474).Moderate identities(63%–64%) were displayed between Si185/333-1 and He185/333.Si185/333-1 had similar structure to Sp185/333.A signal-peptide,a gly-rich region and a his-rich region were found in its secondary structure.RGD motif was found in gly-rich region at position 116–118aa.There was no transmembrane region in Si185/333-1.The element pattern of Si185/333-1 is different from any available pattern that identified in Sp185/333.Si185/333-1 clustered together with pattern C Sp185/333 in phylogenetic tree.The Si185/333-1 m RNA could be detected in tissues including peristomial membrane,coelomocytes,muscle of Aristotles lantern,gut and tube feet,with the highest expression level detected in peristomial membrane and a relatively low expression in ovary and testis.The temporal expression of Si185/333-1 in peristomial membrane and coelomocytes were up-regulated after bacterial,β-D-glucan and ds RNA challenges,reaching the maximum at 12 h post-stimulation.The up-regulation was more obvious in coelomocytes,and bacterial challenge triggered the highest response.These results proved that 185/333-1 gene was involved in the immune defense of S.intermedius,while more studies were necessary for its function in S.intermedius immunity. 展开更多

Validation of Analytical Method of Irbesartan Plasma in Vitro by High Performance Liquid Chromatography-Fluorescence

Irbesartan is an antihypertensive drug whose concentration in blood is very small so it requires a sensitive method of analysis, selective and valid for analysis. In this study, it is carried out optimization of analytical conditions and validation for the analysis of irbesartan in plasma. Chromatography was performed on a C 18 column (250 × 4.6 mm, 5 μm) under isocratic elution with acetonitrile-0.1% formic acid (46:54 v/v), pH 3.75. Detection was made at excitation 250 nm and emission 370 nm and analyses were run at a flow-rate of 1.0 mL/min at a temperature of 40 ℃. Losartan potassium was used as internal standard. Plasma extraction was done by deproteination with acetonitrile, mixed with vortex for 30 seconds, then centrifuged it at 10,000 rpm for 10 rain. In plasma validation, the recovery was 96.22%, and the lower limit of quantification (LLOQ) in plasma was 2 ng/mL. The method also fulfill the criteria for accuracy and precision intra and inter day by normal values (%Diff) not exceed ± 15%. On the stability study, irbesartan in plasma temperature -20 ℃has been stable for 28 days. 展开更多

In this work, experiment investigations and simulation studies of a waveguide-based microwave plasma source operated at 2.45 GHz in atmospheric pressure helium gas are presented. The plasma source has been designed in conjunction with a gas spectrograph as a speciation analysis tool. Emitted He spectra are observed for gas flow rates between (0.2-1) L/min and microwave power in the range (15-150) W. The results obtained demonstrate maximum excitation temperature of about 3800 K and gas temperature of about 2000 K. Mixtures containing small amounts of mercury atoms are considered and the resonant emission of Hg atoms at 253.56 nm is detected. The spectroscopic measurements are completed with a collisional radiative model delivering the electron density and temperature, the amplitude of the electric microwave field, and the population of the excited atomic states for a given absorbed power and gas temperature, The electromagnetic field distribution in the plasma source is obtained by solving Maxwell's equations. Electric field strength of several 10^5 V/m is obtained that agrees well with the results of the collisional radiative model. The calculated and measured line intensity ratios of He spectral lines agree within 300/0-40%. 展开更多

Two-color resonant two-photon mass-analyzed threshold ionization (MATI) spectroscopy was used to record the vibrationally resolved cation spectra of the selected rotamers of p- ethoxyphenol. The adiabatic ionization energies of the trans and cis rotamers are determined to be 61565±5 and 61670±5 cm^-1, which are less than that of p-methoxyphenol by 645 and 643 cm^-1, respectively. Analysis on the MATI spectra of the selected rotamers of p-ethoxyphenol cation shows that the relative orientation of the ethoxy group has little effect on the in-plane ring vibrations. The low-frequency OC2H5 bending vibrations appear to be active for both forms of the cation. 展开更多

Based on X-ray diffraction,optical microscopy and scanning electron microscopy analysis of the Dachang gold ore,it is showed that the sulfide of ore is the main carrier minerals of gold. A majority of gold is embedded in pyrite and tetrahedrite as the form of inclusion and a small amount intergrowth with pyrite and gangue mineral,occasionally the presence is in form of monomer natural gold. The main factors which influence the recovery rate are sulfide mineral particle size,and the fine grained sulfide is beneficial for the dissociation and flotation of gold bearing minerals. The monomer dissociation degree of gold bearing minerals can reach91. 3%,when the grinding fineness is less than 0. 074 mm and grain level accounts as 80%. It is not conducive to the flotation of sulfide if the grinding fineness is low or high. It is difficult to completely dissociate the monomer if there is a small amount of pyrite and arsenopyrite in the ore. Therefore,before leaching the gold,it must conduct pretreatment to reach the ideal recovery rate of the gold,like roasting oxidation,pressure oxidation and biological oxidation. The fine microscopic gold has little influence on the gold recovery rate. 展开更多

A gas liquid centrifugal separator is widely used in industry on account of its simple geometry and little maintenance. These separators have considerable advantages over filters, scrubbers or precipitators in term of compact design, lower pressure drop and higher capacity. A gas liquid centrifugal separator is a device that utilizes centrifugal forces and low pressure caused by rotational motion to separate liquid from gas by density differences. Efficient and reliable separation is required for the optimum operation. These separators are often operated at less than peak efficiency due to the entrainment of separated liquid through an outlet pipe which is closely associated with the very complicated flow phenomena involved. Design parameters such as length of the separation space, vane exit angle, inlet to outlet diameter ratio, models for separation efficiency and pressure drop as a function of physical dimensions are not available in literature. This leaves the designer with very little to go on except known designs and experimentation. The aim of present study is to perform a parametric study to get higher efficiency for gas-liquid separator. A parametric study has been carded out with the help of CFD tools to analyze a separation performance of a centrifugal separator by varying the length of separator space. The best design parameters are analyzed based upon obtained results, tangential velocities, vortices, total pressure losses. From the present study several attempts are made to improve the performance of conventional centrifugal separators. 展开更多

In this paper, it is shown the COMSOL Multiphysics as a fairly robust and simple program, due to the existence of a graphical environment, to perform simulations with good accuracy. Results are compared with other simulation analysis, focusing on the surface plasmon resonance (SPR) phenomena for refractive index sensing in a D-type optical fiber, where the characteristics of the material layers, in terms of the type and thickness, and the residual fiber cladding thickness are optimized. 展开更多

Three earthward flowing magnetic flux ropes observed in the duskside plasma sheet at geocentric solar magnetospheric coordinate X~–55 Re by P1 and P2 of acceleration,reconnection,turbulence and electrodynamics of moon’s interaction with the sun mission during 13:00–15:00 UT on July 3,2012,were studied.The morphologies of the flux ropes were studied in detail based on Grad-Shfranov reconstruction method and electronic pitch angle distribution data.It is found that(1)the flux rope cross-sectional dimensions are 1.0 Re×0.78 Re,1.3 Re×0.78 Re,and 2.5 Re×1.25 Re,respectively.The magnetic field lines were asymmetric about the center with field line compression on both sides of the current sheet at the leading region;(2)the electron energy flux data presented asymmetry with larger electron flux and lower temperature in the precursor region.The flux ropes were blocked by the resistance of compressed particle density in the front central plasma sheet and the enhanced magnetic field on its sides;and(3)it is found that the flux rope has a layered structure.From inside out,event 1 can be divided into three regions,namely electronic depletion core region,closed field line region,and the caudal area possible with fields connected with the ionosphere.It suggests that the flux ropes cannot merge with the tail magnetic field lines near the lunar orbit.Especially,the flux rope asymmetrical shape reflects the different reconnection processes that caused it on both sides of the magnetic structure.The events shown in this paper support the multiple X-line magnetic reconnection model for flux ropes with in situ observations. 展开更多
